BACKGROUND: Leptospirosis is a global zoonotic disease affecting humans, companion animals, and domestic livestock. Clinical presentation of disease is dependent on complex interactions between host species and pathogenic species, serovars, and strains of Leptospira. This is exemplified by L. borgpetersenii serovar Hardjo strains HB203 and JB197, which differ very little by genome content but cause divergent clinical disease phenotypes in the hamster model of leptospirosis. Vaccination is commonly used to protect dogs against leptospirosis, however, memory immune responses induced by canine Leptospira vaccines have not been studied. The region of Antioquia in northeastern Colombia has the highest number of reported leptospirosis cases in the country. A toolbox for chemotaxis assay adapted to Leptospira spp. has emerged in the recent years: soft agar assay, capillary assay, and videomicroscopy tracking. Those methods allow to demonstrate chemotaxis defect, identify diverse chemoattractant, or decipher motile behavior quantitatively. These experiments have demonstrated a role of motility and potentially chemotaxis in leptospirosis pathogenesis. We describe extensively the methods and provide the key steps to use this toolbox.

This article reports an incidental finding of leptospirosis during a special consultation, which was initiated due to the pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). The focus on SARS-CoV‑2 and the uncharacteristic symptoms of these two diseases make it much more difficult to find the correct diagnosis. Outbreaks are possible at any territory with a landscape harbouring natural reservoirs of Leptospyras and inhabited by small moitsaure-loving rodents. Leptospirosis peak season is summer. The main goal of this study was to obtain new isolates of Leptospira spp. from sheep. A total of 10 kidney samples and 44 blood samples were collected from sheep slaughtered in Pelotas, Southern Brazil. One isolate was obtained which was identified by 16S rRNA gene sequencing and serogrouping to be Leptospira noguchii serogroup Autumnalis. We report a human case of leptospirosis in which the spirochete was detected by dark-field microscopy examination of cerebrospinal fluid (CSF) and isolated from both CSF and blood. Leptospira fainei was identified by sequencing the 16S rDNA gene, which had been amplified by polymerase chain reaction. This case confirms the role of L. fainei as a human pathogen and extends its distribution to southern Europe.

Meningeal involvement in leptospiral infection is quite common, usually mild and often overlooked. In contrast, cases of isolated involvement of the central nervous system, including aseptic meningitis, have been reported only rarely. A case of a patient with acute aseptic meningitis caused by Leptospira australis serovar bratislava is reported. This is believed to be the first report of aseptic meningitis due to Leptospira australis. This case indicates the need to consider human leptospirosis in the differential diagnosis of aseptic meningitis.
