Bicarbonate correction of ketoacidosis alters host-pathogen interactions and alleviates mucormycosis.

Patients with diabetic ketoacidosis (DKA) are uniquely predisposed to mucormycosis, an angioinvasive fungal infection with high mortality. Previously, we demonstrated that Rhizopus invades the endothelium via binding of fungal CotH proteins to the host receptor GRP78. Here, we report that surface expression of GRP78 is increased in endothelial cells exposed to physiological concentrations of β-hydroxy butyrate (BHB), glucose, and iron that are similar to those found in DKA patients. Additionally, expression of R. oryzae CotH was increased within hours of incubation with DKA-associated concentr

Host-Pathogen Interactions in Measles Virus Replication and Anti-Viral Immunity.

The measles virus (MeV) is a contagious pathogenic RNA virus of the family Paramyxoviridae, genus Morbillivirus, that can cause serious symptoms and even fetal complications. Here, we summarize current molecular advances in MeV research, and emphasize the connection between host cells and MeV replication. Although measles has reemerged recently, the potential for its eradication is promising with significant progress in our understanding of the molecular mechanisms of its replication and host-pathogen interactions.

Rice OsPAD4 functions differently from Arabidopsis AtPAD4 in host-pathogen interactions.

The extensively studied Arabidopsis phytoalexin deficient 4 (AtPAD4) gene plays an important role in Arabidopsis disease resistance; however, the function of its sequence ortholog in rice is unknown. Here, we show that rice OsPAD4 appears not to be the functional ortholog of AtPAD4 in host-pathogen interactions, and that the OsPAD4 encodes a plasma membrane protein but that AtPAD4 encodes a cytoplasmic and nuclear protein. Suppression of OsPAD4 by RNA interference (RNAi) increased rice susceptibility to the biotrophic pathogen Xanthomonas oryzae pv. oryzae (Xoo), which causes bacteria blight d

New insights into host-pathogen interactions during Entamoeba histolytica liver infection.

Amoebiasis is the third worldwide disease due to a parasite. The causative agent of this disease, the unicellular eukaryote Entamoeba histolytica, causes dysentery and liver abscesses associated with inflammation and human cell death. During liver invasion, before entering the parenchyma, E. histolytica trophozoites are in contact with liver sinusoidal endothelial cells (LSEC). We present data characterizing human LSEC responses to interaction with E. histolytica and identifying amoebic factors involved in the process of cell death in this cell culture model potentially relevant for early step

Single-cell RNA sequencing unravels mosquito biology and host-pathogen interactions.

Single-cell RNA sequencing (scRNA-seq) is fundamentally reshaping the research paradigm of mosquito biology and host-pathogen interactions via its high-resolution transcriptomic profiling capability. This review first outlines the technical workflow and data analysis strategies of scRNA-seq, then highlights key applications of scRNA-seq in constructing mosquito cell atlases, deciphering post-blood-meal transcriptional remodeling, sexual dimorphism, and receptor coexpression patterns. Furthermore, the role of scRNA-seq in revealing the cellular tropism of mosquito-borne pathogens, host immune r

Host-pathogen interactions in tuberculosis: from immunological mechanisms to translational applications.

Tuberculosis (TB), which is caused by Mycobacterium tuberculosis (Mtb), remains the leading cause of death from infectious diseases globally. The complexity of host-Mtb interactions poses a fundamental challenge to developing effective prevention and control measures. Recent advances in multi-omics, spatiotemporal biology, and single-cell technologies have yielded transformative insights into Mtb infection dynamics and its intricate crosstalk with the host immune system. In this review, we will first introduce the current knowledge of Mtb-host immunological interplay and discuss it from three

Playing Telephone: How Secondary Messengers Influence Host-Pathogen Interactions in Tuberculosis.

Secondary messengers are small, diffusible signaling molecules that transmit information from environmental cues detected at the cell surface by extracellular signaling molecules (primary messengers) to effector proteins, thereby enabling an appropriate cellular response. These molecules include cyclic nucleotides, alarmones, and lipid-derived metabolites and are ubiquitous regulators, influencing processes such as growth, metabolism, and neurotransmission in mammalian cells, as well as chemotaxis, biofilm formation, and metabolism in prokaryotes. Mycobacterium tuberculosis encodes an extensiv

Host-Pathogen Interactions Influencing Zoonotic Spillover Potential and Transmission in Humans.

Emerging infectious diseases of zoonotic origin are an ever-increasing public health risk and economic burden. The factors that determine if and when an animal virus is able to spill over into the human population with sufficient success to achieve ongoing transmission in humans are complex and dynamic. We are currently unable to fully predict which pathogens may appear in humans, where and with what impact. In this review, we highlight current knowledge of the key host-pathogen interactions known to influence zoonotic spillover potential and transmission in humans, with a particular focus on

Brucella Phagocytosis Mediated by Pathogen-Host Interactions and Their Intracellular Survival.

