Isolation and sequencing of insecticidal peptides from the primitive hunting spider, Plectreurys tristis (Simon).

About 50 peptide toxins were purified from venom of the primitive hunting spider, Plectreurys tristis. Bioassay by injection into larval Heliothis virescens (tobacco budworm) allowed selection of nine toxins for analysis of amino acid sequences. Total sequences were determined for six of the more insecticidal peptides (46-49 amino acids) and three contained free carboxyl-terminal amino acids. These toxins (plectoxins) are paralytic and/or lethal when injected into insect pests such as the lepidopterous larvae H. virescens, Spodoptera exigua (beet armyworm), and Manduca sexta (tobacco hornworm)

Purification and characterization of two classes of neurotoxins from the funnel web spider, Agelenopsis aperta.

Two classes of paralytic toxins were isolated from the venom of Agelenopsis aperta and their chemical and larvicidal properties characterized. Five acylpolyamine toxins (alpha-agatoxins) of molecular masses 452, 488, 489, 504, and 505 Da produce immediate but reversible paralysis in Manduca sexta following injection. Six insecticidal peptides (mu-agatoxins) produce a gradual but irreversible paralysis. The complete amino acid sequences (36-38 residues) of the mu-agatoxins are presented. These peptides contain eight half-cystines and are quite similar in sequence. At least four of these toxins

Architecture of the insect tracheal system driven by spatially varying limitation of oxygen and carbon dioxide transport.

Insects transport respiratory gases through a system of air-filled tubes (tracheae) that branch extensively to reach individual cells. A century of research has focused primarily on how tracheal systems deliver oxygen, often overlooking the complementary challenge of removing carbon dioxide. Here, we develop and analyse a model of simultaneous O₂ and CO₂ transport, which we parametrize with morphological and metabolic data. The model reveals a fundamental asymmetry: oxygen transport is most limited by the tissue gap between tracheoles and mitochondria; carbon dioxide transport, by contrast, is

Hawkmoths regulate flight torques with their abdomen for yaw control.

Many animals use body parts such as tails to stabilize posture while moving at high speed. In flying insects, leg or abdominal inertia can influence flight posture. In the hawkmoth Manduca sexta, the abdomen contributes ∼50% of the total body weight and it can therefore serve to inertially redirect flight forces. How do torques generated by the wings and abdomen interact for flight control? We studied the yaw optomotor response of M. sexta by using a torque sensor attached to their thorax. In response to yaw visual motion, the abdomen moved antiphase with the stimulus, head and total torque. B

Structural damping renders the hawkmoth exoskeleton mechanically insensitive to non-sinusoidal deformations.

Muscles act through elastic and dissipative elements to mediate movement, which can introduce dissipation and filtering which are important for energetics and control. The high power requirements of flapping flight can be reduced by an insect's exoskeleton, which acts as a spring with frequency-independent material properties under purely sinusoidal deformation. However, this purely sinusoidal dynamic regime does not encompass the asymmetric wing strokes of many insects or non-periodic deformations induced by external perturbations. As such, it remains unknown whether a frequency-independent m

Artificial nighttime lighting impacts visual ecology links between flowers, pollinators and predators.

The nighttime environment is being altered rapidly over large areas worldwide through introduction of artificial lighting, from streetlights and other sources. This is predicted to impact the visual ecology of many organisms, affecting both their intra- and interspecific interactions. Here, we show the effects of different artificial light sources on multiple aspects of hawkmoth visual ecology, including their perception of floral signals for pollination, the potential for intraspecific sexual signalling, and the effectiveness of their visual defences against avian predators. Light sources fal

Comparative properties of mammalian and insect carbonic anhydrases: effects of potassium and chloride on the rate of carbon dioxide hydration.

1. Carbonic anhydrase (E.C.4.2.1.1) catalysed CO2 hydration was studied with enzymes from mammalian and insect sources at CO2 concentrations of 7.6-30.8 mM. 2. At 0.01-0.15 M, potassium chloride (KCl) or choline chloride (ChCl) markedly inhibited all 8 mammalian enzymes studied. 3. Inhibition by KCl is always greater than that associated with ChCl. 4. KCl non-competitively inhibits and choline chloride competitively inhibits bovine carbonic anhydrase. 5. Carbonic anhydrase obtained from fat body, integumentary epithelium and midgut tissues of larval tobacco hornworms, Manduca sexta, is greatly

Characterization of morphological and biological aspects of venomous caterpillars of the genus Lonomia Walker (Lepidoptera: Saturniidae) in Colombia.

The genus Lonomia Walker, 1855 (Lepidoptera: Saturniidae) is of particular interest to the medical community, since the scoli of these caterpillars harbor a venom that induces hemorrhaging in humans. In Colombia, deadly encounters with Lonomia achelous (Cramer, 1777), have been reported since 2000. There is little information on the main biological and ecological aspects of this genus to help better understand and develop prevention strategies. This study aimed to describe morphological and biological aspects (especially of immature stages) of four recently reported species of Lonomia in Colom

3D reconstruction of larval and adult brain neuropils of two giant silk moth species: Hyalophora cecropia and Antheraea pernyi.

