Genome-Wide Analysis of CsCAX Genes and Functional Characterization of CsCAX3 Revealing Its Negative Role in Citrus Bacterial Disease Resistance.

Cation/proton antiporters (CAXs) are key membrane transporters involved in plant development and stress adaptation. In this study, five CsCAX genes (CsCAX1-CsCAX5) were identified from the Citrus sinensis genome through comprehensive bioinformatic analysis. All CsCAX proteins are hydrophobic with 10-11 transmembrane domains, predominantly composed of α-helices and random coils, and are localized to either the plasma membrane or vacuole. Phylogenetic analysis classified them into two subfamilies (IA and IB). Promoter prediction identified stress- and hormone-responsive cis-elements (e.g., LTR,

Silencing of V-ATPase-E gene causes midgut apoptosis of Diaphorina citri and affects its acquisition of Huanglongbing pathogen.

The Asian citrus psyllid, Diaphorina citri Kuwayama, is among the most important pests of citrus. It is the main vector of the Huanglongbing (HLB) pathogen Candidatus Liberibacter asiaticus (CLas), which causes severe losses in citrus crops. Control of D. citri is therefore of paramount importance to reduce the spread of HLB. In this regard, using RNA interference (RNAi) to silence target genes is a useful strategy to control psyllids. In this study, using RNAi, we examined the biological functions of the V-ATPase subunit E (V-ATP-E) gene of D. citri, including its effect on acquisition of CLa

Simple template-based reverse transcription-recombinase polymerase amplification assay for routine diagnosis of citrus tristeza virus.

This study reports a simple template-based reverse transcription-polymerase amplification assay (ST-RT-RPA) for detection of citrus tristeza virus (CTV) from crude plant extract lysed in NaOH:EDTA (1:1) without the need of tedious RNA isolation. The developed assay showed versatility in its usage as amplification can be performed at wide temperature range (14°C to 42°C) and incubation time (4 to 32 min), although the best conditions were 38°C for 30 min. The developed ST-RT-RPA assay could detect the CTV up to 10-8 dilution of crude plant extract of NaOH:EDTA and up to 0.01 fg µl-1 of RNA of C

A Combinatorial Single-Molecule Real-Time and Illumina Sequencing Analysis of Postembryonic Gene Expression in the Asian Citrus Psyllid Diaphorina citri.

Huanglongbing (HLB) is a systemic plant disease caused by 'Candidatus Liberibacter asiaticus (CLas)' and transmitted by Diaphorina citri. D. citri acquires the CLas bacteria in the nymph stage and transmits it in the adult stage, indicating that molting from the nymph to adult stages is crucial for HLB transmission. However, the available D. citri reference genomes are incomplete, and gene function studies have been limited to date. In the current research, PacBio single-molecule real-time (SMRT) and Illumina sequencing were performed to investigate the transcriptome of D. citri nymphs and adu

Effect of Huanglongbing on the Volatile Organic Compound Profile of Fruit Juice and Peel Oil in 'Ray Ruby' Grapefruit.

Along with orange and mandarin, grapefruit production in Florida has declined sharply due to Huanglongbing (HLB), or citrus greening disease, caused by Candidatus Liberibacter asiaticus (CLas). HLB affects the volatile profiles of juice and peel oil in oranges, but there is limited information on grapefruit. In this research, 'Ray Ruby' grapefruit were harvested in 2020 and 2021 from healthy (HLB-) and HLB-affected (HLB+) trees. Peel oil was extracted by hydrodistillation, and the volatiles were analyzed by direct injection of the oil samples into gas chromatography-mass spectrometry (GC-MS).

Disrupting Mating Behavior of Diaphorina citri (Liviidae).

