Protection against cancer by dietary IP6 and inositol.

Inositol hexaphosphate (IP(6)) is a naturally occurring polyphosphorylated carbohydrate, abundantly present in many plant sources and in certain high-fiber diets, such as cereals and legumes. In addition to being found in plants, IP(6) is contained in almost all mammalian cells, although in much smaller amounts, where it is important in regulating vital cellular functions such as signal transduction, cell proliferation, and differentiation. For a long time IP(6) has been recognized as a natural antioxidant. Recently IP(6) has received much attention for its role in cancer prevention and contro

Sevoflurane stimulates inositol 1,4,5-trisphosphate in skeletal muscle.

UNLABELLED: Inositol 1,4,5-triphosphate (IP(3)) plays an important role in excitation-contraction coupling and malignant hyperthermia in skeletal muscle. We investigated whether sevoflurane affects IP(3) formation in L(6) skeletal muscle cells and studied the mechanisms that modulate IP(3). Sevoflurane stimulated IP(3) production from a basal level of 78.4 +/- 6.1 to 730.0 +/- 53.1 pmol. mg. protein(-1) in 2 mM of sevoflurane in a dose-dependent manner. A dose of 10 microM of U73122 (a phospholipase C antagonist) significantly decreased 0.8 mM of sevoflurane-stimulated IP(3) production from 38

Dietary myo-inositol therapy in hyperglycemia-induced embryopathy.

Dysmorphogenesis in diabetic mothers occurs more frequently than in the general population. This phenomenon is believed to be caused by the teratogenic effects of metabolic fuel mixtures with associated membrane injury and aberrations in the biochemical constituents. The present experiment was designed to determine: 1) if hyperglycemia-induced membrane injury is associated with intracellular and/or extracellular lipid disturbances; 2) if supplemental myo-inositol therapy prevents hyperglycemia-induced embryopathy; 3) if a correlation exists between dietary myo-inositol, serum and tissue levels

Inositol 1,4,5-trisphosphate compartmentalization in tracheal smooth muscle.

Pool sizes of inositol phosphate species in myo-[3H]inositol-labeled porcine tracheal smooth muscle were determined under three conditions: (a) unstimulated; (b) stimulated with carbachol; (c) atropine-relaxed from a carbachol contraction. In unstimulated muscle, the inositol 1,4,5-trisphosphate (Ins(1,4,5)P3) content was 14 pmol/100 nmol lipid P1. This is equivalent to a mean [Ins(1,4,5)P3] of about 3 microM (in total cellular water), a level about 30-fold in excess of that required for Ca2+ release from Ins(1,4,5)P3-sensitive sarcoplasmic reticulum (SR). Reduced parasitemia observed with erythrocytes containing inositol hexaphosphate.

Chemicals entrapped in erythrocytes by hypotonic hemolysis can be assessed for possible antiparasitic activity both in vivo and in vitro, regardless of whether they are able to diffuse into erythrocytes readily. Inositol hexaphosphate, a highly charged compound, produced a dramatic lowering of the percentage of cells infected by Babesia microti in vivo and both B. microti and Plasmodium falciparum in vitro. Several possible mechanisms for this observation are discussed.

Inositols in Polycystic Ovary Syndrome: An Overview on the Advances.

This review details the physiologic roles of two insulin sensitizers, myo-inositol (MI) and d-chiro-inositol (DCI). In the human ovary, MI is a second messenger of follicle-stimulating hormone (FSH) and DCI is an aromatase inhibitor. These activities allow a treatment for polycystic ovary syndrome (PCOS) to be defined based on the combined administration of MI and DCI, where the best MI:DCI ratio is 40:1. Moreover, MI enhances the effect of metformin and clomiphene on the fertility of PCOS women seeking pregnancy. As impaired intestinal transport may lead to unsuccessful inositol treatment, we

Biosynthesis and possible functions of inositol pyrophosphates in plants.

Inositol phosphates (InsPs) are intricately tied to lipid signaling, as at least one portion of the inositol phosphate signaling pool is derived from hydrolysis of the lipid precursor, phosphatidyl inositol (4,5) bisphosphate. The focus of this review is on the inositol pyrophosphates, which are a novel group of InsP signaling molecules containing diphosphate or triphosphate chains (i.e., PPx) attached to the inositol ring. These PPx-InsPs are emerging as critical players in the integration of cellular metabolism and stress signaling in non-plant eukaryotes. Most eukaryotes synthesize the prec

An assay procedure for inositol pentaphosphate in avian erythrocytes.

A method to determine the concentration of inositol pentaphosphate (IP5) in chicken erythrocytes is described. Lysates of washed erythrocytes were treated with trichloroacetic acid to remove protein and were chromatographed on Dowex-1 chloride columns. After eluting the other organic phosphates with .3 N HCl, IP5 was eluted with .5 N HCl and quantitated by phosphorus assay. The mean for 12 chickens was 15.4 +/- 2.71 micromoles/g hemoglobin. The method should be useful for the assay of large numbers of samples.

