Plasma Levels of Intact Parathyroid Hormone and Congestion Burden in Heart Failure: Clinical Correlations and Prognostic Role.

Circulating parathyroid hormone (PTH) concentrations increase in heart failure (HF) and are related to disease severity. The relationship between PTH and congestion is still a matter of debate. The objective of this analysis was to evaluate the role of PTH as a marker of congestion and prognosis in HF. We enrolled 228 patients with HF. Intact PTH concentrations and HYDRA score (constituted by: B-type natriuretic peptide, blood urea nitrogen−creatinine ratio, estimated plasma volume status, and hydration status) were evaluated. The study endpoint was all-cause mortality. PTH levels were higher

In vitro differential developmental toxicity of vitamin A congeners.

Several forms of vitamin A were tested in the in vitro hydra assay for their developmental toxicity hazard potential and site of action on progressive ontogenesis. Retinol, retinyl acetate, retinaldehyde, all trans retinoic acid, and 13 cis retinoic acid were tested fully, and each was established clearly as being able to perturb development of artificial hydra "embryos" at, or near, adult toxic treatment levels. All forms of vitamin A tested interfered with differentiation, but, although the alcohol, acetate, and aldehyde forms (group I) prevented the initial stages of differentiation from oc

Complexities of smoking education.

Cigarette smoking is a many-headed Hydra, which grows new smokers' heads as fast as they are lopped off. Efforts to change smokers' behavior usually reach a point of diminishing returns, as evidenced by experiences in smoking withdrawal clinics and in public anti-smoking education campaigns. Obviously, many smokers are refactory to anti-smoking education, a situation which has generated a great deal of pharmacological, neurochemical, psychological, and sociological research. The National Clearinghouse for Smoking and Health in the U.S.A. has used a "gradualist" or permissive approach in anti-s

Self-organization of an organizer: Whole-body regeneration from reaggregated cells in cnidarians.

Cnidarians like the freshwater polyp Hydra and the sea anemone Nematostella, are famous for their enormous capacity to regenerate missing head or feet upon bisection. Classical transplantation experiments have demonstrated that the hypostome, the oral tip of the freshwater polyp Hydra, acts as an axial organizer. Likewise, transplantation of the blastopore lip of an early Nematostella gastrula stage embryo to an aboral position leads to ectopic head formation. Following molecular analyses have shown that Wnt signaling is the key component of this organizer activity. Moreover, when dissociated

Spontaneous body wall contractions stabilize the fluid microenvironment that shapes host-microbe associations.

The freshwater polyp Hydra is a popular biological model system; however, we still do not understand one of its most salient behaviors, the generation of spontaneous body wall contractions. Here, by applying experimental fluid dynamics analysis and mathematical modeling, we provide functional evidence that spontaneous contractions of body walls enhance the transport of chemical compounds from and to the tissue surface where symbiotic bacteria reside. Experimentally, a reduction in the frequency of spontaneous body wall contractions is associated with a changed composition of the colonizing mic

Pulmonary Deposition of Radionucleotide-Labeled Palivizumab: Proof-of-Concept Study.

OBJECTIVE: Current prevention and/or treatment options for respiratory syncytial virus (RSV) infections are limited as no vaccine is available. Prophylaxis with palivizumab is very expensive and requires multiple intramuscular injections over the RSV season. Here we present proof-of-concept data using nebulized palivizumab delivery as a promising new approach for the prevention or treatment of severe RSV infections, documenting both aerosol characteristics and pulmonary deposition patterns in the lungs of lambs. DESIGN: Prospective animal study. SETTING: Biosecurity Control Level 2-designate

Hybrid Diffusion Model for Stable, Affinity-Driven, Receptor-Aware Peptide Generation.

