Familial hemophagocytic lymphohistiocytosis is a disorder which presents with fever, pancytopenia, liver dysfunction and also an increase in non-malignant histiocytes with prominent hemophagocytosis in various organs. It is usually difficult to distinguish from other hemophagocytic syndrome in the absence of family history. It rarely manifests in adults. Chemotherapy is usually indicated. Here, we report the occurrence of this disorder in two brothers in their twenties.

Histiocytic sarcoma is an extremely rare malignant neoplastic proliferation of the haematopoietic cells. Very few cases have been reported in the paediatric age group. Imaging features have been rarely described in the literature. It can involve any region of the body; however, it most commonly involves the lymph nodes. Its imaging appearance can mimic lymphoproliferative disorders; however, with the advent of new immunohistochemical markers, the diagnosis of HS has become more reliable. We report an unusual case of primary bone involvement by HS with multiple lesions in the facial bones of a

Histiocytic sarcoma is an exceptionally rare and highly aggressive neoplasm that exhibits the morphological and immunophenotypic hallmarks of mature histiocytes. It is characterized typically by extranodal presentation and a dismal clinical course, particularly in patients with disseminated disease. A history of hematolymphoid disorder can be identified in a subset of patients, suggesting transdifferentiation of a preexisting hematolymphoid neoplasm in its pathogenesis. A 38-year-old male patient presented for consultation following the detection of a gastric mass during a routine health exami

The kidney pathology of monoclonal gammopathy of renal significance varies greatly. In this report, we present a woman in her 20s with nephrotic syndrome and monoclonal immunoglobulin G kappa (serum and urine) without diabetes. She had a family history of nephrotic syndrome as well as hematologic and connective tissue disorders. A kidney biopsy showed nodular glomerulosclerosis, with the glomerular capillary full of histiocytes, which were strongly positive for kappa, not lambda. Immunoelectron microscopy revealed that histiocytes had infiltrated the glomerular subendothelial space, and enlarg

Hemophagocytic lymphohistiocytosis [HLH] is a reactive disorder characterized by generalised non-malignant histiocytic proliferation with prominent hemophagocytosis. It may be either primary [familial (FLH)] or secondary [infection or malignancy associated]. Organisms incriminated for infection associated hemophagocytic syndrome (IAHS) include viruses, bacteria, spirochetes, fungi and parasites. Reports of IAHS associated with tuberculosis in neonates are rare. The authors report a case of perinatal tuberculosis presenting as hemophagocytic lymphohistiocytosis.

The authors describe a rare tumor with overlapping features of true histiocytic sarcoma and interdigitating dendritic cell sarcoma occurring in infancy. A 3-month-old boy presented with hepatosplenomegaly and abdominal distention. He presented a diagnostic challenge at initial presentation despite extensive investigations. On follow-up at age 16 months he had progressive splenomegaly, thrombocytopenia, and worsening coagulopathy. A diagnosis of histiocytic sarcoma with interdigitating dendritic cell differentiation was established after an open biopsy of liver and spleen. A repeat liver biopsy

OBJECTIVE: To describe and characterize histiocytic sarcoma (HS) first detected in the eyes of dogs using the large database at the comparative ocular pathology laboratory of Wisconsin (COPLOW).
METHODS: Cases diagnosed as HS were selected from the COPLOW database. Slides were reviewed to describe the cellular morphology, localize the tumor within the globe, record the tumor distribution and measure the size of the tumor. Further sections were taken to perform immunohistochemistry for Melan-A, CD18 and S-100, and for ferric iron staining. The following clinical information was recorded: breed

Two cases of mediastinal germ cell tumors associated with primary hemoproliferative disorders are reported. The first case was a 23-year-old man who presented simultaneously with a mediastinal immature teratoma with focal yolk sac differentiation and a cytologically benign histiocytic proliferation associated with refractory thrombocytopenia. In the second case, an unsuspected mediastinal immature teratoma with focal yolk sac and hepatic differentiation was found postmortem in a 33-year-old man who had died of fulminant malignant histiocytosis. Although the histiocytosis in the former case fai

BACKGROUND: This report presents a case of esophageal and nodal metastasis in a primary pulmonary histiocytic sarcoma in a dog. Histiocytic sarcoma is a malignant neoplasm originating from histiocytic cells, characterized by its aggressive nature, high metastatic potential and high mortality rate. Pulmonary involvement occasionally results in pleural effusion and regional lymph node involvement, such as the tracheobronchial, sternal, and mediastinal lymph nodes. Thoracic radiography is the preferred complementary diagnostic test when respiratory disorders are suspected, as it can identify pulm

A new monoclonal antibody, Y1/82A, was raised against phytohaemagglutinin activated peripheral blood mononuclear cells. Using an immunohistochemical technique it was shown that Y1/82A reacts against peripheral blood and bone marrow monocytes and resident macrophages from essentially all human tissues. Y1/82A bound to determinants present in leukaemic cells from patients with acute myelomonocytic leukaemia and acute monocytic leukaemia, but not to neoplastic cells from patients with malignant lymphoproliferative disorders or malignant epithelial tumours. Y1/82A failed to react with other cell t

