Impact of Yeast Types on Quality Characteristics and Storage Stability of Industrial Kefir.

Kefir is a fermented dairy product that contains many types of lactic acid bacteria and different types of yeast. Yeasts in kefir culture are important in creating the unique properties of kefir. When yeasts are not included in kefir production, the unique properties of kefir do not emerge. In this study, the effects of various yeast microorganisms that can cause negative effects, such as swelling and deformation, in industrial kefir production on product quality characteristics were investigated. For this purpose, a commercial yeast-free kefir culture was supplemented with selected yeasts: Kl

Yeast bioaugmentation fosters medium-chain carboxylates and polyhydroxyalkanoates production from cheese whey.

This study investigated polyhydroxyalkanoates (PHA) production from carboxylic acids obtained by yeast-assisted cheese whey (CW) fermentation. A three-stage hybrid strategy was employed. First, a lactose-fermenting yeast (Kluyveromyces marxianus) was inoculated into raw or sterilized CW to produce ethanol and/or lactic acid. Second, these intermediates were converted to carboxylic acids, with an emphasis on caproic acid, using a chain-elongating microbial culture. Batch and fermenter experiments under different operating conditions produced four distinct fermentation products with total carbox

Bifunctional Enzymes for Deoxynivalenol Degradation in Prokaryotic and Eukaryotic Chassis.

Deoxynivalenol (DON), a prevalent mycotoxin produced by Fusarium spp., represents a persistent threat to global food and feed safety. Two innovative strategies were developed herein for efficient DON detoxification. Coupling the genes encoding the aldo-keto reductase AKR13B2 and the pyrroloquinoline quinone-dependent DON dehydrogenase DepA using a flexible linker enabled rational engineering of the bifunctional fusion enzyme BGA (AKR13B2-[GGGGS]2-DepA). This design facilitated proximity-enhanced sequential conversion of DON to 3-keto-DON and the markedly less toxic 3-epi-DON. BGA exhibited sig

Fluorescence-based screening for engineered aldo-keto reductase KmAKR with improved catalytic performance and extended substrate scope.

BACKGROUND: Aldo-keto reductases-catalyzed transformations of ketones to chiral alcohols have become an established biocatalytic process step in the pharmaceutical industry. Previously, we have discovered an aldo-keto reductase (AKR) from Kluyveromyces marxianus that is active to the aliphatic tert-butyl 6-substituted (5R/S)-hydroxy-3-oxohexanoates, but it is inactive to aromatic ketones. In order to acquire an excellent KmAKRmutant for ensuring the simultaneous improvement of activity-thermostability toward tert-butyl 6-cyano-(5R)-hydroxy-3-oxohexanoate ((5R)-1) and broadening the universal a

Specific growth rates and growth stoichiometries of Saccharomycotina yeasts on ethanol as sole carbon and energy substrate.

Emerging low-emission production technologies make ethanol an interesting substrate for yeast biotechnology, but information on growth rates and biomass yields of yeasts on ethanol is scarce. Strains of 52 Saccharomycotina yeasts were screened for growth on ethanol. The 21 fastest strains, among which representatives of the Phaffomycetales order were overrepresented, showed specific growth rates in ethanol-grown shake-flask cultures between 0.12 and 0.46 h-1. Seven strains were studied in aerobic, ethanol-limited chemostats (dilution rate 0.10 h-1). Saccharomyces cerevisiae and Kluyveromyces l

Probiotics supplements on fattening guinea pigs: effects on carcass traits and non-carcass components.

The growing demand for animal protein is driving the search for natural alternatives, such as probiotics, following the ban on growth-promoting antibiotics due to bacterial resistance. The objective of this study was to evaluate the effect of including probiotics made from agro-industrial waste fermented with Lactobacillus acidophilus and Kluyveromyces fragilis and their combination on carcass traits, non-carcass components and colour and pH evolution during muscle-to-meat conversion of 90-day-old male guinea pigs. A total of 40 male guinea pigs were randomly assigned to one of four groups: (a

Microbial communities and volatile profile of Queijo de Azeitão PDO cheese, a traditional Mediterranean thistle-curdled cheese from Portugal.

The production of ovine or caprine milk cheeses with thistle rennet is a common practice in the Mediterranean basin. The aim of the present study was to obtain information on bacteria and yeast communities harboured by Queijo de Azeitão PDO cheese through viable counting and, for the first time, via metataxonomic analysis. Moreover, solid phase microextraction (SPME) technique was applied to characterize Queijo de Azeitão PDO cheese volatile compounds. Nine cheese samples were collected from three different artisan producers located in Portugal. The results of physico-chemical analyses showed

Evaluation of lactulose synthesis using immobilized β-galactosidase in different reactor configurations.

