Integrated in silico and in vivo larvicidal evaluation of compounds targeting juvenile hormone for malaria vector control.

Malaria remains a major global health challenge, particularly in sub-Saharan Africa, where it is a leading cause of morbidity and mortality. Inhibition of the mosquito juvenile hormone binding protein (MJHBP) is essential for developing selective and effective insecticides against malaria vectors. We performed virtual screening on 2,874 compounds from Agrochemical Insecticides Library using Python-based workflow in RDKit and Pandas libraries. The resultant compounds were subjected to molecular docking study against MJHBP (pdb:5V13). Further evaluation was performed through molecular mechanics

Juvenile hormone regulates the maturation of sexually dimorphic naive ethanol olfactory preference in Drosophila melanogaster.

The molecular mechanisms underlying the maturation of innate reward behaviours remain poorly understood. We have identified a sexually dimorphic innate reward behaviour in Drosophila melanogaster that varies depending on the age, sex and mating status in young adults. Our results suggest that the rewarding neuropeptide corazonin, the transcription factor apontic and juvenile hormone signalling regulate naive ethanol olfactory preference responses during early stages of adulthood. Pharmacological blockade of juvenile hormone via precocene increases naive ethanol olfactory preference in females

Culex quinquefasciatus Mosquitoes Exposed to a Juvenile Hormone Analog may Take a Bloodmeal While Gravid.

Blood avidity in female mosquitoes has been shown to be regulated by cycles of hormone production that determine both egg development and distinct behaviors. Specifically, juvenile hormone (JH) drives early egg development until a bloodmeal is acquired, and JH titers are positively correlated with active host seeking and blood feeding behaviors. After a bloodmeal, JH levels fall, and female mosquitoes become refractory to host seeking and biting. While JH analogs (JHAs) are commonly used as larvicides for mosquito control, the effects of these compounds on adult mosquitoes are not well underst

CO2 narcosis induces a metabolic shift mediated via juvenile hormone in Bombus impatiens gynes.

Carbon dioxide (CO2) has pleiotropic effects on insect physiology and behavior. Although diverse, many impacts are related to changes in metabolism and reallocation of macronutrients. Here we examined the metabolic shift induced by CO2 and its regulation using Bombus impatiens. CO2 applied to bumble bee gynes induces bypass of diapause and transition into reproduction. We analyzed ovary activation and macronutrient amounts in four tissues/body parts (fat body, thorax, ovaries, and crop) at three timepoints following CO2 administration. To tease apart the effects of CO2 on reproduction and meta

Identification and characterization of juvenile hormone esterase gene from the yellow fever mosquito, Aedes aegypti.

Juvenile hormone esterase (JHE) plays an important role in regulating juvenile hormone titers. Recent sequencing and annotation of the Aedes aegypti genome identified ten putative jhe gene sequences. Analysis of these ten putative jhe gene sequences showed that only three of them, EAT43357, EAT43353 and EAT43354 contained GQSAG motif and showed high sequence similarity with the sequences of jhe genes identified from other insect species. To determine which putative jhe gene(s) code for functional JHE, the mRNA profiles of EAT43357, EAT43353 and EAT43354 were measured during the final instar la

Kr-h1 Encoding Juvenile Hormone Transcription Factor Impacts Reproductive Functions in Coccinella septempunctata.

The gene encoding juvenile hormone transcription factor Krüppel homolog 1 (Kr-h1) was studied for its effects on ovary development and reproduction in the ladybug beetle, Coccinella septempunctata. Kr-h1 expression in C. septempunctata was evaluated in females supplied with a juvenile hormone (JH) amended diet using quantitative PCR, and the function of Kr-h1 in female reproduction was assessed using RNAi technology. Expression of Kr-h1 in females supplied with a JH diet was significantly higher at 5 and 10 d than in females not supplied with JH. At 5 and 10 d post-injection with Kr-h1-dsRNA,

Precocene-I mediated reduction of juvenile hormone titers and ovarian activity is ephemeral in a bumble bee.

