Microplastics containing plasticizers affect chicken reproduction via disturbing ovary DNA homologous recombination activity.

Microplastics (MPs) alone or carrying organic contaminants (OCs) have been proven to exhibit a wide range of toxic effects on aquatic animals. Nevertheless, the impacts and mechanisms for co-exposure to MPs and OCs on vertebrate animals remain obscure. This study investigated the impact of clean and phthalate esters (PAEs)-spiked MPs on chicken by observing the egg-laying behavior, analyzing the levels of PAEs and their metabolites in eggs, as well as exploring the gene expression in ovaries. Results showed that 16 eggs were collected from the 10 %-spiked MPs group during depuration period tho

Targeting IP6 signaling to destabilize homologous recombination proteins to overcome PARP inhibitor resistance.

Homologous recombination (HR) is crucial for maintaining genomic integrity and is tightly regulated, yet the role of ubiquitin-dependent degradation in HR proteins remains poorly understood. Through high-throughput screening for compounds that modulate the DNA replication stress response, we identify ML367 and its derivative, UNI418. Kinase profiling and detail molecular analyses reveal that UNI418 inhibits PIKfyve and PIP5K1C, reducing inositol hexaphosphate (IP6) levels and triggering Cul4A-dependent degradation of RAD51, CtIP, and CHK1. Further analysis identifies WDR5 as a DCAF protein tha

STAT5A-RNF8 signaling regulates homologous recombination repair and cisplatin responsiveness in cervical cancer.

Cisplatin resistance remains a major limitation in the treatment of cervical cancer, with enhanced DNA damage repair contributing to reduced therapeutic sensitivity. The role of STAT5A in coordinating DNA double-strand break repair under therapeutic stress remains unclear. Here, we investigated the role of STAT5A in cisplatin responsiveness using clinical tissue analysis, CRISPR/Cas9-based genetic models, DNA repair assays, and xenograft models. STAT5A depletion sensitized cervical cancer cells to cisplatin, whereas STAT5A overexpression reduced cisplatin sensitivity. Mechanistically, STAT5A s

Inflammatory kinase TBK1 suppresses homologous recombination DNA repair to sensitize tumors to chemotherapy.

Inflammation exerts context-dependent influences on tumor progression and therapeutic response. Although chemotherapy remains a cornerstone of cancer treatment, its functional interplay with inflammatory signaling is still incompletely understood. Here, we identify TANK-binding kinase 1 (TBK1) as a critical modulator of chemotherapeutic efficacy through its impact on DNA damage repair. TBK1 activation potentiates cancer-cell death induced by chemotherapeutic agents by promoting DNA damage and impairing homologous recombination (HR) repair. This effect occurs independently of canonical inflamma

GNL3 SUMOylation is essential for DNA double-strand break repair by homologous recombination.

DNA double-strand break repair via homologous recombination is critical for maintaining genomic integrity and requires proper DNA end resection. Here, we identify GNL3, a nucleolar GTP-binding protein, as a key regulator of homologous recombination in human cells via SUMOylation-dependent control of DNA end resection. Expression of wild-type GNL3, but not the SUMO-defective K196R mutant, abolished DNA damage induced by knockdown of endogenous GNL3. GNL3 interacts with the BLM-DNA2 helicase-nuclease complex, promoting DNA end resection and subsequent RPA and RAD51 loading. This interaction requ

Functional characterization of BRCA1 variants of unknown significance using homologous recombination repair assays.

Inherited mutations in BRCA1 are among the primary causes of hereditary breast and ovarian cancer (HBOC). Genetic testing has identified numerous pathogenic and benign mutations in BRCA1, but also thousands of variants of uncertain significance (VUS) with unclear functional consequences. Accurate cancer risk prediction for carriers of these VUS requires functional assays to assess their pathogenicity. In this study, we analyzed 16 BRCA1 VUS detected at the Hereditary Cancer Genetic Counseling Laboratory in Salamanca, Spain, with the goal of providing additional data to guide their reclassifica

SLX1 Inhibition Enhances Olaparib Sensitivity by Impairing Homologous Recombination Repair in Breast Cancer.

