Development and Validation of a Multienzyme Isothermal Rapid Amplification-Based Fluorescence Assay for Detection of Megalocytivirus pagrus 1.

Megalocytivirus pagrus 1 is a major viral pathogen that causes high mortality and economic losses in aquaculture worldwide. Although PCR-based diagnostics are highly sensitive and specific, their equipment dependence and long assay times limit field applicability. In this study, a digital multienzyme isothermal rapid amplification with exonuclease probe (digital MIRA-EXO) assay was developed and validated for rapid and specific detection of red sea bream iridovirus (RSIV), infectious spleen and kidney necrosis virus (ISKNV), and turbot reddish body iridovirus (TRBIV). The digital MIRA-EXO assa

Viral susceptibility, transfection and growth of SPB--a fish neural progenitor cell line from the brain of snubnose pompano, Trachinotus blochii (Lacépède).

This study investigates the susceptibilities of the SPB cell line to fish viruses including giant seaperch iridovirus (GSIV-K1), red sea bream iridovirus (RSIV-Ku), grouper nervous necrosis virus (GNNV-K1), chum salmon reovirus (CSV) and eel herpesvirus (HVA). GSIV-K1, RSIV-Ku and CSV replicated well in SPB cells, with a significant cytopathic effect and virus production. However, the cells were HVA and GNNV refractory. To examine the ability of SPB cells to stably express foreign protein, expression vectors encoding GNNV B1 and B2 fused to enhanced green fluorescent protein (EGFP) and GSIV OR

Sex-specific survival and absence of Wolbachia-mediated protection during invertebrate iridescent virus 31 infection in Armadillidium vulgare.

Invertebrate Iridescent Virus 31 (IIV-31, or Iridovirus armadillidium1) is a large double-stranded DNA virus that induces iridescence and high mortality in terrestrial isopods. Despite its broad distribution, its pathogenic mechanisms and interactions with host immunity and endosymbionts are poorly understood. We examined the effects of IIV-31 infection on survival, hemocyte concentration and viability, septicemia, and viral load in Armadillidium vulgare, while testing for protective effects of the feminizing endosymbiont Wolbachia (wVulC strain). Virgin adults from asymbiotic (males and femal

SGIV envelope protein VP088 facilitated virus replication via interacting with other viral proteins and promoting p62-dependent autophagic degradation of TBK1.

Singapore grouper iridovirus (SGIV), a novel member of the genus Ranavirus, family Iridoviridae, frequently causes a severe disease with high mortality in grouper aquaculture. Although previous findings have demonstrated that SGIV envelope protein VP088 was crucial for its infectivity, the underlying mechanism still remained uncertain. Here, we screened the potential viral proteins that interacted with VP088 during SGIV infection using GFP pull-down assay. Co-immunoprecipitation (Co-IP) assays verified the interactions between VP088 and VP018, VP068, or VP156 in vitro. Moreover, confocal micro

An intestinal Eimeria infection in juvenile Asian seabass (Lates calcarifer) cultured in Vietnam--a first report.

This is the first report of an intestinal Eimeria infection in Asian seabass (Lates calcarifer) at the histopathological and ultrastructural levels. The Eimeria infection was often associated with severe pathology and significant mortality in the absence of other pathogens. This showed that it is an important disease of juvenile L. calcarifer in small scale nurseries in Vietnam. Heavy infection and high prevalence levels of the Eimeria infection are suspected to be linked to the low daily water exchange rates practised in these nurseries. Although systemic iridovirus infection was concurrently

Involvement of fish signal transducer and activator of transcription 3 (STAT3) in SGIV replication and virus induced paraptosis.

Signal transducer and activator of transcription 3 (STAT3) is an important transcription factor which plays crucial roles in immune regulation, inflammation, cell proliferation, transformation, and other physiological processes of the organism. In this study, a novel STAT3 gene from orange spotted grouper (Ec-STAT3) was cloned and characterized. Bioinformatic analysis revealed that full-length of Ec-STAT3 was 3105-bp long and contained a 280-bp 5'UTR, a 470-bp 3'UTR, and a 2355-bp open reading frame (ORF) that encoded a 784-amino acid peptide. The deduced protein of Ec-STAT3 showed 98% identit

Genotypic Characterization of Infectious Spleen and Kidney Necrosis Virus (ISKNV) in Southeast Asian Aquaculture.

