IFN-gamma AU-rich element removal promotes chronic IFN-gamma expression and autoimmunity in mice.

We generated a mouse model with a 162 nt AU-rich element (ARE) region deletion in the 3' untranslated region (3'UTR) of the interferon-gamma (IFN-γ) gene that results in chronic circulating serum IFN-γ levels. Mice homozygous for the ARE deletion (ARE-Del) (-/-) present both serologic and cellular abnormalities typical of patients with systemic lupus erythematosus (SLE). ARE-Del(-/-) mice display increased numbers of pDCs in bone marrow and spleen. Addition of IFN-γ to Flt3-ligand (Flt3L) treated in vitro bone marrow cultures results in a 2-fold increase in pDCs with concurrent increases in IR

Bcl11a orchestrates multilineage integrity in aging hematopoiesis by regulating multipotent progenitor fate decisions.

Aging hematopoiesis exhibits progressive myeloid skewing and impaired lymphopoiesis, driving age-related blood disorders. We show that multipotent progenitors (MPPs) mediate this lineage imbalance. Aging expands myeloid-biased MPP3 cells while functionally compromising MPP4 lymphoid potential, collectively skewing hematopoietic output toward the myeloid lineage. We identify Bcl11a as a dosage-sensitive regulator of MPP fate: Bcl11a suppresses Fer to restrain premature myeloid differentiation in MPP3, while activating the Irf8-Ebf1 axis to license lymphoid specification. Strikingly, sustained B

Regulation of myelopoiesis by the transcription factor IRF8.

Interferon regulatory factor-8 (IRF8) is a transcription factor expressed in hematopoietic cells, particularly in mononuclear phagocytes [monocytes/macrophages and dendritic cells (DCs)] and their progenitors. Various studies have demonstrated that IRF8 is essential for the development of monocytes, DCs, eosinophils, and basophils. Conversely, IRF8 suppresses the generation of neutrophils. Accordingly, Irf8 (-/-) mice develop immunodeficiency and a chronic myeloid leukemia (CML)-like disease. Mutations and loss of expression of the human IRF8 gene are also associated with immunodeficiency and

T-bet-dependent ILC1- and NK cell-derived IFN-γ mediates cDC1-dependent host resistance against Toxoplasma gondii.

Host resistance against intracellular pathogens requires a rapid IFN-γ mediated immune response. We reveal that T-bet-dependent production of IFN-γ is essential for the maintenance of inflammatory DCs at the site of infection with a common protozoan parasite, Toxoplasma gondii. A detailed analysis of the cellular sources for T-bet-dependent IFN-γ identified that ILC1s and to a lesser degree NK, but not TH1 cells, were involved in the regulation of inflammatory DCs via IFN-γ. Mechanistically, we established that T-bet dependent innate IFN-γ is critical for the induction of IRF8, an essential tr

Interferon-β induction heterogeneity during KSHV infection is correlated to levels and activation of the transcription factors ATF2 and RelA, and not IRF3.

Careful regulation of type I interferons (IFN) like IFN-β is vital for balancing tissue damage and protection against infections. Heterogeneity in type I IFN expression among virally infected cells is a common phenomenon that may help limit IFN responses, but the source of this heterogeneity is poorly understood. We previously found that during Kaposi's sarcoma-associated herpesvirus replication, type I IFN induction was limited to a small percentage of infected cells. This heterogeneity was not explained by viral gene expression. Here, we used a fluorescent reporter and fluorescence activated

CD11c-Cre driven deletion of Irf8 reveals the effect of somatic mosaicism in a mouse model of SLE.

The pathogenesis of systemic lupus erythematosus (SLE) is caused by a complex mix of genetic factors that lead to dysregulation of the immune response. Mild susceptibility or resistance factors can tilt the scale towards or against pathology. Here, we present evidence for the Irf8 gene as a lupus protective factor in conditions of haploinsufficiency or mosaicism. We targeted Irf8 expression in mice deficient in Fcgr2b, a well characterized mouse model of SLE. As is the case in human SLE, hyperresponsive B cells and dendritic cells (DCs) are causal factors at various stages of disease in Fcgr2b

Shared and distinct functions of the transcription factors IRF4 and IRF8 in myeloid cell development.

Interferon regulatory factor (IRF) 8 and IRF4 are structurally-related, hematopoietic cell-specific transcription factors that cooperatively regulate the differentiation of dendritic cells and B cells. Whilst in myeloid cells IRF8 is known to modulate growth and differentiation, the role of IRF4 is poorly understood. In this study, we show that IRF4 has activities similar to IRF8 in regulating myeloid cell development. The ectopic expression of IRF4 in myeloid progenitor cells in vitro inhibits cell growth, promotes macrophages, but hinders granulocytic cell differentiation. We also show that

Interferon Regulatory Factors as Potential Therapeutic Targets in Cardiovascular Disease: Focusing on Vascular Inflammation.

