BACKGROUND: Environmentally sustainable management of plant fungal diseases requires green agrochemicals that combine high efficacy with novel modes-of-action. Rice blast caused by Magnaporthe oryzae is a destructive crop fungal disease, and developing botanical fungicides with new antifungal mechanisms is urgently needed for its green control.
RESULTS: In this study, we report that an ethanol-petroleum ether crude extract of Hydnocarpus hainanensis seeds (designated DF2) strongly suppresses infection of the rice blast fungus M. oryzae. Bioactivity-guided fractionation of DF2 identified hydno

The transcription factor FgSR functions as a central regulator of ergosterol biosynthesis and 14α-demethylation inhibitor (DMI) sensitivity in Fusarium graminearum, with its orthologous proteins being evolutionarily conserved within Sordariomycetes and Leotiomycetes fungi. In this study, we demonstrate that sterol regulators (SRs) differentially modulate DMI sensitivity across these fungal classes through divergent strategies involving key metabolic genes. In Magnaporthe oryzae, MoSR regulates DMI sensitivity through two distinct mechanisms: it directly modulates the transcription of ergostero

In our ongoing screening culture fluid extracts of Gloeoporus (Caloporus) dichrous strain 83065 inhibited the germination of Magnaporthe grisea and Fusarium graminearum spores. While isolating the active metabolites two new caloporosides, caloporoside G and caloporoside H, in addition to the known caloporoside derivatives F-16438G, caloporoside A, and 2-hydroxy-6-(16-hydroxyheptadecyl)benzoic acid were obtained.

We characterized the genetic structure of 609 strains of Pyricularia oryzae, the fungal pathogen causing rice blast disease, in three main regions in Vietnam using microsatellites (SSR) markers. From the 447 distinct multilocus genotypes identified, six genetic clusters were defined, all of them showing elevated genetic and genotypic diversities. Four of these clusters were related to rice-attacking lineages already described at the worldwide scale, whereas the two remaining clusters were endemic to Vietnam. Strains were unevenly distributed into the six clusters depending on their groups of r

Weeds, pests, and pathogens are among the pre-harvest constraints in rice farming across rice-growing countries. For weed management, manual weeding and herbicides are widely practiced. Among the herbicides, glyphosate [N-(phosphonomethyl) glycine] is a broad-spectrum systemic chemical extensively used in agriculture. Being a competitive structural analog to phosphoenolpyruvate, it selectively inhibits the conserved 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) enzyme required for the biosynthesis of aromatic amino acids and essential metabolites in eukaryotes and prokaryotes. Rice blast, caused by the filamentous ascomycete Pyricularia oryzae, is one of the most devastating diseases of rice. Four genetic clusters were previously identified, and three have a large geographic distribution. Asia is the center of diversity and the origin of most migrations to other continents, and sexual reproduction persisted only in the South China-Laos-North Thailand region, which was identified as the putative center of origin of all P. oryzae populations on rice. Despite the importance of rice blast disease, little is known about the diversity and the population structure of the p

BACKGROUND: The plant growth is influenced by multiple interactions with biotic (microbial) and abiotic components in their surroundings. These microbial interactions have both positive and negative effects on plant. Plant growth promoting bacterial (PGPR) interaction could result in positive growth under normal as well as in stress conditions.
METHODS: Here, we have screened two PGPR's and determined their potential in induction of specific gene in host plant to overcome the adverse effect of biotic stress caused by Magnaporthe grisea, a fungal pathogen that cause blast in rice. We demonstra

The rice-Magnaporthe oryzae pathosystem is presented as a host-pathogen ferroptosis model. Multi-omics insights connect ferroptosis to several cellular processes, such as iron homeostasis, reactive oxygen species (ROS)/redox imbalance, and polyunsaturated fatty acid-containing phospholipid (PUFA-PL) peroxidation, under multiple regulators in rice and M. oryzae, respectively. Interestingly, ferroptosis drives two opposite biological outcomes: fungal infection-related development and rice immunity.

Serendipita indica, a multifunctional and useful endophyte fungus, has been intensively investigated in promoting plant growth and resistance towards biotic and abiotic stress. Multiple chitinases from microorganisms or plants have been identified to have a high antifungal activity as a biological control. However, chitinase of S. indica still needs to be characterized. We functionally characterized a chitinase (SiChi) in S. indica. The result showed that the purified SiChi protein confers high chitinase activity; importantly, SiChi inhibits the conidial germination of Magnaporthe oryzae and F

Rice blast is a devastating disease worldwide, threatening rice production and food security. The blast fungus Magnaporthe oryzae invades the host via the appressorium, a specialized pressure-generating structure that generates enormous turgor pressure to penetrate the host cuticle. However, owing to ongoing evolution of fungicide resistance, it is vitally important to identify new targets and fungicides. Here, we show that Trs85, a subunit of the transport protein particle III complex, is essential for appressorium-mediated infection in M. oryzae. We explain how Trs85 regulates autophagy thro

