Stabilities of hydroxyl radical spin adducts of PBN-type spin traps.

The stability of the hydroxyl spin adduct of nine different PBN-type spin traps has been examined in phosphate buffer solutions of various pH. The hydroxyl adduct is produced by short illumination of hydrogen peroxide with UV light in the presence of spin trap and the decay of its EPR signal followed. The stability measured by the half life of the first-order decay is strongly dependent on the pH of the solution and the structure of the aromatic ring used in the trap. All hydroxyl adducts are more stable in acidic media. tert-Butyl hydroaminoxyl is detected as a degradation product of the hydr

Characterization of hydroxyl free radical mediated damage to plasmid pBR322 DNA.

We have investigated hydroxyl free radical mediated damage to pBR322 DNA produced by ascorbate/iron and oxygen in a phosphate-buffered in vitro system. An observed lag phase in DNA nicking suggests a multi-target model of hydroxyl free radical attack on DNA. In the present report we further examine the model system and show that there is a "heat labile" component of the ascorbate/iron system which can be completely restored by the readdition of ascorbate. These observations have allowed us to rule out the possibility that intermediates build up in the reaction and act independently of ascorbat

Visualizing hydroxyl radical by a dual-channel NIR fluorescence chemosensor within biosystem.

Hydroxyl radicals (•OH) are the predominant oxidative species in living systems and are associated with various diseases and physiological disorders. Because of its extremely short lifetime and inherently low steady-state concentration, real-time detection of •OH in environmental and biological systems remains highly challenging, despite its critical regulatory function. In the present study, we report the design and successful synthesis of a novel dual-channel fluorescent probe, HR-YT, for sensitive •OH monitoring. The probe exhibits a fast response along with excellent sensitivity, achieving

H2O2-Responsive Nanocatalysts for Synergistic Hydroxyl and Chlorine Radical-Mediated Tumor Therapy.

Free radicals with reactive chemical properties can combat tumors without inducing drug resistance. Reactive oxygen species (ROS) have been widely applied in tumor therapy; however, the prevalent hypoxic conditions within tumors severely hinder the generation of ROS, leading to suboptimal antitumor efficacy. Herein, we designed an endogenous H2O2-triggered chlorine radical nanogenerator (Cl•) and hydroxyl radicals(•OH), which have better kinetic advantages and higher catalytic activity in catalyzing the H2O2 reaction. In vitro cytotoxicity tests revealed that the nano-urchins induced marked ca

Nanobubble Reactivity: Evaluating Hydroxyl Radical Generation (or Lack Thereof) under Ambient Conditions.

Nanobubble (NB) generation of reactive oxygen species (ROS), especially hydroxyl radical (·OH), has been controversial. In this work, we extensively characterize NBs in solution, with a focus on ROS generation (as ·OH), through a number of methods including degradation of ·OH-specific target compounds, electron paramagnetic resonance (EPR), and a fluorescence-based indicator. Generated NBs exhibit consistent physical characteristics (size, surface potential, and concentration) when compared with previous studies. For conditions described, which are considered as high O2 NB concentrations, no d

Significant Contribution of Solid Organic Matter for Hydroxyl Radical Production during Oxygenation.

Dark formation of hydroxyl radicals (•OH) from soil/sediment oxygenation has been increasingly reported, and solid Fe(II) is considered as the main electron donor for O2 activation. However, the role of solid organic matter (SOM) in •OH production is not clear, although it represents an important electron pool in the subsurface. In this study, •OH production from oxygenation of reduced solid humic acid (HAred) was investigated at pH 7.0. •OH production is linearly correlated with the electrons released from HAred suspension. Solid HAred transferred electrons rapidly to O2 via the surface-reduc

Hydroxyl radical is the major causative factor in stress-induced gastric ulceration.

