Metabolite-Mediated Alleviation of Iron Deficiency and Growth Promotion of Malus hupehensis by Bacillus licheniformis LCDD6 in Calcareous Soil.

Calcareous soils are typically deficient in essential nutrients such as iron, phosphorus, and potassium, which frequently results in nutrient deficiency in fruit trees. Bacillus licheniformis LCDD6 markedly enhanced Malus hupehensis seedling growth and plant iron nutrition in calcareous soil. This study aimed to elucidate the mechanism underlying these beneficial effects of strain LCDD6 under iron deficiency. Transcriptomic analysis revealed that iron deficiency induced metabolic reprogramming in strain LCDD6, characterized by a significant upregulation of genes involved in the biosynthesis of

Molecular cloning and functional characterization of MhHEC2-like genes in Malus halliana reveals it enhances Fe (iron) deficiency tolerance.

The basic helix-loop-helix (bHLH) family, as one of the largest families of transcription factors (TFs) in plants, plays crucial roles in regulating growth, development, and abiotic stress responses. However, studies on the association of the bHLH genes with apple iron (Fe) deficiency are limited. Here, multiple bHLH genes that responded to Fe deficiency were selected for quantitative real-time PCR in Malus halliana. The results showed that the expression of HEC2-like gene exerted more values compared to other genes under Fe deficiency stress, but the mechanism by which it regulates Fe deficie

Overexpression of a Novel Apple NAC Transcription Factor Gene, MdNAC1, Confers the Dwarf Phenotype in Transgenic Apple (Malus domestica).

Plant height is an important trait for fruit trees. The dwarf characteristic is commonly associated with highly efficient fruit production, a major objective when breeding for apple (Malus domestica). We studied the function of MdNAC1, a novel NAC transcription factor (TF) gene in apple related to plant dwarfing. Localized primarily to the nucleus, MdNAC1 has transcriptional activity in yeast cells. Overexpression of the gene results in a dwarf phenotype in transgenic apple plants. Their reduction in size is manifested by shorter, thinner stems and roots, and a smaller leaf area. The transgeni

Cold-responsive interaction between MdRAD23D1 and MdMYB15 confers cold stress tolerance via the CBF pathway in apple (Malus domestica).

Low temperature is a major environmental factor that impairs plant growth and development, posing a significant threat to crop yield and quality. RAD23 (RADIATION SENSITIVE23) proteins belong to the UBL-UBA (Uiquitin-like-ubiquitin associated) family and function as shuttle factors in the UPS (ubiquitin proteasome system). Although UBL-UBA proteins are known regulators of plant stress responses, the function and mechanism of RAD23 in apple under cold stress are poorly understood. Here, we demonstrated that MdRAD23D1 is induced by 4 °C and positively regulates cold tolerance. Silencing MdRAD23D

Identification of sugar transporter MrSTP20 responding to high-salt stress and hexoses induction in apple rootstock (Malus robusta Rehd).

BACKGROUND: Sugar distribution regulation, especially in the roots, is an important pathway for plants to cope with abiotic stress. Sugar transport proteins (STPs) mediate the transmembrane transport of hexoses such as glucose and fructose; however, the molecular mechanism underlying the response to salt stress remains unclear. RESULTS: Here, we identified 22 STP genes in apple (Malus domestica) using genome-wide screening. Under salt stress, 17 genes were detected in the roots of the apple rootstock Malus robusta Rehd; of them, MrSTP3, MrSTP7, MrSTP14, and MrSTP20 showed stronger positive re

Malus rootstocks affect copper accumulation and tolerance in trees by regulating copper mobility, physiological responses, and gene expression patterns.

We investigated the roles of rootstocks in Cu accumulation and tolerance in Malus plants by grafting 'Hanfu' (HF) scions onto M. baccata (Mb) and M. prunifolia (Mp) rootstocks, which have different Cu tolerances. The grafts were exposed to basal or excess Cu for 20 d. Excess Cu-treated HF/Mb had less biomass, and pronounced root architecture deformation and leaf ultrastructure damage than excess Cu-challenged HF/Mp. Root Cu concentrations and bio-concentration factor (BCF) were higher in HF/Mp than HF/Mb, whereas HF/Mb had higher stem and leaf Cu concentrations than HF/Mp. Excess Cu lowered ro

H2S pretreatment mitigates the alkaline salt stress on Malus hupehensis roots by regulating Na+/K+ homeostasis and oxidative stress.

