c9, t11-Conjugated linoleic acid supplementation improves cognitive memory in ω-3 polyunsaturated fatty acid-deficient mice.

Long-chain polyunsaturated fatty acids (LC-PUFAs) are vital for brain health, with cis9, trans11-conjugated linoleic acid (c9, t11-CLA) showing neuroprotective effects. This study investigated the impact of c9, t11-CLA on offspring cognitive development in an ω-3 PUFA deficiency model. c9, t11-CLA supplementation during gestation and lactation improved episodic-like memory and learning ability, as evidenced by a 63% increase in novel object recognition time and reduced Morris water maze latency. c9, t11-CLA altered brain lipid profiles and promoted myelination in the prefrontal cortex of pubes

Oleic acid attenuates trans-10,cis-12 conjugated linoleic acid-mediated inflammatory gene expression in human adipocytes.

The weight loss supplement conjugated linoleic acid (CLA) consists of an equal mixture of trans-10,cis-12 (10,12) and cis-9,trans-11 (9,11) isomers. However, high levels of mixed CLA isomers, or the 10,12 isomer, causes chronic inflammation, lipodystrophy, or insulin resistance. We previously demonstrated that 10,12 CLA decreases de novo lipid synthesis along with the abundance and activity of stearoyl-CoA desaturase (SCD)-1, a δ-9 desaturase essential for the synthesis of monounsaturated fatty acids (MUFA). Thus, we hypothesized that the 10,12 CLA-mediated decrease in SCD-1, with the subseque

Plasma Linoleic Acid Is Associated With Pediatric Sepsis Phenotype and Acute Kidney Injury.

OBJECTIVES: Linoleic acid (LA) is the most abundant polyunsaturated fatty acid in diet, and it is a precursor to inflammatory lipid mediators called oxylipins. The role of LA and its oxylipins in pediatric sepsis and organ injury is uncertain. Recently, pediatric sepsis phenotypes were described, with phenotype D characterized by the highest proportion of acute kidney injury (AKI), multiple organ failure, and risk of death. We aimed to test the hypothesis LA may play a role in sepsis-associated organ dysfunction. We therefore investigated whether increasing plasma LA and LA-derived lipoxygenas

Linoleic Acid Potentiates Response to Chemotherapy in Biliary Tract Cancer Through RARγ Activation.

Biliary tract cancer (BTC) remains highly challenging owing to its poor prognosis. Although gemcitabine plus cisplatin (Gem/Cis) represents a standard chemotherapy option, treatment resistance significantly limits the outcomes. Metabolic reprogramming influences therapeutic response; however, the specific metabolic determinants of chemosensitivity in BTC are poorly defined. Using untargeted metabolomics followed by targeted validation, we found that plasma linoleic acid (LA) levels were significantly elevated in patients with chemotherapy-responsive BTC. LA supplementation led to tumor accumul

Olive oil and linoleic acid as potential acaricidal agents against Psoroptes cuniculi mites.

Psoroptes cuniculi mite is an important ectoparasite in rabbit breeding. Extra virgin olive oil (EVOO) is a vegetable oil with anti-inflammatory and regenerative properties, and linoleic acid (LA) also has beneficial properties for health. The activity against P. cuniculi mites of EVOO was evaluated at concentrations of 1, 5, 10, 30, 60 and 100 %; LA was evaluated at 50, 100, 150, 200, 300 and 400 mg/mL. The effectiveness was evaluated by the immersion technique using 11 mites and four replicates, and post-treatment mortality was analyzed after 24 and 48 h. Also, the composition of EVOO was de

Estimating the therapeutic potential of NSAIDs and linoleic acid-isomers supplementation against neuroinflammation.

Nonsteroidal anti-inflammatory drugs (NSAIDs) regulate inflammation, which is associated with their role in preventing neurodegenerative diseases in epidemiological studies. It has sparked interest in their unconventional application for reducing neuroinflammation, opening up new avenues in biomedical research. However, given the pharmacological drawbacks of NSAIDs, the development of formulations with naturally antioxidant/anti-inflammatory dietary fatty acids has been demonstrated to be advantageous for the clinical translation of anti-inflammatory-based therapies. It includes improved blood

SARS-CoV-2 spike variants differ in their allosteric responses to linoleic acid.

The SARS-CoV-2 spike protein contains a functionally important fatty acid (FA) binding site, which is also found in some other coronaviruses, e.g. SARS-CoV and MERS-CoV. The occupancy of the FA site by linoleic acid (LA) reduces infectivity by 'locking' the spike in a less infectious conformation. Here, we use dynamical-nonequilibrium molecular dynamics (D-NEMD) simulations to compare the allosteric responses of spike variants to LA removal. D-NEMD simulations show that the FA site is coupled to other functional regions of the protein, e.g. the receptor-binding motif (RBM), N-terminal domain (

Activation of Hair Cell Growth Factors by Linoleic Acid in Malva verticillata Seed.

