Videographic analysis of flight behaviours of host-seeking Anopheles arabiensis towards BG-Malaria trap.

The BG-Malaria trap (BGM) is an adaptation of the well-known BG-Sentinel trap (BGS) with greater trapping efficiencies for anopheline and culicine mosquitoes. Its continued optimization requires greater understanding of mosquito flight behaviors near it. We used three high-resolution infrared cameras (68 frames/second) to track flight behaviors of laboratory-reared Anopheles arabiensis females in vicinity of the BGM in comparison with BGS. Additional comparisons were done for BGM at 20, 40 and 80cm heights, and for BGMs baited with Ifakara blend plus CO2, CO2 alone, or no bait. More mosquitoes

Blood meal analysis of Anopheles vectors of simian malaria based on laboratory and field studies.

Blood feeding and host-seeking behaviors of a mosquito play an imperative role in determining its vectorial capacity in transmitting pathogens. Unfortunately, limited information is available regarding blood feeding behavior of Anopheles species in Malaysia. Collection of resting Anopheles mosquitoes for blood meal analysis poses a great challenge especially for forest dwelling mosquitoes. Therefore, a laboratory-based study was conducted to evaluate the potential use of mosquitoes caught using human landing catch (HLC) for blood meal analysis, and subsequently to document blood feeding behavi

Mutagenesis of the orco odorant receptor co-receptor impairs olfactory function in the malaria vector Anopheles coluzzii.

Mosquitoes rely heavily on their olfactory systems for host seeking, selection of oviposition sites, and avoiding predators and other environmental dangers. Of these behaviors, the preferential selection of a human blood-meal host drives the vectorial capacity of anthropophilic female Anopheles coluzzii mosquitoes. Olfactory receptor neurons (ORNs) are dispersed across several appendages on the head and express an obligate odorant receptor co-receptor (Orco) coupled with a "tuning" odorant receptor (OR) to form heteromeric, odor-gated ion channels in the membrane of these neurons. To examine t

Experience-dependent olfactory behaviors of the parasitic nematode Heligmosomoides polygyrus.

Parasitic nematodes of humans and livestock cause extensive disease and economic loss worldwide. Many parasitic nematodes infect hosts as third-stage larvae, called iL3s. iL3s vary in their infection route: some infect by skin penetration, others by passive ingestion. Skin-penetrating iL3s actively search for hosts using host-emitted olfactory cues, but the extent to which passively ingested iL3s respond to olfactory cues was largely unknown. Here, we examined the olfactory behaviors of the passively ingested murine gastrointestinal parasite Heligmosomoides polygyrus. H. polygyrus iL3s were th

Host-seeking strategies of mosquito disease vectors.

Disease transmission by arthropods normally requires at least 2 host contacts. During the first, a pathogen (nematode, protozoan, or virus) is acquired along with the blood from an infected vertebrate host. The pathogen penetrates the vector's midgut and infects a variety of tissues, where replication may occur during an extrinsic incubation period lasting 3-30, days depending on vector and parasite physiology and ambient temperature. Following salivary-gland infection, the pathogen is usually transmitted to additional susceptible vertebrate hosts during future probing or blood feeding. The ho

Olfactory regulation of mosquito-host interactions.

Mosquitoes that act as disease vectors rely upon olfactory cues to direct several important behaviors that are fundamentally involved in establishing their overall vectorial capacity. Of these, the propensity to select humans for blood feeding is arguably the most important of these olfactory driven behaviors in so far as it significantly contributes to the ability of these mosquitoes to transmit pathogens that cause diseases such as dengue, yellow fever and most significantly human malaria. Here, we review significant advances in behavioral, physiological and molecular investigations into mos

Nutrition, malnutrition, and leishmaniasis.