While PARP inhibitors like Olaparib are effective against BRCA1-deficient breast cancers, their efficacy in BRCA1-proficient tumors depends on the functional status of homologous recombination (HR) repair. Here, we identify the structure-specific endonuclease SLX1 as a key regulator of HR and a determinant of Olaparib sensitivity in BRCA1-intact breast cancer. SLX1 is frequently upregulated in breast cancer and associated with poor prognosis. Functional studies revealed that SLX1 promotes RAD51-mediated HR repair of DNA double-strand breaks. Consequently, SLX1 depletion reduces HR efficiency,

Attenuation of a local Mycobacterium avium subsp paratuberculosis virulent strain by homologous recombination.

Paratuberculosis (PTB) is a disease that affects many countries around the world. One of the strategies that could contribute to its control is the development of effective vaccines that prevent the dissemination of Mycobacterium avium subsp. paratuberculosis, the causal agent. Previously, we showed the humoral and cellular immune response induced by a novel vaccine candidate against PTB based on the inactivated Map6611 virulent strain. In the present study, we independently deleted two operons, mce4 and lprG-p55, possibly associated with Map virulence. Map6611_Δmce4 and Map6611_ΔlprG-p55 viru

Gene editing of NCF1 loci is associated with homologous recombination and chromosomal rearrangements.

CRISPR-based genome editing of pseudogene-associated disorders, such as p47phox-deficient chronic granulomatous disease (p47 CGD), is challenged by chromosomal rearrangements due to presence of multiple targets. We report that interactions between highly homologous sequences that are localized on the same chromosome contribute substantially to post-editing chromosomal rearrangements. We successfully employed editing approaches at the NCF1 gene and its pseudogenes, NCF1B and NCF1C, in a human cell line model of p47 CGD and in patient-derived human hematopoietic stem and progenitor cells. Upon g

Copy number alteration features in pan-cancer homologous recombination deficiency prediction and biology.

Homologous recombination deficiency (HRD) renders cancer cells vulnerable to unrepaired double-strand breaks and is an important therapeutic target as exemplified by the clinical efficacy of poly ADP-ribose polymerase (PARP) inhibitors as well as the platinum chemotherapy drugs applied to HRD patients. However, it remains a challenge to predict HRD status precisely and economically. Copy number alteration (CNA), as a pervasive trait of human cancers, can be extracted from a variety of data sources, including whole genome sequencing (WGS), SNP array, and panel sequencing, and thus can be easily

European experts consensus: BRCA/homologous recombination deficiency testing in first-line ovarian cancer.

BACKGROUND: Homologous recombination repair (HRR) enables fault-free repair of double-stranded DNA breaks. HRR deficiency is predicted to occur in around half of high-grade serous ovarian carcinomas. Ovarian cancers harbouring HRR deficiency typically exhibit sensitivity to poly-ADP ribose polymerase inhibitors (PARPi). Current guidelines recommend a range of approaches for genetic testing to identify predictors of sensitivity to PARPi in ovarian cancer and to identify genetic predisposition. DESIGN: To establish a European-wide consensus for genetic testing (including the genetic care pathwa

Exploring the Frequency of Homologous Recombination DNA Repair Dysfunction in Multiple Cancer Types.

Dysfunctional homologous recombination DNA repair (HRR), frequently due to BRCA mutations, is a determinant of sensitivity to platinum chemotherapy and poly(ADP-ribose) polymerase inhibitors (PARPi). In cultures of ovarian cancer cells, we have previously shown that HRR function, based upon RAD51 foci quantification, correlated with growth inhibition ex vivo induced by rucaparib (a PARPi) and 12-month survival following platinum chemotherapy. The aim of this study was to determine the feasibility of measuring HRR dysfunction (HRD) in other tumours, in order to estimate the frequency and hence

And-1 is required for homologous recombination repair by regulating DNA end resection.

