Interaction of chromium(III) complexes with model lipid bilayers: implications on cellular uptake.

To understand molecular cytotoxicity of chromium(III) and how it affects the stability of biological membranes, studies on the interaction of chromium(III) complexes aquapentaminechromium complex (complex I) and trans- [Cr(5-methoxysalcyclohex) (H(2)O) (2)] ClO(4) (complex II) with model biomembranes have been carried out. Langmuir films of dimyristoylphosphatidylcholine (DMPC), dipalmitoylphosphatidic acid (DPPA), dioctadecyldimethylammoniumbromide (DOMA) at air/water interface interacting with the chromium(III) complexes have been characterized using the surface pressure-molecular area (π-A)

Islet amyloid polypeptide forms rigid lipid-protein amyloid fibrils on supported phospholipid bilayers.

Islet amyloid polypeptide (IAPP) forms fibrillar amyloid deposits in the pancreatic islets of Langerhans of patients with type 2 diabetes mellitus, and its misfolding and aggregation are thought to contribute to beta-cell death. Increasing evidence suggests that IAPP fibrillization is strongly influenced by lipid membranes and, vice versa, that the membrane architecture and integrity are severely affected by amyloid growth. Here, we report direct fluorescence microscopic observations of the morphological transformations accompanying IAPP fibrillization on the surface of supported lipid membran

Controlling spatial distribution of functional lipids in a supported lipid bilayer prepared from vesicles.

Conjugating biomolecules, such as antibodies, to bioconjugate moieties on lipid surfaces is a powerful tool for engineering the surface of diverse biomaterials, including cells and nanoparticles. We developed supported lipid bilayers (SLBs) presenting well-defined spatial distributions of functional moieties as models for precisely engineered functional biomolecular-lipid surfaces. We used quartz crystal microbalance with dissipation (QCM-D) and atomic force microscopy (AFM) to determine how vesicles containing a mixture of 1,2-dipalmitoyl-sn-glycero-3-phosphatidylcholine (DPPC) and 1,2-distea

Structural Analysis of Gel-Phase-Like Subdomains Formed in Lipid-Raft-Mimetic Bilayers.

Lipid rafts are ordered membrane domains composed of sphingomyelin (SM) and cholesterol (chol). They serve as platforms for membrane-based signal transduction. To date, the structural properties of lipid rafts have been investigated using SM/chol/unsaturated-lipid ternary bilayers, which undergo phase separation into SM/chol-rich liquid-ordered (Lo) and unsaturated-lipid-rich liquid-disordered (Ld) phases. Although some previous studies have suggested structural heterogeneity within the Lo phase, the internal organization of lipid-raft-mimetic Lo phases has been largely unexplored owing to the

Influence of Substrate on Supported Lipid Bilayers: Membrane Adhesion, Stretching, Pores, and Remodeling.

Model membranes offer a simplified representation of biomembranes, allowing for controlled structural and functional studies without the inherent complexity of biological systems. Due to the confined space and strong interactions with the support, supported lipid bilayers (SLBs) exhibit markedly distinct properties compared to those of their free-standing counterparts. However, the nano- and microscale origins of these differences, structural effects, and the related biophysical consequences remain incompletely understood. Using a combination of spectroscopy and micromanipulation techniques, w

Insights into the interactions of zinc-phthalocyanines with lipid bilayers for liposomal formulations.

Phthalocyanines (Pcs) are promising photosensitizers for photodynamic therapy (PDT). Inclusion into liposomes helps increase their bioavailability due to their hydrophobic nature. In order to elucidate the molecular interactions between Pcs and lipids employed in liposomal formulations, comprehensive computational simulation studies of zinc-phthalocyanine (ZnPc) and its tetraamino derivative (TaZnPc) were carried out. Initially, unbiased molecular dynamics (MD) simulations were performed. It was found that both Pcs freely insert into the pure DPPC bilayer in the liquid phase (55 °C). However,

Dantrolene inhibition of ryanodine channels (RyR2) in artificial lipid bilayers depends on FKBP12.6.

Dantrolene is a neutral hydantoin that is clinically used as a skeletal muscle relaxant to prevent overactivation of the skeletal muscle calcium release channel (RyR1) in response to volatile anesthetics. Dantrolene has aroused considerable recent interest as a lead compound for stabilizing calcium release due to overactive cardiac calcium release channels (RyR2) in heart failure. Previously, we found that dantrolene produces up to a 45% inhibition RyR2 with an IC50 of 160 nM, and that this inhibition requires the physiological association between RyR2 and CaM. In this study, we tested the hyp

Structure and dynamics of the drug-bound bacterial transporter EmrE in lipid bilayers.

The dimeric transporter, EmrE, effluxes polyaromatic cationic drugs in a proton-coupled manner to confer multidrug resistance in bacteria. Although the protein is known to adopt an antiparallel asymmetric topology, its high-resolution drug-bound structure is so far unknown, limiting our understanding of the molecular basis of promiscuous transport. Here we report an experimental structure of drug-bound EmrE in phospholipid bilayers, determined using 19F and 1H solid-state NMR and a fluorinated substrate, tetra(4-fluorophenyl) phosphonium (F4-TPP+). The drug-binding site, constrained by 214 pro

The Transmembrane Conformation of the Influenza B Virus M2 Protein in Lipid Bilayers.