Adriamycin inhibits inositol 1,4,5-trisphosphate 3-kinase activity in vitro and blocks formation of inositol 1,3,4,5-tetrakisphosphate in stimulated Jurkat T-lymphocytes. Does inositol 1,3,4,5-tetrakisphosphate play a role in Ca(2+)-entry?

Effects of the cytostatic drug adriamycin on inositol polyphosphate metabolism were analyzed in a human T-cell line (Jurkat) using a recently developed anion-exchange high performance liquid chromatography/post-column complexometric dye system. Treatment of intact T-cells with adriamycin prior to stimulation with an anti-CD3 monoclonal antibody induced a dose- and time-dependent decrease in the intracellular level of inositol 1,3,4,5-tetrakisphosphate (complete inhibition after 2 h at 10 microM adriamycin) and an increase in the level of inositol 1,3,4-trisphosphate without significantly chang

Decreased levels of N-Acetylaspartyglutamate, myo-inositol, and syllo-inositol, in cortical brain regions of women exposed to adverse childhood experiences.

Adverse Childhood Experiences (ACE), including abuse and neglect, can have lasting negative effects on health, decreasing lifespan and increasing the risk of chronic diseases. While research on ACE's impact on brain biochemistry is limited, Magnetic Resonance Spectroscopy (MRS) provides a non-invasive way to study these alterations. This study aims to identify neurochemical patterns linked to ACE exposure using J-edited MRS methods. 43 female participants (18 Low-ACE and 25 High-ACE), aged 19 to 31, were recruited. ACE exposure was assessed using the Maltreatment and Abuse Chronology of Exposu

Myo-inositol and D-chiro-inositol (40:1) reverse histological and functional features of polycystic ovary syndrome in a mouse model.

Mice exposed to continuous light undergo functional and histological changes that mimic those of human Polycystic Ovary Syndrome (PCOS). We herein induced the syndrome by exposing 30-day-old females to 10 weeks of permanent light. Ovarian morphology and histology, as well as reproductive parameters (time of observed pregnancy/delivery) were investigated. Ovaries of PCOS-modeled mice showed lack of tertiary follicles and corpora lutea, altered ovarian architecture, and increased thickness of the theca layer. When mice were returned to a normal light-dark regimen for 10 days, a slight, spontaneo

Proteome-wide quantification of inositol pyrophosphate-protein interactions.

Inositol polyphosphates (InsPs) and inositol pyrophosphates (PP-InsPs) are highly phosphorylated signaling molecules involved in diverse cellular processes. To resolve discrete signaling events mediated by these structurally related metabolites, a mass spectrometry-based approach was developed to derive apparent binding constants on a proteome-wide scale. The method employs chemically synthesized affinity reagents for inositol hexakisphosphate (InsP6) and the inositol pyrophosphates 1PP-InsP5, 5PP-InsP5, and 1,5(PP)2-InsP4 (InsP8). Concentration-dependent affinity enrichment combined with tand

Inositol biosynthesis is inversely regulated by glycolytic activity.

Inositol is an essential metabolite required for membrane biogenesis, cell signaling and trafficking, and gene expression. Its importance is underscored by the fact that inositol depletion leads to death in eukaryotic cells ranging from yeast to human. Perturbation of inositol homeostasis is implicated in numerous human disorders. In yeast, inositol is synthesized de novo from glucose-6-phosphate (G-6-P) through a two-step pathway controlled by the rate-limiting enzyme myo-inositol phosphate synthase (MIPS), encoded by INO1. INO1 expression is tightly controlled by the Henry regulatory circuit

Untangling Inositol (Pyro)Phosphate Biology Through Emerging Technologies.

Inositol phosphates (InsPs) and inositol pyrophosphates (PP-InsPs) constitute a remarkably diverse yet evolutionarily conserved signaling network generated through combinatorial phosphorylation of myo-inositol. Despite their widespread presence across eukaryotes, progress in understanding their biology was long constrained by major analytical challenges arising from their extreme charge density, structural similarity, and isomer diversity. The advent of new analytical approaches is now transforming the field. High-resolution analytical platforms, including capillary-electrophoresis mass spectr

Cellular adaptations of fission yeast to inositol starvation.

UNLABELLED: Inositol is an essential nutrient and a precursor of phosphorylated metabolites that play important roles in cell physiology. Here we characterize the impact of intracellular inositol deficiency, taking advantage of fission yeast Schizosaccharomyces pombe, an obligate inositol auxotroph, as a model system. We find that cells deprived of inositol ceased growth after two or three rounds of division, with genome content indicative of a G2 arrest. Inositol starvation elicited (i) widespread changes in the expression of genes involved in lipid metabolism and inositol polyphosphate dynam

Myo-Inositol in Pregnancy: Implications for Acne Management.

BACKGROUND: Acne vulgaris is common during pregnancy and presents unique therapeutic challenges due to fetal safety considerations and restrictions on conventional acne medications. Adjunctive strategies that target hormonal and metabolic contributors to acne without teratogenic risk are therefore of particular relevance to dermatologic care. OBJECTIVE: To review the evidence supporting myo-inositol as an adjunctive option for pregnancy-related acne and contextualize its dermatologic relevance within associated metabolic, endocrine, and psychosocial conditions.
