Use of a feline cell line in the syncytia infectivity assay for the detection of bovine leukemia virus infection in cattle.

This report describes a modified syncytia infectivity assay (SIA) for the direct detection of bovine leukemia virus (BLV) in blood lymphocytes of cattle, using transformed feline (CC81) cells as the indicator system. The data show that the syncytia present in cultures of CC81 cells inoculated with BLV-infected cells are specific and arise through a mechanism similar to that responsible for the phenomenon of "late" polykaryocytosis described in other virus systems. The susceptibility of the CC81 cells to the syncytia-inducing effect of BLV-infected cells is comparable with that of early passage

Substitution of feline leukemia virus long terminal repeat sequences into murine leukemia virus alters the pattern of insertional activation and identifies new common insertion sites.

The recombinant retrovirus, MoFe2-MuLV (MoFe2), was constructed by replacing the U3 region of Moloney murine leukemia virus (M-MuLV) with homologous sequences from the FeLV-945 LTR. NIH/Swiss mice neonatally inoculated with MoFe2 developed T-cell lymphomas of immature thymocyte surface phenotype. MoFe2 integrated infrequently (0 to 9%) near common insertion sites (CISs) previously identified for either parent virus. Using three different strategies, CISs in MoFe2-induced tumors were identified at six loci, none of which had been previously reported as CISs in tumors induced by either parent vi

Presence of Endogenous Viral Elements Negatively Correlates with Feline Leukemia Virus Susceptibility in Puma and Domestic Cat Cells.

While feline leukemia virus (FeLV) has been shown to infect felid species other than the endemic domestic cat host, differences in FeLV susceptibility among species has not been evaluated. Previous reports have noted a negative correlation between endogenous FeLV (enFeLV) copy number and exogenous FeLV (exFeLV) infection outcomes in domestic cats. Since felids outside the genus Felis do not harbor enFeLV genomes, we hypothesized absence of enFeLV results in more severe disease consequences in felid species lacking these genomic elements. We infected primary fibroblasts isolated from domestic c

Lymphokines modulate the growth and survival of thymic tumor cells containing a novel feline leukemia virus/Notch2 variant.

Tumorigenesis occurs through a multistep process initiated by genetic lesions and facilitated by endogenous and external growth/survival signals. In many malignancies, specific oncogenic mutations correlate with phenotypic characteristics, inferring lineage-specific pathogenic mechanisms. To characterize these relationships in a unique feline tumor, we studied primary cells and two-cell lines independently-derived from a thymic lymphoma that contained and actively expressed a novel feline leukemia virus (FeLV) recombinant with transduced host Notch2 sequences. All three tumor cell populations

Molecular Characterization of Feline Leukemia Virus from Cats in Shelters in the Metropolitan Region of Recife, Pernambuco, Northeast Brazil.

This research aimed to conduct an epidemiological-molecular study of feline leukemia virus (FeLV) infection in cats from shelters in the Metropolitan Region of Recife (MRR), Pernambuco, Northeast Brazil. Blood samples from 260 cats from 18 shelters located in 5 municipalities within the MRR were analyzed. DNA samples from peripheral blood were subjected to Polymerase Chain Reaction (PCR) to detect FeLV proviral DNA. PCR-positive samples were sequenced and analyzed using BLAST software for phylogenetic tree construction. The prevalence of infection in cats was 9.6% (25/260). Among the 18 sample

Laboratory Changes Supporting a Diagnosis of Immune-Mediated Anemia in Cats With Initial Suspicion of Progressive Feline Leukemia Virus.

BACKGROUND: Progressive feline leukemia virus (FeLV) infection and anemia of suspected immune-mediated origin (ASIMO) are associated with similar hematologic abnormalities, including anemia, neutropenia, and lymphocytosis. Cats with ASIMO may test positive for FeLV using point-of-care (POC) ELISA tests. OBJECTIVE: To describe laboratory changes that may result in the misclassification of cats with ASIMO as having progressive FeLV. METHODS: Cats with ASIMO and laboratory results mimicking FeLV infection were retrospectively identified via searches of laboratory and clinical records. Inclusion

Prospective Investigation of Feline Leukemia Virus Infection in Stray Cats Subjected to a Trap-Neuter-Return Program in Switzerland.