Lactulose is a disaccharide composed of galactose and fructose. In the human body, it functions as a prebiotic, promoting the growth of beneficial bacteria, such as bifidobacteria and lactobacilli, in the gastrointestinal tract. Lactulose is produced via a transgalactosylation reaction between lactose and fructose, catalyzed by β-galactosidase. Prominent β-galactosidases used in lactulose synthesis include those from Aspergillus oryzae and Kluyveromyces lactis. Various methodologies have been developed to enhance the efficiency of enzymatic lactulose production. The objective of this study was

Comparative fermentability of a glucose/xylose enriched enzymatic slurry using different yeast strains.

Lignocellulosic biomass represents a sustainable energy source as ethanol fuel demand increases; process integration and efficient C6/C5 sugars utilization are essential for achieving techno-economically viable production. Here, alkaline-sulfite pretreatment combined with disc refining was applied to sugarcane bagasse for conserving most of cellulose and hemicellulose fractions. Then, β-glucosidase supplementation of cellulase at rational enzyme dosage (18 FPU/g biomass) was established for the enzymatic hydrolysis of the pretreated solids, achieving approximately 75% conversion yield for both

A unique metabolic gene cluster regulates lactose and galactose metabolism in the yeast Candida intermedia.

Lactose assimilation is a relatively rare trait in yeasts, and Kluyveromyces yeast species have long served as model organisms for studying lactose metabolism. Meanwhile, the metabolic strategies of most other lactose-assimilating yeasts remain unknown. In this work, we have elucidated the genetic determinants of the superior lactose-growing yeast Candida intermedia. Through genomic and transcriptomic analyses, we identified three interdependent gene clusters responsible for the metabolism of lactose and its hydrolysis product galactose: the conserved LAC cluster (LAC12, LAC4) for lactose upta

Improvement of third generation bioethanol production from photoheterotrophically cultivated Chlorella vulgaris in the presence of carrot pomace and onion peel waste.

Microalgae are promising feedstocks, holding great potential for bioethanol production. However, microalgal bioethanol production requires the use of different approaches in order to optimize the process. In this study, the bioethanol production capacities of local yeasts (Saccharomyces cerevisiae, Candida boidinii and Kluyveromyces marxianus) from Chlorella vulgaris biomass cultivated with 0.5 g/L carrot pomace sugar was investigated. Onion peel, a common waste product, was used for investigating its potential in improving yeast growth and bioethanol production by K. marxianus. Parameters suc

Molecular Dynamics Simulation-Assisted Tuning of Key Sites to Alter the Substrate Specificity of the Aldo-Keto Reductase KmAKR.

Aldo-keto reductase (AKR) is an important biocatalyst for the synthesis of chiral alcohols; however, its inability to catalyze bulky substrates severely limits its industrial applications. Previously, the T23V/Q213A mutant of AKR from Kluyveromyces marxianus (KmAKR) exhibited an extended substrate scope, but it still showed poor catalytic activity toward some valuable aliphatic and aromatic ketones. Here, we developed a computer-assisted strategy to virtually screen a mutation library constructed from residues 23 and 213. Guided by the binding free energy calculated from high-throughput molecu

Sec14 family of lipid transfer proteins in yeasts.

The hydrophobicity of lipids prevents their free movement across the cytoplasm. To achieve highly heterogeneous and precisely regulated lipid distribution in different cellular membranes, lipids are transported by lipid transfer proteins (LTPs) in addition to their transport by vesicles. Sec14 family is one of the most extensively studied groups of LTPs. Here we provide an overview of Sec14 family of LTPs in the most studied yeast Saccharomyces cerevisiae as well as in other selected non-Saccharomyces yeasts-Schizosaccharomyces pombe, Kluyveromyces lactis, Candida albicans, Candida glabrata, C

Manogepix (APX001A) Displays Potent In Vitro Activity against Human Pathogenic Yeast, but with an Unexpected Correlation to Fluconazole MICs.

Manogepix (APX001A) is the active moiety of the novel drug candidate fosmanogepix (APX001). We previously reported the broad-spectrum activity of manogepix but also observed a correlation between increased manogepix and fluconazole MICs. Here, we extended this study and included isolates with acquired fluconazole resistance. Isolates (n = 835) were identified using CHROMagar, matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS), and, when needed, internal transcribed spacer (ITS) sequencing.