The role of the metal-catalyzed production of hydroxyl radicals (OH.) on gastric ulceration caused by restraint-cold stress in rat was studied. Stress causes a 50% increase in the thiobarbituric acid reactive species (TBARS) as a measure of the lipid peroxidation, nearly 70% increase in protein oxidation as measured by its carbonyl content and about 40% decrease in the glutathione content of the fundic stomach, suggesting oxidative damage by stress. Stress also causes a time-dependent increase in the mitochondrial superoxide dismutase activity and a decrease in the peroxidase activity, both of

Evidence of hydroxyl free radical generation by calcium overload in rat myocardium.

Although calcium (Ca2+) is important in cardiac dysfunction and has also been reported as a source of oxidative toxicity, the connection between Ca2+ overload and oxygen free radicals in the myocardium is not clear. We have investigated whether Ca2+ overload generates hydroxyl free radicals in rat ventricle. HPLC with electrochemical detection was used to measure the levels of 2,3- and 2,5-dihydroxybenzoic acid (DHBA) formed when the hydroxyl free radical reacts with salicylate. Ringer's solution containing salicylic acid (0.5 nmol microL-1 min-1) was infused through a microdialysis probe in t

Hydroxyl radical formation in hyperglycemic rats during middle cerebral artery occlusion/reperfusion.

Preexisting hyperglycemia is associated with enhanced reperfusion injury in the postischemic rat brain. The goal of this study was to evaluate whether the hyperglycemic exacerbation of brain injury is associated with enhanced generation of hydroxyl radicals in rats subjected to middle cerebral artery occlusion (2 h), followed by reperfusion (2 h). Magnetic resonance images revealed the exacerbation of focal brain injury in hyperglycemic rats. The salicylate trapping method was used in conjunction with microdialysis to continuously estimate hydroxyl radical production by measurement of the stab

Brain temperature alters hydroxyl radical production during cerebral ischemia/reperfusion in rats.

Selective neuronal cell death in the CA1 pyramidal cells of the hippocampus and neurons of the dorsolateral striatum as a consequence of brain ischemia/reperfusion (IR) can be ameliorated with brain hypothermia. Since postischemic injury is mediated partially by chemical production of reactive oxygen species (ROS), decreased ROS production may be one of the mechanisms responsible for cerebral protection by hypothermia. To determine if ischemic brain temperature alters ROS production, reversible IR was produced in rats by occlusion of both carotid arteries with hemorrhagic hypotension. After 15

[Hydroxyl radical generation and post-ischemic reperfusion brain damage implications for neuroprotection].

We report the effects of the free radical scavengers, salicylic acid and bifemerane HC1, on the survival of hippocampal neurons in the Mongolin gerbil model of ischemia-reperfusion brain damage. In addition to performing histological examinations, we used the salicylate-trapping method to measure the amounts of hydroxyl radicals generated in the cerebral cortex and the hippocampus. Ischemia-reperfusion significantly reduced the number of neurons in the CA1 region of the hippocampus. The decrease was largely prevented by pretreating the animals with either salicylate, 20 mg/kg, or bifemerane HC

Iron supplementation generates hydroxyl radical in vivo. An ESR spin-trapping investigation.

Electron spin resonance (ESR) spectroscopy has been used to investigate hydroxyl radical generation in rats with chronic dietary iron loading. A secondary radical spin-trapping technique was used where hydroxyl radical forms methyl radical upon reaction with DMSO. The methyl radical was then detected by ESR spectroscopy as its adduct with the spin trap alpha-phenyl-N-t-butylnitrone (PBN). This adduct was detected in the bile of rats 10 wk after being fed an iron-loading diet and 40 min after the i.p. injection of the spin trap PBN dissolved in DMSO. Bile samples were collected into a solution

Myocardial microdialysis of salicylic acid to detect hydroxyl radical generation during ischemia.