Feline leukemia virus (FeLV) remains a serious concern in some countries despite advances in diagnostics and vaccines. FeLV-infected cats often have reduced lifespans due to FeLV-associated diseases. The infection is transmitted through social interactions. While Northern European countries have reported a decrease in FeLV among pet cats, Switzerland's rates remain stagnant at 2.7% (2016/17: 95% CI 1.4-5.2%). Research on FeLV in Swiss stray cats has been lacking, even though these animals could serve as a virus reservoir. Sampling stray cats that do not receive regular veterinary care can be c

Distinctive receptor binding properties of the surface glycoprotein of a natural feline leukemia virus isolate with unusual disease spectrum.

BACKGROUND: Feline leukemia virus (FeLV)-945, a member of the FeLV-A subgroup, was previously isolated from a cohort of naturally infected cats. An unusual multicentric lymphoma of non-T-cell origin was observed in natural and experimental infection with FeLV-945. Previous studies implicated the FeLV-945 surface glycoprotein (SU) as a determinant of disease outcome by an as yet unknown mechanism. The present studies demonstrate that FeLV-945 SU confers distinctive properties of binding to the cell surface receptor. RESULTS: Virions bearing the FeLV-945 Env protein were observed to bind the ce

Lymphocyte mitogen reactivity and enumeration of circulating B- and T-cells during feline leukemia virus infection in the cat.

Mitogen-induced blast transformation of peripheral blood lymphocytes and quantitative changes in circulating T- and B-cells were studied serially in cats inoculated with feline leukemia virus (FeLV). Concanavalin A-induced blast transformation sharply declined beginning at 5 weeks post inoculation (Pl) in FeLV-infected cats when compared to age-matched uninfected control cats. Similar but less consistent changes were seen in responses to pokeweed mitogen-induced stimulation. In most infected kittens this defect persisted until they died from thymic lymphosarcoma, 15-24 weeks Pl. An early lymph

Molecular Detection of Feline Leukemia Virus in Oral, Conjunctival, and Rectal Mucosae Provides Results Comparable to Detection in Blood.

Feline leukemia virus (FeLV) infection causes immunosuppression, degeneration of the hematopoietic system, and fatal neoplasms. FeLV transmission occurs mainly by close social contact of infected and susceptible cats. Developing procedures for the diagnosis of feline retroviruses is crucial to reduce negative impacts on cat health and increase the number of animals tested. Blood collection requires physical or chemical restraint and is usually a stressful procedure for cats. Our objective was to evaluate the use of samples obtained from oral, conjunctival, and rectal mucosae for the molecular

Subtle mutational changes in the SU protein of a natural feline leukemia virus subgroup A isolate alter disease spectrum.

FeLV-945 is a representative isolate of the natural feline leukemia virus (FeLV) variant predominant in non-T-cell malignant, proliferative, and degenerative diseases in a geographic cohort. The FeLV-945 surface glycoprotein (SU) is closely related to natural horizontally transmissible FeLV subgroup A (FeLV-A) but was found to differ from a prototype to a larger extent than the members of FeLV-A differ among themselves. The sequence differences included point mutations restricted largely to the functional domains of SU, i.e., VRA, VRB, and PRR. Despite the sequence differences in these critica

Heme and sensory neuropathy: insights from novel mutations in the heme exporter feline leukemia virus subgroup C receptor 1.

Hereditary sensory and autonomic neuropathies (HSANs) are a group of clinically and genetically heterogeneous disorders of the peripheral nervous system mainly characterized by impaired nociception and autonomic dysfunction. We previously identified heme metabolism as a novel pathway contributing to sensory neurons maintenance and nociception. Indeed, we reported mutations in the feline leukemia virus subgroup C receptor 1 (FLVCR1) gene in individuals affected by HSAN. FLVCR1 gene encodes for 2 heme export proteins, FLVCR1a (plasma membrane) and FLVCR1b (mitochondria), crucially involved in th

Cross-species transmission of host adaptation of feline leukemia virus between domestic cats and the wild felid Leopardus guigna.