Vitamin B(1) (thiamine) is an essential nutrient for humans. Vitamin B(1) deficiency causes beriberi, which disturbs the central nervous and circulatory systems. In countries in which rice (Oryza sativa) is a major food, thiamine deficiency is prevalent because polishing of rice removes most of the thiamine in the grain. We demonstrate here that thiamine, in addition to its nutritional value, induces systemic acquired resistance (SAR) in plants. Thiamine-treated rice, Arabidopsis (Arabidopsis thaliana), and vegetable crop plants showed resistance to fungal, bacterial, and viral infections. Thi

Bacillus velezensis YL2021 has extensive antimicrobial activities against phytopathogens, and its genome harbors a catechol-type siderophore biosynthesis gene cluster. Here, we describe the characterization of siderophores produced by strain YL2021 and its antimicrobial activity in vitro and in vivo. A few types of siderophores were detected by chrome azurol S plates coupled with Arnow's test, purified, and identified by reversed-phase high-performance liquid chromatography. We found that strain YL2021 can produce different antimicrobial compounds under low-iron M9 medium or iron-sufficient Lu

Blast disease is a notorious fungal disease leading to dramatic yield losses on major food crops such as rice and wheat. The causal agent, Pyricularia oryzae, encompasses different lineages, each having a different host range. Host shifts are suspected to have occurred in this species from Setaria spp. to rice and from Lolium spp. to wheat. The emergence of blast disease on maize in Iran was observed for the first time in the north of the country in 2012. We later identified blast disease in two additional regions of Iran: Gilan in 2013 and Golestan in 2016. Epidemics on the weed barnyard gras

Genetic transformation plays an imperative role in our understanding of the biology in unicellular yeasts and filamentous fungi, such as Saccharomyces cerevisiae, Aspergillus nidulans, Cryphonectria parasitica, and Magnaporthe oryzae. It also helps to understand the virulence and drug resistance mechanisms of the pathogenic fungus Cryptococcus that causes cryptococcosis in health and immunocompromised individuals. Since the first attempt at DNA transformation in this fungus by Edman in 1992, various methods and techniques have been developed to introduce DNA into this organism and improve the

Plant diseases cause enormous economic losses in agriculture and threaten global food security, and application of agrochemicals is an important method of crop disease control. Exploration of disease-resistance mechanisms and synthesis of highly bioactive agrochemicals are thus important research objectives. Here, we show that propranolol, a phosphatidate phosphatase (Pah) inhibitor, effectively suppresses fungal growth, sporulation, sexual reproduction, and infection of diverse plants. The MoPah1 enzyme activity of the rice blast fungus Magnaporthe oryzae is inhibited by propranolol. Alterati

We examined the genomes of 100 isolates of Magnaporthe oryzae (Pyricularia oryzae), the causal agent of rice blast disease. We grouped current field populations of M. oryzae into three major globally distributed groups. A genetically diverse group, clade 1, which may represent a group of closely related lineages, contains isolates of both mating types. Two well-separated clades, clades 2 and 3, appear to have arisen as clonal lineages distinct from the genetically diverse clade. Examination of genes involved in mating pathways identified clade-specific diversification of several genes with ort

Rice blast is a highly destructive crop disease that requires the interplay of three essential factors: the virulent blast fungus, the susceptible rice plant, and favorable environmental conditions. Although previous studies have focused mainly on the pathogen and rice, recent research has shed light on the molecular mechanisms by which the blast fungus and environmental conditions regulate host resistance and contribute to blast disease outbreaks. This review summarizes significant achievements in understanding the sophisticated modulation of blast resistance by Magnaporthe oryzae effectors a

Rice blast, caused by Magnaporthe oryzae, remains a major constraint to rice (Oryza sativa L.) production, and durable resistance will require broader use of genetic diversity beyond commonly studied elite cultivars. We present chromosome-scale genome assemblies of two agronomically significant but genetically divergent rice cultivars: CT21375-F4-4 (hereafter, CT21) and Moroberekan. Despite their genetic divergence, both cultivars show consistently strong resistance to blast under multiyear field conditions. We combined field phenotyping with genome-wide protein-to-genome alignment, predicted

Bacterial blight and blast, caused by Xanthomonas oryzae pv. oryzae and Magnaporthe oryzae, respectively, are the two most devastating diseases of rice (Oryza sativa L.). Although the diseases were first documented in the 17th-19th centuries, systematic research on their pathology and host resistance began in the early 20th century and was significantly accelerated following the development of near-isogenic lines (NILs) carrying single resistance genes in universally susceptible backgrounds in the 1980s. Sets of NILs for bacterial blight (IRBB) and International Rice Research Institute (IRRI)-

Plants encounter various fungi during their life history, with some of them pathogenic, some beneficial, and some neither, forming complex plant-fungi interactions. Endophytic fungi are a type of fungi that live inside the plant, and are often beneficial to the host. In this study, the endophytic fungi of green foxtail (Setaria viridis) were explored for their defensive role against the pathogenic fungus Magnaporthe oryzae, which causes rice blast, one of the most devastating crop diseases. The endophytic fungi were isolated, taxonomically identified, fermented, and chemically extracted. The f
