In vivo bioluminescence imaging of murine xenograft cancer models with a red-shifted thermostable luciferase.

PURPOSE: Conventional in vivo bioluminescence imaging using wild-type green-emitting luciferase is limited by absorption and scattering of the bioluminescent signal through tissues. Imaging methods using a red-shifted thermostable luciferase from Photinus pyralis were optimized to improve the sensitivity and image resolution. In vivo bioluminescence imaging performance of red- and green-emitting luciferases were compared in two different xenograft mouse models for cancer. METHODS: Human hepatoblastoma cell line (HepG2) and human acute monocytic leukemia cell line (Thp1) cells were genetically

Development of a luciferase-based reporter system to monitor Bifidobacterium breve UCC2003 persistence in mice.

BACKGROUND: Probiotics such as bifidobacteria have been shown to maintain a healthy intestinal microbial balance and help protect against infections. However, despite these benefits, bifidobacteria still remain poorly understood at the biochemical, physiological and especially the genetic level. Herein we describe, for the first time, the development of a non-invasive luciferase-based reporter system for real-time tracking of Bifidobacterium species in vivo. RESULTS: The reporter vector pLuxMC1 is based on the recently described theta-type plasmid pBC1 from B. catenatulatum 1 and the luxABCDE

A luciferase immunoprecipitation systems assay enhances the sensitivity and specificity of diagnosis of Strongyloides stercoralis infection.

BACKGROUND: We investigated whether luciferase immunoprecipitation systems (LIPS) can be the basis for a more rapid, specific, and standardized assay for the diagnosis of Strongyloides stercoralis infection. METHODS: A LIPS assay was developed based on immunoglobulin (Ig) G or IgG4 antibody to a recombinant Strongyloides antigen (NIE) and was compared with an NIE enzyme-linked immunosorbent assay (ELISA). A second antigen, S. stercoralis immunoreactive antigen (SsIR), was tested alone and in combination with NIE. The assays were tested using serum samples from patients with parasitologically

A cell-based luciferase assay amenable to high-throughput screening of inhibitors of arenavirus budding.

Several arenaviruses cause hemorrhagic fever (HF) disease in humans for which there are no licensed vaccines, and current therapy is limited to the use of ribavirin (Rib) that is only partially effective and associated with significant side effects. In addition, compelling evidence indicates that the prototypic arenavirus lymphocytic choriomeningitis virus (LCMV) is a neglected human pathogen of clinical significance. Therefore, it is important to develop novel and effective anti-arenaviral drugs. The arenavirus Z protein is the driving force of arenavirus budding, and PPPY and PTAP late (L) d

Profiling Norovirus Antibody Specificity and Function Using Luciferase Immunoprecipitation System (LIPS) Assays.

The luciferase immunoprecipitation system (LIPS) assay is a liquid-phase immunoassay that quantitates antigen-specific serum antibodies by measuring luminescence emitted by reporter enzyme Renilla luciferase (Ruc) fused to an antigen of interest. The LIPS assay can be utilized as a high-throughput and sensitive serological method for profiling serum antibodies recognizing diverse antigens. Initially adapted to assess the presence of antibodies to noroviruses, it was later modified to measure the ability of antibodies to block norovirus carbohydrate binding, a well-established correlate with ne

Cell-Based Luciferase Assay for Testing SARS-CoV-2 3CL Protease Inhibitors.

A cell-based screening system for viral protease inhibitors was developed using firefly luciferase fragment complementation and validated on the SARS-CoV-2 3CLpro model. The optimal luciferase variant incorporating the VLQSGF proteolytic site (Luc III) retained 88% of its native activity. A critical requirement for system performance was the use of an extended nsp4-nsp6 fragment of the viral polyprotein rather than the mature protease, underscoring the importance of the native context for 3CLpro activity. The bicistronic construct pCAG-Luc-III-IRES-nsp4-6 enables coordinated expression of the

Real-time tracking of vaccinia virus entry using a split luciferase complementation assay.

