The impact of within-host coinfection interactions on between-host parasite transmission dynamics varies with spatial scale.

Within-host interactions among coinfecting parasites can have major consequences for individual infection risk and disease severity. However, the impact of these within-host interactions on between-host parasite transmission, and the spatial scales over which they occur, remain unknown. We developed and apply a novel spatially explicit analysis to parasite infection data from a wild wood mouse (Apodemus sylvaticus) population. We previously demonstrated a strong within-host negative interaction between two wood mouse gastrointestinal parasites, the nematode Heligmosomoides polygyrus and the co

Immunopathological Changes Caused by Oesophagostomum radiatum in Calves: Insights into Host-Parasite Interactions.

The intensity and prevalence of different gastrointestinal nematode species vary across regions worldwide. Oesophagostomum radiatum commonly shows a high occurrence in young cattle. O. radiatum causes anaemia, hypoproteinaemia, and immunopathological changes in the large intestine wall, impairing calves' body weight gain. This study aimed to assess the impact of natural O. radiatum infection on haematological parameters and immune responses in 23 Nellore calves, considering sex-based differences. Assessments included Oesophagostomum egg count (EPG), worm count, packed cell volume (PCV), total

Genomic insights into host and parasite interactions during intracellular infection by Toxoplasma gondii.

To gain insights into the molecular interactions of an intracellular pathogen and its host cell, we studied the gene expression and chromatin states of human fibroblasts infected with the Apicomplexan parasite Toxoplasma gondii. We show a striking activation of host cell genes that regulate a number of cellular processes, some of which are protective of the host cell, others likely to be advantageous to the pathogen. The simultaneous capture of host and parasite genomic information allowed us to gain insights into the regulation of the T. gondii genome. We show how chromatin accessibility and

Host-parasite interactions which influence the virulence of Trypanosoma (Trypanozoon) brucei brucei organisms.

Subclones were prepared in mice from T. b. brucei ILTat 1.4 parasites. Subclones which did not differentiate to stumpy forms in mice were highly virulent and did not stimulate detectable antibody responses. A subclone which did give rise to stumpy forms in mice, was less virulent and did stimulate an antibody response specific for the trypanosome surface glycoprotein. Clones and subclones of T.b. brucei parasites which did not give rise to stumpy forms in mice, did give rise to stumpy forms in Bovidae. Plasma from cattle infected with those parasites did not stimulate differentiation of T.b. b

New tools for exploring parasite biology and elucidating host-pathogen interactions in cryptosporidiosis.

SUMMARYCryptosporidiosis is a major public health concern, the extent of which has only truly been appreciated within the last decade. Cryptosporidium research has undergone a renaissance, with new insights into population structure, species diversity, and evolution of the parasite driven by the advent of genetic transformation techniques and novel models for culture in vitro and in vivo. Here, we summarize the impact of these advances on our understanding of this important parasite. In the initial section, we focus on what we have learned about host range and infectivity from comparative geno

Host-parasite interactions during Entamoeba histolytica infection: an ex vivo human intestinal model.

Amebiasis, caused by the protozoan parasite Entamoeba histolytica, remains a significant public health problem in specific endemic regions. Current experimental models have limitations in accurately replicating human pathogenesis, highlighting the need for alternative platforms that preserve tissue complexity and physiological context. This study aimed to evaluate E. histolytica infection using an ex vivo human precision-cut intestinal slice (PCIS) model by analyzing histological and molecular changes over time (1, 3, 12, 24, and 48 h post-infection). Tissue integrity and parasite invasion wer

Mammalian placental explants: A tool for studying host-parasite interactions and placental biology.

The placenta plays a critical role in host-pathogen interactions. Thus, ex vivo infection of mammalian placental explants is an excellent and simple method to study the mechanisms of cellular and tissue invasion by different pathogens in different mammalian species. These explants can be maintained in culture for several days, preserving the tissue architecture and resembling in-utero conditions under more physiological conditions than their isolated counterparts in isolated cell culture models. In addition, placental explants not only allow us to study how the placenta responds and defends it

Hepatic tissue culture model for study of host-parasite interactions in alveolar echinococcosis.

An in vitro model for growth and differentiation of the metacestode tissue of the tapeworm Echinococcus multilocularis is described. This model simulates the organotropism of the parasite toward the liver of the intermediate host. In the presence of collagen-embedded primary hepatocytes from rats and humans, which can be kept in culture for 2 to 3 months, the parasitic vesicles grew by exogenous budding and multiplied about 12-fold within 3 weeks. In contrast, without the hepatocytes, the metacestodes rapidly degenerated. Development of protoscolices was seen only in the presence of rat hepato

Divergent co-transcriptomes of different host cells infected with Toxoplasma gondii reveal cell type-specific host-parasite interactions.

The apicomplexan parasite Toxoplasma gondii infects various cell types in avian and mammalian hosts including humans. Infection of immunocompetent hosts is mostly asymptomatic or benign, but leads to development of largely dormant bradyzoites that persist predominantly within neurons and muscle cells. Here we have analyzed the impact of the host cell type on the co-transcriptomes of host and parasite using high-throughput RNA sequencing. Murine cortical neurons and astrocytes, skeletal muscle cells (SkMCs) and fibroblasts differed by more than 16,200 differentially expressed genes (DEGs) befor

Host-parasite interaction: parasite-derived and -induced proteases that degrade human extracellular matrix.