Infectious spleen and kidney necrosis virus (ISKNV) is a species within the genus Megalocytivirus (family Iridoviridae), which causes high mortality disease in many freshwater and marine fish species. ISKNV was first reported in Asia and is an emerging threat to aquaculture with increasing global distribution, in part due to its presence in ornamental fish with clinical and subclinical infections. The species ISKNV includes three genotypes: red seabream iridovirus (RSIV), turbot reddish body iridovirus (TRBIV), and ISKNV. There is an increasing overlap in the recognized range of susceptible fi

A comprehensive analysis of the genomic and proteomic profiles of a megalocytivirus isolated from Larimichthys crocea.

INTRODUCTION: The prevalence of viral diseases has posed significant challenges to the sustainable development of large yellow croaker (Larimichthys crocea) aquaculture, with megalocytivirus being one of the primary viral pathogens affecting this species. There have been two proteomic and genomic studies regarding two members of the genus Megalocytivirus: the spotted knifejaw iridovirus (SKIV) and the infectious spleen and kidney necrosis virus (ISKNV). However, both studies were conducted more than 10 years ago. To further investigate the pathogenesis of megalocytivirus, we sequenced the geno

Scale Drop Disease Virus (SDDV) infection induces intestinal dysbiosis and secondary Vibrio expansion in Asian seabass (Lates calcarifer).

Scale drop disease virus (SDDV) is a major viral pathogen causing high mortality in farmed Asian seabass (Lates calcarifer), yet its impacts on host-associated microbiota remain poorly understood. In this study, juvenile L. calcarifer were intraperitoneally challenged with 107 viral copies (0.1 ml SDDV), resulting in 94.2% cumulative mortality over a 24-day experimental period. A total of 36 intestinal content samples were collected at defined time points relative to challenge (dpc) from clinically healthy (H) and sick (S) fish, representing five stages: pre-infection baseline (H0 dpc), early

Megalocytivirus infection in cultured Nile tilapia Oreochromis niloticus.

Megalocytiviruses, such as infectious spleen and kidney necrosis virus (ISKNV), induce lethal systemic diseases in both ornamental and food fish species. In this study, we investigated an epizootic affecting Nile tilapia Oreochromis niloticus cultured in the US Midwest. Diseased fish displayed lethargy, gill pallor, and distension of the coelomic cavity due to ascites. Histopathological examination revealed a severe systemic abundance of intravascular megalocytes that were especially prominent in the gills, kidney, spleen, liver, and intestinal submucosa. The first report of cathepsin A gene in a teleost (rock bream, Oplegnathus fasciatus): An investigation of immune functions upon infection with several pathogens.

We isolated and characterised a cDNA encoding the lysosomal protective protein (serine protease) cathepsin A (CTSA) from rock bream (Oplegnathus fasciatus). The full-length rock bream CTSA (RbCTSA) cDNA (1814 bp) contains an open reading frame of 1419 bp that encodes 472 amino acids. Alignment of multiple CTSA protein sequences revealed that the active site serine and histidine residues were well-conserved among the other CTSA sequences. RbCTSA is highly expressed in the peripheral blood leukocytes, kidney, spleen, liver, intestine, gill, heart, brain, stomach, and eye. RbCTSA expression was a

An epizootic of patent iridescent virus disease in multiple species of blackflies in Chiapas, Mexico.

Simulium blackfly larvae (Diptera: Simuliidae) were collected from rivers and streams at 500-1500 m a.s.l. in Chiapas State of southern Mexico. Among 45 sites surveyed over an area of 2300 km2 (around 15 degrees 15'N 92 degrees 20'W), some Simulium larvae from three sites were opalescent violet-blue, interpreted as patent infection with invertebrate iridescent virus (IIV). Dissection confirmed the presence of putative Iridovirus particles, 130nm diameter, but no IIV isolates were obtained from homogenates injected into Galleria mellonella (L) larvae (Lepidoptera: Pyralidae). A robust nanobody-magnetic bead immunoassay for food safety monitoring: Sensitive detection of viral contamination in giant salamander.