Leishmaniasis is a vector-borne infectious disease with a long history of infecting humans and other animals. It is a known emerging or resurging disease. The host nutritional state has an indispensable role in defense against pathogens. The host defense system disorganization as a result of undernutrition is responsible for asymptomatic infections and even severe diseases. Host susceptibility and pathophysiologic severity to infection can be aggravated owing to undernourishment in a number of pathways, and infection also may aggravate preexisting poor nutrition or further increase host suscep

Genomic analysis identifies candidate supergene architecture for host-feeding behavior in wild Aedes aegypti dengue vector mosquitoes.

BACKGROUND: Aedes aegypti is the primary arboviral vector globally, and its affinity to feed on human hosts is a key determinant of transmission intensity. Despite the epidemiological importance of anthropophily, the genomic architecture underlying the molecular basis of host-feeding behavior remains understudied. METHODS: Here, we present an integrated genome-wide association study (GWAS) and structural variant analysis of host-feeding behavior in 21 field-collected Ae. aegypti females phenotyped as human-blood-fed (HF; n = 11) or non-human-blood-fed (NHF; n = 10). Beyond detoxification: xenobiotic-metabolizing enzymes rewire mosquito sensory perception.

Mosquito olfaction regulates host-seeking, ecological adaptation, and vectorial capacity. Beyond the classical receptor-centric paradigm, we argue that xenobiotic-metabolizing enzymes form a dynamic perireceptor layer that actively sculpts odor identity, signal intensity, and duration. This metabolism gain control mechanism may integrate olfactory sensitivity and behavioral plasticity within a unified sensory framework.

Molecular characterization and evolution of a chemosensory receptor gene family in three notorious rice planthoppers, Nilaparvata lugens, Sogatella furcifera and Laodelphax striatellus, based on genome and transcriptome analyses.

BACKGROUND: The white-backed planthopper (WBPH) Sogatella furcifera, the brown planthopper (BPH) Nilaparvata lugens, and the small brown planthopper (SBPH) Laodelphax striatellus (Hemiptera: Delphacidae) are rice pests that damage rice plants by sap-sucking and by transmitting viruses. Host-seeking behavior involves chemosensory receptor genes that include odorant receptors (ORs), ionotropic receptors (IRs) and gustatory receptors (GRs). RESULTS: We used genome and transcriptome data to identify 141 ORs, 28 GRs and 25 IRs in BPH; 135 ORs, 18 GRs and 16 IRs in WBPH; and 37 ORs, 14 GRs and 6 IR

Culex quinquefasciatus Mosquitoes Exposed to a Juvenile Hormone Analog may Take a Bloodmeal While Gravid.

Blood avidity in female mosquitoes has been shown to be regulated by cycles of hormone production that determine both egg development and distinct behaviors. Specifically, juvenile hormone (JH) drives early egg development until a bloodmeal is acquired, and JH titers are positively correlated with active host seeking and blood feeding behaviors. After a bloodmeal, JH levels fall, and female mosquitoes become refractory to host seeking and biting. While JH analogs (JHAs) are commonly used as larvicides for mosquito control, the effects of these compounds on adult mosquitoes are not well underst

Temperature and time of host-seeking activity impact the efficacy of chemical control interventions targeting the West Nile virus vector, Culex tarsalis.

West Nile virus (WNV) is the leading mosquito-borne disease causing-pathogen in the United States. Concerningly, there are no prophylactics or drug treatments for WNV and public health programs rely heavily on vector control efforts to lessen disease incidence. Insecticides can be effective in reducing vector numbers if implemented strategically, but can diminish in efficacy and promote insecticide resistance otherwise. Vector control programs which employ mass-fogging applications of insecticides, often conduct these methods during the late-night hours, when diel temperatures are coldest, and

Terror in the dirt: Sensory determinants of host seeking in soil-transmitted mammalian-parasitic nematodes.

Infection with gastrointestinal parasitic nematodes is a major cause of chronic morbidity and economic burden around the world, particularly in low-resource settings. Some parasitic nematode species, including the human-parasitic threadworm Strongyloides stercoralis and human-parasitic hookworms in the genera Ancylostoma and Necator, feature a soil-dwelling infective larval stage that seeks out hosts for infection using a variety of host-emitted sensory cues. Here, we review our current understanding of the behavioral responses of soil-dwelling infective larvae to host-emitted sensory cues, an

Attractiveness of vertebrate hosts to Culex pipiens (Diptera: Culicidae) and other mosquitoes in Israel.