The convergence of biotechnology and artificial intelligence has the potential to transform drug development, especially in the field of therapeutic peptide design. Peptides are short chains of amino acids with diverse therapeutic applications that offer several advantages over small molecular drugs, such as targeted therapy and minimal side effects. However, limited oral bioavailability and enzymatic degradation have limited their effectiveness. With advances in deep learning techniques, innovative approaches to peptide design have become possible. In this work, we demonstrate HYDRA, a hybrid

Ancestral roles of atypical cadherins in planar cell polarity.

Fat, Fat-like, and Dachsous family cadherins are giant proteins that regulate planar cell polarity (PCP) and cell adhesion in bilaterians. Their evolutionary origin can be traced back to prebilaterian species, but their ancestral function(s) are unknown. We identified Fat-like and Dachsous cadherins in Hydra, a member of phylum Cnidaria a sister group of bilaterian. We found Hydra does not possess a true Fat homolog, but has homologs of Fat-like (HyFatl) and Dachsous (HyDs) that localize at the apical membrane of ectodermal epithelial cells and are planar polarized perpendicular to the oral-ab

Mechanisms underlying toxicity induced by CdTe quantum dots determined in an invertebrate model organism.

A systematic and thorough quantitative analysis of the in vivo effects of inorganic nanoparticles is extremely important for the design of functional nanomaterials for diagnostic and therapeutic applications, better understanding of their non-specificity toward tissues and cell types, and for assessments of their toxicity. This study was undertaken to examine the impact of CdTe quantum dots (QDs) on an invertebrate freshwater model organism, Hydra vulgaris, for assessment of long term toxicity effects. The continuous exposure of living polyps to sub-lethal doses of QDs caused time and dose dep

Accurate vibrational hydrogen-bond shift predictions with multicomponent DFT.

In this work we explore the use of multicomponent methods for the computational simulation of anharmonic OH vibrational shifts. Multicomponent methodologies have become popular over the last years, but still are limited in their application range. However, by enabling the simultaneous quantum treatment of protonic and electronic wave functions/densities, they hold promise for the treatment of anharmonic effects and proton vibrations in general. This potential has only been probed but not fully realized so far. This study investigates the performance of Nuclear-Electronic Orbital Density Functi

Neuroanatomical dimensions in major depression linked to cognition, adverse life events, self-harm, metabolomics and genetics.

BACKGROUND: Major depressive disorder (MDD) is a leading cause of disability worldwide, yet its diagnosis relies on clinical symptoms alone. METHODS: Using the semi-supervised machine learning algorithm, Heterogeneity through Discriminative Analysis (HYDRA), we had identified two neuroanatomical dimensions in deeply phenotyped (i.e., comprehensively assessed across neuroimaging, clinical, and behavioural domains), medication-free participants with MDD from the COORDINATE-MDD consortium. In the present study, we apply this pre-trained HYDRA model to the UK Biobank (UKB) to validate these dimen

Parkinson's disease subtypes and their association with probable rapid eye movement sleep behavior disorder severity: a brainstem tractography and machine learning investigation.

Rapid Eye Movement (REM) sleep behavior disorder (RBD) affects nearly half of Parkinson's disease (PD) patients. However, the structural heterogeneity within the brainstem, which regulates REM sleep, remains largely unexplored in PD. Our objective was to identify distinct PD subtypes based on microstructural characteristics in the brainstem and examine their associations with the severity of RBD. Data, including diffusion tensor imaging and REM sleep behavior disorder screening questionnaire (RBDSQ) responses, were obtained from 124 PD patients and 61 healthy controls through the Parkinson's P

Seasonal environmental change and sex change in a cnidarian.

In various animal species with distinct sexes, sequential hermaphroditism occurs whereby adults change their sex. The role of environmental factors in triggering sex change events is still relatively poorly understood, despite that climate-driven sex-ratio distortion may increase extinction risk. Here, we investigated the effect of seasonal temperature fluctuations on the expression of sex-related genes and sex change in the gonochoristic freshwater cnidarian, brown hydra (Hydra oligactis). Individuals of this species reproduce clonally throughout much of the year but switch to sexual reproduc

Comparison of Different HIV-1 Resistance Interpretation Tools for Next-Generation Sequencing in Italy.

