A missense mutation in Grm6 reduces but does not eliminate mGluR6 expression or rod depolarizing bipolar cell function.

GRM6 encodes the metabotropic glutamate receptor 6 (mGluR6) used by retinal depolarizing bipolar cells (DBCs). Mutations in GRM6 lead to DBC dysfunction and underlie the human condition autosomal recessive complete congenital stationary night blindness. Mouse mutants for Grm6 are important models for this condition. Here we report a new Grm6 mutant, identified in an electroretinogram (ERG) screen of mice maintained at The Jackson Laboratory. The Grm6nob8 mouse has a reduced-amplitude b-wave component of the ERG, which reflects light-evoked DBC activity. Sequencing identified a missense mutatio

Glycation and receptors: coming of AGE in the midst of danger.

The formation of advanced glycation end products (AGEs), first described in food science, is driven by hyperglycaemia (diabetes mellitus) and the modifications on long-lived proteins (natural ageing); AGE formation is enhanced by oxidative stress and inflammation, which are factors that accompany high-AGE settings of diabetes mellitus and ageing. Exogenous tobacco, ultra-processed foods, pollutants and alcohol contribute to AGE formation. Evidence demonstrates higher concentrations of AGEs in the blood and tissues of patients with diabetes mellitus than in samples from healthy control individu

Rapid detection and simultaneous antibiotic susceptibility analysis of Yersinia pestis directly from clinical specimens by use of reporter phage.

Yersinia pestis is a tier 1 agent due to its contagious pneumopathogenicity, extremely rapid progression, and high mortality rate. As the disease is usually fatal without appropriate therapy, rapid detection from clinical matrices is critical to patient outcomes. We previously engineered the diagnostic phage ΦA1122 with luxAB to create a "light-tagged" reporter phage. ΦA1122::luxAB rapidly detects Y. pestis in pure culture and human serum by transducing a bioluminescent signal response. In this report, we assessed the analytical specificity of the reporter phage and investigated diagnostic uti

Insect phototransduction: illuminating pathways to precision Pest management.

Insects rely on their visual systems to perform critical behaviors such as host location and phototaxis, and phototransduction, the conversion of photons into electrical signals, constitutes the core mechanism underlying visual function. Most research focuses on model insects like fruit flies, while common agricultural pests remain underexplored. This review integrates current knowledge of insect visual phototransduction to identify molecular and neural targets for precision pest management. This review systematically outlines the complete phototransduction pathway in insect vision: compound e

Aging and Light Stress Result in Overlapping and Unique Gene Expression Changes in Photoreceptors.

Advanced age is one of the leading risk factors for vision loss and eye disease. Photoreceptors are the primary sensory neurons of the eye. The extended photoreceptor cell lifespan, in addition to its high metabolic needs due to phototransduction, makes it critical for these neurons to continually respond to the stresses associated with aging by mounting an appropriate gene expression response. Here, we sought to untangle the more general neuronal age-dependent transcriptional signature of photoreceptors with that induced by light stress. To do this, we aged flies or exposed them to various du

Light control of the duration of the daily melatonin signal under long and short days in the Soay ram. Role of inhibition and entrainment.

Light acts in two ways to control the duration of the nocturnal melatonin rhythm. It inhibits the production of melatonin from the pineal gland and it entrains the underlying circadian rhythm generators located in the suprachiasmatic nuclei. To investigate the role of these two mechanisms under long and short days, four experiments were carried out using groups of adult Soay rams (n = 6-8). The animals were housed in individual pens in light-controlled rooms and entrained to long (LD 16:8) or short (LD 8:16) days for at least 8 wk. The treatments were as follows: (i) dark period extended by 4

Possible neuroprotective mechanism of human neuroglobin.

Neuroglobin (Ngb) is a newly discovered hexacoordinate globin that is expressed in vertebrate brain and can reversibly bind oxygen. Expression of Ngb increases in response to oxygen deprivation and protects neurons from hypoxia in vitro and in vivo. Recent work on human Ngb has shed light on the mechanism of this neuroprotection by human Ngb, as discussed in this review. Human ferric Ngb has been found to act as a guanine nucleotide dissociation inhibitor for the alpha subunit of heterotrimeric G proteins. Moreover, other Ngb-binding proteins also have been identified. These findings suggest t

Characterization of postsynaptic glutamate transporter functionality in the zebrafish retinal first synapse across different wavelengths.

In the zebrafish retina, incident light undergoes wavelength-dependent processing encompassing mechanisms such as color opponency, contrast enhancement, and motion detection prior to neural transmission to the brain proper. In darkness, photoreceptors continuously release glutamate into the synaptic cleft, a process that diminishes in response to increased light intensity, thereby conveying visual signals to ON and OFF bipolar cells. Specifically, in zebrafish, the ON pathway signal transduction is mediated by metabotropic glutamate receptor 6b (mGluR6b) and Excitatory Amino Acid Transporters

Calcium signaling and genetic rare diseases: An auditory perspective.

Deafness is a highly heterogeneous disorder which stems, for 50%, from genetic origins. Sensory transduction relies mainly on sensory hair cells of the cochlea, in the inner ear. Calcium is key for the function of these cells and acts as a fundamental signal transduction. Its homeostasis depends on three factors: the calcium influx, through the mechanotransduction channel at the apical pole of the hair cell as well as the voltage-gated calcium channel at the base of the cells; the calcium buffering via Ca2+-binding proteins in the cytoplasm, but also in organelles such as mitochondria and the

Mel1b and Mel1c melatonin receptors mediate green light-induced secretion of growth hormone in chick adenohypophysis cells via the AC/PKA and ERK1/2 signalling pathways.

