Elemental sulfur reduction in the deep-sea vent thermophile, Thermovibrio ammonificans.

The reduction of elemental sulfur is an important energy-conserving pathway in prokaryotes inhabiting geothermal environments, where sulfur respiration contributes to sulfur biogeochemical cycling. Despite this, the pathways through which elemental sulfur is reduced to hydrogen sulfide remain unclear in most microorganisms. We integrated growth experiments using Thermovibrio ammonificans, a deep-sea vent thermophile that conserves energy from the oxidation of hydrogen and reduction of both nitrate and elemental sulfur, with comparative transcriptomic and proteomic approaches, coupled with scan

Circatidal control of gene expression in the deep-sea hot vent shrimp Rimicaris leurokolos.

Biological clocks are a ubiquitous feature of all life, enabling the use of natural environmental cycles to track time. Although studies on circadian rhythms have contributed greatly to the knowledge of chronobiology, biological rhythms in dark biospheres such as the deep sea remain poorly understood. Here, based on a free-running experiment in the laboratory, we reveal potentially endogenous rhythms in the gene expression of the deep-sea hydrothermal vent shrimp Rimicaris leurokolos. Oscillations with approximately 12 h periods, probably reflecting tidal influence, greatly prevail over others

Geochemical Mobility of Elements in Antarctic Environments Affected by CO2-Rich Hydrothermal Fluids: Astrobiological Implications.

Hydrothermal systems are widespread in our solar system. Identification of alteration mineral assemblages on Mars and potentially in ocean worlds such as Enceladus suggests the existence of extensive hydrothermal fluid-igneous rock interactions of astrobiological interest in different planetary bodies. Here, we studied the terrestrial analog Cerro Caliente, a band of geothermal alterations located in the glaciovolcanic environment of Deception Island (Antarctica), with the aim of determining the mobility of major chemical elements (e.g., alkalis, phosphorus) and its implications in the habitab

Multi-scale design of the structure and mechanical performance of the deep-sea hydrothermal mussel (Bathymodiolus aduloides) shell.

Deep-sea hydrothermal vents are renowned for being among the most extreme environments on Earth. However, the mussel shells found in these vent sites demonstrate remarkable productivity, despite being subjected to high pressure as well as unusual levels of heavy metals, pH, temperature, CO2, and sulphides. To comprehend how these mussels endure such extreme conditions, a systematic comparative study was conducted, focusing on the unique chemical composition, structural designs, and mechanical properties of hydrothermal vent mussels (Bathymodiolus aduloides) in comparison to shallow-water musse

Resource partitioning among brachiopods and bivalves at ancient hydrocarbon seeps: A hypothesis.

Brachiopods were thought to have dominated deep-sea hydrothermal vents and hydrocarbon seeps for most of the Paleozoic and Mesozoic, and were believed to have been outcompeted and replaced by chemosymbiotic bivalves during the Late Cretaceous. But recent findings of bivalve-rich seep deposits of Paleozoic and Mesozoic age have questioned this paradigm. By tabulating the generic diversity of the dominant brachiopod and bivalve clades-dimerelloid brachiopods and chemosymbiotic bivalves-from hydrocarbon seeps through the Phanerozoic, we show that their evolutionary trajectories are largely unrela

Integrated multi-approaches reveal unique metabolic mechanisms of Vestimentifera to adapt to deep sea.

BACKGROUND: Vestimentiferan tubeworms are deep-sea colonizers, in which chemoautotrophic symbiosis was first observed. These animals are gutless and depend on endosymbiotic bacteria for organic compound synthesis and nutrition supply. Taxonomically, vestimentiferans belong to Siboglinidae and Annelida. Compared with other siboglinids, vestimentiferans are distinguished by high tolerance of the prevailing hydrogen sulfide in hydrothermal vents, rapid growth in local habitats, and a physical structure consisting of a thick chitinous tube. The metabolic mechanisms contributing to these features r

Infrared stimulation of mitochondrial metabolism in deep-sea yeast expands the energetic framework of hydrothermal ecosystems.