BACKGROUND: Next-generation sequencing (NGS) is gradually replacing Sanger sequencing for HIV genotypic drug resistance testing (GRT). This work evaluated the concordance among different NGS-GRT interpretation tools in a real-life setting. METHODS: Routine NGS-GRT data were generated from viral RNA at 11 Italian laboratories with the AD4SEQ HIV-1 Solution v2 commercial kit. NGS results were interpreted by the SmartVir system provided by the kit and by two online tools (HyDRA Web and Stanford HIVdb). NGS-GRT was considered valid when the coverage was >100 reads (100×) at each PR/RT/IN resistan

Validation of publicly-available software used in analyzing NGS data for HIV-1 drug resistance mutations and transmission networks in a Washington, DC, Cohort.

The DC Cohort is an ongoing longitudinal observational study of persons living with HIV. To better understand HIV-1 drug resistance and potential transmission clusters among these participants, we performed targeted, paired-end next-generation sequencing (NGS) of protease, reverse transcriptase and integrase amplicons. We elected to use free, publicly-available software (HyDRA Web, Stanford HIVdb and HIV-TRACE) for data analyses so that laboratory personnel without extensive bioinformatics expertise could use it; making the approach accessible and affordable for labs worldwide. With more labor

Two distinct subtypes of obsessive compulsive disorder revealed by heterogeneity through discriminative analysis.

Neurobiological heterogeneity in obsessive compulsive disorder (OCD) is understudied leading to conflicting neuroimaging findings. Therefore, we investigated objective neuroanatomical subtypes of OCD by adopting a newly proposed method based on gray matter volumes (GMVs). GMVs were derived from T1-weighted anatomical images of patients with OCD (n = 100) and matched healthy controls (HCs; n = 106). We first inquired whether patients with OCD presented higher interindividual variability HCs in terms of GMVs. Then, we identified distinct subtypes of OCD by adopting heterogeneity through discrimi

Connectome Gradient-Based Subtyping of Major Depressive Disorder Reveals Distinct Neurobiological and Transcriptomic Signatures.

BACKGROUND/OBJECTIVE: Major depressive disorder (MDD) exhibits significant heterogeneity, and identifying these distinct biological subtypes aids clinical intervention. In this study, we employed functional gradient and Hydra clustering methods to distinguish subtypes of depression. METHOD: Imaging data were derived from a large-scale, multicenter MDD imaging database, comprising 1067 patients with MDD and 907 healthy controls (HCs). Based on the principal functional gradient, the Hydra method was employed to distinguish subtypes of MDD. We employed the Mann-Whitney U-test to compare function

Subtypes of insomnia revealed by the heterogeneity of neuroanatomical patterns: A structural MRI study.

The current conflicting neuroimaging findings of insomnia disorder (ID) may be attributed to heterogeneity in ID. The present study aims to clarify the high heterogeneity in ID and examine the objective neurobiological subtypes of ID by using a novel machine learning method based on gray matter volumes (GMVs). We recruited 56 patients with ID and 73 healthy controls (HCs). The T1-weighted anatomical images were obtained for each participant. We investigated whether the ID has higher interindividual heterogeneity in GMVs. Then, we used a heterogeneous machine learning algorithm by discriminativ

Primary body axes of vertebrates: generation of a near-Cartesian coordinate system and the role of Spemann-type organizer.

A rationale for the complex-appearing generation of the primary body axes in vertebrates can be obtained if this process is divided into two parts. First, an ancestral system is responsible for the anteroposterior (AP) patterning of the brain and the positioning of the heart. The blastopore (marginal zone) acts as a source region that generates primary AP-positional information for the brain, a process that is largely independent of the organizer. This evolutionary old system was once organizing the single axis of radial-symmetric ancestors. Second, the trunk is assumed to be an evolutionary l
