Lipogenesis-driven EGFR palmitoylation enables metastatic immune evasion in triple-negative breast cancer.

Metastasis is the leading cause of death in triple-negative breast cancer (TNBC), yet how tumor cells evade immune recognition during dissemination remain unclear. Here, we show that de novo lipogenesis drives metastatic immune evasion through FASN-mediated palmitoylation of EGFR. This modification establishes a lipid-dependent membrane signaling scaffold that sustains PI3K-AKT-mTOR signaling independently of MAPK compensation, thereby suppressing MHC-I antigen presentation through post-translational regulation. As a result, metastatic-competent cells escape CD8+ T cell surveillance and coloni

GCN5 drives MASLD progression through LXRα/SREBP1c signaling pathway-mediated de novo lipogenesis.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a global health concern that affects nearly one-quarter of the world's population. General control non-repressed protein 5 (GCN5), a histone acetyltransferase (HAT), has been implicated in the progression of several diseases, but its role in MASLD remains unclear. Here, we provide the experimental evidence that progressive human and male murine MASLD is driven by GCN5, but not by p300/CREB binding protein associated factor (PCAF) activation. Hepatocyte-specific GCN5 overexpression accelerates MASLD progression, whereas its abl

TGFβ activity stabilizes ACC1 to increase de novo lipogenesis in metabolic liver disease.

Metabolic liver disease arises due to dysregulated signaling between hepatocytes and non-parenchymal cells (NPCs). Through parallel RNA sequencing screens in diet-induced and genetic mouse models, backdropped by human transcriptomic data, we identified latent TGFβ binding protein-3 (LTBP3) - a regulator of TGFβ secretion - as a novel contributor to metabolic liver disease pathogenesis. GalNAc-conjugated Ltbp3 ASO reduced hepatic triglyceride accumulation in diet-induced metabolic liver disease mouse models, which was phenocopied in mice lacking hepatocyte TGFβ activity, but surprisingly not in

LncRNA MEG3 Restrains Hepatic Lipogenesis via the FOXO1 Signaling Pathway in HepG2 Cells.

Nonalcoholic fatty liver disease (NAFLD) become a main public health concern, and is characterized by lipid accumulation in the hepatocytes. We found that overexpression of lncRNA MEG3 significantly reduced the expression of FOXO1, ACC1, and FAS, and subsequently decreased the lipid accumulation in HepG2 cells. Moreover, inhibition of lncRNA MEG3 could increase the lipid accumulation and the mRNA and protein levels of FOXO1, ACC1, and FAS. Further study showed that lncRNA MEG3 regulates the lipogenesis process by inhibiting the entry of FOXO1 into the nucleus translocation. Our study demonstra

Leukemia inhibitory factor suppresses hepatic de novo lipogenesis and induces cachexia in mice.

Cancer cachexia is a systemic metabolic syndrome characterized by involuntary weight loss, and muscle and adipose tissue wasting. Mechanisms underlying cachexia remain poorly understood. Leukemia inhibitory factor (LIF), a multi-functional cytokine, has been suggested as a cachexia-inducing factor. In a transgenic mouse model with conditional LIF expression, systemic elevation of LIF induces cachexia. LIF overexpression decreases de novo lipogenesis and disrupts lipid homeostasis in the liver. Liver-specific LIF receptor knockout attenuates LIF-induced cachexia, suggesting that LIF-induced fun

Aberrant iron distribution via hepatocyte-stellate cell axis drives liver lipogenesis and fibrosis.

Hepatocytes have important roles in liver iron homeostasis, abnormalities in which are tightly associated with liver steatosis and fibrosis. Here, we show that non-alcoholic fatty liver disease (NAFLD) and steatohepatitis (NASH) are characterized by iron-deficient hepatocytes and iron overload in hepatic stellate cells (HSCs). Iron deficiency enhances hepatocyte lipogenesis and insulin resistance through HIF2α-ATF4 signaling. Elevated secretion of iron-containing hepatocyte extracellular vesicles (EVs), which are normally cleared by Kupffer cells, accounts for hepatocyte iron deficiency and HS

Relationship between de novo lipogenesis and serum sex hormone binding globulin in humans.

OBJECTIVE: Obesity and liver fat are associated with decreased levels of serum sex hormone binding globulin (SHBG). Laboratory studies suggest that hepatic de novo lipogenesis (DNL) is involved in the downregulation of SHBG synthesis. The aim of the present study was to address the role of DNL on serum SHBG in humans. DESIGN: A cross-sectional study examining the association between DNL, measured by stable isotopes, and serum SHBG, stratified by sex. PARTICIPANTS: Healthy men (n = 34) and women (n = 21) were combined from two cross-sectional studies. Forty-two per cent of participants had he

Metabolic convergence on lipogenesis in RAS, BCR-ABL, and MYC-driven lymphoid malignancies.

