Analysis of chromosome damage for biodosimetry using imaging flow cytometry.

The dicentric chromosome assay (DCA), which involves counting the frequency of dicentric chromosomes in mitotic lymphocytes and converting it to a dose-estimation for ionizing radiation exposure, is considered to be the gold standard for radiation biodosimetry. Furthermore, for emergency response, the DCA has been adapted for triage by simplifying the scoring method [1]. With the development of new technologies such as the imaging flow cytometer, it may now be possible to adapt this microscope-based method to an automated cytometry method. This technology allows the sensitivity of microscopy t

Label-Free Near-Infrared Image Cytometry for Chlorophyll Quantification and Carotenoid Prediction with Multiparametric Biophysicochemical Profiling.

Microalgae offer a promising platform for carotenoid manufacturing due to their productivity, tunable selectivity, and compatibility with large-scale bioprocessing. Yet, their cellular responses exhibit highly heterogeneous and asynchronous pigment and morphological transitions under fluctuating light stress, making precise prediction and process optimization challenging. Existing analytical methods cannot resolve these transitions in living cells because they either require destructive sampling or lack spectral specificity. In this study, we introduce near-infrared (nIR) image cytometry (NIC)

Image cytometry and topographical analysis of proliferation of endothelial cells in vitro during Bartonella (Rochalimaea) infection.

Bartonella quintana and Bartonella henselae are clinically associated with proliferative neovascular lesions. The effect of Bartonella infection on human endothelial cells was evaluated in vitro by quantitative image analysis. Particular emphasis is placed on reporting the methodologies employed. Human umbilical vein endothelial cells were infected in vitro with the two Bartonella species. Cell proliferation (cell density), cell morphology (cell surface, form and elongation factors) and spatial reorganization (global topographical analysis and hierarchical cluster detection) were monitored ove

[Image analytic DNA cytometry--a possibility for assessment of prognosis after resection of ductal pancreatic cancers?].

In this study we analysed DNA-ploidy as a potential prognostic parameter in ductal pancreatic carcinoma. Paraffin embedded histological material, obtained by resection from 34 patients with a ductal pancreatic carcinoma, was selected for analysis. Tumor areas within the paraffin embedded material were identified by HE-stained reference sections. One 50 microns section was dewaxed, rehydrated and mechanically and enzymatically prepared to form a suspension of 10000 cells/ml. One milliliter of the suspension, which contained bare nuclei with small rests of cytoplasma, was centrifuged on glass sl

A Comparative Study of Liquid-Based Cytology and DNA Image Cytometry in the Diagnosis of Serous Effusion.

BACKGROUND: Liquid-based cytology is one of the most useful methods to diagnose a patient with serous effusion, especially when malignancy is suspected. As an alternative to the use of liquid-based cytology only, the serous effusion can be further processed using the technique of DNA image cytometry, which may augment diagnostic utility. The aim of this study was to compare the diagnostic yields of liquid-based cytology, DNA image cytometry, and both in combination, regardless of serous-effusion etiology. METHODS: We conducted a descriptive study on patients with serous effusions from July 20

DNA ploidy and S-phase fraction by image and flow cytometry in breast cancer fine-needle cytopunctures.

The aim of this study was to achieve a better definition of the respective indications of flow cytometry (FCM) and image cytometry (ICM) in clinical practice by comparing their efficiency in a series of 104 primary breast carcinomas that were sampled by means of fine-needle cytopuncture and analyzed by both methods. The comparison involved the DNA content and the estimation of S-phase fraction (SPF). For this purpose, a manual rectangular model was used for ICM, and Modfit software was used for FCM. With respect to DNA ploidy, the concordance rate between the two methods was 87% (DNA diploid v

Differential effects of depleting agents on cytoplasmic and nuclear non-protein sulphydryls: a fluorescence image cytometry study.

The intracellular distribution of glutathione (GSH) was measured by a quantitative image cytometry method, using the sulphydryl-reactive agent mercury orange. This readily forms fluorescent adducts with GSH and other non-protein sulphydryls (NPSH), but reacts much more slowly with protein sulphydryls. Under optimum staining conditions mean integrated mercury orange fluorescence per cell was closely correlated with a standard biochemical assay for GSH. Use of the DNA dye DAPI as a counterstain allowed measurement of nuclear NPSH. The mean nuclear-cytoplasmic ratio was 0.57 +/- 0.05. Isolation o

Comparison of ploidy analysis by flow cytometry and image analysis in hydatidiform mole and non-molar abortion.