Feline leukemia virus (FeLV) is widespread in domestic cats and frequently spills over into wild felid populations, causing severe outbreaks in nondomestic felid populations. Domestic cats carry both endogenous FeLV (enFeLV) and exogenous variants of FeLV (exFeLV), which can recombine to generate novel variants with alternate receptor usage. In contrast, most nondomestic felids, including the guigna, do not harbor enFeLV. This study applied amplification of the FeLV envelope gene combined with Illumina sequencing to characterize FeLV envelope gene diversity and transmission dynamics in guignas

Effect of antineoplastic drugs on the expression of Bcl-2 and Bcl-xL genes in the feline T-cell leukemia cell line.

The Bcl-2 gene is the first member of a rapidly expanding family of genes that regulate apoptosis. Bcl-2 has been shown to repress cell death triggered by a diverse array of stimuli including chemotherapy and gamma-irradiation. Chemotherapy of feline lymphoma is generally carried out with antineoplastic drugs, which are reported to induce apoptosis in tumor cells. However, the precise apoptotic signals, induced by chemotherapeutic drugs against feline tumors have not been fully characterized. Therefore, we have evaluated the expression of Bcl-2 and Bcl-xL in FT-1 upon in vitro treatment with t

Innovative Colorimetric Neutral Red-Based Loop-Mediated Isothermal Amplification (NR-LAMP) Assay: Transforming Rapid and Affordable Feline Leukemia Virus Detection.

Feline leukemia virus (FeLV) is a retrovirus that globally affects both domestic and wild cats, leading to the development of leukemia, lymphoma, and immunosuppression. However, it is important to note that the daily antigen test may yield false negative results. In this study, we successfully developed the first colorimetric loop-mediated isothermal amplification (LAMP) associated with neutral red (NR-LAMP) for the detection of FeLV. The NR-LAMP assay exhibited high sensitivity and efficiency compared to the routine polymerase chain reaction (PCR) reference method. To ensure specificity, a no

Retroviral DNA--the silent winner: blood transfusion containing latent feline leukemia provirus causes infection and disease in naïve recipient cats.

BACKGROUND: The feline leukemia virus (FeLV) is a gamma-retrovirus of domestic cats that was discovered half a century ago. Cats that are infected with FeLV may develop a progressive infection resulting in persistent viremia, immunodeficiency, tumors, anemia and death. A significant number of cats mount a protective immune response that suppresses viremia; these cats develop a regressive infection characterized by the absence of viral replication and the presence of low levels of proviral DNA. The biological importance of these latter provirus carriers is largely unknown. RESULTS: Here, we de

Prevalence of feline leukemia virus infection and associated diseases in a Portuguese domestic cat population: A 4.5-year cross-sectional study.

In a 2019 Pan-European Study, Portugal exhibited the highest prevalence of Feline Leukemia Virus (FeLV) infection (8.8%). Following the coronavirus disease 2019 (COVID-19) pandemic, it is crucial to evaluate how the prevalence of FeLV has evolved. FeLV infection is associated with the highest morbidity rates, primarily due to the increased incidence of diseases that compromise the health of cat populations, which varies according to the lifestyle and background of the cats studied. This study aimed (1) to estimate the prevalence and temporal trends of FeLV and FIV infections among cats present

A Novel, Safe, Non-Adjuvanted Alphavirus Replicon-Based Vaccine Expressing the Feline Leukemia Virus Envelope Protein Protects Against Virulent FeLV Challenge.

Background/Objectives: A number of different vaccines against feline leukemia virus (FeLV) are available; however, there is continuous debate regarding the efficacy advantages of adjuvanted vaccines versus the potential safety advantages of non-adjuvanted vaccines. Methods: For this reason, we developed a non-adjuvanted vaccine based on a replicon RNA particle (RP) expressing the FeLV gp85 envelope protein, which possesses the safety of a non-adjuvanted vaccine while consistently providing high efficacy. Results: In two efficacy studies, a high-level of protection against virulent FeLV challen