Parasitic protozoa are among the most important pathogens worldwide. Diseases such as malaria, leishmaniasis, amoebiasis, giardiasis, trichomoniasis, and trypanosomiasis affect millions of people. Humans are constantly threatened by infections caused by these pathogens. Parasites engage a plethora of surface and secreted molecules to attach to and enter mammalian cells. The secretion of lytic enzymes by parasites into host organs mediates critical interactions because of the invasion and destruction of interstitial tissues, enabling parasite migration to other sites within the hosts. Extracell

Ursolic acid induces apoptosis and disrupts host-parasite interactions in Theileria annulata-infected cells.

This study explores the anti-proliferative and anti-parasitic properties of ursolic acid (UA) in Theileria annulata-infected bovine (TA) cells. Dose-response analyses determined an IC50 value of approximately 5 μg/mL for UA, demonstrating selective cytotoxicity toward infected cells with minimal impact on healthy cells. UA treatment induced pronounced morphological alterations and apoptosis in TA cells, as evidenced by light microscopy and a time-dependent increase in cell mortality. Notably, UA exhibited consistent efficacy against both buparvaquone (BPQ)-sensitive and BPQ-resistant TA cell l

Toxoplasma gondii VIP1 mediates parasitophorous vacuole-host endoplasmic reticulum interactions to facilitate parasite development.

Membrane contact sites (MCS) are areas of close apposition between organelles without membrane fusion, allowing for exchange of biomolecules. The endoplasmic reticulum (ER) forms many MCS via two proteins, vesicle-associated membrane protein-associated proteins A and B (VAPA and VAPB). The obligate intracellular parasite Toxoplasma gondii resides within mammalian cells in a parasitophorous vacuole (PV), which closely contacts the host ER at distances compatible with MCS. However, the proteins mediating this interaction remain largely unknown. Here, using molecular and microscopy approaches, we

LIFE CYCLE VARIATION AND HOST-PARASITE INTERACTIONS OF FOUR CONGENERIC SPECIES OF TURTLE ACANTHOCEPHALANS.

Turtle acanthocephalans in the genus Neoechinorhynchus have complex life cycles and are thought to involve an ostracod intermediate host and a turtle definitive host. For Neoechinorhynchus emydis, an additional snail paratenic host is part of its life cycle. However, how snails become infected and whether other species of Neoechinorhynchus can be found in snails remains unclear. This study was conducted to understand the survival, development, and host use of 4 species of turtle acanthocephalans (Neoechinorhynchus chrysemydis, N. emydis, Neoechinorhynchus emyditoides, and Neoechinorhynchus pse

Mouse models for pathogenic African trypanosomes: unravelling the immunology of host-parasite-vector interactions.

African trypanosomiasis is a parasitic disease that affects a variety of mammals, including humans, on the sub-Saharan African continent. To understand the diverse parameters that govern the host-parasite-vector interactions, mouse models for the disease have proven to be a cornerstone. Despite the fact that most trypanosomes cannot be considered natural pathogens for rodents, experimental infections in mice have shed a tremendous amount of light on the general biology of these parasites and their interaction with and evasion of the mammalian immune system. Different aspects including inflamma

The role of somatostatin in schistosomiasis: a basis for immunomodulation in host-parasite interactions?

This review explores the possible modulatory role of the neuropeptide somatostatin in the outcome of Schistosoma-caused morbidity in man. Somatostatin could play an important role in Schistosoma mansoni-man interactions via its influence on intersystem signalling; therapeutically, via its direct effect on Schistosoma-caused morbidity (fibrosis, granuloma size, portal hypertension, variceal bleeding); and via immunomodulation of Schistosoma-induced inflammatory responses in the liver and intestines. In schistosomiasis-endemic regions two interesting patterns of infection emerge. First, the inte

Host defense or parasite cue: Skin secretions mediate interactions between amphibians and their parasites.

Amphibian skin secretions (substances produced by the amphibian plus microbiota) plausibly act as a first line of defense against parasite/pathogen attack, but may also provide chemical cues for pathogens. To clarify the role of skin secretions in host-parasite interactions, we conducted experiments using cane toads (Rhinella marina) and their lungworms (Rhabdias pseudosphaerocephala) from the range-core and invasion-front of the introduced anurans' range in Australia. Depending on the geographical area, toad skin secretions can reduce the longevity and infection success of parasite larvae, or

Parasite responses to resource provisioning can be altered by within-host co-infection interactions.

Anthropogenic changes to the environment significantly impact wildlife infectious diseases by modifying food resources and impacting host-parasite interactions through changes in host demography, behaviour and immune defences. Supplemental resource provisioning has been found to both enhance and mitigate parasite transmission; however, the role of co-infecting parasites in mediating these effects remains understudied. We developed a mathematical model to explore these dynamics, motivated by the empirical system of wood mice (Apodemus sylvaticus) infected with the nematode Heligmosomoides polyg