The Chinese giant salamander is a valued food source of high-quality protein; however, the prevalence of Giant Salamander Iridovirus (GSIV) poses a significant threat to the safety and stability of the aquaculture food supply chain. Rapid and sensitive screening of viral pathogens in complex food matrices is therefore critical. We developed a bridge-free double-nanobody sandwich ELISA using magnetic beads (MBs) for detection of GSIV. AlphaFold 3 modeling suggested that covalent immobilization via conserved framework lysines may promote an optimal "upright" nanobody orientation, significantly e

Isolation and characterisation of a novel Bohle-like virus from two frog species in the Darwin rural area, Australia.

Twelve captive magnificent tree frogs Litoria splendida and 2 green tree frogs L. caerulea on a property in the Darwin rural area (Northern Territory, Australia) either died or were euthanased after becoming lethargic or developing skin lesions. Samples from both species of frog were submitted for histopathology and virus isolation. An irido-like virus was cultured from tissue samples taken from both species and was characterised using electron microscopy, restriction enzyme digests and nucleic acid amplification and sequencing. The isolates were determined to belong to the genus Ranavirus, we

Enhanced insecticidal activity of Chilo iridescent virus expressing an insect specific neurotoxin.

Previously we have generated a recombinant Chilo iridescent virus (CIV) by inserting the green fluorescent protein gene (gfp) into the CIV 157L open reading frame (ORF) locus and showed that this recombinant (rCIV-Δ157L-gfp) was fully infectious both in cell culture as well as in insect larvae. This study opened up a new avenue for increasing the speed of kill of CIV and other iridoviruses by inserting virulence or toxin genes into the viral genome. In the current study we constructed a recombinant CIV (rCIV-Δ157L/gfp-AaIT) where the 157L ORF was replaced with both the AaIT neurotoxin gene fro

Molecular Identification and mRNA Expression Profiles of Galectin-9 Gene in Red Sea Bream (Pagrus major) Infected with Pathogens.

Galectin (Gal) is a member of a family of β-galactoside-binding lectin. The members of this family play important roles in the recognition of carbohydrate ligands and in various other biological processes. In this study, we identified the gene encoding Gal-9 in Pagrus major (PmGal-9) and analyzed its expression in various tissues after pathogen challenge. Alignment analysis revealed that the two galactose-binding lectin domains of the deduced protein were highly conserved among all the teleosts. Phylogenetic analysis revealed that PmGal-9 is most closely related to the Gal-9 gene of gilthead s

Mosquito Iridescent Virus: New Records from Nature and Infections Using Strelkovimermis spiculatus (Mermithidae) as a Vector Under Laboratory Conditions.

Iridoviridae is a DNA virus family that affects both vertebrates and invertebrates. Immature aquatic stages of many dipteran species infected with iridovirus have been found in different places worldwide. The most represented genera of the Culicidae family are Aedes and Psorophora. To date, sixteen species of Aedes naturally infected with iridoviruses have been reported. Moreover, there are four records for the genus Psorophora, one for Culiseta, and two for Culex. In this paper, we report two new mosquito species as natural hosts of iridoviridae in Argentina: Aedes albifasciatus (Macquart) an

Establishment and application of a novel tilapia head kidney cell line (THK-03) as an in vitro aquatic toxicology model: Evaluating heavy metal immunotoxicity and virus responses.

The head kidney, a primary immune organ in teleosts, serves as a critical source for immunocompetent in vitro models. Here, we established THK-03, a continuous cell line derived from the head kidney of Nile tilapia (Oreochromis niloticus), and systematically evaluated its applicability in environmental toxicology and viral pathogenesis. THK-03 exhibited stable proliferation in L-15 medium at 25 °C, maintaining epithelial morphology for over 100 passages. Species authentication by 18S rRNA sequencing and functional validation through high transfection efficiency confirmed its reliability. The c