Juvenile hormones (JHs) are key insect endocrine signals regulating pre-adult development and adult life history strategies, physiology, and behavior. The importance of this endocrine signal has led to the development of insecticides targeting JH signaling pathways. While effective against target pests, they can also affect beneficial insects, such as pollinators and natural pest enemies, many of which are hymenopterans. Research on JH, which is crucial for understanding the physiology, behavior, and organization of social insects, requires effective means to manipulate JH signaling pathways.

Juvenile Fibroadenoma in a Prepubertal Girl With Idiopathic Precocious Puberty: A Case Report Challenging Hormonal Paradigms.

Juvenile fibroadenoma (JF) is a rare benign breast tumor that predominantly occurs in adolescent and young adult females. However, its occurrence in prepubertal girls is exceedingly rare and poses diagnostic and therapeutic challenges. We present a case of a six-year-old girl with a rapidly growing breast mass, ultimately diagnosed as juvenile fibroadenoma. This case highlights the importance of considering JF in the differential diagnosis of breast masses in pediatric patients, even in very young children. The clinical presentation, diagnostic workup, and management of JF are discussed, along

Membrane-localized MET engages PVR to mediate extranuclear juvenile hormone signaling in Aedes aegypti.

Although Methoprene-tolerant (MET) is well established as the intracellular receptor mediating the genomic actions of juvenile hormone (JH) in insects, the identity of the receptor responsible for initiating extranuclear JH responses has remained elusive. In the yellow fever mosquito Aedes aegypti, we identify the platelet-derived growth factor- and vascular endothelial growth factor-receptor related (PVR) protein as a key mediator of membrane-initiated JH signaling. JH treatment induces robust PVR phosphorylation in the fat body of adult female mosquitoes. Disruption of PVR function suppresse

Histone deacetylases synergistically regulate juvenile hormone signaling in the yellow fever mosquito, Aedes aegypti.

Controlling Aedes aegypti mosquitoes is crucial for managing mosquito-transmitted diseases like dengue, zika, chikungunya, and yellow fever. One of the efficient methods to control mosquitoes is to block their progression from the larval to the adult stage. Juvenile hormones (JH) maintain the larval stage and ensure proper developmental timing for transitioning from larval-pupal-adult stages. Our previous studies showed that histone deacetylases (HDACs) regulate JH signaling and metamorphosis in the red flour beetle Tribolium castaneum. However, the role of HDACs in regulating JH signaling in

A long-term growth hormone treatment stimulates growth and lipolysis in gilthead sea bream juveniles.

The enhancement of the endocrine growth hormone (GH)/insulin-like growth factor I (IGF-I) system by the treatment with a sustained release formulation of a recombinant bovine GH (rBGH), is a good strategy to investigate growth optimization in aquaculture fish species. To further deepen into the knowledge of rBGH effects in fish and to estimate the growth potential of juveniles of gilthead sea bream, the present work evaluated rBGH injection on growth, GH/IGF-I axis and lipid metabolism modulation, and explored the conservation of GH effects provoked by the in vivo treatment using in vitro mode

The Effects of Acute Neonatal Pain on Expression of Corticotropin-Releasing Hormone and Juvenile Anxiety in a Rodent Model.

Premature infants in the neonatal intensive care unit (NICU) may be subjected to numerous painful procedures without analgesics. One necessary, though acutely painful, procedure is the use of heel lances to monitor blood composition. The current study examined the acute effects of neonatal pain on maternal behavior as well as amygdalar and hypothalamic activation, and the long-term effects of neonatal pain on later-life anxiety-like behavior, using a rodent model. Neonatal manipulations consisted of either painful needle pricks or non-painful tactile stimulation in subjects' left plantar paw s

Transcriptional regulation of juvenile hormone-mediated induction of Krüppel homolog 1, a repressor of insect metamorphosis.

The Krüppel homolog 1 gene (Kr-h1) has been proposed to play a key role in the repression of insect metamorphosis. Kr-h1 is assumed to be induced by juvenile hormone (JH) via a JH receptor, methoprene-tolerant (Met), but the mechanism of induction is unclear. To elucidate the molecular mechanism of Kr-h1 induction, we first cloned cDNAs encoding Kr-h1 (BmKr-h1) and Met (BmMet1 and BmMet2) homologs from Bombyx mori. In a B. mori cell line, BmKr-h1 was rapidly induced by subnanomolar levels of natural JHs. Reporter assays identified a JH response element (kJHRE), comprising 141 nucleotides, loca

Changes in growth hormone receptor gene expression during therapy in children with juvenile idiopathic arthritis.