BACKGROUND: Metabolic reprogramming is a central feature in many cancer subtypes and a hallmark of cancer. Many therapeutic strategies attempt to exploit this feature, often having unintended side effects on normal metabolic programs and limited efficacy due to integrative nature of metabolic substrate sourcing. Although the initiating oncogenic lesion may vary, tumor cells in lymphoid malignancies often share similar environments and potentially similar metabolic profiles. We examined cells from mouse models of MYC-, RAS-, and BCR-ABL-driven lymphoid malignancies and find a convergence on de

Blood meal drives de novo lipogenesis in the fat body of Rhodnius prolixus.

In insects, lipids are stored in the fat body mainly as triacylglycerol. Lipids can be directly provided by digestion and incorporated from the hemolymph, or synthesized de novo from other substrates such as carbohydrates and amino acids. The first step in de novo lipid synthesis is catalyzed by acetyl-CoA carboxylase (ACC), which carboxylates acetyl-CoA to form malonyl-CoA. Rhodnius prolixus is a hematophagous insect vector of Chagas disease and feeds exclusively on large and infrequent blood meals. Adult females slowly digest the blood and concomitantly accumulate lipids in the fat body. In

Role of Dietary Fructose and Hepatic De Novo Lipogenesis in Fatty Liver Disease.

Nonalcoholic fatty liver disease (NAFLD) is a liver manifestation of metabolic syndrome. Overconsumption of high-fat diet (HFD) and increased intake of sugar-sweetened beverages are major risk factors for development of NAFLD. Today the most commonly consumed sugar is high fructose corn syrup. Hepatic lipids may be derived from dietary intake, esterification of plasma free fatty acids (FFA) or hepatic de novo lipogenesis (DNL). A central abnormality in NAFLD is enhanced DNL. Hepatic DNL is increased in individuals with NAFLD, while the contribution of dietary fat and plasma FFA to hepatic lipi

Hepatocyte SLCO4C1 is a cAMP uptake transporter for inhibiting lipogenesis and a therapeutic target for MASLD.

Metabolic dysfunction-associated steatotic liver disease (MASLD), the most prevalent cause of chronic liver disease worldwide, currently lacks precision medicine treatments. SLCO4C1 acts as a transporter for endogenous compounds and xenobiotics. This study aims to investigate whether SLCO4C1 plays a regulatory role in MASLD pathogenesis and to elucidate the underlying mechanisms. Using human, mouse and cellular models, we found that hepatocyte SLCO4C1 is upregulated in MASLD patients, where it serves as a key cAMP transporter dependent on Gln463, suppressing lipogenesis through the PKA-CREB-SR

Elevation of hepatic de novo lipogenesis in mice with overnutrition is dependent on multiple substrates.

Increased de novo lipogenesis (DNL) contributes to hyperlipidemia, MASLD, and ASCVD in insulin-resistant subjects. However, multiple pathways support lipogenesis and few have sought to quantify the contributions of the discrete metabolic pathways that contribute to lipogenesis. In this study, antisense oligonucleotides (ASOs) targeting glucokinase (Gck), lactate dehydrogenase A (Ldha), and glutamic-pyruvic transaminase 2 (Gpt2) were utilized to restrict substrate flux from lipogenic precursors in C57BL6/J mice, comparing controls (CO) and chronic overnutrition (ON). In CO mice, ASO treatments

Direct Hepatocyte Insulin Signaling Is Required for Lipogenesis but Is Dispensable for the Suppression of Glucose Production.

During insulin-resistant states such as type II diabetes mellitus (T2DM), insulin fails to suppress hepatic glucose production (HGP) yet promotes lipid synthesis. This metabolic state has been termed "selective insulin resistance" to indicate a defect in one arm of the insulin-signaling cascade, potentially downstream of Akt. Here we demonstrate that Akt-dependent activation of mTORC1 and inhibition of Foxo1 are required and sufficient for de novo lipogenesis, suggesting that hepatic insulin signaling is likely to be intact in insulin-resistant states. Moreover, cell-nonautonomous suppression

Adipose tissue loss in adjuvant arthritis is associated with a decrease in lipogenesis, but not with an increase in lipolysis.