Determination of DNA ploidy is useful in the diagnosis and classification of hydatidiform mole. Most reports of ploidy analysis in molar tissue have used DNA flow cytometry. Although image analysis cytometry offers theoretical advantages over flow cytometry, there have been few reports of ploidy analysis by image analysis in hydatidiform mole. We selected 47 cases and measured DNA ploidy by flow cytometry and image analysis cytometry in complete hydatidiform mole, partial hydatidiform mole and non-molar abortion. The two cytometry modalities were compared using kappa statistics. There was reas

Three-dimensional imaging flow cytometry: current trends and future perspectives.

Imaging flow cytometry (IFC) combines microscopic techniques and flow cytometry, enabling high-throughput, population-scale single-cell analysis with rich spatial detail and high molecular specificity. To date, most established IFC platforms are fundamentally built around 2D imaging, which is inherently limited in its ability to capture the complex 3D morphology, intracellular organization, and spatially distributed molecular features of single cells under flow. Driven by the growing demand for volumetric cellular information, IFC has progressively evolved from 2D imaging toward 3D imaging flo

Multidimensional profiling of cancer microenvironments in FFPE tissues by imaging mass cytometry.

In recent years, significant advancements have been achieved in the development of multiplex imaging methodologies for immunophenotyping, enabling a comprehensive characterization of the complexity of tumor microenvironments. Imaging mass cytometry combines the detection of over 40 cellular targets with spatial information, enabling the identification of not only which cells are present in a tissue but also their localization relative to each other. Here, we present an easy-to-implement imaging mass cytometry workflow that ranges from antibody selection and testing to running a full panel. Add

Development of a 2-dimensional atlas of the human kidney with imaging mass cytometry.

An incomplete understanding of the biology of the human kidney, including the relative abundances of and interactions between intrinsic and immune cells, has long constrained the development of therapies for kidney disease. The small amount of tissue obtained by renal biopsy has previously limited the ability to use patient samples for discovery purposes. Imaging mass cytometry (IMC) is an ideal technology for quantitative interrogation of scarce samples, permitting concurrent analysis of more than 40 markers on a single tissue section. Using a validated panel of metal-conjugated antibodies de

A Map of Human Type 1 Diabetes Progression by Imaging Mass Cytometry.

Type 1 diabetes (T1D) results from the autoimmune destruction of insulin-producing β cells. A comprehensive picture of the changes during T1D development is lacking due to limited sample availability, inability to sample longitudinally, and the paucity of technologies enabling comprehensive tissue profiling. Here, we analyzed 1,581 islets from 12 human donors, including eight with T1D, using imaging mass cytometry (IMC). IMC enabled simultaneous measurement of 35 biomarkers with single-cell and spatial resolution. We performed pseudotime analysis of islets through T1D progression from snapshot

Visualization and quantification of cytotoxicity mediated by antibodies using imaging flow cytometry.

Conventional approaches for the detection of antibody dependent cell-mediated cytotoxicity (ADCC) activity rely on quantification of the release of traceable compounds from target cells or flow cytometry analysis of population-wide phenomena. We report a new method for the direct imaging and quantification of ADCC of cancer cells. The proposed method using imaging flow cytometry combines the statistical power of flow cytometry with the analytical advantages of cell imaging, providing a novel and more comprehensive perspective of effector/target cell interactions during ADCC events. Easy-to-use imaging-cytometry assay to analyze chitin patterns in yeasts.

BACKGROUND & AIM: Pathogenic fungi are a major threat to public health, and fungal infections are becoming increasingly common and treatment resistant. Chitin, a component of the fungal cell wall, modifies host immunity and contributes to antifungal resistance. Moreover, chitin content is regulated by chitin synthases and chitinases. However, the specific roles and mechanisms remain unclear. In this study, we developed a cytometric imaging assay to quantify chitin content and identify the distribution of chitin in the yeast cell wall. METHODS: The Candida albicans SC5314 and Nakaseomyces glab
