Splenic B cells function as immunogenic antigen-presenting cells for the induction of effector T cells.

Previous studies performed in our laboratory have revealed that an ordered, sequential, tricellular interaction is obligatory for the antigen-driven induction of a specific effector memory T cell. Thus, it was found that antigen-pulsed peritoneal macrophages signal, in spleen cells, the generation of antigen-specific initiator lymphocytes. These lymphocytes, following injection to syngeneic recipients, recruit, in the draining lymph nodes, "virgin" antigen-reactive T lymphocytes. Although the nature of the first and last cell in the interacting sequence was well characterized, the identity of

Mechanisms of systemic autoimmunity in murine models of SLE.

Our laboratory has utilized spontaneous and experimentally induced models of systemic autoimmunity in mice in order to elucidate the cellular deficiencies in immunoregulation that are essential to this process. In the spontaneously autoimmune mouse strains, genetic defects in T and B cell tolerance are the primary abnormalities that drive the syndrome. The induced chronic graft-vs-host model depends on abnormal T-B interactions resulting from allogeneic recognition of major histocompatibility complex (MHC) class II. Future investigations will target the biochemistry of the loss of tolerance an

In vitro stimulation of human tonsillar subepithelial B cells: requirement for interaction with activated T cells.

Human tonsillar subepithelial B cells, which are a marginal zone-equivalent B cell subset, respond readily to T-independent type 2 antigens, but not to polyclonal B cell activators in vitro. In this study, subepithelial (SE) B cells were induced to proliferate and mature into plasma cells when co-cultured with activated T cells. The response of SE B cells was not observed when co-cultures were carried out in transwell chambers or in the presence of blocking anti-LFA-1 antibodies, demonstrating the need for a close T-B cell interaction. The presence of soluble CD40 also prevented the B cell res

In vitro effect of prednisolone on peripheral blood suppressor T cell activity in patients with alcoholic hepatitis.

An immunological process is suggested to play some role in the pathogenesis of alcoholic hepatitis; however, its nature has not been clarified. In the present study, we examined, in 30 patients with alcoholic liver disease, an in vitro effect of prednisolone on suppressor T cell activity to see whether an altered cellular immunoregulatory mechanism is affected by corticosteroid. Suppressor T cell activity, which was induced by concanavalin A (Con A), was assessed by inhibition of phytohemagglutinin-stimulated blast transformation of autologous lymphocytes, and prednisolone (10 micrograms/ml) w

Abnormalities of B-cell activation and immunoregulation in splenectomized patients.

Using a reverse hemolytic plaque assay as the effector system, we studied B-lymphocyte function in 12 patients after posttraumatic splenectomy, as well as in 25 normal individuals. The time interval between the splenectomy and the immunological studies varied between 2 days and 7 years. Compared to normal individuals, the splenectomized patients had markedly elevated numbers of spontaneous immunoglobulin-secreting cells (ISC) and severely decreased responses to the polyclonal activator pokeweed mitogen. A tendency towards normalization of these abnormalities, especially the high spontaneous IS

The pathogenesis of Graves' disease.

Graves' disease may prove to be due to a genetic defect in immune surveillance or control, which then permits a randomly mutating thyroid-directed clone of T-lymphocytes to survive, interact with previously normal thyroid cell membrane antigen, and set up a localized cell-mediated immune response. The T-lymphocytes so interacting then cooperate with and direct groups of B-lymphocytes, which consequently produce TSI's: the stimulation of the thyroid parenchymal cells is a direct result of TSI stimulation. PHA stimulates Graves' lymphocytes to produce TSI's by first stimulating T-lymphocytes, wh

[Immunological aspects of pulmonary granulomatosis with special reference to sarcoidosis].

A wide variety of exogenous or endogenous aggressors exhibit the capacity to induce pulmonary granulomatous lesions. This lesion is characterized by exacerbation of phagocytic function and tentative development of a specific immune response. In many clinical situations there is a well developed circulating antibody response with deficient cell-mediated immunity of variable degree. Finally, it is argued that the granulomatous lesion may be due to abnormal cooperation between T and B lymphocytes.