Deep-sea hydrothermal vents are archetypal chemosynthetic ecosystems, yet the intense geothermal near-infrared (NIR) radiation emitted by black smoker chimneys remains a largely unexplored environmental factor. While infrared (IR)-driven photosynthesis has been described in vent-associated bacteria, whether deep-sea eukaryotes can biologically respond to geothermal NIR radiation has remained unknown. Here, we demonstrate that the vent yeast Rhodotorula mucilaginosa F-J1 exhibits wavelength-specific growth stimulation under NIR exposure through a previously unrecognized mitochondria-centered me

Nitrogen Oxides in Early Earth's Atmosphere as Electron Acceptors for Life's Emergence.

We quantify the amount of nitrogen oxides (NOx) produced through lightning and photochemical processes in the Hadean atmosphere to be available in the Hadean ocean for the emergence of life. Atmospherically generated nitrate (NO3-) and nitrite (NO2-) are the most attractive high-potential electron acceptors for pulling and enabling crucial redox reactions of autotrophic metabolic pathways at submarine alkaline hydrothermal vents. The Hadean atmosphere, dominated by CO2 and N2, will produce nitric oxide (NO) when shocked by lightning. Photochemical reactions involving NO and H2O vapor will then

In situ deep-sea surface-enhanced Raman scattering sensor reveals cyanide as a key factor in cold seep nitrogen cycling.

Deep-sea hydrothermal vents and cold seeps, typical ecosystems characterized by abundant carbon but limited nitrogen, support thriving microbial activities. The source of nitrogen has long been a key focus in scientific research. Although cultivation experiments have confirmed the presence of diverse nitrogen-fixing microorganisms, direct in situ evidence has been lacking. Therefore, a deep-sea in situ surface-enhanced Raman scattering sensor has been developed. It can adapt to extreme environment (≥350°C and ≥2000-m depth and LOD < 10-7 M) and has excellent deep-sea application potential. Cru

Highly structured populations of copepods at risk to deep-sea mining: Integration of genomic data with demogenetic and biophysical modelling.

Copepoda is the most abundant taxon in deep-sea hydrothermal vents, where hard substrate is available. Despite the increasing interest in seafloor massive sulphides exploitation, there have been no population genomic studies conducted on vent meiofauna, which are known to contribute over 50% to metazoan biodiversity at vents. To bridge this knowledge gap, restriction-site-associated DNA sequencing, specifically 2b-RADseq, was used to retrieve thousands of genome-wide single-nucleotide polymorphisms (SNPs) from abundant populations of the vent-obligate copepod Stygiopontius lauensis from the La

Frankenstein or a Submarine Alkaline Vent: Who is Responsible for Abiogenesis?: Part 2: As life is now, so it must have been in the beginning.

We argued in Part 1 of this series that because all living systems are extremely far-from-equilibrium dynamic confections of matter, they must necessarily be driven to that state by the conversion of chemically specific external disequilibria into specific internal disequilibria. Such conversions require task-specific macromolecular engines. We here argue that the same is not only true of life at its emergence; it is the enabling cause of that emergence; although here the external driving disequilibria, and the conversion engines needed must have been abiotic. We argue further that the initial

Adaptation strategies of iron-oxidizing bacteria Gallionella and Zetaproteobacteria crossing the marine-freshwater barrier.

Iron-oxidizing Betaproteobacteria and Zetaproteobacteria are generally associated with freshwater and marine environments, respectively. Despite repeated cross-environment observations of these taxa, there has been no focused exploration of the genomes of marine Gallionella (Betaproteobacteria) to understand transitions between freshwater and marine habitats. Consequently, their roles in these environments remain uncertain. Here, we present strong evidence for the co-occurrence of Gallionella and Zetaproteobacteria at deep-sea hydrothermal vents at the Arctic Mid-Ocean Ridges through metagenom

Hawaiian Geothermal Fumaroles Contain Diverse and Novel Viruses.

