Immunity responses as checkpoints for efficient transmission of begomoviruses by whiteflies.

Begomoviruses are a group of single stranded DNA plant viruses exclusively transmitted by the sweet potato whitefly Bemisia tabaci in a persistent, circulative manner. After acquisition from plant phloem, this group of viruses circulate and are retained within the whitefly, interacting with tissues, cells and molecular pathways for maintaining the safety of the infective intact virions, by exploiting cellular mechanisms and avoiding degradation by the insect immune responses. During retention, the virions are internalized in the midgut cells, exit and spend hours-days in the hemolymph and cros

Progress on the role of eicosanoids in insect immunity.

Eicosanoids are synthesized from arachidonic acid, which are oxygenated metabolites of C20 polyunsaturated fatty acids through cyclooxygenases (COXs) or lipoxygenases (LOXs). Eicosanoids include prostaglandins (PGs), thromboxanes (TXs), epoxyeicosatrienoic acids (EETs) and leukotrienes (LTs), etc. Eicosanoids mediate a number of physiological processes in insects, such as Malpighian tubule and ovary development, or immune response. They play the important roles in regulating cellular immune processes to clear bacteria, fungi, parasites and viruses by hemocytes and the humoral immune pathways.

Untargeted metagenomic sequencing identifies Toscana virus in patients with idiopathic meningitis, southern Spain, 2015 to 2019.

BackgroundVarious pathogens, including bacteria, fungi, parasites, and viruses can lead to meningitis. Among viruses causing meningitis, Toscana virus (TOSV), a phlebovirus, is transmitted through sandfly bites. TOSV infection may be suspected if patients with enterovirus- and herpesvirus-negative aseptic (non-bacterial) meningitis recall recent insect bites. Other epidemiological factors (season, rural area) may be considered. The broad range of possible meningitis aetiologies poses considerable diagnosis challenges. Untargeted metagenomic next-generation sequencing (mNGS) can potentially ide

A recombinant AeDNA containing the insect-specific toxin, BmK IT1, displayed an increasing pathogenicity on Aedes albopictus.

The Aedes aegypti densovirus (AeDNV) has previously shown potential in mosquito control. To improve its efficacy as a biopesticide, the gene for an excitatory insect-specific toxin from Buthus martensii Karsch (BmK IT1) was inserted into the AeDNV genome and cloned into pUCA plasmid. The coding sequence for green fluorescent protein was ligated to the C-terminus of the BmK IT1 gene as a screening marker. Recombinant and helper plasmids were cotransfected into C6/36 cells; wild-type viruses were the controls. The recombinant viruses were identified and quantified by real-time polymerase chain r

Begomoviruses: what is the secret(s) of their success?

Begomoviruses constitute an extremely successful group of emerging plant viruses transmitted by whiteflies of the Bemisia tabaci complex. Hosts include important vegetable, root, and fiber crops grown in the tropics and subtropics. Factors contributing to the ever-increasing diversity and success of begomoviruses include their predisposition to recombine their genomes, interaction with DNA satellites recruited throughout their evolution, presence of wild plants as a virus reservoir and a source of speciation, and extreme polyphagia and continuous movement of the insect vectors to temperate reg

Leveraging transcriptome sequence read archives for virus detection in wild and colony populations of triatomines (Hemiptera: Reduviidae: Triatominae).

Triatomines are infamous as vectors of the parasite Trypanosoma cruzi, the causative agent of Chagas disease. However, climate-driven range expansion and urbanization adaptation of triatomine populations, coupled with their highly diverse feeding strategies (vertebrate haematophagy, kleptohaematophagy, and coprophagy), and has elevated interest in triatomines as potential arboviral vectors. Information on the triatomine virome is scant, with prior records including only eight insect-specific viruses: Triatoma virus (TrV) and Rhodnius prolixus viruses 1-7. Here, we leverage publicly available t

Laboratory infection of the sandfly Phlebotomus papatasi Scopoli (Diptera, Psychodidae) with three Phleboviruses.

Membrane feeding and intrathoracic inoculation techniques were used to infect Phlebotomus papatasi sandflies with Phlebotomus fever group viruses. Sandflies were orally infected with Pacui virus, and were inoculated with Pacui, Sicilian sandfly fever and Naples sandfly fever viruses. Pacui virus did not multiply in these insects after oral infection, but an 8 X 10(4)-fold increase in virus concentration occurred within four days of intrathoracic inoculation of this virus. The infection rate was 100%. Sicilian sandfly fever virus multiplied in P. papatasi after infection by inoculation, althoug

Chronic bee paralysis virus exploits host antimicrobial peptides and alters gut microbiota composition to facilitate viral infection.

The significance of gut microbiota in regulating animal immune response to viral infection is increasingly recognized. However, how chronic bee paralysis virus (CBPV) exploits host immune to disturb microbiota for its proliferation remains elusive. Through histopathological examination, we discovered that the hindgut harbored the highest level of CBPV, and displayed visible signs of damages. The metagenomic analysis showed that a notable reduction in the levels of Snodgrassella alvi and Lactobacillus apis, and a significant increase in the abundance of the opportunistic pathogens such as Enter

MicroRNA response in insect salivary glands to plant virus infection.