The Brucella species is the causative agent of brucellosis in humans and animals. So far, brucellosis has caused considerable economic losses and serious public health threats. Furthermore, Brucella is classified as a category B bioterrorism agent. Although the mortality of brucellosis is low, the pathogens are persistent in mammalian hosts and result in chronic infection. Brucella is a facultative intracellular bacterium; hence, it has to invade different professional and non-professional phagocytes through the host phagocytosis mechanism to establish its lifecycle. The phagocytosis of Brucel

Host-Pathogen Interactions of Chlamydia trachomatis in Porcine Oviduct Epithelial Cells.

Chlamydia trachomatis (Ct) causes the most prevalent bacterial sexually transmitted disease leading to ectopic pregnancy and infertility. Swine not only have many similarities to humans, but they are also susceptible to Ct. Despite these benefits and the ease of access to primary tissue from this food animal, in vitro research in swine has been underutilized. This study will provide basic understanding of the Ct host-pathogen interactions in porcine oviduct epithelial cells (pOECs)-the counterparts of human Fallopian tube epithelial cells. Using NanoString technology, flow cytometry, and confo

Mining host-pathogen protein interactions to characterize Burkholderia mallei infectivity mechanisms.

Burkholderia pathogenicity relies on protein virulence factors to control and promote bacterial internalization, survival, and replication within eukaryotic host cells. We recently used yeast two-hybrid (Y2H) screening to identify a small set of novel Burkholderia proteins that were shown to attenuate disease progression in an aerosol infection animal model using the virulent Burkholderia mallei ATCC 23344 strain. Here, we performed an extended analysis of primarily nine B. mallei virulence factors and their interactions with human proteins to map out how the bacteria can influence and alter h

Host-pathogen interactions in epidermolysis bullosa patients colonized with Staphylococcus aureus.

Patients with the genetic blistering disease epidermolysis bullosa (EB) often have chronic wounds that can become colonized by different bacteria, especially the opportunistic pathogen Staphylococcus aureus. We therefore determined the S. aureus colonization rates in EB patients from the Netherlands by collecting swabs from their anterior nares, throats and wounds. Within a period of ∼2 years, more than 90% of the sampled chronic wounds of EB patients were found to be colonized by S. aureus. Molecular typing revealed that EB patients were not colonized by a single S. aureus type. Rather the S.

Review paper: Host-pathogen interactions in the kidney during chronic leptospirosis.

Pathogenic species of Leptospira cause leptospirosis, a global zoonotic disease. Leptospira colonize renal tubules of chronically infected maintenance hosts, from where they are shed in urine to the environment and survive in suitable moist conditions. Transmission of disease to new hosts is facilitated by contact with contaminated urine or water sources, because Leptospira can penetrate broken skin or mucosal surfaces of new hosts. Infection of new hosts may be asymptomatic, as with chronically infected maintenance hosts, or may result in an acute disease process in which clinical signs can i

Proteomic profiling of serologic response to Candida albicans during host-commensal and host-pathogen interactions.

Candida albicans is a commensal inhabitant of the normal human microflora that can become pathogenic and invade almost all body sites and organs in response to both host-mediated and fungus-mediated mechanisms. Serologic responses to C. albicans that underlie its dichotomist relationship with the host (host-commensal and host-pathogen interactions) display a high degree of heterogeneity, resulting in distinct serum anti-Candida antibody signatures (molecular fingerprints of anti-Candida antibodies in serum) that can be used to discriminate commensal colonization from invasive disease. We descr

Vector competence across Aedes aegypti populations shaped by host-preference adaptation and host-pathogen interactions.

Aedes aegypti is the principal vector for yellow fever, dengue, Zika, and chikungunya viruses. Although this mosquito continues to expand its global range, its intrinsic ability to acquire, support the replication of, and transmit specific viruses-known as vector competence (VC)-varies across populations. A mechanistic understanding of this variation is a key goal of vector biology and is central to devising interventions to contain epidemics. In this review, we examine how evolutionary history fundamentally shaped VC in the sylvatic and human-adapted subspecies of Ae. aegypti. We discuss how

Role of myeloid cells expressing CD169/Siglec-1 in host-pathogen interactions.

CD169/Siglec-1 is a type-I transmembrane lectin receptor expressed on subcapsular sinus macrophages and activated monocytes, which plays a key role in innate immune surveillance and orchestration of adaptive immunity. As a sialic acid-binding immunoglobulin-like lectin (Siglec), CD169 recognizes sialylated gangliosides and glycoconjugates present on surface of several viral envelopes, including HIV-1, SARS-CoV-2, and Ebola virus as well as sialylated bacterial capsules such as those of Neisseria meningitidis and Campylobacter jejuni. Moreover, different viruses were shown to be captured by CD1

Respiratory enterovirus D68: virology, clinical surveillance, host-pathogen interactions, and therapeutic prospects.

Enterovirus D68 (EV-D68), a unique enterovirus resembling human rhinoviruses, was long considered to cause only sporadic outbreaks of mild, self-limiting respiratory infections mainly in children. However, over the past decade, EV-D68 has exhibited a biennial outbreak pattern across multiple regions worldwide, coinciding with an increased incidence of severe respiratory illnesses and cases of acute flaccid myelitis (AFM) in children. The immune system plays a crucial role in providing rapid and effective defense. Nonetheless, our knowledge of the complex interactions between EV-D68 and the hos