Microbial communities of geothermal habitats are central to understanding the evolution of life on Earth. Metagenomics has provided insight into the role of viruses in shaping microbial diversity of complex environments. However, identification of novel viruses is constrained by lack of marker genes and low nucleotide similarities between related viral taxa. While microbial and viral diversity have been explored in terrestrial hot springs and hydrothermal vent systems, other volcanic features remain underexplored. Fumaroles (steam vents) are geothermal features that heat groundwater with magma

Early holobiont-level divergence and adaptation across seafloor geochemical contrasts in the deep-sea squat lobster Shinkaia crosnieri.

Deep-sea hydrothermal vents and methane seeps host unique animal communities sustained by chemosynthetic symbioses, yet how biological diversification proceeds across these geochemically distinct and geographically fragmented habitats remains poorly understood. Previous studies have largely focused on either host genomic adaptation or symbiont diversity as drivers of isolation, leaving unresolved how environmental geochemistry and host-microbe associations jointly shape evolutionary trajectories in chemosynthetic ecosystems. Here, we addressed this gap by combining chromosome-level host genomi

Prokaryotic Novelty and Metabolic Diversity in the Tagoro Shallow Hydrothermal System (Canary Islands, Spain).

The shallow hydrothermal system of the Tagoro submarine volcano (Canary Islands, Spain) has been discharging low-temperature, nutrient-rich, acidic fluids high in CO2 and H2S since March 2012. Despite influencing local microbial communities, the effects of these hydrothermal fluids on community structure and diversity across spatial gradients remain poorly understood. Using 16S rRNA gene sequencing, we observed significant differences in prokaryotic community structure from epipelagic waters to benthic habitats affected by hydrothermal activity. Prokaryotic richness and diversity decreased clo

Arctic deep-sea hydrothermal microbiomes as a natural niche for novel antimicrobial peptides.

BACKGROUND: The escalating threat of antimicrobial resistance (AMR) has created an urgent need for new antimicrobial agents. Antimicrobial peptides (AMPs) are promising alternatives to conventional antibiotics due to their broad-spectrum activity and reduced risk of resistance development. While most AMP discovery efforts have focused on terrestrial microbes, extreme environments remain largely untapped. Deep-sea hydrothermal vent biofilms, such as those from the Arctic Mid-Ocean Ridges (AMOR), are unique ecosystems characterized by high pressure, temperature gradients, and chemical extremes.

Characterization of biogenic iron oxides collected by the newly designed liquid culture method using diffusion chambers.

We designed a new culture method for neutrophilic iron-oxidizing bacteria using liquid medium (i) to study the formation and mineralogical characteristics of biogenic iron oxides (BIOS) and (ii) to apply BIOS to various scientific and engineering applications. An iron-oxidizing bacterium, Mariprofundus ferrooxydans PV-1(T) (ATCC, BAA-1020), was cultured using a set of diffusion chambers to prepare a broad anoxic-oxic interface, upon which BIOS formation is typically observed in natural environments. Iron oxide precipitates were generated in parallel with bacterial growth. A scanning electron m

Difference in sulfur regulation mechanism between tube-dwelling and free-moving polychaetes sympatrically inhabiting deep-sea hydrothermal chimneys.

The environment around deep sea hydrothermal vents is characterized by an abundance of sulfur compounds, including toxic hydrogen sulfide. However, numerous communities of various invertebrates are found in it. It is suggested that invertebrates in the vicinity of hydrothermal vents detoxify sulfur compounds by biosynthesis of taurine-related compounds in the body. On the other hand, the vent endemic polychaete Alvinella pompejana has spherocrystals composed of sulfur and other metals in its digestive tract. It was considered that the spherocrystals contribute to the regulation of sulfur in bo

Kinetics of CO2 reduction across Fe(Ni)S precipitates: the role of pH gradients.

Alkaline hydrothermal vents have been proposed as geochemical reactors that could sustain prebiotic carbon fixation via natural pH and redox gradients. Here, we investigate the kinetics of CO2 reduction to formic acid across Fe(Ni)S precipitates in a microfluidic analogue of a hydrothermal vent. By combining controlled experiments with a minimal kinetic model, we demonstrate that formate production depends sensitively on both the absolute pH of the ocean-side compartment and the magnitude of the pH differential across the mineral interface. The model, which incorporates electron transfer throu