Host preferences of mosquitoes, mainly Culex pipiens L. and Aedes caspius (Pallas), were studied in Israel using live baits, chemical attractants, light, and suction traps. The feeding habits of Cx. pipiens were opportunistic. This species used a wide range of mammalian and avian hosts; a calf, chickens, turkeys, and sheep were the most attractive baits. A. caspius fed mainly on large mammals and was most attracted to calf and sheep baits. Although sheep were attractive, few Cx. pipiens and no Ae. caspius engorged after entering the traps. In addition to defensive behavior and the mechanical p

Survival of adults and development stages of Sarcoptes scabiei var. canis when off the host.

All life-stages of Sarcoptes scabiei var. canis survive in the hosts' environment for several days to several weeks depending on r.h. and temperature. Survival of larvae was comparable to males; survival of nymphs was comparable to females. Females and nymphs generally survived longer than larvae and males. Low temperature (10-15 degrees) and high r.h. prolonged survival of all life stages. At 10-15 degrees C, females and nymphs survived 1-3 weeks at 97% r.h., 1-2 weeks at 75% r.h. and 5-8 days at 45% r.h. At 20-25 degrees C, survival was significantly reduced but all life-stages survived at l

Feeding site selection in Rhodnius prolixus nymphs, American trypanosomiasis vector.

Thermal cues are fundamental for host-seeking in kissing bugs, primary vectors of American trypanosomiasis. Host surface temperature may reflect underlying vascular richness, potentially guiding feeding site selection. In this study, we investigated whether Rhodnius prolixus nymphs preferentially select specific regions of a mammalian host and whether local skin temperature influences this choice. Nymphs from first to fifth instars were allowed to feed on anesthetized mice, and insect densities were recorded across distinct body regions (head, lateral abdomen, paws and tail). Mouse body temper

Host preference of field-derived Schistosoma mansoni is influenced by snail host compatibility and infection status.

Schistosome parasites cause a chronic inflammatory disease in humans, and recent studies have emphasized the importance of control programs for understanding the aquatic phases of schistosomiasis transmission. The host-seeking behavior of larval schistosomes (miracidia) for their snail intermediate hosts plays a critical role in parasite transmission. Using field-derived strains of Kenyan snails and parasites, we tested two main hypotheses: (1) Parasites prefer the most compatible host, and (2) parasites avoid hosts that are already infected. We tested preference to three Biomphalaria host sna

Blood-feeding drive inhibition of Aedes sierrensis (Diptera: Culicidae) induced by the parasite Lambornella clarki (Ciliophora: Tetrahymenidae).

Female Aedes sierrensis (Ludlow) infected with the parasitic ciliate Lambornella clarki Corliss and Coats were significantly less responsive toward a human host than uninfected females. The proportion of infected females that were inhibited in host-seeking and blood-feeding remained high in all daily exposures to the host. Those that did respond took approximately twice as long to land on the host and 25% longer to complete probing as did uninfected females. In contrast, nearly all uninfected females more than 5 d old rapidly responded to and blood fed on the host. Uninseminated females were m

Neurotransmitters and molecular chaperones interactions in cerebral malaria: Is there a missing link?

Plasmodium falciparum is responsible for the most severe and deadliest human malaria infection. The most serious complication of this infection is cerebral malaria. Among the proposed hypotheses that seek to explain the manifestation of the neurological syndrome in cerebral malaria is the vascular occlusion/sequestration/mechanic hypothesis, the cytokine storm or inflammatory theory, or a combination of both. Unfortunately, despite the increasing volume of scientific information on cerebral malaria, our understanding of its pathophysiologic mechanism(s) is still very limited. In a bid to maint
