PDGFRβ in Stem Cell Microenvironments: Cross-Tissue Evidence and Hypotheses for the Limbal Stem Cell Niche.

Stem cells reside within specialized microenvironments, termed niches, composed of vascular networks, stromal cells, extracellular matrix (ECM), immune components, and neural elements, which collectively regulate quiescence, self-renewal, and lineage commitment of stem cells. Platelet-derived growth factor receptor-β (PDGFRβ), a class III receptor tyrosine kinase (RTK) predominantly expressed in perivascular and mesenchymal stromal cells, has been associated with vascular-stromal organization across multiple stem cell systems. Rather than acting mainly as a direct determinant of stemness-assoc

Therapeutic effects of adipose mesenchymal stem cell-derived exosomes on limbal stem cells deficiency in cats.

Corneal transparency is maintained by the limbus and its stem cells, and damage or deficiency in this region leads to conjunctivalization and subsequent vision impairment. This study aims to evaluate the therapeutic effects of feline adipose mesenchymal stem cell-derived exosomes (fAD-MSCExo) in the treatment of limbal stem cell deficiency in cats. For this purpose, 30 animals presenting with severe conjunctivalization were included in the study. Cats were randomly assigned to two groups: Group 1 (n = 15), serving as the control group and receiving steroid treatment, Group 2 (n = 15), receivin

In vitro generation of pancreatic endocrine cells from human adult fibroblast-like limbal stem cells.

Stem cells might provide unlimited supply of transplantable cells for β-cell replacement therapy in diabetes. The human limbus is a highly specialized region hosting a well-recognized population of epithelial stem cells, which sustain the continuous renewal of the cornea, and the recently identified stromal fibroblast-like stem cells (f-LSCs), with apparent broader plasticity. However, the lack of specific molecular markers for the identification of the multipotent limbal subpopulation has so far limited the investigation of their differentiation potential. In this study we show that the human

Optimizing outcomes in keratolimbal allograft for limbal stem cell deficiency.

PURPOSE OF REVIEW: The present review will focus on recent advancements affecting outcomes in keratolimbal allograft (KLAL). RECENT FINDINGS: Recent advancements involve methodologies for donor selection, eye bank preparation of KLAL tissue, risk stratification, and immunosuppression to prevent both early and late immunologic rejection. SUMMARY: Keratolimbal allograft (KLAL) remains an essential technique for addressing severe limbal stem cell deficiency in patients without access to a suitable living donor eye (whether autologous or allogeneic). While outcomes of KLAL have traditionally lag

A Review of Contact Lens-Induced Limbal Stem Cell Deficiency.

Limbal stem cell deficiency (LSCD) is a pathologic condition caused by the dysfunction and destruction of stem cells, stem cell precursors and limbal cell niche in the corneal epithelium, leading to severe conjunctivalization of the cornea. Etiologies for LSCD span from congenital (aniridia), traumatic (chemical or thermal injuries), autoimmune (Stevens-Johnson syndrome) and iatrogenic disease to contact lens (CL) wear. Of these, CL wear is the least understood and is often a subclinical cause of LSCD. Even with recent advances in LSCD research, limitations persist in establishing the pathogen

3D printed biomimetic bilayer limbal implants for regeneration of the corneal structure in limbal stem cell deficiency.

Limbal stem cell deficiency (LSCD) causes vision loss and is often treated by simple corneal epithelial cell transplantation with poor long-term efficiency. Here, we present a biomimetic bilayer limbal implant using digital light processing 3D printing technology with gelatin methacrylate (GelMA) and poly (ethylene glycol) diacrylate (PEGDA) bioinks containing corneal epithelial cells (CECs) and corneal stromal stem cells (CSSCs), which can transplant CECs and improve the limbal niche simultaneously. The GelMA/PEGDA hydrogel possessed robust mechanical properties to support surgical transplant

Reconsidering pterygium in the context of focal limbal stem cell dysfunction.

Limbal stem cell deficiency (LSCD) is traditionally regarded as a rare ocular surface disorder characterised by impaired corneal epithelial regeneration. Emerging evidence suggests that pterygium, a common fibrovascular lesion extending onto the cornea, may involve focal limbal epithelial or niche dysfunction. This narrative review examines clinical, histopathological and biological evidence relevant to the relationship between pterygium and focal limbal dysfunction. The peripheral light focusing hypothesis provides a biologically plausible explanation for the characteristic nasal predilection

Limbal stem cell deficiency: a dreaded complication of chickenpox in children.

We report a rare case of Limbal Stem Cell Deficiency (LSCD) in a 10-year-old child following chickenpox. The child presented with persistent ocular symptoms, including pain, redness, and photophobia, two months after the resolution of systemic chickenpox. Ophthalmic examination revealed conjunctival hyperemia, 360° conjunctivalization, corneal neovascularization, and an epithelial defect. LSCD, characterized by corneal neovascularization and conjunctivalization, can occur after viral infections, including herpes zoster. To our knowledge, this is the first reported case of LSCD following chicke

[Research progress on imagine diagnosis of limbal stem cell deficiency].

Limbal stem cell deficiency (LSCD) is an ocular surface disease resulting from a reduction and/or dysfunction of limbal stem cells. The symptoms of LSCD are non-specific and can be difficult to distinguish from other ocular surface diseases through slit-lamp examination. Impression cytology is currently considered the gold standard for LSCD diagnosis; however,it is a qualitative method with low sensitivity. Nonetheless, emerging imaging techniques offer quantitative diagnosis and staging of LSCD. This review article examines four imaging methods and their associated parameters for diagnosing LS
