Regulation of a hair follicle keratin intermediate filament gene promoter.

During hair growth, cortical cells emerging from the proliferative follicle bulb rapidly undergo a differentiation program and synthesise large amounts of hair keratin proteins. To identify some of the controls that specify expression of hair genes we have defined the minimal promoter of the wool keratin intermediate filament gene K2.10. The region of this gene spanning nucleotides -350 to +53 was sufficient to direct expression of the lacZ gene to the follicle cortex of transgenic mice but deletion of nucleotides -350 to -150 led to a complete loss of promoter activity. When a four base subst

Intermediate filament expression by normal and diseased human corneal epithelium.

Cicatricial conjunctivitis may be a sequel to systemic disorders (eg, Stevens-Johnson syndrome, cicatricial pemphigoid) or local disorders such as chemical burns. The cicatrisation is often associated with corneal epithelial changes that cause visual loss. These have been attributed to encroachment of the conjunctival epithelium over the cornea. However, the epithelial anomalies are poorly understood. We investigated the corneal epithelial changes in cicatricial conjunctivitis with an immunohistochemical study of intermediate filaments in normal and pathological specimens. Our results show tha

Common epithelial ovarian tumors. Immunohistochemical intermediate filament profiles.

The authors studied 79 common epithelial ovarian tumors in order to ascertain the intermediate filament profiles in formalin-fixed and methacarn-fixed, paraffin-embedded surgical pathology materials. Ultra-structural correlations were attempted with several tumors. All categories of common benign and malignant epithelial tumors were examined. Antibodies used in the study included antikeratins (AE1/AE3, 35BH11, 34BE12), carcinoembryonic antigen (CEA), and vimentin. All ovarian epithelial tumors expressed keratin in uniform fashion, except high molecular weight keratin (34BE12) which was focal.

Coexpression of intermediate filament polypeptides in human fetal and adult tissues.

Tissues from human fetuses (12 to 14 weeks) were studied by using immunohistochemical methods, with special emphasis on coexpression of intermediate filaments. Well-characterized antibodies, monoclonal as well as polyclonal were used. Indirect immunoperoxidase staining disclosed simultaneous expression of cytokeratin and vimentin, or vimentin and desmin in several tissues, whereas some other tissues coexpressed three classes of intermediate filaments, i.e., cytokeratin, vimentin, and desmin. Coexpression of cytokeratin and vimentin was seen in immature tubules of the kidney localized in the bl

Disorganisation of intermediate filament structure in alcoholic and other liver diseases.

The distribution of Mallory body antigens JMB1 and 2 was examined in 82 human fresh diagnostic needle liver biopsies and 28 necropsies by the indirect immunoperoxidase technique using 2 monoclonal antibodies (anti-JMB1 and 2) against Mallory bodies. The JMB1 antigen was detectable in bile duct epithelium and in hepatocytes of histologically normal livers. It was also found in all Mallory bodies in various hepatic disorders. This antigen was markedly increased in the cytoplasm of all liver cells in acute alcoholic hepatitis superimposed on alcoholic cirrhosis, in most cases of acute alcoholic h

Expression of intermediate filament proteins in thyroid gland and thyroid tumors.

The presence of intermediate filament proteins of cytokeratin/prekeratin type and vimentin type was evaluated in non-neoplastic thyroid glands and in different types of thyroid neoplasms. Follicular epithelium of both normal and goitrous thyroids showed a strong reaction with anticytokeratin antibodies that widely cross-react with various simple epithelia. On the other hand, in normal thyroid, there were only occasionally (in one of 12 cases) solitary cells reacting with antibodies to epidermal prekeratin. In nodular goiters, such cells were often seen (eight of 18), especially among the linin

Mechanical Probing of the Intermediate Filament-Rich Caenorhabditis Elegans Intestine.

It is commonly accepted that intermediate filaments have an important mechanical function. This function relies not only on intrinsic material properties but is also determined by dynamic interactions with other cytoskeletal filament systems, distinct cell adhesion sites, and cellular organelles which are fine-tuned by multiple signaling pathways. While aspects of these properties and processes can be studied in vitro, their full complexity can only be understood in a viable tissue context. Yet, suitable and easily accessible model systems for monitoring tissue mechanics at high precision are

Expression of intermediate filament proteins in benign lesions of the oral mucosa.

Immunohistochemistry with monospecific antibodies was used to study the expression patterns of cytokeratins (Cks) and vimentin in non-dysplastic lesions of the oral cavity, including lichen planus and fibromas. In hyperplastic lesions, Ck expression did not deviate significantly from the normal non-keratinizing squamous epithelium of the oral cavity. Hyperkeratotic lesions showed pronounced aberrations in their Ck profile. These lesions were characterized by extended expression of the keratinization marker Ck 10, the basal cell Ck 14 and the hyperproliferation-associated Ck 16 in the suprabasa

Expression of the intermediate filament nestin during rodent tooth development.

