Luteolin in Inflammatory Bowel Disease and Colorectal Cancer: A Disease Continuum Perspective.

Inflammatory bowel disease (IBD) is a chronic intestinal inflammatory condition that may progress to colorectal cancer (CRC), presenting significant challenges to global health. With shifts in lifestyle, the incidence of both conditions continues to rise, underscoring the urgent need for effective treatments. While traditional therapies can be effective, their high recurrence rates and associated adverse reactions limit their broader application. Luteolin, a flavonoid derived from natural plants, has emerged as a promising focus in both IBD and CRC research due to its multi-target therapeutic

Phase 2 study of palmitoylethanolamide combined with luteoline in frontotemporal dementia patients.

Frontotemporal dementia is a devastating neurodegenerative disorder for which no pharmacological treatments have been approved. Neuroinflammation plays a central role in driving the pathogenic mechanisms underlying frontotemporal dementia. In the last few years, co-ultramicronized palmitoylethanolamide combined with luteoline has emerged as a potential therapeutic molecule in neurodegenerative disorders pathogenically related to frontotemporal dementia, for its demonstrated strong anti-inflammatory and neuroprotective properties. Here we wanted to determine whether treatment with co-ultramicro

PknB and STP as potential targets of luteolin in combating Trueperella pyogenes infections.

Trueperella pyogenes (T. pyogenes) is a significant opportunistic pathogen that causes suppurative infection in many animals, as well as humans. Considering the strong drug resistance of T. pyogenes, the development of novel antibacterial drugs and drug targets to combat infections is necessary. Serine/threonine protein kinases (STKs) and serine/threonine phosphatases (STPs) play pivotal roles in the physiological processes, pathogenesis, and resistance of several important bacterial pathogens, indicating their potential as antimicrobial drug targets. In this study, we aimed to investigate the

Cerium-Luteolin Nanocomplexes in Managing Inflammation-Related Diseases by Antioxidant and Immunoregulation.

Oxidative stress, characterized by an imbalance between reactive oxygen species (ROS) production and the antioxidant defense system, plays a pivotal role in inflammation-related diseases. Excessive ROS levels can induce cellular damage and impair normal physiological functions, triggering the release of inflammatory mediators and exacerbating the inflammatory response, ultimately leading to irreversible tissue damage. In this study, we synthesized cerium ion-luteolin nanocomplexes (CeLutNCs) by coordinating Ce ions with the natural product luteolin, aiming to develop a therapeutic agent with e

Inhibitory effects of luteolin on titanium particle-induced osteolysis in a mouse model.

Wear particles liberated from the surfaces of an implanted prosthesis are associated with peri-implant osteolysis and subsequent aseptic loosening. In the latter wear particle-induced inflammation and osteoclastogenesis have been identified as critical factors, and their inhibition as important steps in the treatment of affected patients, such as those undergoing total hip replacement. In this study the ability of luteolin to inhibit both titanium (Ti) particle-induced osteoclastogenesis in vitro and osteolysis in a murine calvaria Ti particle-induced model of osteolysis was examined. The resu

Luteolin Mitigates D-Galactose-Induced Brain Ageing in Rats: SIRT1-Mediated Neuroprotection.

Luteolin is an essential natural polyphenol found in a variety of plants. Numerous studies have supported its protective role in neurodegenerative diseases, yet the research for its therapeutic utility in D-galactose (D-gal)-induced brain ageing is still lacking. In this study, the potential neuroprotective impact of luteolin against D-gal-induced brain ageing was explored. Forty rats were randomly divided into four groups: control, luteolin, D-gal, and luteolin-administered D-gal groups. All groups were subjected to behavioural, cholinergic function, and hippocampal mitochondrial respiration

Luteolin ameliorates rat myocardial ischaemia-reperfusion injury through activation of peroxiredoxin II.

BACKGROUND AND PURPOSE: Antioxidants provide a promising therapeutic effect for the cardiovascular disease. Luteolin, a polyphenolic bioflavonoid, is known to confer cardioprotection, although the underlying mechanisms, especially the role of luteolin on the antioxidant enzymes, such as the peroxiredoxin family, remain unknown. EXPERIMENTAL APPROACH: We measured the effects of luteolin on myocardial ischaemia/reperfusion (MI/R) injury in vivo (Sprague-Dawley rats) and in vitro, together with the underlying mechanisms, with a focus on signalling by peroxiredoxins. H9c2 cells were used to asses

Luteolin restricts dengue virus replication through inhibition of the proprotein convertase furin.

