Mechanisms of muscular electrophysiological and mitochondrial dysfunction following exposure to malathion, an organophosphorus pesticide.

Muscle dysfunction in acute organophosphorus (OP) poisoning is a cause of death in human. The present study was conducted to identify the mechanism of action of OP in terms of muscle mitochondrial dysfunction. Electromyography (EMG) was conducted on rats exposed to the acute oral dose of malathion (400 mg/kg) that could inhibit acetylcholinesterase activity up to 70%. The function of mitochondrial respiratory chain and the rate of production of reactive oxygen species (ROS) from intact mitochondria were measured. The bioenergetic pathways were studied by measurement of adenosine triphosphate (

Permethrin and malathion resistance in head lice: results of ex vivo and molecular assays.

BACKGROUND: Treatment of head lice infestation relies on the application of topical insecticides. Overuse of these products has led to the emergence of resistance to pyrethroids and malathion worldwide. Permethrin resistance in head lice is mostly conferred by the knockdown resistance (kdr) trait. OBJECTIVE: To evaluate the occurrence of permethrin- and malathion-resistant head lice in Paris. METHODS: A prospective survey was conducted in 74 elementary schools. Live lice collected on schoolchildren were randomly selected and submitted to ex vivo bioassays or underwent individual DNA extracti

Effects of malathion metabolites on degranulation of and mediator release by human and rat basophilic cells.

In the present study, the effects of malathion and malathion derivatives on histamine and beta-hexosaminidase release by RBL-1 cells, rat peritoneal mast cells (RPMC), and human peripheral blood basophils (HPBB) and cutaneous mast calls were examined. One hour of incubation of RBL-1 cells with all organophosphate compounds tested, except for malathion and malathion monoacid, led to an increase in histamine release. beta-Hexosaminidase, an enzyme released by basophilic cells and a biochemical marker of degranulation, was not released from RBL-1 cells after 1 h of exposure to organophosphate com

Impact of protein deficiency and exposure to hexachlorocyclohexane or malathion on lipid metabolism in pregnant rats.

Dietary intake of three oral doses of hexachlorocyclohexane (HCH) (60 mg/kg body wt) or malathion (500 mg/kg) by normal and protein-deficient diet fed pregnant rats on the 6th, 10th and 14th day of gestation resulted in the impairment of lipid metabolism, viz. hypercholesterolemia and hypertriglyceridemia. The cholesterol, triglyceride and phospholipid contents in serum, brain, liver, kidney and uterus were increased significantly by HCH and malathion exposure, irrespective of the protein content in the diet. The incorporation of [1,2-14C]acetate into the hepatic lipids was stimulated by both

A large-scale field trial of malathion as an insecticide for antimalarial work in Southern Uganda.

Malathion shows promise as a substitute for chlorinated-hydrocarbon insecticides in the control of malaria whenever the latter are unsuitable because of Anopheles resistance or other reasons. A field trial of malathion was carried out in 1963-64, covering an area of about 500 km(2) with a population of about 26 000, in Masaka District, southern Uganda. All houses and animal shelters were sprayed with malathion at 2 g/m(2) at roughly 4-month intervals. The average combined densities of the females of the two main malaria vectors, Anopheles funestus and An. gambiae, fell from an average of 66 pe

Potential role of a newly AChE reactivator in the depressive-like behavior induced by malathion.

AChE inhibition caused by exposure to organophosphate (OP) compounds is strongly related to behavioural disorders such as depression. Malathion is an OP that already has a relationship between its exposure and behavioural changes, although few data still have its effects in a longer exposure protocol. In addition, intoxication therapy is based on the use of atropine-oxime which still has its controversial efficacy depending on the type of compound. For this, (3Z)-5-Chloro-3-(hydroxyimino)indolin-2-one (Cℓ-HIN), a compound that has properties of isatin and oxime in its structure, have shown rea

In vitro selection of a single-stranded DNA molecular recognition element for the pesticide malathion.

Many large-scale applications of the organophosphate pesticide malathion have led to widespread environmental contamination. Concentrations are found in the environment well above those which are harmful to humans and environmental organisms. No current method of detection for this pesticide is rapid, cost-effective, and specific for malathion. Therefore, we utilized a stringent Systematic Evolution of Ligands by Exponential Enrichment (SELEX) process to identify a Molecular Recognition Element (MRE) for malathion. This MRE was identified from a large ssDNA library and has an equilibrium disso

Cross-resistance to insecticides in a malathion-resistant strain of Ceratitis capitata (Diptera: Tephritidae).

Resistance to malathion has been reported in field populations of the Mediterranean fruit fly, Ceratitis capitata (Wiedemann) (Diptera: Tephritidae), in areas of Spain where an intensive use of this insecticide was maintained for several years. The main goal of this study was to determine whether resistance to malathion confers cross-resistance to different types of insecticides. Susceptibility bioassays showed that the malathion-resistant W-4Km strain (176-fold more resistant to malathion than the susceptible C strain) has moderate levels of cross-resistance (three- to 16-fold) to other organ

Generation of a mutagenized organophosphorus hydrolase for the biodegradation of the organophosphate pesticides malathion and demeton-S.