UNLABELLED: Most arboviruses rely on insect vectors for transmission, with salivary glands serving as a critical gateway for viral spread to new hosts. MicroRNAs (miRNAs) are key regulators of gene expression, yet their roles in salivary glands during virus infection remain poorly understood. Using the small brown planthopper (SBPH)-rice stripe virus (RSV) system, we sequenced small RNAs from salivary glands of nonviruliferous and viruliferous insects and identified 5,909 known miRNAs from 201 families. Of these, 1,143 miRNAs were differentially expressed upon RSV infection, including 1,090 up

Iflaviruses in arthropods: when small is mighty.

Iflaviruses are arthropod-infecting viruses classified in the family Iflaviridae. The most well-studied iflaviruses are responsible for deleterious overt (symptomatic) infections in economically important insect species like honeybees and silkworms. The fact that iflaviruses are also found in mass reared insects such as crickets and flies, makes their study relevant for the food and feed industries and for biological control applications (e.g., mass reared insects for sterile insect techniques). In recent years it was found that iflaviruses are not restricted to insects, but are widely spread

A New Clade of Insect-Specific Flaviviruses from Australian Anopheles Mosquitoes Displays Species-Specific Host Restriction.

Flaviviruses are arthropod-borne viruses found worldwide and are responsible for significant human and veterinary diseases, including dengue, Zika, and West Nile fever. Some flaviviruses are insect specific and replicate only in mosquitoes. We report a genetically divergent group of insect-specific flaviviruses from Anopheles mosquitoes that do not replicate in arthropod cell lines or heterologous Anopheles species, exhibiting unprecedented specialization for their host species. Determination of the complete sequences of the RNA genomes of three of these viruses, Karumba virus (KRBV), Haslams

Quantification of saliva production in Musca domestica L. (Diptera: Muscidae) and the impact of MdSGHV infection.

Musca domestica salivary gland hypertrophy virus (MdSGHV) affects house flies by enlarging salivary glands, impeding ovary development in females, and mating behavior in both males and females. It is not known if this virus impacts the quantity of saliva produced by house flies. This study aimed to establish baseline saliva quantities in healthy M. domestica across sexes and ages and examine how MdSGHV infection influences saliva output in 5-day-old males. Results reveal that healthy female Musca domestica produce more saliva on average than males and that saliva production among both sexes de

Africanized honey bees in Colombia exhibit high prevalence but low level of infestation of Varroa mites and low prevalence of pathogenic viruses.

The global spread of the ectoparasitic mite Varroa destructor has promoted the spread and virulence of highly infectious honey bee viruses. This phenomenon is considered the leading cause for the increased number of colony losses experienced by the mite-susceptible European honey bee populations in the Northern hemisphere. Most of the honey bee populations in Central and South America are Africanized honey bees (AHBs), which are considered more resistant to Varroa compared to European honey bees. However, the relationship between Varroa levels and the spread of honey bee viruses in AHBs remain

Investigation of viral and parasitic pathogens in Bombus terrestris (Hymenoptera: Apidae) populations.

In recent years, with the increase in both abiotic and biotic stress factors, there has been a significant decline in honey bee and bumblebee populations worldwide, which in turn threatens pollination services that are crucial for agricultural production. This study aimed to determine the prevalence and incidence of viral and parasitic pathogens in Bombus terrestris (Linnaeus, 1758) individuals obtained from 23 wild-caught queen bumblebees collected in Ordu Province, Türkiye. For this purpose, analyses conducted on bumblebee samples collected from different businesses revealed that the Acute B

Metagenomic sequencing characterizes a wide diversity of viruses in field mosquito samples in Nigeria.

Mosquito vectors are a tremendous public health threat. One in six diseases worldwide is vector-borne transmitted mainly by mosquitoes. In the last couple of years, there have been active Yellow fever virus (YFV) outbreaks in many settings in Nigeria, and nationwide, entomological surveillance has been a significant effort geared towards understanding these outbreaks. In this study, we used a metagenomic sequencing approach to characterize viruses present in vector samples collected during various outbreaks of Yellow fever (YF) in Nigeria between 2017 and 2020. Mosquito samples were grouped in

Antiviral Treatment Reveals a Cooperative Pathogenicity of Baculovirus and Iflavirus in Spodoptera exigua, a Lepidopteran Insect.

NPVThe beet armyworm, Spodoptera exigua, is a serious insect pest infesting various vegetable crops. Two infectious insect viruses, baculovirus and iflavirus, are known to induce epizootics in S. exigua populations. Indeed, some laboratory colonies have appeared to be covertly infected by these viruses. Diagnostic PCR tests detected two different viruses: Spodoptera exigua multiple nucleopolyhedrosis virus (SeMNPV) and iflaviruses (SeIfV1 and SeIfV2). Viral extract from dead larvae of S. exigua could infect Sf9 cells and produce occlusion bodies (OBs). Feeding OBs to asymptomatic larvae of S.

Comparative studies of transmission mode and localisation patterns of common RNA viruses in Queensland fruit fly (Bactrocera tryoni) reveal most are vertically transmitted.

RNA viruses are common in tephritid fruit flies including the Queensland fruit fly, Australia's most significant horticultural pest. For many their transmission, tissue tropism and load across host development remain unexplored. Yet these factors are important for host biology, ecology and pest management. We investigated Bactrocera tryoni orbivirus (OV), Bactrocera tryoni xinmovirus (XV), Bactrocera tryoni toti-like virus (TLV) and Bactrocera tryoni iflavirus species 2 (IVsp.2) that commonly coinfect B. tryoni laboratory populations.
