Responses to GABA by isolated insect neuronal somata: pharmacology and modulation by a benzodiazepine and a barbiturate.

Mechanically dissociated neuronal somata from the thoracic ganglia of Locusta migratoria and Schistocerca gregaria were viable in vitro for hours and were current- and voltage-clamped to record the responses evoked by brief pressure applications of gamma-aminobutyric acid (GABA) in the presence of various modulators. The application of GABA and muscimol, but not baclofen, produced a hyperpolarization and concurrent increase in the membrane conductance. The current underlying this response reversed at -65 mV, was evoked in all cells tested and showed outward rectification. In 6 of 74 Locusta ne

Duplicated paralog of insulin receptor functions distinctively in locust reproduction for high fecundity.

Insulin receptor (InR) mediates the highly conserved insulin/insulin-like growth factor signaling pathway that regulates a broad range of life-history traits. Most insects encode 2 InR paralogs, but their functional divergence and redundancy remain to be explored. Here, we pursued the functionality of 2 InRs, LmInR1, and LmInR2, in the migratory locust Locusta migratoria. LmInR1 is clustered into the clade of ancestral InR, and LmInR2 belongs to the clade of InR gene duplication. While LmInR1 expression was at the highest levels in nymphal stage and adult ovary, LmInR2 was predominantly expres

Multiple Mammalian Cytokines and Erythropoietin-Mimetic Peptides Protect Insect Neurons via Phylogenetically Conserved Cytokine Receptor-Like Factor 3 (CRLF3).

Erythropoietin (Epo) and its non-erythropoietic splice variant EV-3 have demonstrated potent neuroprotective effects across species, although the respective mechanisms are variable and incompletely understood. Unlike vertebrates, insects lack both Epo and the classical Epo receptor but express Cytokine Receptor-Like Factor 3 (CRLF3), a conserved type I receptor that serves as a neuroprotective receptor for Epo and EV-3 in insects and human iPSC-derived neuron-like cells. Insects, which express CRLF3 but lack all other group 1 type I cytokine receptors, represent a suitable model to study the f

Knockdown of the corazonin gene reveals its critical role in the control of gregarious characteristics in the desert locust.

The two plague locusts, Schistocerca gregaria and Locusta migratoria, exhibit density-dependent phase polyphenism. Nymphs occurring at low population densities (solitarious forms) are uniformly colored and match their body color to the background color of their habitat, whereas those occurring at high population densities (gregarious) develop black patterns. An injection of the neuropeptide, corazonin (Crz) has been shown to induce black patterns in locusts and affect the classical morphometric ratio, F/C (F, hind femur length; C, maximum head width). We herein identified and cloned the CRZ ge

Enhancement of short-term synaptic plasticity by prior environmental stress.

All chemical synapses can rapidly up- or downregulate the strength of their connections to reshape the postsynaptic signal, thereby stressing the informational importance of specific neural pathways. It is also true that an organism's environment can exert a powerful influence on all aspects of neural circuitry. We investigated the effect of a prior high-temperature stress on the short-term plasticity of a neuromuscular synapse in the hindleg tibial extensor muscle of Locusta migratoria. We found that the prior stress acted to precondition the synapse by increasing the upper temperature limit

Discovery of 2-Cinnamoylbenzimidazoles as Novel Insect and Fungal Trehalose-6-Phosphate Phosphatase Inhibitors.

Trehalose-6-phosphate phosphatase (TPP), a key enzyme in trehalose biosynthesis, represents a promising target for pest and fungal control due to the absence of TPP in higher animals. However, few TPP inhibitors have been reported, and broad-spectrum agents targeting both insects and fungi remain scarce. In this study, 2-cinnamoylbenzimidazole was identified as a dual-targeting inhibitor scaffold against insect and fungal TPPs. Structure-guided optimization by pocket size yielded 5-1 (Ki = 1.3 μM) for insect TPP and 5-3 (Ki = 17.3 μM) for fungal TPP. Molecular docking revealed that 2-cinnamoyl

High-level expression and purification of active scorpion long-chain neurotoxin BjαIT from Pichia pastoris.