Severe economic damage from citrus greening disease, caused by 'Candidatus Liberibacter asiaticus' bacteria, has stimulated development of methods to reduce mating and reproduction in populations of its insect vector, Diaphorina citri (Hemiptera: Liviidae). Male D. citri find mating partners by walking on host plants, intermittently producing vibrational calls that stimulate duetting replies by receptive females. The replies provide orientational feedback, assisting the search process. To test a hypothesis that D. citri mating can be disrupted using vibrational signals that compete with and/or

Disruption of Vector Host Preference with Plant Volatiles May Reduce Spread of Insect-Transmitted Plant Pathogens.

Plant pathogens can manipulate the odor of their host; the odor of an infected plant is often attractive to the plant pathogen vector. It has been suggested that this odor-mediated manipulation attracts vectors and may contribute to spread of disease; however, this requires further broad demonstration among vector-pathogen systems. In addition, disruption of this indirect chemical communication between the pathogen and the vector has not been attempted. We present a model that demonstrates how a phytophathogen (Candidatus Liberibacter asiaticus) can increase its spread by indirectly manipulati

Asymptomatic spread of huanglongbing and implications for disease control.

Huanglongbing (HLB) is a bacterial infection of citrus trees transmitted by the Asian citrus psyllid Diaphorina citri. Mitigation of HLB has focused on spraying of insecticides to reduce the psyllid population and removal of trees when they first show symptoms of the disease. These interventions have been only marginally effective, because symptoms of HLB do not appear on leaves for months to years after initial infection. Limited knowledge about disease spread during the asymptomatic phase is exemplified by the heretofore unknown length of time from initial infection of newly developing clust

Foliar Applications of Microbe-Associated Molecular Pattern Molecules Enhance Citrus Resistance Against Huanglongbing.

Citrus Huanglongbing (HLB) associated with 'Candidatus Liberibacter asiaticus' (CLas), a biotrophic pathogen, is a devastating citrus diseases worldwide. Enhancing host plant resistance through elicitor treatments has been exploited for crop disease management. HLB was suggested to be a pathogen-triggered immune disease in HLB susceptible varieties. Here, we hypothesized that boosting citrus immunity to CLas with biotic elicitors including microbe-associated molecular patterns (MAMPs) can increase citrus resistance to HLB. In greenhouse assays, foliar spray of chitosan, harpins, or lipopolysac

Lemon Terpenes Influence Behavior of the African Citrus Triozid Trioza erytreae (Hemiptera: Triozidae).

The African citrus triozid, Trioza erytreae Del Guercio (Hemiptera: Triozidae) is one of the primary vectors of the bacterium Candidatus Liberibacter spp. which causes citrus greening, a disease of global economic importance in citrus production. Despite its economic importance, little is known about its chemical ecology. Here, we used behavioral assays and chemical analysis to study the chemical basis of interaction between T. erytreae and one of its preferred host plants, Citrus jambhiri. In dual choice Y-tube olfactometer assays, lemon leaf odors attracted females but not males compared to

Dual-gRNA CRISPR/Cas9 Deletion of CsDMR6 in Sweet Orange Supported by Improved In Vitro Regeneration.

Huanglongbing (HLB), caused by Candidatus Liberibacter spp., remains the most destructive disease affecting citrus worldwide. To support host-directed genome-editing strategies aimed at reducing susceptibility, we optimized key regeneration steps in Citrus sinensis and validated a dual-gRNA CRISPR/Cas9 approach targeting the susceptibility gene CsDMR6. Juvenile explants of 'Valencia' and hybrid genotypes (CsH1-CsH3) were successfully established in vitro, and shoot elongation was markedly improved by supplementing Citrus Shoot Multiplication (CiSM) medium with 1 mg L-1 GA3. Ubiquitination-driven proteostasis remodeling underlies oxidative stress and actin degradation in citrus huanglongbing.

Citrus huanglongbing, caused by Candidatus Liberibacter asiaticus (CLas), is associated with severe metabolic disruption, chronic immune activation, and progressive decline of citrus trees, but the posttranslational mechanisms underlying these responses remain poorly understood. Here, we combined ubiquitinated peptide immunoaffinity enrichment with 4D label-free quantitative proteomics to profile protein abundance and protein ubiquitination in infected sweet orange leaves. Infection caused extensive proteome remodeling and a strong global increase in protein ubiquitination. Integrated analysis

Proteomic and transcriptomic analyses of saliva and salivary glands from the Asian citrus psyllid, Diaphorina citri.

