The pair of fatty acid degradation regulator (FadR)-characterized pseudo-palindromic motifs spacing with 11 bp contribute unequally to the DNA binding ability of the HTH-type transcriptional regulator Rv0494 from Mycobacterium tuberculosis H37Rv.

Transcriptional regulation is an effective way for bacteria to respond to various environmental challenges, such as carbon source scarcity. Acting as a transcription factor belonging to the fatty acid degradation regulator (FadR) subfamily, the HTH-type transcriptional regulator Rv0494 is reported to function in persistence, starvation response and regulation of very-long-chain fatty acid synthesis in Mycobacterium tuberculosis (Mtu), a notorious pathogen that causes tuberculosis. However, DNA sequence dependence of Rv0494, especially the contributions of the pair of FadR-characterized palindr

Interactome of Glyceraldehyde-3-Phosphate Dehydrogenase Points to the Existence of Metabolons in Paracoccidioides lutzii.

Paracoccidioides is a dimorphic fungus, the causative agent of paracoccidioidomycosis. The disease is endemic within Latin America and prevalent in Brazil. The treatment is based on azoles, sulfonamides and amphotericin B. The seeking for new treatment approaches is a real necessity for neglected infections. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is an essential glycolytic enzyme, well known for its multitude of functions within cells, therefore categorized as a moonlight protein. To our knowledge, this is the first approach performed on the Paracoccidioides genus regarding the descr

Substrate Specificity and Kinetic Mechanism of the NAD+-Dependent Deacylase from Mycobacterium tuberculosis, Rv1151c (Mt-Sirt).

The pathogen Mycobacterium tuberculosis (Mtb) relies on extensive metabolic remodeling and post-translational regulation of its proteins to retain persistence in its host, yet these biochemical functions remain largely uncharacterized, including the actions of lysine deacylases. Here, we characterized Rv1151c, the sole annotated NAD+-dependent deacylase in Mtb, which we designate Mt-Sirt. Using synthetic substrates, peptide libraries, and mutational analysis, we showed that Mt-Sirt exhibits strong selectivity for peptide substrates containing Nε-succinyl-lysine and long-chain fatty-acyl-lysine

Proteogenomic Biomarker Profiling for Predicting Radiolabeled Immunotherapy Response in Resistant Prostate Cancer.

Treatment resistance prevents patients with preoperative chemoradiotherapy or targeted radiolabeled immunotherapy from achieving a good result, which remains a major challenge in the prostate cancer (PCa) area. A novel integrative framework combining a machine learning workflow with proteogenomic profiling was used to identify predictive ultrasound biomarkers and classify patient response to radiolabeled immunotherapy in high-risk PCa patients who are treatment resistant. The deep stacked autoencoder (DSAE) model, combined with Extreme Gradient Boosting, was designed for feature refinement and

Extrapulmonary locations of mycobacterium tuberculosis DNA during latent infection.

BACKGROUND: One-third of the world's population has latent infection with Mycobacterium tuberculosis, and 10%-15% of cases of reactivation occur at extrapulmonary sites without active pulmonary tuberculosis. METHODS: To establish the frequency and location of mycobacterial DNA, organ specimens from 49 individuals who died from causes other than tuberculosis were studied by means of polymerase chain reaction (PCR), PCR plus DNA hybridization, in situ PCR, real-time PCR, and spoligotyping. RESULTS: Lung specimens from most subjects (36) were positive for M. tuberculosis, as were specimens from

Pathway of glucose catabolism in Caulobacter crescentus.

Glucose when present as a sole organic carbon source in a mineral salts medium is dissimilated by Caulobacter crescentus ATCC 15252 (strain CB-2) by the Entner-Doudoroff pathway throughout the culture cycle (exponential, transition, and stationary phase). Most of the available glucose that is present at the onset of exponential growth is assimilated by the cells during the transition phase or the period associated with stalk cell development. Swarmer cell development is minimized during this phase. During this same period the pH drops from 6.1 to 4.9 as a result of an abundant excretion of ace

Halting Tuberculosis in Its Tracks: Advances and Strategies for Discovering Therapeutic Targets in Mycobacterium tuberculosis.

