Lupeol- and Cis-Vaccenic Acid-Containing Bioactive Fraction of Mondia whitei Induces Vacuolation-Associated Cytotoxicity Through Autophagy-Lysosome Perturbation and Caspase-Dependent Apoptosis in Human Colorectal Adenocarcinoma Cells.

Plant-derived natural products are essential and prominent contributors to drug discovery, especially as anticancer agents. Medicinal plants used in alternative therapy are often obscured by poor definitions of the underlying mechanisms behind their bioactivity. In this study, we investigated the cytotoxic and mechanistic effects of an enriched bioactive fraction derived from Mondia whitei. Bioactivity-guided fractionation was performed using C18 solid-phase extraction. The cytotoxic effects were evaluated using the MTT assay, while cellular morphology and mechanistic pathways were examined th

Action of Betulinic Acid in the Inhibition of Efflux Pump NorA in Staphylococcus aureus Strains: In Vitro and In Silico Approaches.

Antibiotic resistance poses a serious challenge to public health, particularly in the case of Staphylococcus aureus, a Gram-positive bacterium that employs multiple resistance mechanisms, including efflux pumps such as NorA, which extrude antimicrobial compounds from the cell and reduce antibiotic efficacy. Therefore, the search for substances capable of inhibiting these mechanisms represents a promising strategy to combat bacterial resistance. Betulinic acid (BA), a pentacyclic triterpene of the lupane type, commonly found in different parts of plants, has demonstrated various pharmacological

Development of an LC-MS Method for the Analysis of Birch (Betula sp.) Bark Bioactives Extracted with Biosolvents.

Birch (Betula sp.) bark is a well-known natural source of betulin (Bet) and betulinic acid (BAc), both of which have several bioactive properties. The evaluation of the extraction performance, relative to these lupane-type triterpenoids, provided by different biosolvents requires the development of a high-resolution and high-sensitivity liquid chromatography-mass spectrometry (LC-MS) approach that is also compatible with challenging extractants such as natural deep eutectic solvents (NADESs). In this work, an LC-MS method was developed and analytically characterized prior to its application fo

Antimicrobial and antiparasitic potential of lupeol: antifungal effect on the Candida parapsilosis species complex and nematicidal activity against Caenorhabditis elegans.

Introduction. There is growing concern about infections caused by non-albicans Candida species, including species of the Candida parapsilosis complex - which have seen a considerable increase in cases during the COVID-19 pandemic - in addition to concern about nematode resistance to currently used anthelmintics.Gap Statement. Lupeol is a triterpenoid phytosterol that has a wide range of biological activities, although its antifungal and antiparasitic potential is still poorly explored. Additionally, its effect on the biofilm of the C. parapsilosis species complex has not yet been studied.Aim.

Cytotoxic Potential of Betulinic Acid Fatty Esters and Their Liposomal Formulations: Targeting Breast, Colon, and Lung Cancer Cell Lines.

Betulinic acid is a lupane-type pentacyclic triterpene mostly found in birch bark and thoroughly explored for its wide range of pharmacological activities. Despite its impressive biological potential, its low bioavailability has challenged many researchers to develop different formulations for achieving better in vitro and in vivo effects. We previously reported the synthesis of fatty acid esters of betulinic acid using butyric, stearic, and palmitic acids (But-BA, St-BA, and Pal-BA) and included them in surfaced-modified liposomes (But-BA-Lip, St-BA-Lip, Pal-BA-Lip).