In many countries afflicted with dengue fever, traditional medicines are widely used as panaceas for illness, and here we describe the systematic evaluation of a widely known natural product, luteolin, originating from the "heat clearing" class of herbs. We show that luteolin inhibits the replication of all four serotypes of dengue virus, but the selectivity of the inhibition was weak. In addition, ADE-mediated dengue virus infection of human cell lines and primary PBMCs was inhibited. In a time-of-drug-addition study, luteolin was found to reduce infectious virus particle formation, but not v

Luteolin's Potential in Managing Osteoporosis and Bone Metabolism Disorders: Preclinical Insights.

Bone homeostasis is a process of dynamic regulation between bone resorption and bone formation, and is the foundation for normal skeletal development in the human body. Once this balance is disrupted, it will lead to bone loss and decreased bone density, profoundly affecting the occurrence and development of bone metabolism-related diseases. For example, common clinical bone metabolic disorders such as osteoporosis, avascular necrosis of the femoral head, and alveolar bone resorption. However, current treatments for bone metabolism-related diseases often have certain limitations, which has dri

Evaluating the inhibitory potential of natural compound luteolin on human lysozyme fibrillation.

Numerous pathophysiological conditions known as amyloidosis, have been connected to protein misfolding leading to aggregation of proteins. Inhibition of cytotoxic aggregates or disaggregation of the preformed fibrils is thus one of the important strategies in the prevention of such diseases. Growing interest and exploration of identification of small molecules mainly natural compounds can prevent or delay amyloid fibril formation. We examined the mechanism of interaction and inhibition of human lysozyme (HL) aggregates with luteolin (LT). Biophysical and computational approaches have been empl

Liver fibrosis in mice induced by carbon tetrachloride and its reversion by luteolin.

Hepatic fibrosis is effusive wound healing process in which excessive connective tissue builds up in the liver. Because specific treatments to stop progressive fibrosis of the liver are not available, we have investigated the effects of luteolin on carbon tetrachloride (CCl(4))-induced hepatic fibrosis. Male Balb/C mice were treated with CCl(4) (0.4 ml/kg) intraperitoneally (i.p.), twice a week for 6 weeks. Luteolin was administered i.p. once daily for next 2 weeks, in doses of 10, 25, and 50 mg/kg of body weight. The CCl(4) control group has been observed for spontaneous reversion of fibrosis

Trimethyltin chloride triggers ferroptosis in myocardial injury: mitochondria-dependent protection by luteolin.

Trimethyltin chloride (TMT), a poison from water and soil, has been linked to bradycardia. Ferroptosis, an iron-driven type of regulated cell death, is critically involved in cardiovascular diseases. However, the mechanism of TMT-induced myocardial injury remains unclear. This study aims to investigate the toxicological mechanism of TMT-induced myocardial injury and evaluate the protective effect of Lut. In this study, RNA sequencing identified Gpx4 as the key ferroptosis-related gene of TMT-induced cardiomyopathy. In vitro, TMT induced ferroptosis, which was reversed by Ferrostatin-1. Notably

Photoacidity and ESIPT in luteolin: acidity-vibrational frequency relationships across electronic states.

The photochemical antioxidant properties of small organic molecules in electronically excited states have become an important topic in modern photochemistry and redox biology. Photoexcitation can trigger ultrafast proton and hydrogen transfer processes, leading to substantial changes in acidity and redox behavior. This study systematically explores the excited-state proton/hydrogen transfer pathways, acidity, and photophysical characteristics of luteolin (LU), a compound containing multiple potential deprotonation sites, using density functional theory (DFT) and time-dependent DFT (TD-DFT) cal

Luteolin alleviates PCOS by inhibiting AR/STAT3/NLRP3-mediated granulosa cell pyroptosis.

BACKGROUND: Polycystic ovary syndrome (PCOS), a common endocrine-metabolic disorder, is driven by hyperandrogenism and chronic low-grade inflammation that impair follicular development. Granulosa cell pyroptosis is increasingly recognized as a key pathogenic mechanism in PCOS. Luteolin (LUT), a natural flavonoid found in many traditional medicinal plants, exhibits potent anti-inflammatory properties. However, its role in regulating granulosa cell pyroptosis within the context of PCOS has not been elucidated. METHODS: We established a dehydroepiandrosterone (DHEA)-induced PCOS rat model to eva
