An ERG channel inhibitor from the scorpion Buthus eupeus.

The isolation of the peptide inhibitor of M-type K(+) current, BeKm-1, from the venom of the Central Asian scorpion Buthus eupeus has been described previously (Fillipov A. K., Kozlov, S. A., Pluzhnikov, K. A., Grishin, E. V., and Brown, D. A. (1996) FEBS Lett. 384, 277-280). Here we report the cloning, expression, and selectivity of BeKm-1. A full-length cDNA of 365 nucleotides encoding the precursor of BeKm-1 was isolated using the rapid amplification of cDNA ends polymerase chain reaction technique from mRNA obtained from scorpion telsons. Sequence analysis of the cDNA revealed that the pre

Severe Form of ßIV-Spectrin Deficiency With Mitochondrial Dysfunction and Cardiomyopathy-A Case Report.

ßIV-spectrin is a protein of the spectrin family which is involved in the organization of the cytoskeleton structure and is found in high quantity in the axon initial segment and the nodes of Ranvier. Together with ankyrin G, ßIV-spectrin is responsible for the clustering of KCNQ2/3-potassium channels and NaV-sodium channels. Loss or reduction of ßIV-spectrin causes a destabilization of the cytoskeleton and an impairment in the generation of the action potential, which leads to neuronal degeneration. Furthermore, ßIV-spectrin has been described to play an important role in the maintenance of t

Neurocardiac pathologies associated with potassium channelopathies.

Voltage-gated potassium channels are expressed throughout the human body and are essential for physiological functions. These include delayed rectifiers, A-type channels, outward rectifiers, and inward rectifiers. They impact electrical function in the heart (repolarization) and brain (repolarization and stabilization of the resting membrane potential). KCNQx and KCNHx encode Kv7.x and Kv11.x proteins, which form delayed rectifier potassium channels. KCNQx and KCNHx channelopathies are associated with both cardiac and neuronal pathologies. These include electrocardiographic abnormalities, card

Assessment of gabapentin efficacy in patients with KCNQ2-developmental epileptic encephalopathy.

KCNQ2‑related developmental epileptic encephalopathy (KCNQ2‑DEE) is characterized by intractable epilepsy, cognitive impairment, and behavioral problems, highlighting the urgent need for precision therapies. This study aimed to promote the clinical application of gabapentin, a known Kv7 channel opener, in KCNQ2‑DEE by evaluating its efficacy in four patients carrying different KCNQ2 variants. Three patients with S6‑segment variants (p.A294V, p.F305I, p.F305del) showed clinical improvement in both seizure control and cognition following add‑on gabapentin. In contrast, the patient with the S4 va

Why we urgently need improved seizure and epilepsy therapies for children and neonates.

In contrast to epilepsy in adolescents and adults, neonatal seizures and early onset epilepsy poses unique challenges with significant repercussion for treatment choices. Most importantly, high seizure burden and epileptic encephalopathy are associated with developmental, behavioural and cognitive problems. The causes are multifactorial and include etiology, seizure burden, epileptic encephalopathy, but also antiseizure medication. In contrast to adults and older children only very few drugs have been licenced for infants and neonates, and after a long delay. Very recently, extrapolation of ad

Discovery of fluoxakalner, a novel and preferential Kv7.2/7.3 channel opener for antinociception in mice.

Activating neuronal M-current (Kv7.2/7.3) channels constitutes a promising therapeutic strategy for antinociception and epilepsy through hyperpolarizing membrane potential and reducing neuronal hyperexcitability. However, retigabine (RTG), the first-in-class Kv7 opener, was withdrawn from clinical use due to its oxidative metabolites that cause tissue discoloration and toxicity. To mitigate this clinical risk, we synthesized the N-{4-[(4-fluorobenzyl)oxy]-2,6-dimethylphenyl}-2-phenylacetamide, designated fluoxakalner, by modifying the primary amine moiety of RTG through introducing two methyl

In Vivo/In vitro metabolic profiling and evaluation of key metabolites of QO-83, a novel KCNQ channel opener with antiepileptic potential.

KCNQ2/3 voltage-gated potassium channels constitute validated therapeutic targets for epilepsy by regulating neuronal hyperexcitability. QO-83, a novel KCNQ channel opener exhibiting potent activity and channel subtype selectivity, was undergoing preclinical studies as an antiepileptic candidate drug. This study integrated in vivo metabolism studies in rats and Beagles with in vitro models of human and rat hepatic systems using liquid chromatography-mass spectrometry (LC-MS). Key metabolites were further evaluated for channel open activity in cells and pharmacokinetics/pharmacodynamics (PK/PD)

Function of KCNQ2 channels at nodes of Ranvier of lumbar spinal ventral nerves of rats.

Previous immunohistochemical studies have shown the expression of KCNQ2 channels at nodes of Ranvier (NRs) of myelinated nerves. However, functions of these channels at NRs remain elusive. In the present study, we addressed this issue by directly applying whole-cell patch-clamp recordings at NRs of rat lumbar spinal ventral nerves in ex vivo preparations. We show that depolarizing voltages evoke large non-inactivating outward currents at NRs, which are partially inhibited by KCNQ channel blocker linopirdine and potentiated by KCNQ channel activator retigabine. Furthermore, linopirdine signific

Niemann-Pick Type C Disease Reveals a Link between Lysosomal Cholesterol and PtdIns(4,5)P2 That Regulates Neuronal Excitability.

There is increasing evidence that the lysosome is involved in the pathogenesis of a variety of neurodegenerative disorders. Thus, mechanisms that link lysosome dysfunction to the disruption of neuronal homeostasis offer opportunities to understand the molecular underpinnings of neurodegeneration and potentially identify specific therapeutic targets. Here, using a monogenic neurodegenerative disorder, NPC1 disease, we demonstrate that reduced cholesterol efflux from lysosomes aberrantly modifies neuronal firing patterns. The molecular mechanism linking alterations in lysosomal cholesterol egres

KCNQ (Kv7) potassium channel activators as bronchodilators: combination with a β2-adrenergic agonist enhances relaxation of rat airways.

KCNQ (Kv7 family) potassium (K(+)) channels were recently found in airway smooth muscle cells (ASMCs) from rodent and human bronchioles. In the present study, we evaluated expression of KCNQ channels and their role in constriction/relaxation of rat airways. Real-time RT-PCR analysis revealed expression of KCNQ4 > KCNQ5 > KCNQ1 > KCNQ2 > KCNQ3, and patch-clamp electrophysiology detected KCNQ currents in rat ASMCs. In precision-cut lung slices, the KCNQ channel activator retigabine induced a concentration-dependent relaxation of small bronchioles preconstricted with methacholine (MeCh; EC50 = 3.

Voltage-gated potassium channels control extended access cocaine seeking: a role for nucleus accumbens astrocytes.

Dopaminergic signaling in the nucleus accumbens shell (NAc) regulates neuronal activity relevant to reward-related learning, including cocaine-associated behaviors. Although astrocytes respond to dopamine and cocaine with structural changes, the impact of dopamine and cocaine on astrocyte functional plasticity has not been widely studied. Specifically, behavioral implications of voltage-gated channel activity in the canonically non-excitable astrocytes are not known. We characterized potassium channel function in NAc astrocytes following exposure to exogenous dopamine or cocaine self-administr
