Effects of histamine and cimetidine on the levels of serotonin and 5-hydroxyindoleacetic acid in various parts of the digestive tract and in the blood and brain of rats.

In the investigations on male Wistar rats it was demonstrated that histamine (0.05 and 0.5 mg/kg) decreased the serotonin level, without affecting the level of 5-HIAA in the stomach and duodenum. Contrary to this, cimetidine (15, 75 and 150 mg/kg) raised slightly the level of serotonin and decreased the 5-HIAA level in the stomach and duodenum. In the jejunum histamine in the lower dose raised the levels of serotonin and 5-HIAA, and in the higher dose it decreased only the concentration of serotonin. Cimetidine, on the other hand, only in the highest dose increased the serotonin level and decr

Effect of Lanreotide Depot/Autogel on Urinary 5-Hydroxyindoleacetic Acid and Plasma Chromogranin A Biomarkers in Nonfunctional Metastatic Enteropancreatic Neuroendocrine Tumors.

BACKGROUND: Urinary 5-hydroxyindoleacetic acid (5-HIAA) is an established biomarker in neuroendocrine tumors and carcinoid syndrome; however, its role in nonfunctional neuroendocrine tumors is not defined. We present post hoc data on urinary 5-HIAA and plasma chromogranin A (CgA) from the CLARINET study. METHODS: Patients with well- or moderately differentiated, nonfunctioning, locally advanced or metastatic enteropancreatic neuroendocrine tumors were randomized to deep subcutaneous lanreotide depot/autogel 120 mg or placebo once every 28 days for 96 weeks. Tumor response, evaluated centrally

Pharmacological properties of new cyclic derivatives of phenylsuccinimide and their influence on noradrenaline, dopamine, serotonin, 5-hydroxyindoleacetic acid, gamma-aminobutyric acid concentrations and monoaminooxidase activity in animal brains.

Three new cyclic derivatives of phenylsuccinimide (I-10, I-11 and IL-7) were subjected to preliminary pharmacological studies in animals. Minimal peripheral action and inhibitory influence on the CNS were noted (only IL-7) was found to stimulate CNS). Neurotoxic effects were similar to those of the classical succinimides . Anticonvulsant activity and behavioral effects of tested compounds are discussed in the light of their influence on neurotransmitters.

Developmental changes in serotonin and 5-hydroxyindoleacetic acid concentrations and opiate receptor binding in rat spinal cord following neonatal 5,7-dihydroxytryptamine treatment.

Normal levels of serotonin and 5-hydroxyindoleacetic (5-HIAA) were measured in the developing rat spinal cord and brain stem using HPLC. Serotonin, 5-HIAA and 3H-naloxone binding were monitored following neonatal 5,7-dihydroxytryptamine (5,7-DHT) treatment. Serotonin in the spinal cord develops biphasically with peaks at gestation day 21 (E21) and postnatal day 8 (P8) while 5-HIAA peaks at P3 and between P8 and P11. In the brain stem, serotonin gradually rises from E15 to adult levels which are reached by P32 while 5-HIAA rises sharply to P15 and thereafter decreases to adult levels by P32. 5,

Postbiotic of Bifidobacterium breve i1088 alleviates type 2 diabetes by promoting GLP-1 secretion: identification of 5-hydroxyindoleacetic acid as a key active metabolite.

OBJECTIVE: The pathogenesis of type 2 diabetes mellitus (T2DM) is intrinsically linked to the suboptimal secretion of glucagon-like peptide-1 (GLP-1). While postbiotics have emerged as a compelling therapeutic modality for metabolic modulation, the precise bioactive effectors and their underlying mechanisms remain largely elusive. This study aimed to evaluate the effect of Bifidobacterium breve i1088 (B breve i1088) lysate on GLP-1 secretion and identify its key active molecules. METHODS: The GLP-1 secretory activity of i1088 lysate was assessed in human enteroendocrine NCI-H716 cells. Subseq

Changes in plasma levels of serotonin and 5-hydroxyindoleacetic acid and population of serotonin-secreting cells in small and large intestine of Wistar rats in hypo- and hyperandrogenemia.

We studied the effects of hypo- and hyperandrogenemia on the plasma levels of serotonin and its major metabolite 5-hydroxyindoleacetic acid as well as on the size of EC1 cell pool in the small and large intestine of adult male Wistar rats in the control group, after orchiectomy, and after administration of long-acting testosterone Omnadren 250. Orchiectomy did not change the levels of serotonin, 5-hydroxyindoleacetic acid, and the number of AN-1-cells in the epithelium of the small and large intestine. Administration of Omnadren 250 increased serum level of free testosterone and reduced plasma

Carcinoid heart disease: the role of urinary 5-hydroxyindoleacetic acid excretion and plasma levels of atrial natriuretic peptide, transforming growth factor-beta and fibroblast growth factor.

BACKGROUND: Serotonin excretion plays a role in the development of carcinoid heart disease (CHD), but the exact pathogenesis is not known. In the current study, the authors evaluated 24-hour urinary 5-hydroxyindoleacetic acid (5-HIAA) excretion, as well as plasma levels of transforming growth factor-beta (TGF-beta), fibroblast growth factor (FGF), and atrial natriuretic peptide (ANP) in patients with and without CHD determined by ultrasound examination. METHODS: Urine and plasma samples were obtained for 37 patients and cardiac ultrasound was performed during follow-up in 1999 and 2000. Media

Development and validation of dispersive liquid-liquid microextraction coupled with LC-MS/MS for plasma 5-hydroxyindoleacetic acid quantification with application as a biomarker in major depressive disorder.