[T and B cell immune reactions in inflamed tissue].

The T and B lymphocytes in secondary lymphoid organs, such as the spleen and lymph nodes, normally reliably protect our body from infectious diseases; however, in rheumatoid arthritis they infiltrate tissues and substantially contribute to tissue destruction in rheumatic joints by production of inflammatory chemokines and autoreactive antibodies. It was previously unclear whether these lymphocytes infiltrate tissues as fully differentiated effector cells from neighboring lymph nodes or whether they are locally generated. A recent study has now shown that T and B cells actively cooperate togeth

[Peculiarities of dephosphorylation reaction catalyzed by purified protein tyrosine phosphatase CD45 from thymocytes of intact and exposed to ionising radiation rats].

This paper is devoted to analysis of parameters of catalytical activity of CD45, the major transmembrane proteintyrosine phosphatase (PTP-ase) of the lymphocytes, isolated from plasma membranes of thymocytes of control and 0.5 Gy irradiated rats. CD45 catalytic features were evaluated using 0.2 mM sodium vanadate as the inhibitor and paranitrophenylphosphate (1-8 mM) and phosphotyrosine (1-6 mM) as, respectively, nonspecific and specific substrates. With the former, irradiation was shown to cause a decrease in Vmax but an increase in affinity. With phosphotyrosine both Vmax and affinity decrea

Role of complement in induction of antibody production in vivo. Effect of cobra factor and other C3-reactive agents on thymus-dependent and thymus-independent antibody responses.

In an in vivo study in mice, suppression by the C3-cleaving protein of cobra venom (CoF), and other C3-reactive agents (zymosan, aggregated IgG, anti-C3 antibodies, and type III pneumococcal polysaccharide) of the thymus-dependent antibody responses to sheep erythrocytes, ovalbumin, and human IgG was demonstrated. The thymus-independent antibody response to polyvinyl-pyrrolidone was however unaffected by CoF. These and other published observations suggest that there may be a requirement for functional C3 in induction of thymus-dependent but not thymus-independent antibody production. A model f

Physicochemical characterization and immunomodulatory activity of sulfated Chinese yam polysaccharide.

A sulfated Chinese yam polysaccharide (S-CYP) was obtained from non-starch Chinese yam polysaccharide (CYP) by chlorosulfonic acid-pyridine (CSA-Pyr) method. The physicochemical characteristics and immunological activity of S-CYP were evaluated in this study. Results showed that sulfated modification changed the physicochemical properties of Chinese yam polysaccharide. Body weights and thymus index of the mice in polysaccharides group were increased compared with the cyclophosphamide (Cy)-induced immunosuppressed mice in model group, and could restore the splenomegaly that caused by Cy. Immunoregulatory defects in a family with selective IgA deficiency.

Defects in regulation of the humoral immune system were examined in a family with selective IgA deficiency. Two patients (mother and son) were clinically well and had selective abnormalities of B lymphocytes that can differentiate into IgA plasma cells plus specific T cell suppression of IgA production. One patient (daughter) had several clinical problems and received nine monthly transfusions of normal plasma. Prior to therapy, this patient had abnormal immunoregulation of both B and T lymphocyte population that cooperated in the formation of all cases of immunoglobulin-producing cells. After

Cyclosporin A promotes spontaneous outgrowth in vitro of Epstein-Barr virus-induced B-cell lines.

Cyclosporin A (CSA) is a fungal metabolite which exerts profound effects on the immune system and has potential as a selective immuno-suppressive agent. Clinical trials with human renal allograft recipients have confirmed this potential but there have been disturbing reports of lymphoma in a significant number of patients. Despite extensive animal studies, the specificity of this drug for human lymphocyte subpopulations is largely unknown. We demonstrate here that in vitro CSA has no apparent effect on Epstein-Barr virus (EBV)-induced B-lymphocyte activation, but totally inhibits the T-cell de

[The structure of immunoregulatory cells as a reflection of their interaction in infectious diseases].