UNLABELLED: A recombinant vaccinia virus (vA4-HiBiT) was constructed with a short, split luciferase HiBiT tag at the C-terminus of the A4 core protein open reading frame. When HeLa or A549 cells stably expressing LgBiT were infected with vA4-HiBiT, the interaction of the split luciferase fragments reconstituted NanoLuc enzyme activity. Upon addition of a membrane-permeable luciferase substrate, a strong luminescence signal indicated core entry into the cytoplasm within 5 min after infection and was proportional to virus multiplicity within the range of 0.5-80 PFU/cell. This direct entry assay

Utility of the immunoglobulin E crosslinking-induced luciferase expression assay in buckwheat allergy diagnosis.

BACKGROUND: Buckwheat allergy carries a high risk of anaphylaxis. Although the oral food challenge (OFC) is the diagnostic gold standard, it can provoke severe reactions, leading many patients to adopt elimination diets. Conventional markers, including buckwheat-specific IgE (Bw-sIgE) and skin prick testing, have limited accuracy. The IgE crosslinking-induced luciferase expression (EXiLE) assay is a serum-based in vitro cell-activation test that has shown high accuracy for other food allergies. We evaluated its utility for buckwheat allergy. METHODS: This multicenter observational study in Ja

Split-Luciferase Reassembly Assay to Measure Endoplasmic Reticulum-Mitochondria Contacts in Live Cells.

Endoplasmic reticulum (ER)-mitochondria contact sites play a critical role in cell health and homeostasis, such as the regulation of Ca2+ and lipid homeostasis, mitochondrial dynamics, autophagosome and mitophagosome biogenesis, and apoptosis. Failure to maintain normal ER-mitochondrial coupling is implicated in many neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, amyotrophic lateral sclerosis, and hereditary spastic paraplegia. It is of considerable significance to explore how the dysregulation of ER-mitochondrial contacts could lead to cell death and whether repa

The serodiagnositic value of Chlamydia trachomatis antigens in antibody detection using luciferase immunosorbent assay.

INTRODUCTION: Among the different antigens used in the detection of anti-Chlamydia trachomatis antibodies, significant differences in sensitivity and specificity have been observed. Further evaluation of C. trachomatis antigens in antibody detection is urgently needed for the development and application of C. trachomatis serologic assays. METHODS: Chlamydia trachomatis antigens Pgp3, TmeA, InaC, and HSP60 were selected and used in luciferase immunosorbent assay (LISA). The detection results obtained from well-defined C. trachomatis positive and negative samples were compared with the commerci

Protein Pull-down Assay Using HiBiT-tag-dependent Luciferase Activity Measurement.

Co-immunoprecipitation or pull-down assays are frequently used to analyze protein-protein interactions. In these experiments, western blotting is commonly used to detect prey proteins. However, sensitivity and quantification problems remain in this detection system. Recently, the HiBiT-tag-dependent NanoLuc luciferase system was developed as a highly sensitive detection system for small amounts of proteins. In this report, we introduce the method of using HiBiT technology for the detection of prey protein in a pull-down assay. Using this protocol, we demonstrate the formation of a ternary comp

Tracing location by applying Emerald luciferase in an early phase of murine endometriotic lesion formation.

The pathogenesis of endometriosis has not been fully elucidated. We focused on the behavior of the ectopic endometrium, that is, the origin of the endometriotic lesion, before adhering to the peritoneal cavity. To observe lesion formation in the very early phase, we developed a novel endometriosis animal model using bioluminescence technology. We established a new transgenic mouse that expressed Emerald luciferase (ELuc) under the control of the CAG promoter. This transgenic mouse, called the CAG-ELuc mouse, showed strong bioluminescence emission; we succeeded in tracing the lesion location by

Optimized application of the secreted Nano-luciferase reporter system using an affinity purification strategy.

Secreted Nano-luciferase (secNluc) is a newly engineered secreted luciferase that possesses advantages of high structural stability, long half-life, and glow-type kinetics together with high light emission intensity, and thus would become one of the most valuable tools for bioluminescence assays. However, like other secreted luciferases, secNluc has to mix with the components in the conditioned medium surrounding test cells, or in the biological samples such as blood or urine after being secreted. These components may interfere with secNluc-catalyzed bioluminescence reactions and thus limit th

Establishment of ATP-based luciferase viability assay in 96-well plate for Trypanosoma congolense.