As an insect-selective neurotoxin, scorpion long-chain BjαIT is a promising prospect for insecticidal application; however, the difficulty of obtaining natural BjαIT represents the major obstacle preventing analysis of its insecticidal activity against agricultural insect pests. Here, we screened recombinant Pichia pastoris transformants showing high levels of secretory recombinant (r)BjαIT. Secreted rBjαIT was expressed at levels as high as 340 mg/L following methanol induction in a fed-batch reactor, with ∼21 mg of pure rBjαIT obtained from 200-mL fed-batch culture supernatant by Ni2+-nitril

Palytoxin evokes reversible spreading depolarization in the locust CNS.

Spreading depolarization (SD) describes the near-complete depolarization of central nervous system (CNS) neural cells as a consequence of chemical, electrical, or metabolic perturbations. It is well established as the central mechanism underlying insect coma and various mammalian neurological dysfunctions. Despite significant progress in our understanding, the question remains: which cation channel, if any, generates SD in the CNS? Previously, we speculated that the sodium-potassium ATPase (NKA) might function as a large-conductance ion channel to initiate SD in insects, potentially mediated b

Cold acclimation improves chill tolerance in the migratory locust through preservation of ion balance and membrane potential.

Most insects have the ability to alter their cold tolerance in response to temporal temperature fluctuations, and recent studies have shown that insect cold tolerance is closely tied to the ability to maintain transmembrane ion gradients that are important for the maintenance of cell membrane potential (Vm). Several studies have therefore suggested a link between preservation of Vm and cellular survival after cold stress, but none has measured Vm in this context. We tested this hypothesis by acclimating locusts (Locusta migratoria) to high (31°C) and low temperature (11°C) for 4 days before ex

Apoptosis of male germ-line stem cells after laser ablation of their niche.

Male germ-line stem cells (GSCs) and their niche-the apical cells or hub cells-display a unique feature at the apices of insect testicular follicles. In the locust, Locusta migratoria, the niche consists of only one large apical cell surrounded by about 60 GSCs. The apical cell can be readily identified in the intact follicle. Using laser ablation it is feasible to destroy the apical cell exclusively without injuring neighboring GSCs or any other cells. The most immediate effect on GSCs is the loss of their structural polarity. Beginning about 3 h after laser treatment chromatin starts to clum

Hyperkalaemia, not apoptosis, accurately predicts insect chilling injury.

There is a growing appreciation that insect distribution and abundance are associated with the limits of thermal tolerance, but the physiology underlying thermal tolerance remains poorly understood. Many insects, like the migratory locust (Locusta migratoria), suffer a loss of ion and water balance leading to hyperkalaemia (high extracellular [K+]) in the cold that indirectly causes cell death. Cells can die in several ways under stress, and how they die is of critical importance to identifying and understanding the nature of thermal adaptation. Whether apoptotic or necrotic cell death pathway

Concurrent effects of cold and hyperkalaemia cause insect chilling injury.

Chilling injury and death are the ultimate consequence of low temperature exposure for chill susceptible insects, and low temperature tolerance is considered one of the most important factors determining insect distribution patterns. The physiological mechanisms that cause chilling injury are unknown, but chronic cold exposure that causes injury is consistently associated with elevated extracellular [K(+)], and cold tolerant insects possess a greater capacity to maintain ion balance at low temperatures. Here, we use the muscle tissue of the migratory locust (Locusta migratoria) to examine whet

Efficient utilization of aerobic metabolism helps Tibetan locusts conquer hypoxia.

BACKGROUND: Responses to hypoxia have been investigated in many species; however, comparative studies between conspecific geographical populations at different altitudes are rare, especially for invertebrates. The migratory locust, Locusta migratoria, is widely distributed around the world, including on the high-altitude Tibetan Plateau (TP) and the low-altitude North China Plain (NP). TP locusts have inhabited Tibetan Plateau for over 34,000 years and thus probably have evolved superior capacity to cope with hypoxia. RESULTS: Here we compared the hypoxic responses of TP and NP locusts from m

Chemical Ecology and Olfaction in Short-Horned Grasshoppers (Orthoptera: Acrididae).