Hydrogen sulfide (H2S) plays an important role in the plant salt stress response. The main component of salt stress is neutral salt (NaCl); NaHCO3 and Na2CO3 play a key role in soil alkaline due to the influence of pH. Malus hupehensis Rehd. var. pingyiensis Jiang (Pingyi Tiancha, PYTC) is a salt-sensitive apple rootstock. Seedlings of PYTC pretreated with NaHS (an H2S donor) were exposed to an alkaline salt solution, and then the plant growth, root architecture, oxidative damage, Na+/K+ homeostasis and gene expression of MhSOS1 and MhSKOR were investigated. The results showed that NaHS pretre

Expansion to new habitats and a new commercial host (Malus domestica) by Anastrepha ludens (Tephritidae) likely influenced by global warming.

Anastrepha ludens (Mexican fruit-fly) is a highly polyphagous fruit fly species (Tephritidae) attacking wild and commercial fruit from Mexico to Panama. Here we report on a recent habitat and host range expansion as A. ludens lately started to attack apples (Malus domestica) in Mexico, a phenomenon likely influenced by global warming. We document natural infestations in apple-growing regions in the States of Nuevo León and Hidalgo, Mexico where A. ludens has started to attack the cultivars 'Golden Delicious', 'Rayada' and 'Criolla'. No infestations were found in the apple-growing region of Zac

T3SS-dependent differential modulations of the jasmonic acid pathway in susceptible and resistant genotypes of Malus spp. challenged with Erwinia amylovora.

Fire blight is a bacterial disease of Maloideae caused by Erwinia amylovora (Ea). This necrogenic enterobacterium uses a type III secretion system (T3SS) to inject type III effectors into the plant cells to cause disease on its susceptible hosts, including economically important crops like apple and pear. The expressions of marker genes of the salicylic acid (SA) and jasmonic acid (JA) defense regulation pathways were monitored by RT-qPCR in leaves of two apple genotypes, one susceptible and one resistant, challenged with a wild type strain, a T3SS-deficient strain or water. The transcriptiona

Identification of Dirigent (DIR) genes in apple (Malus domestica) displays the role of MdDIR7 in iron (Fe)-deficiency stress tolerance.

The overexpression of MdDIR7 can alleviate osmotic and oxidative damage, increase the expression of MdFRO2 and MdIRT1, and improve plant performance under Fe-deficient conditions. Iron (Fe) is an essential trace element that serves as a critical component in numerous cellular processes, such as photosynthesis, respiration, and nitrogen-sulfur assimilation. Fe deficiency commonly occurs in alkaline and calcareous soils worldwide and severely affects plant growth and development. Members of the Dirigent (DIR) family have been identified and shown to play a role in abiotic stress response in seve

Genome-Wide Identification of the Ca2+-ATPase Gene Family and Functional Analysis of MdACA39 in Resistance to Alternaria alternata in Malus domestica.

The calcium ion-transporting ATPase (Ca2+-ATPase) gene family maintains plant intracellular Ca2+ homeostasis and regulates growth, development and stress immunity; however, its functions remain poorly characterized in Malus domestica. Here, we performed a genome-wide identification of apple Ca2+-ATPase genes and obtained 45 members, which were classified into MdACA (39) and MdECA (6) subfamilies and unevenly distributed on 14 chromosomes. Phylogenetic analysis of Ca2+-ATPase genes from Malus domestica, Arabidopsis thaliana, and Oryza sativa classified these proteins into five subgroups. The AC

Addition of Phenylboronic Acid to Malus domestica Pollen Tubes Alters Calcium Dynamics, Disrupts Actin Filaments and Affects Cell Wall Architecture.

A key role of boron in plants is to cross-link the cell wall pectic polysaccharide rhamnogalacturonan-II (RG-II) through borate diester linkages. Phenylboronic acid (PBA) can form the same reversible ester bonds but cannot cross-link two molecules, so can be used as an antagonist to study the function of boron. This study aimed to evaluate the effect of PBA on apple (Malus domestica) pollen tube growth and the underlying regulatory mechanism. We observed that PBA caused an inhibition of pollen germination, tube growth and led to pollen tube morphological abnormalities. Fluorescent labeling, co