The developing tooth represents a suitable model for understanding the molecular mechanisms involved in induction, morphogenesis and differentiation of organs. It is conceivable that the developmental changes could be reflected in the distribution of different cytoskeletal components and in this report we analyze the expression of the intermediate filament nestin during rodent tooth development at the protein and mRNA levels (by immuno light and electron microscopy, and by in situ hybridization). Nestin is expressed at all stages of tooth development, but the expression levels increase after b

Making heads and tails of intermediate filament assembly, dynamics and networks.

Thus far, intermediate filaments (IFs) have been the least understood of the three cytoskeletal filament systems with regard to their structure, assembly, network formation, and dynamics. This picture is now slowly but definitely changing, as recent in vivo and in vitro experiments, including generation of transgenic animals, have yielded important new data shedding light on the following areas: the molecular architecture of IFs; the role of the highly variable end domains during IF assembly and network formation; the factors that govern whether IF proteins are involved in de novo filament for

The dynamic phosphorylation of the human intermediate filament keratin 1 chain.

The major sites of phosphorylation have been determined on the human keratin intermediate filament keratin 1 (type II) chain expressed in terminally differentiating epidermis. A total of nine phosphate sites were found, involving 1 threonine and 8 serine residues, and were localized to end domain sequences. The sites identified corresponded to major sites of phosphorylation as determined by direct quantitation of O-phosphoserine. Since the tissue was cultured with [32 P] orthophosphate only briefly, labeling occurred primarily by turnover, so that information on the dynamics of phosphorylation

Expression of intermediate filament proteins in normal and diseased thyroid glands.

A total of 67 samples from normal and pathological thyroid glands were stained (as formalin fixed paraffin sections) with a panel of monoclonal antibodies directed against intermediate filament proteins. The study confirmed previous reports of cytokeratin and vimentin coexpression in primary thyroid carcinomas, but coexpression was also detected in normal thyroid and in a range of benign conditions including follicular adenomas, Hashimoto's thyroiditis, and diffuse hyperplasia (thyrotoxicosis). Prekeratin expression was found (using antibodies recognising higher molecular weight cytokeratins)

Type III intermediate filaments in redox interplay: key role of the conserved cysteine residue.

Intermediate filaments (IFs) are cytoskeletal elements involved in mechanotransduction and in the integration of cellular responses. They are versatile structures and their assembly and organization are finely tuned by posttranslational modifications. Among them, type III IFs, mainly vimentin, have been identified as targets of multiple oxidative and electrophilic modifications. A characteristic of most type III IF proteins is the presence in their sequence of a single, conserved cysteine residue (C328 in vimentin), that is a hot spot for these modifications and appears to play a key role in t

Caspase cleavage of keratin 18 and reorganization of intermediate filaments during epithelial cell apoptosis.

Keratins 8 (K8) and 18 (K18) are major components of intermediate filaments (IFs) of simple epithelial cells and tumors derived from such cells. Structural cell changes during apoptosis are mediated by proteases of the caspase family. During apoptosis, K18 IFs reorganize into granular structures enriched for K18 phosphorylated on serine 53. K18, but not K8, generates a proteolytic fragment during drug- and UV light-induced apoptosis; this fragment comigrates with K18 cleaved in vitro by caspase-6, -3, and -7. K18 is cleaved by caspase-6 into NH2-terminal, 26-kD and COOH-terminal, 22-kD fragmen

Antibodies to intermediate filaments in polymyalgia rheumatica and giant cell arteritis: a sequential study.

Serum specimens from 35 patients with polymyalgia rheumatica and giant cell arteritis (PMR/GCA) were obtained sequentially at variable time intervals up to a year from onset of disease. These were tested for antibodies to intermediate filaments by indirect immunofluorescence using HEp2 cells as substrate. Twenty four of 35 (68%) patients' sera at onset of disease were positive at an anti-intermediate filament antibody (AIFA) titre of greater than 1/40 compared with three outs of 19 (15%) control sera. AIFA were predominantly of IgM class, and there was no significant change in AIFA titres on f

Fc-mediated binding of IgG to vimentin-type intermediate filaments in vascular endothelial cells.

Prior studies have shown that vascular endothelial cells bind circulating IgG intracellularly during cell death. We now demonstrate that all endothelial cells have intracellular binding sites for IgG and that these binding sites are exposed to circulating IgG only if the plasma membrane is damaged. The binding sites are located on the cytoskeletal intermediate filaments and can be detected also in other cells containing vimentin-type intermediate filaments. Monoclonal human IgG1 exhibited saturable, high-affinity binding to vimentin-enriched cytoskeletons. Binding was inhibited by Fc fragments

Distribution of intermediate filaments and actin microfilaments in parotid autoimmune sialoadenitis of Sjögren syndrome.

In order to study the importance of myoepithelial cells in the formation of parotid epi-myoepithelial islands of the autoimmune sialoadenitis in Sjögren's syndrome, the distribution of cytoskeletal elements has been investigated by electron and immunofluorescence microscopy. Antibodies to intermediate filament proteins--prekeratins, desmin and vimentin--were used as well as a 'DNase I/anti-DNase I' immunohistochemical procedure to detect actin microfilaments for characterization of myoepithelial cells compared to the other types of parotid epithelial cells.