Animal African trypanosomosis (AAT), caused by Trypanosoma congolense, is widespread throughout sub-Saharan Africa. There are significant concerns related to the current drugs available for the treatment of AAT due to their limited effectiveness across species and their adverse effects. Moreover, drug resistant trypanosomes have recently been reported in the field. High throughput screening (HTS) of large chemical compound library collections is a promising approach for identifying novel drug candidates. While HTS for Trypanozoon trypanosomes, T. brucei sspp. and T. evansi is well established,

Immobilization of bacterial luciferase into poly(N-isopropylacrylamide) film for electrochemical control of a bioluminescence reaction.

A poly(N-isopropylacrylamide) film was modified on an indium-tin oxide electrode in order to immobilize bacterial luciferase (BL) on the electrode surface. By using the modified electrode, flavin mononucleotide (FMN) was electrochemically reduced to FMNH(2), which is one of the substrates of the BL luminescence reaction, to control the bioluminescence reaction by BL. The BL reaction in the modified film could be promoted and controlled by the electrochemical generation of FMNH(2). This BL luminescence system was evaluated as a model system for the inhibitory assay of hydrophobic molecules on p

In vivo imaging of chikungunya virus in mice and Aedes mosquitoes using a Renilla luciferase clone.

Chikungunya virus (CHIKV) is a mosquito-borne alphavirus that in humans causes an acute febrile illness characterized by fever, arthralgia, and rash. It is currently associated with large outbreaks in Asia, Africa, and islands of the Indian Ocean and has been introduced from these tropical regions into Europe, where local transmission has been recorded on two occasions. The underlying basis of the pathogenesis of CHIKV and related alphaviruses that produce similar symptoms remains unclear. By applying new techniques, for example, in vivo imaging in live animals and arthropods, we may improve o

Luciferase-induced immune activation prevents orthotopic K7M2luc osteosarcoma establishment in BALB/c mice.

BACKGROUND: Use of murine orthotopic tumor models dictates utilization of luciferase-expressing cells for in vivo bioluminescence imaging. However, luciferase may trigger immune responses that impair tumor establishment. MATERIALS AND METHODS: We transduced K7M2 cells wild-type (wt) with luciferase gene and evaluated their growth in vitro. We then implanted cells orthotopically in BALB/c mice and upon unsuccessful tumor establishment assessed systemical and local mechanism for tumor growth suppression. RESULTS: All investigated cell lines had a comparative doubling time in vitro at 24 and 48

A rapid and robust luciferase-based reporter system to assess SARS-CoV-2 protease activity.

Despite effective antiviral drugs that have emerged to combat SARS-CoV-2 infections, novel therapeutic strategies are required to better address the ongoing and future evolutions of the virus. Targeting viral proteases, such as the main protease (Mpro), remains a promising approach. Here, we present a rapid and sensitive luminescence-based reporter system, the i-NSP4/5-Gluc2, to assess Mpro activity. This system employs Gaussia luciferase (Gluc) fused to a pro-interleukin 1β (pro-IL-1β) fragment containing a specific Mpro cleavage site. Upon Mpro cleavage, Gluc is released and secreted, genera

Use of Rift Valley Fever Virus Expressing NanoLuc Luciferase for the Assessment of Neutralizing Antibodies and Antivirals.

Rift Valley fever (RVF) is an arboviral zoonotic disease affecting many African countries with the potential to spread to other geographical areas. In this chapter we describe the use of a replication-competent recombinant (r)RVFV expressing NanoLuc Luciferase (Nluc) for in vitro studies. The determination of parameters such as neutralizing antibodies in serum samples, or the antiviral activity of drugs is usually carried out using standard assays based on the assessment of cytopathic effect on cell cultures. The use of a virus encoding a traceable reporter protein allows to correlate the pres