Influenza A and B viruses cause seasonal flu epidemics. The M2 protein of influenza B (BM2) is a membrane-embedded tetrameric proton channel that is essential for the viral lifecycle. BM2 is a functional analog of AM2 but shares only 24% sequence identity for the transmembrane (TM) domain. The structure and function of AM2, which is targeted by two antiviral drugs, have been well characterized. In comparison, much less is known about the structure of BM2 and no drug is so far available to inhibit this protein. Here we use solid-state NMR spectroscopy to investigate the conformation of BM2(1-51

Structural Insights into the Yersinia pestis Outer Membrane Protein Ail in Lipid Bilayers.

Yersinia pestis the causative agent of plague, is highly pathogenic and poses very high risk to public health. The outer membrane protein Ail (Adhesion invasion locus) is one of the most highly expressed proteins on the cell surface of Y. pestis, and a major target for the development of medical countermeasures. Ail is essential for microbial virulence and is critical for promoting the survival of Y. pestis in serum. Structures of Ail have been determined by X-ray diffraction and solution NMR spectroscopy, but the protein's activity is influenced by the detergents in these samples, underscorin

Functional analysis of Kv1.2 and paddle chimera Kv channels in planar lipid bilayers.

Voltage-dependent K(+) (Kv) channels play key roles in shaping electrical signaling in both excitable and nonexcitable cells. These channels open and close in response to the voltage changes across the cell membrane. Many studies have been carried out in order to understand the voltage-sensing mechanism. Our laboratory recently determined the atomic structures of a mammalian Kv channel Kv1.2 and a mutant of Kv1.2 named the 'paddle chimera' channel, in which the voltage sensor paddle was transferred from Kv2.1 to Kv1.2. These two structures provide atomic descriptions of voltage-dependent chann

Surfactant effects of chlorpromazine and imipramine on lipid bilayers containing sphingomyelin and cholesterol.

The surface-active drugs chlorpromazine (CPZ) and imipramine (IP) have been tested on large unilamellar vesicles composed of phosphatidylcholine (PC), sphingomyelin (SM), and cholesterol (Ch) in different proportions. The well-characterized nonionic detergent Triton X-100 (TX) has also been used in parallel experiments. Leakage of vesicular aqueous contents and bilayer solubilization have been measured for each surfactant molecule and vesicle composition. All three surface-active molecules behave in a qualitatively similar way, irrespective of bilayer composition: they induce leakage at concen

ATP-sensitive K+ channels from aortic smooth muscle incorporated into planar lipid bilayers.

Glibenclamide binding sites were identified in a membrane preparation from canine aortic smooth muscle. The dissociation constant for [3H]glibenclamide binding was 10 +/- 2 nM, with a density of 420 +/- 108 fmol/mg protein. The properties of ATP-sensitive potassium (KATP) channels from the same membrane preparation incorporated into planar lipid bilayers were investigated. ATP was a potent inhibitor of the channels with half-maximal inhibition of channel activity by 41 microM ATP. Glibenclamide inhibited channel activity, and cromakalim activated the channel in the presence of ATP. Blockers of

Sodium-induced lipid bilayer phase transition for an ester lipid and phase transition resistance for an ether branched-lipid.

Environmental factors such as temperature and ion concentration can induce the phase transition from the liquid-crystalline to gel phase. In our work, we investigate the effect of temperatures and NaCl concentrations on phase transitions for two model lipids, i.e., an ether linked lipid with methyl branches (phosphatidyl glycerophosphate methyl ester, PGP-Me) and a more common ester phospholipid (dimyristoylphosphatidylcholine, DMPC). At 5.0 M NaCl 30 °C, the temperature closest to DMPC's phase transition temperature (Tm) in pure water, DMPC experiences a phase transition, but PGP-Me does not.

Influences of Vibrio cholerae Lipid A Types on LPS Bilayer Properties.

Lipopolysaccharides (LPS) present in the outer leaflet of Gram-negative bacterial outer membranes protect the bacteria from external threats and influence antibiotic permeability as well as immune system recognition. The structure of lipid A, the anchor of an LPS molecule to the outer membrane, can make direct influences on membrane properties. Particularly, in Vibrio cholerae, a Gram-negative bacterium responsible for cholera, a severe diarrheal disease, modifications of lipid A structures grant antibiotic resistance and are a primary factor that led to the current cholera pandemic. However,

Lipid-Bilayer Dynamics Probed by a Carbon Dot-Phospholipid Conjugate.

Elucidating the dynamic properties of membranes is important for understanding fundamental cellular processes and for shedding light on the interactions of proteins, drugs, and viruses with the cell surface. Dynamic studies of lipid bilayers have been constrained, however, by the relatively small number of pertinent molecular probes and the limited physicochemical properties of the probes. We show that a lipid conjugate comprised of a fluorescent carbon dot (C-dot) covalently attached to a phospholipid constitutes a versatile and effective vehicle for studying bilayer dynamics. The C-dot-modif

Anthrax toxin-induced rupture of artificial lipid bilayer membranes.

We demonstrate experimentally that anthrax toxin complexes rupture artificial lipid bilayer membranes when isolated from the blood of infected animals. When the solution pH is temporally acidified to mimic that process in endosomes, recombinant anthrax toxin forms an irreversibly bound complex, which also destabilizes membranes. The results suggest an alternative mechanism for the translocation of anthrax toxin into the cytoplasm.

Characterization of proton transport across a waveguide-supported lipid bilayer.

Cellular energy transduction processes are often driven by transmembrane ion gradients, and numerous artificial biomembrane systems have been developed that allow for chemically or light-induced charge transport into/out of liposomes. Liposomal architectures, however, are not readily interfaced to a solid-state transducer. Formation of an ion gradient across a planar-supported membrane, "wired" to a substrate electrode, may ultimately allow utilization of the potential energy to drive other electrochemical processes. Described here is a novel conductive polymer/planar waveguide assembly that p