We examined in vivo monitoring of hydroxyl radical (.OH) generation during myocardial ischemia for 30 min by occluding the left anterior descending (LAD) in dog heart using a microdialysis technique. The hydroxyl radical reacts with salicylate and generates 2,3- and 2,5-dihydroxybenzoic acids (DHBA) which can be measured electrochemically in picomole quantity by a high pressure liquid chromatography-electrochemical (HPLC-EC) procedure. When the premature ventricular contraction (PVC) occurred at almost 25 sec intervals, marked elevation of the levels of 2,3- and 2,5-DHBA was observed in the he

Hydroxyl radical formation and lipid peroxidation enhancement by chromium. In vitro study.

Chromium VI compounds have been shown to be carcinogenic in occupationally exposed humans, and to be genotoxic, mutagenic, and carcinogenic in a variety of experimental systems. In contrast, most chromium III compounds are relatively nontoxic, noncarcinogenic, and nonmutagenic. Reduction of Cr6+ leads to reactive intermediates, such as Cr5+, Cr4+, or other radical species. The molecular mechanism for the intracellular Cr6+ reduction has been the focus of recent studies, but the details are still not understood. Our study was initiated to compare the effect of Cr(6+)-hydroxyl radical formation

Free radical activity of synthetic vitreous fibers: iron chelation inhibits hydroxyl radical generation by refractory ceramic fiber.

Synthetic vitreous fibers are in widespread use but the parameters that dictate their carcinogenicity are still a matter of scientific debate. The free radical activities of a panel comprising an asbestos sample and five different respirable synthetic vitreous fiber samples were determined, to address the hypothesis that carcinogenic fibers have greater free radical activity than noncarcinogenic fibers. On the basis of recent inhalation studies, the six samples were divided into three carcinogenic fibers-amphibole asbestos, silicon carbide, and refractory ceramic fiber 1 (designated with the a

Quantification of Plasma-Produced Hydroxyl Radicals in Solution and their Dependence on the pH.

HO radicals are the most important reactive species generated during water treatment by non-thermal plasma devices. In this letter, we report the first quantification of the steady-state concentration and lifetime of plasma-produced hydroxyl radicals in water solutions at pH 3 and 7, and we discuss the differences based on their reactivity with other plasma-generated species. Finally, we show to what extent the use of chemical probes to quantify short-lived reactive species has an influence on the results and that it should be taken into account.

Sialic acid is an essential moiety of mucin as a hydroxyl radical scavenger.

In this work, we examined the antioxidant role of mucin, a typical sialic acid containing high-molecular weight glycoprotein. The function of mucin as a hydroxyl radical (.OH) scavenger was characterized using bovine submaxillary gland mucin (BSM). Non-treated BSM effectively protected DNA from the attack of .OH; however, desialylated BSM lost this potential. Moreover, we estimated the scavenging effects of BSM against .OH generated by UV irradiation of hydrogen peroxide using ESR analysis. Our results indicate that BSM has .OH scavenging ability the and sialic acid in mucin is an essential mo

Hydroxyl radicals production in the vanadium ions/dialuric acid systems.

1. The effects of vanadium ions on the .OH radical production in the presence of and in the absence of dialuric acid were studied. 2. Dialuric acid enhanced deoxyribose degradation. 3. Vanadium ions and vanadium/EDTA complexes decreased the degradation of deoxyribose in the presence and in the absence of dialuric acid. 4. The question as to whether or not free .OH radicals are formed via reaction of vanadium ions with H2O2 in the presence of dialuric acid is discussed. 5. The results are interpreted with a view to the vanadium ability to decrease the toxic effects of dialuric acid.

A new method for the detection of hydroxyl radical production by phagocytic cells.

Benzoic acid, a specific scavenger of hydroxyl radical (OH.) is known to be oxidized as the result of a reaction with OH.. We have determined that the decarboxylation of benzoic acid can be used to detect OH. generated in cell-free systems and human granulocytes. Benzoic acid is oxidized by the xanthine-xanthine oxidase enzyme system. This system is known to generate O2-, H2O2 and OH.. This oxidation is inhibited by superoxide dismutase, catalase and mannitol. Therefore, the oxidation of benzoic acid occurs by a mechanism similar to that reported for the oxidation of methional to ethylene and
