Tunable linear feedback control of urban drainage systems using models defined purely from data.

Real-time and model-predictive control promises to make urban drainage systems (UDS) adaptive, coordinated, and dynamically optimal. Though early implementations are promising, existing control algorithms have drawbacks in computational expense, trust, system-level coordination, and labor cost. Linear feedback control has distinct advantages in computational expense, interpretation, and coordination. However, current methods for building linear feedback controllers require calibrated software models. Here we present an automated method for generating tunable linear feedback controllers that re

Neuronal population models reveal specific linear conductance controllers sufficient to rescue preclinical disease phenotypes.

Preclinical drug candidates are screened for their ability to ameliorate in vitro neuronal electrophysiology, and go/no-go decisions progress drugs to clinical trials based on population means across cells and animals. However, these measures do not mitigate clinical endpoint risk. Population-based modeling captures variability across multiple electrophysiological measures from healthy, disease, and drug phenotypes. We pursued optimizing therapeutic targets by identifying coherent sets of ion channel target modulations for recovering heterogeneous wild-type (WT) population excitability profile

Combining Threshold, Thurstonian and Classical Linear Models in Horse Genetic Evaluations for Endurance Competitions.

The racing time and rank at finish traits are commonly used for endurance horse breeding programs as a measure of their performance. Even so, given the nature of endurance competitions, many horses do not finish the race. However, the exclusion of non placed horses from the dataset could have an influence on the prediction of individual breeding values. The objective of the present paper was to develop a multitrait model including race time (T), rank (R) and placing (P), with different methodologies, to improve the genetic evaluation in endurance competitions in Spain. The database contained 6

Fitting a linear-linear piecewise growth mixture model with unknown knots: A comparison of two common approaches to inference.

A linear-linear piecewise growth mixture model (PGMM) is appropriate for analyzing segmented (disjointed) change in individual behavior over time, where the data come from a mixture of 2 or more latent classes, and the underlying growth trajectories in the different segments of the developmental process within each latent class are linear. A PGMM allows the knot (change point), the time of transition from 1 phase (segment) to another, to be estimated (when it is not known a priori) along with the other model parameters. To assist researchers in deciding which estimation method is most advantag

Comparison of linear and threshold models for genetic evaluation of morphological defects in Nellore cattle.

Morphological defects in beef cattle can compromise animal welfare and productivity, yet they remain underexplored in genetic evaluations. In this study, we assessed the prevalence and estimated genetic parameters for seven morphological defects in Nellore cattle, including depigmentation, feet and legs malformation, chamfer deviations, loin and jaw defects, hump irregularities, and navel abnormalities, using linear and threshold models. Data from over 180,000 animals recorded between 1998 and 2021 were analyzed. Defect prevalence increased over time, likely due to improved phenotyping and bro

FLASH-MM: fast and scalable single-cell differential expression analysis using linear mixed-effects models.

Single-cell RNA sequencing (scRNA-seq) enables detailed comparisons of gene expression across cells and conditions. Single-cell differential expression analysis faces challenges like sample correlation, individual variation, and scalability. We develop a fast and scalable linear mixed-effects model (LMM) estimation algorithm, FLASH-MM, to address these issues. We reformulate aspects of the linear mixed model estimation procedure to make it faster, by reducing computational complexity and memory usage. Simulation studies with scRNA-seq data show that FLASH-MM is accurate, computationally effici

A lossless one-shot distributed algorithm for addressing heterogeneity in multi-site generalized linear models.

OBJECTIVE: We propose Heterogeneity-aware Collaborative One-shot Lossless Algorithm for Generalized Linear Model (COLA-GLM-H), a novel one-shot lossless distributed algorithm that enables the integration of heterogeneous multi-institutional data while relying solely on instituion-level summary information rather than patient-level data. MATERIALS AND METHODS: Generalized Linear Models (GLMs) are widely used in medical research for analyzing diverse outcome types. In multi-institution settings, we demonstrated that the global likelihood can be reconstructed using only institution-level summary

Enhancing the Accuracy of Linear Finite Element Models of Vehicle Structures Considering Spot-Welded Flanges.

Structural engineering simulations have required increasingly complex computational models to replace physical tests accurately. This work focuses on the numerical analysis of vehicle body structures, whose size and complexity make the use of very accurate nonlinear models unfeasible due to the prohibitive computational costs involved. The purpose of this study is to find a new approach to model spot-welded joints in linear finite element models of thin-wall vehicle body structures, improving the accuracy of the model without increasing its complexity. Using a set of simplified nonlinear model

Different approaches to characterize artificial breeding sites of Aedes aegypti using generalized linear mixed models.