Chemoreception plays a crucial role in the reproduction and survival of insects, which often rely on their sense of smell and taste to find partners, suitable habitats, and food sources, and to avoid predators and noxious substances. There is a substantial body of work investigating the chemoreception and chemical ecology of Diptera (flies) and Lepidoptera (moths and butterflies); but less is known about the Orthoptera (grasshoppers, locusts, crickets, and wēta). Within the Orthoptera, the family Acrididae contains about 6700 species of short-horned grasshoppers. Grasshoppers are fascinating o

Identified octopaminergic neurons modulate contractions of locust visceral muscle via adenosine 3',5'-monophosphate (cyclic AMP).

D,L-Octopamine elevates the cyclic AMP content of the lateral oviduct of the locust, Locusta migratoria, in a dose-dependent manner with a threshold of about 10(-8) M. The effect of octopamine is potentiated by the phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (IBMX). The response is specific for octopamine and synephrine with an order of potency being octopamine = synephrine greater than metanephrine greater than tyramine greater than norepinephrine = dopamine = 5-hydroxytryptamine and the effect of octopamine is inhibited by the alpha-adrenergic receptor antagonist phentolamine. Validation of Insect Dopa Decarboxylase as a Potential Insecticide Target and Discovery of Selective Lead Compounds.

Dopa decarboxylase (DDC) is a key rate-limiting enzyme involved in cuticle tanning in insects, catalyzing the conversion of l-dopa to dopamine. Given its essential role in insect survival, DDC represents a potential target for pest control. In this study, we recombinantly expressed and characterized DDC from Drosophila melanogaster and developed a structural model for virtual screening. Quercetin (Que), the first identified inhibitor of insect DDC, was found to have an IC50 value of 9.43 μM. Notably, Que displayed much lower inhibitory activity against human DDC, with an IC50 value of 29.56 μM

Juvenile hormone promotes paracellular transport of yolk proteins via remodeling zonula adherens at tricellular junctions in the follicular epithelium.

Juvenile hormone (JH) acts as a gonadotrophic hormone stimulating insect vitellogenesis and oogenesis. Paracellular transport of yolk proteins through intercellular channels (patency) in the follicular epithelium is a developmentally regulated and evolutionarily conserved process during vitellogenesis. However, the mechanisms underlying patency opening are poorly understood. Using the migratory locust Locusta migratoria as a model system, we report here that JH-regulated remodeling of zonula adherens (ZA), the belt-like adherens junction maintaining physical linking between follicle cells cont

Recombinant production of the insecticidal scorpion toxin BjαIT in Escherichia coli.

Scorpion long-chain insect neurotoxins have important potential application value in agricultural pest control. The difficulty of obtaining natural toxins is the major obstacle preventing analyses of their insecticidal activity against more agricultural insect pests. Here we cloned the insect neurotoxin BjαIT gene into the pET32 expression vector and expressed the resulting thioredoxin (Trx)-BjαIT fusion protein in Escherichia coli. Soluble Trx-BjαIT was expressed at a high level when induced at 18 °C with 0.1 mM isopropyl β-d-1-thiogalactopyranoside, and it was purified by Ni2+-nitriloacetic

Metarhizium anisopliae infection-associated metabolites act upon LmOBP2 to suppress locust feeding.

Entomopathogenic fungi can manipulate host behavior to promote infection success, but the molecular effectors remain largely unknown. Here we show that Metarhizium anisopliae suppresses feeding in the migratory locust Locusta migratoria by downregulating an antenna-enriched odorant-binding protein gene, LmOBP2. Metabolomic profiling of infected hemolymph identified infection-associated increases in indole, taurine, DL-serine and dimethylglycine; injection or exposure to these metabolites reduced antennal LmOBP2 transcript abundance. Recombinant LmOBP2 bound key corn volatiles (e.g., leaf alcoh

[New studies on natural inhibitors of proteolytic enzymes].

New data on proteolytic enzyme inhibitors and mechanisms of their interaction with the enzymes are reviewed. In recent years, a number of new inhibitors comprising families earlier unknown have been described such as proteins from the parasitic nematode Ascaris lumbricoides, ecotin from the periplasm of Escherichia coli, proteins PMP-C and PMP-D from locust Locusta migratoria, and hirustasin from the medicinal leech Hirudo medicinalis. At the same time, some proteins that may be assigned to inhibitors on the basis of their structures were found to perform other (not inhibitory) functions. Thus