Tuberculosis is a global emergency. Despite two decades of intense research to understand and cure the disease, biological uncertainties prevail and hamper the therapeutic progress. Increasing incidences of multidrug-resistant (MDR) and extensively drug resistant (XDR) Mycobacterium tuberculosis strains further complicate tuberculosis control. Modern research, therefore, focuses on development of new antitubercular drugs to treat drug-resistant tuberculosis and shorten the duration of the standard chemotherapy. Fortunately, the recent accelerated approval of delamanid and bedaquiline for use i

Growth and gene expression of Pseudomonas nitroreducens TX1 on octylphenol polyethoxylate surfactants.

Pseudomonas nitroreducens ATCC PTA-6168 (also named TX1) efficiently grows on non-ionic surfactants as the sole source of carbon and energy aerobically. The global gene expression and bacterial stress responses during degradation remain further investigated. This study compared the growth, viability, and transcriptomic profiles of strain TX1 during the log phase when grown on minimal salts basal medium supplemented with 0.5% octylphenol polyethoxylates (OPEOn, commercial name Triton® X-100) vs. in 0.5% succinate. Differentially expressed genes were identified, with 219 upregulated and 22 downr

Glyoxylate detoxification is an essential function of malate synthase required for carbon assimilation in Mycobacterium tuberculosis.

The glyoxylate shunt is a metabolic pathway of bacteria, fungi, and plants used to assimilate even-chain fatty acids (FAs) and has been implicated in persistence of Mycobacterium tuberculosis (Mtb). Recent work, however, showed that the first enzyme of the glyoxylate shunt, isocitrate lyase (ICL), may mediate survival of Mtb during the acute and chronic phases of infection in mice through physiologic functions apart from fatty acid metabolism. Here, we report that malate synthase (MS), the second enzyme of the glyoxylate shunt, is essential for in vitro growth and survival of Mtb on even-chain

Organic acids impede Salmonella infection of chicken macrophage-like cell line (HD11) by modulating itaconate gene expression.

Most human salmonellosis cases are linked to the consumption of poultry products contaminated with Salmonella enterica serovar Typhimurium. In poultry, infections cause intestinal inflammation, decreased weight gain, lower egg production, and potential economic losses for the poultry sector. This study examined the effects of a natural antimicrobial mixture (AuraShield, As) on Salmonella enterica serovar Typhimurium SE10/72 infection of chicken macrophage-like cells (HD11), focusing on infection inhibition, inflammatory response, cell viability, metabolic changes, and oxidative stress. The HD1

Itaconic acid inhibits nontuberculous mycobacterial growth in pH dependent manner while 4-octyl-itaconic acid enhances THP-1 clearance of nontuberculous mycobacteria in vitro.

Increasingly prevalent, nontuberculous mycobacteria (NTM) infections affect approximately 20% of people with cystic fibrosis (CF). Previous studies of CF sputum identified lower levels of the host metabolite itaconate in those infected with NTM. Itaconate can inhibit the growth of M. tuberculosis (MTB) in vitro via the inhibition of the glyoxylate cycle enzyme (ICL), but its impact on NTM is unclear. To test itaconic acid's (IA) effect on NTM growth, laboratory and CF clinical strains of Mycobacterium abscessus and Mycobacterium avium were cultured in 7H9 minimal media supplemented with 1-10 m

Metabolites as regulators of autoimmune diseases.

Immune cell metabolism is essential for regulating immune responses, including activation, differentiation, and function. Through glycolysis and oxidative phosphorylation (OXPHOS), metabolism supplies energy and key intermediates for cell growth and proliferation. Importantly, some metabolites generated during these processes act as signaling molecules that influence immune activity. Autoimmune diseases such as rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE) involve multiple immune cell types, and recent research in immunometabolism has revealed that disrupted metabolic pathwa

Repurposing ethyl bromopyruvate as a broad-spectrum antibacterial.