Hemophagocytic lymphohistiocytosis (HLH) is a rare disorder of the immune system that may rapidly progress into life-threatening complications. There is uncontrolled activation of histiocytes and phagocytic macrophages, resulting in excess secretions of cytokines that manifest into systemic hyperinflammation affecting several organs. HLH may present as primary due to underlying familial or genetic causes or as secondary due to underlying conditions such as malignancies, infections, or autoimmune disorders. This case is of a seven-year-old girl who developed culture-proven MRSA infective endoca

Cutaneous crystal-storing histiocytosis (CSH) is an extremely rare histopathologic finding of histiocytes accumulating crystals of immunoglobulin or paraproteins, often associated with lymphoplasmacytic malignancies. It commonly presents in a wide age range of both female and male adults with a history of a lymphoproliferative disorder. Dermatologic examination typically shows cutaneous nodules, plaques, or papules, which frequently present on the trunk, proximal extremities, head, and neck. Histopathologic examination demonstrates numerous large eosinophilic CD68-positive histiocytes with a w

ALK-positive histiocytosis is a recently described rare histiocytic proliferative disorder of early infancy. When infants present with anemia, thrombocytopenia, and hepatosplenomegaly, this entity should be included in the differential diagnosis along with hemophagocytic lymphohistiocytosis, metabolic/storage diseases, hematopoietic malignancies, and autoimmune thrombocytopenia. We report 2 new cases of ALK-positive histiocytosis, one with kidney involvement and the other with extensive bone marrow involvement in addition to an overt liver disease. Renal involvement by ALK-positive histiocytos

Crystal-storing histiocytosis (CSH) is a rare condition often linked to monoclonal gammopathies and lymphoproliferative disorders. It involves the accumulation of crystallized immunoglobulins within histiocytes and may mimic other diseases, especially when presenting in unusual sites such as the lung. We report on a 51-year-old man with chronic cough and fatigue, initially suspected of having an infectious pulmonary process. Imaging revealed a lesion in the right lung. Bronchial biopsy showed large eosinophilic histiocytes with granular cytoplasm. Immunohistochemistry confirmed histiocytic dif

T-cell proliferative disorders are occasionally associated with Epstein-Barr virus (EBV) infection. This study was designed to assess the clinical features of cutaneous lymphomas positive for EBV genes. Polymerase chain reaction was used to detect EBV genes in tissue or blood samples from patients with malignant lymphoma or related diseases. Of 26 DNA samples tested, EBV genes were detected in two samples, both of which were obtained from the same patient with subcutaneous T-cell lymphoma showing clinical and histologic features of cytophagic histiocytic panniculitis. On the basis of these fin

Canine histiocytic proliferative disorders include reactive diseases such as cutaneous and systemic histiocytosis and neoplastic diseases such as cutaneous histiocytoma and localized and disseminated histiocytic sarcoma (malignant histiocytosis). Their etiology and pathogenesis are unknown. Canine cutaneous and systemic histiocytosis target the skin and subcutis and have similar clinical behavior. Systemic histiocytosis also affects other organ systems. Clinicopathologic and phenotypic features of canine cutaneous and systemic histiocytosis were examined in this study. Canine cutaneous (18 cas

Reactivity for Ki-1 antibody was studied in 145 patients with a large variety of cutaneous disorders. The antigen was consistently expressed and by a high proportion of tumour cells in infiltrates in which atypical cells revealed a 'histiocytic' appearance, i.e. lymphomatoid papulosis (LP), T-immunoblastic lymphoma with the characteristic of true histiocytic lymphoma, Hodgkin's disease, T-blast cell proliferation with giant multivesicular bodies, concurrent LP and mycosis fungoides (MF), and two cases of MF. Ki-1+ cells with the usual morphology of atypical T-cells formed a major component in

A 9-month-old male Great Dane had progressive generalized nodular dermatopathy for several months. There were > 100 raised, alopecic, firm, painful nodules throughout the skin. Aspirates from several lesions yielded moderate numbers of irregularly round or polygonal to spindle-shaped cells with mild to moderate anisocytosis and few inflammatory cells, and the cytologic interpretation was proliferation of mesenchymal or histiocytic cells. On histopathologic examination, nodules were composed of densely packed sheets of round to spindle-shaped cells with mild anisokaryosis and low mitotic activi

Crystal-storing histiocytosis (CSH) is an uncommon histiocytic proliferation reported to involve diverse organs and tissues, but involvement of the central nervous system (CNS) is rare. In most cases CSH is identified in association with underlying lymphoproliferative, plasma cell diseases or rarely with various inflammatory or infectious conditions. CSH is characterized by the cytoplasmic accumulation of crystalline material in histiocytes, most commonly of kappa immunoglobulin light chain. We report a unique case of localized CSH involving the left cerebellum and caudal brain stem in a young