Hair loss by excessive stress from work and lifestyle changes has become a growing concern, particularly among young individuals. However, most drugs for alopecia impose a plethora of side effects. We have found the powerful impact of Malva verticillata seed extracts on alleviating hair loss. This study further isolated effective chemicals in M. verticillata seed extracts by liquid silica gel column chromatography. Under the screening for the growth rate (%) of human follicles dermal papilla cells (HFDPCs), we identified linoleic acid (LA) and oleic acid in n-hexane of M. verticillate (MH)2 fr

Feedback activation of ferrous 5-lipoxygenase during leukotriene synthesis by coexisting linoleic acid.

Ferrous lipoxygenases seem to be activated through a feedback control mechanism via FA hydroperoxides generated from PUFAs by partially existing ferric lipoxygenases. However, during leukotriene synthesis, feedback activation of ferrous 5-lipoxygenase in the presence of arachidonic acid (AA) was not observed. In the present study, we examined the feedback activation of ferrous 5-lipoxygenase in the 5-lipoxygenase/AA system in the presence of linoleic aicd (LA), which is a predominant component of membrane phospholipids. When potato 5-lipoxygenase was incubated with AA and LA in the presence of

Gamma-linoleic acid and ascorbate improves skeletal ossification in offspring of diabetic rats.

Maternal diabetes causes a range of complications in offspring, including reduced skeletal ossification. This study examined whether feeding gamma-linoleic acid (GLA) and ascorbate, alone or in combination, to diabetic pregnant rats improves skeletal development in their offspring. In addition, Ca(2+) concentration was monitored in maternal plasma and fetal tissue, as well as placental mRNA expression of calbindin-D(9k). Female rats rendered diabetic with streptozotocin were fed GLA (500 mg/kg/d), ascorbate (290 mg/kg/d), ascorbyl-GLA (790 mg/kg/d), or GLA and ascorbate (500 and 290 mg/kg/d, r

The Safety of Nebulized Conjugated Linoleic Acid for COVID-19 Respiratory Tract Infections.

Linoleic acid is an essential fatty acid, and in 2007, the US FDA listed conjugated linoleic acid (CLA) isomers as generally recognized as safe (GRAS) for human consumption. The potassium salt of conjugated linoleic acid (K-CLA) exhibits broad-spectrum antimicrobial activity. K-CLA also possesses mucolytic properties. This study reviewed the safety of nebulized K-CLA for COVID-19-related respiratory tract infections early during the pandemic. Among 57 treated subjects, adverse events leading to treatment discontinuation occurred in three patients. Two reported an increased cough, and the other

An oxidized derivative of linoleic acid stimulates dehydroepiandrosterone production by human adrenal cells.

We previously reported that an oxidized derivative of linoleic acid stimulated steroidogenesis in rat adrenal cells. This derivative was also detected in human plasma, and was positively correlated with visceral adiposity and plasma DHEA-S. The present study sought to characterize the effects of this derivative, 12,13-epoxy-9-keto-(10- trans)-octadecenoic acid (EKODE), on steroid production by normal human adrenocortical cells obtained during clinically-indicated adrenalectomy. Cell suspensions were incubated in the presence of varying concentrations of EKODE and ACTH. EKODE (16 microM) signif

The omega-6 fatty acid linoleic acid is associated with risk of gastroschisis: a novel dietary risk factor.

Gastroschisis is a congenital abdominal wall defect, thought by many to represent a disruption in intrauterine blood flow, where there is herniation of abdominal organs. Dietary intake is an important environmental factor that has been implicated in the development of many diseases. Omega-6 polyunsaturated fatty acids (PUFAs) are nutrients that are substrates for eicosanoid and cytokine synthesis and prone to oxidation, and play a role in modulating inflammation, immune function, and vascular system development. This pilot case-control study explored the association of dietary intake of the om

The effect of conjugated linoleic acid on platelet function, platelet fatty acid composition, and blood coagulation in humans.

Despite extensive research on conjugated linoleic acid (CLA) showing multiple beneficial effects in animal models, little is known about the role of dietary CLA in human health. To investigate if the beneficial effects of CLA seen in animal models are relevant to humans, we conducted a study with 17 healthy female volunteers who lived in the Metabolic Research Unit of the Western Human Nutrition Research Center for 93 d. This paper reports only the results from this study that are related to the effects of CLA supplementation on blood coagulation, platelet function, and platelet fatty acid com

13-Hydroxyoctadecadienoic acid is the mitogenic signal for linoleic acid-dependent growth in rat hepatoma 7288CTC in vivo.

Growth of hepatoma 7288CTC in male Buffalo rats is directly dependent on uptake of linoleic acid (LA) from the arterial blood. One to 5% of the LA taken up is converted to 13-hydroxyoctadecadienoic acid (HODE), an agent that enhances epidermal growth factor-dependent mitogenesis. The role of 13-HODE in LA-dependent growth of solid tumors is not known. In this study, we examined LA uptake and 13-HODE formation on growth of tissue-isolated hepatoma 7288CTC in vivo and on [3H]thymidine incorporation and DNA content during perfusion in situ. Fatty acid uptake and metabolite release were determined