The interferon regulatory factor (IRF) family exerts dual regulatory roles in vascular inflammation. Pro-inflammatory IRF1/3/5/7 drive endothelial dysfunction, macrophage M1 polarization, vascular smooth muscle cell (VSMC) transdifferentiation, and adaptive immune amplification via nuclear factor-κB (NF-κB), NOD-like receptor pyrin domain-containing 3 (NLRP3), cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING), and Janus kinase/signal transducer and activator of transcription (JAK/STAT) pathways. Conversely, IRF4/8 mediate anti-inflammatory effects by promoting M2 polarization

The combination of gene perturbation assay and ChIP-chip reveals functional direct target genes for IRF8 in THP-1 cells.

Gene regulatory networks in living cells are controlled by the interaction of multiple cell type-specific transcription regulators with DNA binding sites in target genes. Interferon regulatory factor 8 (IRF8), also known as interferon consensus sequence binding protein (ICSBP), is a transcription factor expressed predominantly in myeloid and lymphoid cell lineages. To find the functional direct target genes of IRF8, the gene expression profiles of siRNA knockdown samples and genome-wide binding locations by ChIP-chip were analyzed in THP-1 myelomonocytic leukemia cells. Consequently, 84 genes

Decoding microglial metabolic cell death and PANoptosis hub genes in subacute phase of traumatic brain injury: a multi-omics and experimental validation study.

Microglia play a central regulatory role in neuroinflammation and metabolic disturbances during the subacute phase of traumatic brain injury (TBI). PANoptosis and metabolic cell death are key drivers of TBI pathological progression; however, the key regulatory genes governing these processes within microglia remain unclear. Bulk and single-cell RNA-seq datasets from mice with subacute TBI were retrieved from the NCBI GEO database. Seven hub genes associated with PANoptosis and metabolic cell death were identified through weighted gene co-expression network analysis (WGCNA) and maximum group ce

Targeting β2-Adrenergic Receptor Stability With Lycorine Reveals a Host-Directed Pathway for Antiviral Defense Against Poxviruses.

Psychological stress is a pervasive yet poorly understood modulator of infectious disease outcomes. Here, we report a mechanistic link between chronic stress and severe viral pathogenesis, identifying the β2-adrenergic receptor (β2-AR) as a critical host factor and therapeutic target. Using a model of chronic restraint stress and cowpox virus (CPXV) infection, we demonstrate that psychological stress significantly exacerbates disease severity and mortality. This increased susceptibility is driven by autophagy-mediated downregulation of β2-AR, a process that is recapitulated during viral infect

High expression of IRF8 in AChR-specific cells regulates the function of B cells.

Myasthenia gravis (MG) is a kind of autoimmune disease. Interferon regulatory factor 8 (IRF8) is an interferon common sequence-binding protein. IRF8 is a key factor involved in the development, maturation, and antimicrobial activity of myeloid cells. Second-generation sequencing of splenocytes at the peak period of disease in experimental autoimmune MG (EAMG) rats and complete Freund's adjuvant (CFA)-treated mice suggested that the expression of IRF8 gene is related to the pathogenesis of EAMG. This study focused on the differential expression of IRF8 in B cell and T cell in EAMG, and the unde

Susceptibility of IFN regulatory factor-1 and IFN consensus sequence binding protein-deficient mice to brucellosis.

IFN-gamma is a key cytokine controlling Brucella infection, and the diverse functions of this cytokine are mediated by IFN regulatory factors (IRFs) such as IRF-1, IRF-2, and IFN consensus sequence binding protein (ICSBP). However, the roles of these three IRFs in Brucella infection have not been investigated. The infection of each IRF-deficient mouse strain provides an opportunity to determine not only the significance of each IRF molecule but also the crucial immune components necessary for host defense during in vivo infection, because respective IRF-deficient mouse strains contain unique i

Crucial role of interferon consensus sequence binding protein, but neither of interferon regulatory factor 1 nor of nitric oxide synthesis for protection against murine listeriosis.

Listeria monocytogenes is widely used as a model to study immune responses against intracellular bacteria. It has been shown that neutrophils and macrophages play an important role to restrict bacterial replication in the early phase of primary infection in mice, and that the cytokines interferon-gamma (IFN-gamma) and tumor necrosis factor-alpha (TNF-alpha) are essential for protection. However, the involved signaling pathways and effector mechanisms are still poorly understood. This study investigated mouse strains deficient for the IFN-dependent transcription factors interferon consensus seq

Histone deacetylase inhibition induces an antitumor IRF8-dependent trained immunity-like status in macrophages.

Epigenetic reprogramming underlies innate immune memory. However, whether trained immunity can be induced via epigenetic intervention to combat tumors, and the key regulators controlling this process, remain unclear. Here, we demonstrate that histone deacetylase inhibitor (HDACi) priming establishes a trained immunity-like state in monocytes/macrophages, conferring potent antitumor effect in multiple models of triple-negative breast cancer (TNBC) in female mice. Interferon regulatory factor 8 (IRF8) is identified as the master regulator of the inflammatory transcriptional program, which is gov