BACKGROUND: As no globally accepted dengue vaccines or specific antiviral therapies are currently available, controlling breeding sites of Aedes aegypti is a target to prevent dengue outbreaks. The present study aimed to characterize outdoor artificial breeding sites in urban households using an exhaustive classification system. METHODS: A cross-sectional entomological survey was carried out in Colón city, Entre Ríos, Argentina, using a two-stage stratified sampling design during March and April 2014. The city was stratified given the degree of urbanization of each block, and blocks and house

Estimating the reliability of repeatedly measured endpoints based on linear mixed-effects models. A tutorial.

There are various settings in which researchers are interested in the assessment of the correlation between repeated measurements that are taken within the same subject (i.e., reliability). For example, the same rating scale may be used to assess the symptom severity of the same patients by multiple physicians, or the same outcome may be measured repeatedly over time in the same patients. Reliability can be estimated in various ways, for example, using the classical Pearson correlation or the intra-class correlation in clustered data. However, contemporary data often have a complex structure t

Enlarged colony housing promotes linear progression of subchondral bone remodeling in joint instability rat models.

Objective: Osteoarthritis (OA) is a disease with high prevalence and an unmet medical need for disease modifying treatments. In rat models, OA-like subchondral bone and cartilage changes can be induced by instability surgery with different severity levels. Factors which determine structural changes additionally comprise the study duration and activity-impacted joint loading. Methods: A medial meniscal tear (MMT) or anterior cruciate ligament transection with partial meniscectomy (ACLT+pMx) was induced unilaterally in rats housed in a rat colony cage (RCC), allowing high activity levels includi

DLMM as a lossless one-shot algorithm for collaborative multi-site distributed linear mixed models.

Linear mixed models are commonly used in healthcare-based association analyses for analyzing multi-site data with heterogeneous site-specific random effects. Due to regulations for protecting patients' privacy, sensitive individual patient data (IPD) typically cannot be shared across sites. We propose an algorithm for fitting distributed linear mixed models (DLMMs) without sharing IPD across sites. This algorithm achieves results identical to those achieved using pooled IPD from multiple sites (i.e., the same effect size and standard error estimates), hence demonstrating the lossless property.

Multi-disease analysis of maternal antibody decay using non-linear mixed models accounting for censoring.

Biomedical studies often generate repeated measures of multiple outcomes on a set of subjects. It may be of interest to develop a biologically intuitive model for the joint evolution of these outcomes while assessing inter-subject heterogeneity. Even though it is common for biological processes to entail non-linear relationships, examples of multivariate non-linear mixed models (MNMMs) are still fairly rare. We contribute to this area by jointly analyzing the maternal antibody decay for measles, mumps, rubella, and varicella, allowing for a different non-linear decay model for each infectious

Human cornea thermo-viscoelastic behavior modelling using standard linear solid model.

BACKGROUND: Corneal biomechanics is of great interest to researchers recently. Clinical findings relate them to corneal diseases and to outcomes of refractive surgery. To have a solid understanding of corneal diseases' progression, it is important to understand corneal biomechanics. Also, they are essential for better explaining outcomes of refractive surgeries and their undesired consequences. There is a difficulty for studying corneal biomechanics in-vivo and multiple limitations arise for ex-vivo studies. Hence mathematical modelling is considered as a proper solution to overcome such obsta

Investigating the wave profiles to the linear quadratic model in mathematical biology.

This study investigates the dynamic behavior of the linear quadratic model (LQM), a fundamental framework in radiation biology that describes cellular response to radiation, particularly in the context of DNA damage and cancer progression. The LQM was originally developed to quantify radiation-induced cell death and repair mechanisms, with a focus on double-stranded DNA breaks, the most critical type of radiation damage. Despite advances in tracking tumor cell dissemination, the mechanisms underlying cancer invasion remain poorly understood. Mathematical modeling, particularly through partial

Simultaneous variable selection and estimation for a partially linear Cox model.

We consider simultaneous variable selection and estimation for a deep neural network-based partially linear Cox model and propose a novel penalized approach. In particular, a two-step iterative algorithm is developed with the use of the minimum information criterion to ensure sparse estimation. The proposed method circumvents the curse of dimensionality while facilitating the interpretability of linear covariate effects on survival, and the algorithm greatly reduces the computational burden by avoiding the need to select the optimal tuning parameters that is usually required by many other popu

Use of the Linear No-threshold (LNT) Model in Radiological Protection: An Update.

The linear no-threshold (LNT) model was introduced into the radiological protection system by the International Commission on Radiological Protection (ICRP) in 1966. The appropriateness of this model is still hotly debated today. Based on a recently published article, we summarize recent results in radiobiology and epidemiology and discuss their impact on the use of the LNT model regarding radiological protection. The scientific results published in radiobiology and epidemiology have strengthened our scientific knowledge of cancer risks associated with low dose and/or low dose-rate radiation e