A sensitive dispersive liquid-liquid microextraction (DLLME) coupled with liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed and validated for plasma 5-hydroxyindoleacetic acid (5-HIAA) quantification as a depression biomarker. Box-Behnken design evaluated four DLLME factors (disperser volume, extraction volume, pH, centrifugation time) where second-order polynomial model identified optimal conditions with R² = 0.9444, achieving 95.1% extraction recovery. Chromatographic separation employed Poroshell 120 EC-C18 column (2.7 μm, 50 × 3.0 mm) with isocratic elution (20

A longitudinal study of the relationships between psychometric test scores, offence history and the plasma concentrations of phenylacetic and 5-hydroxyindoleacetic acids in seven inmates of a prison for the psychiatrically disturbed.

1. The plasma concentrations of phenylacetic (PAA) and 5-hydroxyindoleacetic (5HIAA) acids in seven inmates incarcerated in the Regional Psychiatric Centre (Prairies), Correctional Service of Canada, were assessed each weekday for four weeks (i.e., 20 samples each). Psychometric assessments for hostility, anger, depression and anxiety were also performed daily. Mean differences between subjects in psychometric and biochemical measures were subjected to tests of statistical significance. 2. The subject who was clearly most aggressive by offence history/institutional behavior scored significantl

Endogenous release of neuronal serotonin and 5-hydroxyindoleacetic acid in the caudate-putamen of the rat as revealed by intracerebral dialysis coupled to high-performance liquid chromatography with fluorimetric detection.

Extracellular levels of endogenous serotonin (5-HT) and its major metabolite, 5-hydroxyindoleacetic acid (5-HIAA), were measured in the caudate-putamen of anesthetized and awake rats using intracerebral microdialysis coupled to HPLC with fluorimetric detection. A dialysis probe (of the loop type) was perfused with Ringer solution at 2 microliters/min, and samples collected every 30 or 60 min. Basal indole levels were followed for up to 4 days in both intact and 5,7-dihydroxytryptamine (5,7-DHT) lesioned animals. Immediately after the probe implantation, the striatal 5-HT levels were about 10 t

Effects of estradiol on the serotonin secretion and turnover in the hypothalamus of male tilapia, Oreochromis mossambicus, in vitro.

The effects of estradiol on the secretion and turnover of serotonin in the hypothalamic fragments of male tilapia, Oreochromis mossambicus, were studied using a static incubation system. The quantitative analysis of serotonin and its related metabolite, 5-hydroxyindoleacetic acid, were performed by high-performance liquid chromatography with electrochemical detection. The hypothalamic fragments were incubated with 17 beta-estradiol at a concentration of 2 x 10(-8), 8 x 10(-8), 2 x 10(-7), 4 x 10(-7), or 4 x 10(-6) g/ml. Serotonin metabolism in patients with carcinoid tumors: incidence of 5-hydroxytryptophan-secreting tumors.

The urinary excretion of 5-hydroxyindoleacetic acid, 5-hydroxytryptophan, serotonin, tryptamine, and tyramine and the serum serotonin concentration were determined in 38 consecutive patients with metastatic carcinoid tumors. Four of the 23 patients with the carcinoid syndrome (15%) had markedly elevated excretion of 5-hydroxytryptophan. None of the patients had markedly elevated excretion of tryptamine or tyramine; a substantial number of patients had moderately low urinary excretion of these amines. All of the patients with the carcinoid syndrome had elevated urinary 5-hydroxyindoleacetic aci

Monoamine metabolites in the cerebrospinal fluid in infantile spinal muscular atrophy.

Analyses of tryptophan and tyrosine metabolites in cerebrospinal fluid were performed to examine an abnormality of the monoamine neurone system in severe infantile spinal muscular atrophy (SMA type I). The levels of 5-hydroxyindoleacetic acid and homovanillic acid were significantly lower than those in controls. Decreases in the concentration of 5-hydroxyindoleacetic acid and kynurenine seemed to be related to the severity of SMA type I. These results suggest that the monoamine neurone system may play a role in the pathophysiology of SMA type I.

Do indole markers predict carcinoid heart disease?

Deficiency of tryptophan with elevated serum serotonin and liver dysfunction are the prerequisites for the experimental production of cardiac lesions in the guinea pig model of carcinoid syndrome. To apply the above principles in human subjects with carcinoid disease, various indole markers were compared in patients with or without heart involvement, to a group of normal subjects. In the present study, plasma tryptophan (T), serotonin (5HT), and urinary 5 hydroxyindoleacetic acid (5HIAA) measurements were made in 18 (group 1) patients with carcinoid syndrome and 24 normal individuals (group 2)

The depletion of tryptophan and serotonin in the brain of developing hyperphenylalaninemic rats is abolished by the additional administration of lysine.

In suckling hyperphenylalaninemic (hyper-Phe) rats, all essential amino acids including tryptophan are depleted in the blood. The inadequate supply of Trp to the developing brain leads to a decline of Trp, of serotonin, and of 5-hydroxyindoleacetic acid. The exhaustion of amino acids in both blood and brain can be restored by administration of Lys. Even though Phe is still elevated in blood and brain, Trp, serotonin and 5-hydroxyindoleacetic acid, are no longer depleted in the brain. This observation contradicts the idea that the serotonin deficit in the developing hyper-Phe brain is caused by

Small infarctions of cochlear, retinal, and encephalic tissue in young women.

BACKGROUND AND PURPOSE: Recently, a rare syndrome that involves uniformly the brain, inner ear, and retina in previously healthy young women has been described. Brain biopsies and ophthalmologic examinations disclosed small infarcts as a pathoanatomical substrate of the disease. In previous reports, an autoimmune disorder or a coagulopathy have been suggested as possible etiologies. CASE DESCRIPTIONS: Both patients (aged 22 and 20 years) had brain involvement with neurological and neuropsychological deficits. Multifocal small hyperintensities were shown in magnetic resonance imaging of the br