AIMS: The bacterial organophosphorus hydrolase (OPH) enzyme hydrolyses and detoxifies a broad range of toxic organophosphate pesticides and warfare nerve agents by cleaving the various phosphorus-ester bonds (P-O, P-F, P-CN, P-S); however, OPH hydrolyses these bonds with varying efficiencies. The aim of this study was to generate a variant OPH enzyme with improved hydrolytic efficiency against the poorly hydrolysed P-S class of organophosphates. METHODS AND RESULTS: The gene encoding OPH was sequentially mutated at specific codons by saturation mutagenesis and screened for improved activity a

Malathion detoxification by human hepatic carboxylesterases and its inhibition by isomalathion and other pesticides.

The organophosphorothioate (OPT) pesticide malathion (MAL) in mammals is readily hydrolyzed by mammalian carboxylesterases (CE). The reaction competes with the CYP-catalyzed formation of malaoxon (MOX), the toxic metabolite. Alterations or individual variations in CE activity may result in increased MOX formation, enhancing MAL toxicity. We have characterized the human hepatic CE activity in a panel of 18 human liver microsomes as well as the inhibitory effect of IsoMAL, a major impurity of MAL commercial formulations, parathion (PAR), chlorpyrifos (CPF), and chlorpyrifos-oxon (CPFO). CE activ

Novel insecticide-removal activity against deltamethrin and malathion in a lithic-derived Modestobacter strain.

A large fraction of agricultural pesticides, including insecticides, fails to reach target organisms and instead persists in soil, where residues may accumulate and generate transformation products with unpredictable or enhanced toxicity. Therefore, sustainable mitigation strategies require not only efficient pesticide removal but also effective detoxification. Soil-dwelling bacteria, particularly actinobacteria, possess diverse enzymatic capabilities that can contribute to the transformation of these chemical compounds, thereby influencing their environmental fate. In this study, we evaluated

Formulation of water pollutant discharge limits for malathion based on nonsensitive aquatic organism protection.

The "Integrated Wastewater Discharge Standard" was implemented for water pollutant discharge in China's pesticide industry, which has no control requirements for particular wastewater pollutants in the industry. In the standard, certain pollutants discharge limits are limited strictly or loosely, resulting in practical management implementation difficulties. In view of the highly selective targeting characteristics of organic pesticide active ingredients in fungicides, insecticides, and herbicides, a method for deriving discharge limits based on the water quality criteria for pesticides for th

Effect of malathion on larval competition between Aedes albopictus and Aedes atropalpus (Diptera: Culicidae).

Aedes albopictus (Skuse) and Aedes atropalpus (Coquillett) (Diptera: Culicidae) are container-dwelling mosquito species that are well established in the eastern United States. Interspecific larval competition studies have shown Ae. albopictus to be a superior competitor over many species. A laboratory experiment was conducted in artificial containers to evaluate the effects of malathion on larval interactions between Ae. albopictus and Ae. atropalpus. The survivorship of Ae. albopictus increased with increasing Ae. atropalpus densities in control but decreased with increasing Ae. atropalpus de

In vitro dermal absorption of methyl salicylate, ethyl parathion, and malathion: first responder safety.

In vitro tests with fresh dermatomed (0.3 to 0.4 mm thick) female breast skin and one leg skin specimen were conducted in Bronaugh flow-through Teflon diffusion cells with three chemicals used to simulate chemical warfare agents: 14C-radiolabeled methyl salicylate (MES), ethyl parathion (PT), and malathion (MT), at three dose levels (2, 20, and 200 mM). Tests were conducted at a skin temperature of 29 degrees C using a brief 30-min exposure to the chemical and a 6.5-h receivor collection period. Rapid absorption of all three chemicals was observed, with MES absorbed about 10-fold faster than P

Investigation of the Protective Effect of Morin on Oxidative Stress and Apoptosis in Malathion-Induced Hepatotoxicity in Rats.

Malathion (MAL) is a widely used organophosphate pesticide associated with hepatotoxicity through oxidative stress and apoptosis. Morin, a naturally occurring flavonoid, exhibits potent antioxidant and anti-inflammatory properties. This study investigated oxidative stress, inflammatory, and apoptotic alterations induced by acute and subacute MAL exposure in rats and evaluated the potential protective effects of morin. Fifty 3-month-old male Sprague Dawley rats were randomly assigned to six groups: control, morin (200 mg/kg), subacute MAL (150 mg/kg), acute MAL (300 mg/kg), subacute MAL + morin

An electrochemiluminescence sensor for sensitive detection of malathion based on g-C3N4-CdTe composite nanomaterials.

A self-enhanced electrochemical luminescence (ECL) composite material g-C3N4-CdTe QDs was prepared. The combination of g-C3N4 and CdTe QDs can amplify the ECL signal and improve the stability. Based on this discovery, g-C3N4-CdTe QDs and acetylcholine esterase (AChE) were used to construct an ECL sensor for organophosphorus pesticides (OP) detection. The sensor showed a strong initial ECL signal in PBS containing S2O82-. It is because that g-C3N4 not only acts as a co-reaction promoter to amplify the ECL signal of the CdTe QDs/S2O82- system but also acts as a carrier with large specific surfac

The combination of acute pancreatitis and toxic hepatitis developing secondary to exposure to malathion : a case report.

Organophosphate(OPH) compounds are cholinesteraseinhibiting chemicals used as pesticide. Pancreatitis secondary to malathion toxicity is rare and toxic hepatitis has been reported in only one case. In this paper, we report the case of the combination of acute pancreatitis and toxic hepatitis, which developed in a 30-year old farm worker and the mechanism is discussed in this first report of its kind. Awareness of this complication should prompt earlier investigation because early diagnosis coupled with timely therapeutic measures may improve patient prognosis.