BACKGROUND: High levels of cytokines in juvenile idiopathic arthritis (JIA) can alter target cell sensitivity to growth hormone (GH) leading to short stature in adulthood. We hypothesized that the down-regulation of GH receptor (GHR) gene expression could be involved in growth failure of children with JIA. METHODS: In 18 (12 F and 6 M) prepubertal JIA patients and 13 age- and sex-matched healthy children, we evaluated serum growth-promoting factors and inflammatory indexes. We also measured GHR gene expression, by real-time PCR, in lymphocytes of patients and controls. All parameters were eva

Evaluation of juvenile hormone analogues as rodent feed-through insecticides for control of immature phlebotomine sandflies.

The juvenile hormone analogues methoprene and pyriproxyfen were evaluated as rodent feed-through insecticides for control of immature stages of the sandfly Phlebotomus papatasi Scopoli (Diptera: Psychodidae). The development and survival of P. papatasi second-instar larvae fed faeces from Syrian hamsters, Mesocricetus auratus, that had been fed a diet containing methoprene (0, 9.788, 97.88 or 978.8 p.p.m.) or pyriproxyfen (0, 9.82, 98.2 or 982 p.p.m.) were evaluated. The faeces of methoprene-treated hamsters greatly reduced the percentage of larvae that pupated at all concentrations tested and

Characterization of a juvenile hormone-regulated chymotrypsin-like serine protease gene in Aedes aegypti mosquito.

After female mosquitoes ingest blood from vertebrate hosts, exopeptidases and endopeptidases are required for digesting blood proteins in the midgut into amino acids, which female mosquitoes use to build yolk proteins. These proteases are not always present in the midgut, and their diverse expression patterns suggest that production of these enzymes is highly regulated in order to meet specific physiological demands at various stages. Here we report identification of a serine-type protease, JHA15, in the yellow fever mosquito Aedes aegypti. This protein shares high sequence homology with chymo

Just taller or more bone? The impact of growth hormone on osteogenesis imperfecta and idiopathic juvenile osteoporosis.

There are scant data to date on the use of GH therapy in osteogenesis imperfecta (OI) or in idiopathic juvenile osteoporosis (IJO). In OI, we review three clinical trials (aggregate study population 46), and 13 patient reports from the NCGS, and two independent patient reports. Based on evidence of increased growth rate versus some reported increased fracture rate, we conclude that GH should probably not be used as first-line therapy in OI, pending further data from clinical trials. There are no published reports of the use of GH therapy in IJO and only one patient with documented IJO enrolled

Evidence for an inhibitory role of cyclic AMP in the control of juvenile hormone biosynthesis by cockroach corpora allata.

High levels of cyclic AMP were found in the corpora allata of adult female Diploptera punctata. The adenylate cyclase activator forskolin caused a rapid, reversible and dose-dependent accumulation of cAMP in the corpora allata in vitro. The sensitivity of the corpora allata to forskolin was low when juvenile hormone (JH) synthetic activity was high, and vice versa. Incubation of corpora allata with compounds which cause or mimic elevated intracellular cAMP levels (forskolin, 3-isobutyl-1-methylxanthine, 8-bromo-cAMP) led to a rapid and dose-dependent inhibition of juvenile hormone synthesis. Post-eclosion growth in the Drosophila ejaculatory duct is driven by Juvenile hormone signaling and is essential for male fertility.

The Drosophila Ejaculatory duct (ED) is a secretory tissue of the somatic male reproductive system. The ED is involved in the secretion of seminal fluid components and ED-specific antimicrobial peptides that aid in fertility and the female post-mating response. The ED is composed of secretory epithelial cells surrounded by a layer of innervated contractile muscle. The ED grows in young adult males during the first 24 h post-eclosion, but the cell cycle status of the ED secretory cells and the role of post-eclosion ED growth have been unexplored. Here, we show that secretory cells of the adult