BACKGROUND: The emergence of drug-resistant bacteria is a major hurdle for effective treatment of infections caused by Mycobacterium tuberculosis and ESKAPE pathogens. In comparison with conventional drug discovery, drug repurposing offers an effective yet rapid approach to identifying novel antibiotics. METHODS: Ethyl bromopyruvate was evaluated for its ability to inhibit M. tuberculosis and ESKAPE pathogens using growth inhibition assays. The selectivity index of ethyl bromopyruvate was determined, followed by time-kill kinetics against M. tuberculosis and Staphylococcus aureus. We first te

Transcriptome-Based Comparative Expression Profiling of Sweet Potato during a Compatible Response with Root-Knot Nematode Meloidogyne incognita Infection.

M. incognita, a root-knot nematode (RKN), infects the roots of several important food crops, including sweet potato (Ipomoea batatas Lam.), and severely reduces yields. However, the molecular mechanisms underlying infection remain unclear. Previously, we investigated differential responses to RKN invasion in susceptible and resistant sweet potato cultivars through RNA-seq-based transcriptome analysis. In this study, gene expression similarities and differences were examined in RKN-susceptible sweet potato cultivars during the compatible response to RKN infection. Three susceptible cultivars in

Transcriptomic and proteomic profiling revealed reprogramming of carbon metabolism in acetate-grown human pathogen Candida glabrata.

BACKGROUND: Emergence of Candida glabrata, which causes potential life-threatening invasive candidiasis, has been widely associated with high morbidity and mortality. In order to cause disease in vivo, a robust and highly efficient metabolic adaptation is crucial for the survival of this fungal pathogen in human host. In fact, reprogramming of the carbon metabolism is believed to be indispensable for phagocytosed C. glabrata within glucose deprivation condition during infection. METHODS: In this study, the metabolic responses of C. glabrata under acetate growth condition was explored using hi

Formaldehyde assimilation through coordination of the glyoxylate pathway and the tricarboxylic acid cycle in broad bean roots.

Formaldehyde (HCHO) assimilation in broad bean (Vicia faba L. cv. YD) roots was investigated using 13C-labeled HCHO followed by 13C-NMR analysis. Results revealed that H13CHO was first oxidized to H13COOH in the roots treated with 2 mM H13CHO in a time-dependent manner. Subsequently, a massive signal peak of [2, 4-13C]citrate (Cit) and a signal peak of [2, 3-13C]succinate (Su) were observed in accompany with an enhancement in the signal intensity of [3-13C]Cit. Selective inhibition of enzyme synthesis under conditions of respiratory inhibition.

When Neurospora mycelium is transferred from a medium containing sucrose to one containing acetate as sole source of carbon, a preferential synthesis of many Krebs cycle, glyoxylate cycle, and associated enzymes occurs. Respiration was inhibited during preferential enzyme synthesis in the following ways. (i) The amount of aeration (shaking) was reduced, (ii) cyanide was added to the culture, (iii) the carbon source, acetate, was removed, (iv) a mutant strain was starved of its Krebs cycle intermediates, and (v) respiration was inhibited by mutation. The effect of this respiratory inhibition on

Glyoxylate cycle gene ICL1 is essential for the metabolic flexibility and virulence of Candida glabrata.

The human fungal pathogen Candida glabrata appears to utilise unique stealth, evasion and persistence strategies in subverting the onslaught of host immune response during systemic infection. However, macrophages actively deprive the intracellular fungal pathogen of glucose, and therefore alternative carbon sources probably support the growth and survival of engulfed C. glabrata. The present study aimed to investigate the role of the glyoxylate cycle gene ICL1 in alternative carbon utilisation and its importance for the virulence of C. glabrata. The data showed that disruption of ICL1 rendered