Salivary secretions play critical roles in interactions among insects, insect-vectored pathogens, and host plants. The Asian citrus psyllid Diaphorina citri is a sap-sucking Hemipteran that serves as a vector for Candidatus Liberibacter asiaticus, the causal agent of citrus greening disease ("Huanglongbing" or HLB). D. citri continuously injects saliva into host plants using specialized stylets so as to feed and transmit the HLB pathogen. Knowledge on the composition and function of salivary proteins of this pest is very limited. In this study, proteomic and transcriptomic approaches were adop

Psyllid glycine-rich antimicrobial peptides oligomerize to directly cleave zinc transporter ZnuA on the membranes of citrus Huanglongbing pathogen.

INTRODUCTION: Huanglongbing (HLB), the most devastating citrus disease worldwide, is caused by Candidatus Liberibacter asiaticus (CLas) and transmitted by Asian citrus psyllid, Diaphorina citri. Currently, the potential role of the insect vector's innate immunity against CLas remains poorly understood. OBJECTIVES: This study aimed to identify and characterize a 41-amino acid glycine-rich antimicrobial peptide from D. citri (DcGrAMP1), elucidate its molecular mechanism against CLas and related α-proteobacteria, and evaluate its efficacy in suppressing CLas in HLB-affected citrus under greenhou

Dual targeting of pathogen load and callose deposition improves phloem function and carbon allocation in citrus greening disease.

Huanglongbing (HLB), caused by Candidatus Liberibacter asiaticus (CLas), disrupts sugar transport in citrus due to bacterial proliferation and excessive callose deposition that blocks sieve elements. Reducing the blockage of phloem may reduce symptoms, but it is unclear whether targeting CLas or suppressing callose deposition is more critical, as both contribute to phloem dysfunction. This study evaluated whether an integrated strategy targeting both CLas and callose-mediated phloem blockage could more effectively restore phloem transport, carbohydrate allocation, and productivity in HLB-affec

Physiological and Horticultural Responses of HLB-Affected 'Valencia' Orange Trees to Trunk-Injected Oxytetracycline and Streptomycin Formulations Differing in Adjuvant and pH.

BACKGROUND/OBJECTIVES: Trunk injection of oxytetracycline (OTC) can suppress Candidatus Liberibacter asiaticus (CLas) and improve the productivity of citrus trees affected by huanglongbing (HLB), a devastating bacterial disease, but conventional acidic formulations may cause phytotoxicity and exacerbate trunk injury. This study evaluated a proprietary adjuvant intended to stabilize OTC at higher pH and assessed streptomycin (STM) as an alternative or complement to OTC. METHODS: Two commercial field trials with HLB-affected 'Valencia' sweet orange trees compared a 2 × 4 factorial combination o

High susceptibility of Diaphorina citri first instar nymphs to Bacillus thuringiensis-derived pesticidal proteins from multiple structural groups.

The Asian citrus psyllid, Diaphorina citri, is the primary vector of Candidatus Liberibacter asiaticus, the causative agent of citrus greening disease, which continues to threaten citrus production worldwide. Current management relies heavily on chemical insecticides, despite increasing concerns regarding resistance and environmental impacts. Because pathogen acquisition is most efficient during the nymphal stage, management strategies targeting immature psyllids may improve disease suppression. Although bacterial pesticidal proteins (BPP) have generally shown limited activity against D. citri

DNA Barcode Reference Library for the African Citrus Triozid, Trioza erytreae (Hemiptera: Triozidae): Vector of African Citrus Greening.

Citrus (Citrus spp.) production continues to decline in East Africa, particularly in Kenya and Tanzania, the two major producers in the region.