Analysis of the structure of immunoregulatory cells (IRC) as the reflection of cell interaction in healthy persons and in patients with typhoid fever, tick-borne encephalitis, or chronic opisthorchiasis has shown that the characteristics under study (information entropy, structural information, the coefficient of ecological-genetic correspondence) are highly sensitive and specific, which makes it possible to use them quite effectively for differentiating health from disease and for identifying this disease. The study of IRC structure as the reflection of cell interrelations helps evaluate the

Interaction of human thymus-derived and non-thymus-derived lymphocytes in vitro. Induction of proliferation and antibody synthesis in B lymphocytes by a soluble factor released from antigen-stimulated T lymphocytes.

Relatively pure populations of human T and B lymphocytes were obtained from blood and tonsils using density gradient centrifugation in bovine serum albumin. Antigen alone was incapable of triggering the B lymphocyte into blast transformation or to secrete antibody. However, supernatants from tetanus toxoid-stimulated T cells obtained from immune donors contained a factor mitogenic for B lymphocytes. 50-60% of B cells responded to this lymphocyte mitogenic factor (LMF) by proliferation, loss of C3 reactivity, and change to a secretory state. LMF-stimulated B cells exhibited a three- to fivefold

Lymphoid response of the burn patient.

Clinical observations have long suggested immunologic compromise in burned patients. Resolution of the immune system into cooperative dual components of T-cells mediating delayed hypersensitivity and B-cells mediating antibody responses prompted the present survey of T-cell and B-cell changes after acute thermal burn injury. Adults and children sustaining extensive burn injuries were studied for up to 60 days after injury. T-cell function was assessed by lymphocyte counts, in vitro lymphocyte synthesis of RNA and DNA with and without mitogenic stimulation, and lymphocyte response and stimulato

Mycosis fungoides associated with Mediterranean lymphoma.

Mycosis fungoides was observed in a 71-year-old male with Mediterranean lymphoma, a B-cell malignancy. It is proposed that this association is not incidental since hypergammaglobulinaemia and even monoclonal gammopathies have repeatedly been described in cutaneous T-cell lymphomas. Mediterranean lymphoma might have resulted from (a) helper cell activities of tumor T-lymphocytes, (b) common antigenic stimuli, or (c) deranged T-B cooperation due to concomitant mycosis fungoides.

Increased IgM B cell differentiation lymphokine production by T lymphocytes from patients with primary biliary cirrhosis.

Hypergammaglobulinemia, mainly due to increased serum immunoglobulin M concentrations, is a common and distinctive feature of primary biliary cirrhosis. T-B cell cooperation plays a pivotal role in the regulation of immunoglobulin secretion. In this paper, the production of regulatory B lymphokines by T cells, as well as the functional response of B lymphocytes to these molecules, was investigated in patients with primary biliary cirrhosis. T cells from patients with primary biliary cirrhosis have an enhanced ability to produce lymphokines that regulate the proliferation of B cells and their d

Trends in patterns of treatment of childhood cancer in Los Angeles County.

To assess the proportion of children with cancer who have been managed by mainstream pediatric cancer programs, population-based cancer incidence data for Los Angeles County (LAC) children (under 20 years of age) for the years 1972 through 1987 were linked with patient records of children registered with the two national cooperative pediatric oncology groups, Children's Cancer Study Group and Pediatric Oncology Group. The proportion of children with cancer who were registered by cooperative groups increased markedly over time: 9% of LAC children younger than 15 years of age who were diagnosed

The role of CD40L (gp39)/CD40 in T/B cell interaction and primary immunodeficiency.

The interaction between the CD40 ligand (gp39), expressed by activated T cells, and CD40, constitutively expressed by B cells, is critical for an effective antibody response to T cell dependent antigens. Patients with X-linked hyper IgM (HIM) syndrome fail to express a functional CD40 ligand due to a mutation within the gene for gp39. As a direct consequence, HIM patients, when immunized with T dependent antigens, produce only small amounts of IgM antibody without the development of immunologic memory, amplification and switch from IgM to IgG. Mutations affecting the gene for the HIM syndrome