A previous study showed that melatonin (MEL) membrane receptors 1b (Mel1b) and Mel1c promoted the secretion of growth hormone (GH) in chick adenohypophysis cells under monochromatic green light. However, the intracellular signalling pathways of these two receptors are unclear. Therefore, cultured adenohypophysis cells derived from chickens exposed to monochromatic green light were treated with MEL, Mel1b- and Mel1c-specific blockers, protein kinase A (PKA) inhibitors and adenylate cyclase (AC), or AC activator in vitro to explore the signal transduction mechanism that promote the secretion of

Cell-nonautonomous function of ceramidase in photoreceptor homeostasis.

Neutral ceramidase, a key enzyme of sphingolipid metabolism, hydrolyzes ceramide to sphingosine. These sphingolipids are critical structural components of cell membranes and act as second messengers in diverse signal transduction cascades. Here, we have isolated and characterized functional null mutants of Drosophila ceramidase. We show that secreted ceramidase functions in a cell-nonautonomous manner to maintain photoreceptor homeostasis. In the absence of ceramidase, photoreceptors degenerate in a light-dependent manner, are defective in normal endocytic turnover of rhodopsin, and do not res

Light acclimation, retrograde signalling, cell death and immune defences in plants.

This review confronts the classical view of plant immune defence and light acclimation with recently published data. Earlier findings have linked plant immune defences to nucleotide-binding site leucine-rich repeat (NBS-LRR)-dependent recognition of pathogen effectors and to the role of plasma membrane-localized NADPH-dependent oxidoreductase (AtRbohD), reactive oxygen species (ROS) and salicylic acid (SA). However, recent results suggest that plant immune defence also depends on the absorption of excessive light energy and photorespiration. Rapid changes in light intensity and quality often c

A role for the ciliary marginal zone in the melanopsin-dependent intrinsic pupillary light reflex.

Maintenance of pupillary constriction in light-adapted rodents has traditionally been thought to involve a reflex between retina, brain and iris, with recent work identifying the melanopsin-expressing intrinsically photosensitive retinal ganglion cells (ipRGCs) as the major conduits for retinal input to the brain. There is also a less well-understood phenomenon whereby the iris of some mammals, including mice, will constrict to light when either the eye, or the iris itself is physically isolated from the brain. The intrinsic pupillary light reflex (iPLR) is the term given to pupil constriction

Cnidocyte discharge is regulated by light and opsin-mediated phototransduction.

BACKGROUND: Cnidocytes, the eponymous cell type of the Cnidaria, facilitate both sensory and secretory functions and are among the most complex animal cell types known. In addition to their structural complexity, cnidocytes display complex sensory attributes, integrating both chemical and mechanical cues from the environment into their discharge behavior. Despite more than a century of work aimed at understanding the sensory biology of cnidocytes, the specific sensory receptor genes that regulate their function remain unknown. RESULTS: Here we report that light also regulates cnidocyte functi

Photoactuated Micropumps Enable Oscillatory Fluidics and Autonomous Locomotion.

Oscillatory fluid flows underpin transport, mixing, and signal propagation in living systems, yet artificial microfluidic platforms rely almost extensively on external power or chemically driven gradients to reproduce such dynamics. Here, we introduce a fuel-free strategy based on photo-mechanical transduction, in which light is directly converted into mechanical deformation of soft matter to generate reversible and oscillatory fluid flow without gradients. We demonstrated that azobenzene-doped liquid crystal films convert photochemical isomerization into amplified contractile-expansive motion

Neuronal regulation of photo-induced pineal photoreceptor proteins in carp Catla catla.

In the present in vitro study on the pineal in carp Catla catla, specific agonist and antagonists of receptors for different neuronal signals and regulators of intra-cellular Ca(++) and cAMP were used to gather basic information on the neuronal signal transduction cascade mechanisms in the photo-induced expression of rod-like opsin and alpha-transducin-like proteins in any fish pineal. Western-blot analysis followed by quantitative analysis of respective immunoblot data for both the proteins revealed that photo-induced expression of each protein was stimulated by cholinergic (both nicotinic an

Imaging of jasmonic acid binding sites in tissue.

Hormones regulate the mechanism of plant growth and development, senescence, and plants' adaptation to the environment; studies of the molecular mechanisms of plant hormone action are necessary for the understanding of these complex phenomena. However, there is no measurable signal for the hormone signal transduction process. We synthesized and applied a quantum dot-based fluorescent probe for the labeling of jasmonic acid (JA) binding sites in plants. This labeling probe was obtained by coupling mercaptoethylamine-modified CdTe quantum dots with JA using N-hydroxysuccinimide (NHS) as a coupli

TRP2: a candidate transduction channel for mammalian pheromone sensory signaling.

The vomeronasal organ (VNO) of terrestrial vertebrates plays a key role in the detection of pheromones, chemicals released by animals that elicit stereotyped sexual and aggressive behaviors among conspecifics. Sensory transduction in the VNO appears unrelated to that in the vertebrate olfactory and visual systems: the putative pheromone receptors of the VNO are evolutionarily independent from the odorant receptors and, in contrast to vertebrate visual and olfactory transduction, vomeronasal transduction is unlikely to be mediated by cyclic-nucleotide-gated channels. We hypothesized that sensor

Ultraviolet radiation-induced apoptosis in keratinocytes: on the role of cytosolic factors.

Epidemiological and experimental evidences have established solar ultraviolet (UV) radiation as the leading cause of skin cancers. Specifically, the frequency of non-melanoma skin cancer, one of the malignancies with the most rapidly increasing incidence, is directly related to the total exposure to solar UV light. As part of a general effort to elucidate the components of cellular signal transduction pathways, the mechanisms of cellular responses to UV radiation have received considerable attention over the last few years. These efforts were driven mainly by the conviction that understanding