Homologous recombination (HR) is a major mechanism to repair DNA double-strand breaks (DSBs). Although tumor suppressor CtIP is critical for DSB end resection, a key initial event of HR repair, the mechanism regulating the recruitment of CtIP to DSB sites remains largely unknown. Here, we show that acidic nucleoplasmic DNA-binding protein 1 (And-1) forms complexes with CtIP as well as other repair proteins, and is essential for HR repair by regulating DSB end resection. Furthermore, And-1 is recruited to DNA DSB sites in a manner dependent on MDC1, BRCA1 and ATM, down-regulation of And-1 impai

Homologous recombination and human health: the roles of BRCA1, BRCA2, and associated proteins.

Homologous recombination (HR) is a major pathway for the repair of DNA double-strand breaks in mammalian cells, the defining step of which is homologous strand exchange directed by the RAD51 protein. The physiological importance of HR is underscored by the observation of genomic instability in HR-deficient cells and, importantly, the association of cancer predisposition and developmental defects with mutations in HR genes. The tumor suppressors BRCA1 and BRCA2, key players at different stages of HR, are frequently mutated in familial breast and ovarian cancers. Other HR proteins, including PAL

Requirement of heterogeneous nuclear ribonucleoprotein C for BRCA gene expression and homologous recombination.

BACKGROUND: Heterogeneous nuclear ribonucleoprotein C1/C2 (hnRNP C) is a core component of 40S ribonucleoprotein particles that bind pre-mRNAs and influence their processing, stability and export. Breast cancer tumor suppressors BRCA1, BRCA2 and PALB2 form a complex and play key roles in homologous recombination (HR), DNA double strand break (DSB) repair and cell cycle regulation following DNA damage. METHODS: PALB2 nucleoprotein complexes were isolated using tandem affinity purification from nuclease-solubilized nuclear fraction. Immunofluorescence was used for localization studies of protei

Repeat expansion by homologous recombination in the mouse germ line at palindromic sequences.

Genetic instability can be induced by unusual DNA structures and sequence repeats. We have previously demonstrated that a large palindrome in the mouse germ line derived from transgene integration is extremely unstable and undergoes stabilizing rearrangements at high frequency, often through deletions that produce asymmetry. We have now characterized other palindrome rearrangements that arise from complex homologous recombination events. The structure of the recombinants is consistent with homologous recombination occurring by a noncrossover gene conversion mechanism in which a break induced i

Anti-(U1)snRNP autoantibodies inhibit homologous pairing activity of the human recombination complex.

The co-purification of the U1snRNP particle with a high-molecular-weight human homologous pairing activity has been observed consistently. Using human autoimmune sera directed against various snRNPs, it has been found that autoantibody binding to antigenic determinants specifically associated with the U1snRNP particle inhibits the formation of paired DNA molecules by the human homologous pairing activity. Immunoprecipitation of U1snRNP with anti-(U1)RNP autoantibodies significantly reduced the homologous pairing activity in these fractions. NaDodSO4-PAGE analysis of immunoprecipitated samples

Adenovirus homologous recombination does not require expression of the immediate-early E1a gene.

To investigate whether early genes other than those involved directly in DNA replication are required for efficient adenovirus recombination, pairs of viruses with deletions in E1a, E1b 496R, E1b 196R, or E4 and containing differing restriction site markers were used to infect both permissive and non- or semipermissive cells. Recombination was assayed among intracellular and extracellular genomes by restriction digestion and blot hybridization. Recombination was delayed in infections of nonpermissive cells with E1a- viruses until a time consistent with the late onset of DNA replication charact

Homologous recombination repair germline variants and subsequent neoplasm risk among childhood cancer survivors.

Childhood cancer radiation therapy (RT) increases subsequent neoplasm risk. Radiation dose may modulate DNA damage responses, but the small sample sizes of prior human studies of homologous recombination repair hampered dose-specific investigations. We pooled data for 12 180 survivors (8339 from the Childhood Cancer Survivor Study and 3841 from the St Jude Lifetime Cohort) to estimate associations between deleterious homologous recombination repair variants and RT-related subsequent neoplasms (most commonly breast cancer, meningioma, thyroid cancer, and sarcoma) using conditional logistic regr