Adjuvant-induced arthritis is a model of rheumatoid arthritis that induces cachexia. In other cachectic situations, there is an increase in lipolysis resulting in a loss of adipose tissue mass. The aim of this work was to analyse the effect of chronic arthritis, induced by adjuvant injection, on white adipose tissue (WAT). For this purpose, rats were killed 10 days after adjuvant injection, when the first external symptoms appeared, on days 15 and 22 when the external signs of the illness reach their severest level. As arthritis decreases food intake, a pair-fed group was also included. Serum

Suppression of de novo lipogenesis and dietary PUFA supplementation inhibit prostate cancer progression.

Prostate cancer progression is characterized by dysregulated lipid metabolism, with fatty acid synthase (FASN), the rate-limiting step in de novo lipogenesis (DNL), resulting in significant accumulation of saturated lipids. Here, we investigate whether pharmacologic FASN inhibition creates a metabolic state that increases reliance on exogenous polyunsaturated fatty acids (PUFAs). Inhibition of FASN profoundly alters membrane phospholipid composition, driving compensatory incorporation of PUFAs into membrane phospholipids, thus increasing susceptibility to lipid peroxidation and oxidative damag

Lactose and sucrose each stimulate hepatic de novo lipogenesis: A randomized crossover trial.

BACKGROUND: Fructose-containing sugars can exaggerate postprandial lipemia and stimulate hepatic de novo lipogenesis (DNL) relative to glucose-based carbohydrates. Despite being consumed by most of the global population, the effects of galactose-containing sugars on hepatic DNL are currently unknown. OBJECTIVES: To assess the effects of lactose ingestion on lipemia and DNL. METHODS: Twenty-four adults without obesity (12 male and 12 female) completed 3 laboratory visits in a randomized, crossover design [33 ± 14 d washout; mean ± SD]. During laboratory visits, participants consumed beverages

Muscle-derived ANXA2 promotes hepatic steatosis by activating SREBP1c-mediated de novo lipogenesis.

BACKGROUND & AIMS: Hepatic steatosis, a hallmark of many liver diseases, is primarily driven by metabolic dysfunction; however, the extrahepatic regulatory mechanisms remain poorly understood. This study aimed to investigate muscle-liver crosstalk and to identify the underlying mechanisms of hepatic steatosis in patients with muscular dystrophy. METHODS: We used mouse models of muscular dystrophy, fibroadipogenic progenitor (FAP)-specific annexin A2 (ANXA2) knockout mice, and muscle biopsy samples from patients with muscular dystrophy. The correlation between serum ANXA2 levels and lipid accu

Proanthocyanidin and mitoglitazone suppress lipogenesis by targeting ferroptosis in metabolic dysfunction-associated steatohepatitis.

Metabolic dysfunction-associated steatohepatitis (MASH) can progress to liver cirrhosis, increasing mortality risk. The study investigates the role of ferroptosis-an inflammatory cell death mechanism-in MASH and evaluates the therapeutic effects of mitoglitazone and proanthocyanidin in targeting ferroptosis to mitigate MASH progression. Forty male albino mice were divided into five groups (n = 8): normal control (NC) fed a standard chow diet and given 2% DMSO; MASH group was maintained on MASH protocol (high fructose-high fat diet); mitoglitazone (Mito) group was kept on MASH protocol and give

De novo lipogenesis protects dormant breast cancer cells from ferroptosis and promotes metastasis.

Dormant disseminated tumor cells (DTCs) remain viable for years to decades before establishing a clinically overt metastatic lesion. DTCs are known to be highly resilient and able to overcome the multiple biological hurdles imposed along the metastatic cascade. However, the specific metabolic adaptations of dormant DTCs remain to be elucidated. Here, we reveal that dormant DTCs upregulate de novo lipogenesis and favor the activation and incorporation of monounsaturated fatty acids (MUFAs) to their cellular membranes through the activation of acyl-coenzyme A synthetase long-chain family member

PANX1-mediated ATP release confers FAM3A's suppression effects on hepatic gluconeogenesis and lipogenesis.

BACKGROUND: Extracellular adenosine triphosphate (ATP) is an important signal molecule. In previous studies, intensive research had revealed the crucial roles of family with sequence similarity 3 member A (FAM3A) in controlling hepatic glucolipid metabolism, islet β cell function, adipocyte differentiation, blood pressure, and other biological and pathophysiological processes. Although mitochondrial protein FAM3A plays crucial roles in the regulation of glucolipid metabolism via stimulating ATP release to activate P2 receptor pathways, its mechanism in promoting ATP release in hepatocytes rema