We present a detailed analysis of the brain anatomy of two saturniid species, the cecropia silk moth, Hyalophora cecropia, and the Chinese oak silk moth, Antheraea pernyi, including 3D reconstructions of the major brain neuropils in the larva and in male and female adults. The 3D reconstructions, prepared from high-resolution optical sections, showed that the corresponding neuropils of these saturniid species are virtually identical. Similarities between the two species include a pronounced sexual dimorphism in the adults in the form of a male-specific assembly of markedly enlarged glomeruli f

Tobacco mosaic virus inoculation inhibits wound-induced jasmonic acid-mediated responses within but not between plants.

Jasmonic acid (JA) and salicylic acid (SA) have both been implicated as important signal molecules mediating induced defenses of Nicotiana tabacum L. against herbivores and pathogens. Since the application of SA to a wound site can inhibit both wound-induced JA and a defense response that it elicits, namely nicotine production, we determined if tobacco mosaic virus (TMV) inoculation, with its associated endogenous systemic increase in SA, reduces a plant's ability to increase JA and nicotine levels in response to mechanical damage, and evaluated the consequences of these interactions for the a

Identification of mosquito sterol carrier protein-2 inhibitors.

A mosquito sterol carrier protein-2, AeSCP-2, has been shown to aid in the uptake of cholesterol in mosquito cells. The discovery of chemical inhibitors of AeSCP-2 is reported here. AeSCP-2 inhibitors (SCPIs) belong to several chemotypes of hydrophobic compounds. Those inhibitors competed with cholesterol for AeSCP-2, binding with relatively high binding affinities. In cultured insect cells, SCPIs reduced cholesterol uptake by as much as 30% at 1-5 microM concentrations. SCPIs were potent larvicides to the yellow fever mosquito, Aedes aegypti, and to the tobacco hornworm, Manduca sexta, with 5

Inbreeding increases susceptibility to powdery mildew (Oidium neolycopersici) infestation in horsenettle (Solanum carolinense L).

BACKGROUND: While Tai Chi Chuan (TCC) is widely recognized for its physical and mental health benefits, its mindful components are often overlooked in traditional training. Mindfulness-enhanced Tai Chi Chuan (MTCC) has been proposed to address this limitation, yet little research has explored its long-term impact on exercise motivation and physical fitness. This study extends previous findings by investigating the sustained effects of a 24-week MTCC intervention in improving physical and mental health outcomes among college students, with a focus on long-term sustainability and motivation main

Metarhizium fight club: Within-host competitive exclusion and resource partitioning.

Both Metarhizium robertsii ARSEF 2575 (Mr2575) and Metarhizium anisopliae ARSEF 549 (Ma549) infect a range of insects whilst also interacting with plants; however, little is known about the traits that affect the competitive ability of different strains. We examined the interactions between Mr2575 and Ma549 in culture and during co-infection of plants (Arabidopsis thaliana) and insects. Mr2575 outcompetes Ma549 under nutrient-limiting conditions, including root exudates, giving it a priority advantage on Arabidopsis roots. However, during co-infection of Manduca sexta or Drosophila melanogaste

Instability in a coiled-coil signaling helix is conserved for signal transduction in soluble guanylyl cyclase.

How nitric oxide (NO) activates its primary receptor, α1/β1 soluble guanylyl cyclase (sGC or GC-1), remains unknown. Likewise, how stimulatory compounds enhance sGC activity is poorly understood, hampering development of new treatments for cardiovascular disease. NO binding to ferrous heme near the N-terminus in sGC activates cyclase activity near the C-terminus, yielding cGMP production and physiological response. CO binding can also stimulate sGC, but only weakly in the absence of stimulatory small-molecule compounds, which together lead to full activation. How ligand binding enhances cataly

An integrated analysis of phenotypic selection on insect body size and development time.

Most studies of phenotypic selection do not estimate selection or fitness surfaces for multiple components of fitness within a unified statistical framework. This makes it difficult or impossible to assess how selection operates on traits through variation in multiple components of fitness. We describe a new generation of aster models that can evaluate phenotypic selection by accounting for timing of life-history transitions and their effect on population growth rate, in addition to survival and reproductive output. We use this approach to estimate selection on body size and development time f

The flying insect thoracic cuticle is heterogenous in structure and in thickness-dependent modulus gradation.

The thorax is a specialized structure central to insect flight. In the thorax, flight muscles are surrounded by a thin layer of cuticle. The structure, composition, and material properties of this chitinous structure may influence the efficiency of the thorax in flight. However, these properties, as well as their variation throughout the thorax and between insect taxa, are not known. We provide a multi-faceted assessment of thorax cuticle for fliers with asynchronous (honey bee; Apis mellifera) and synchronous (hawkmoth; Manduca sexta) muscles. These muscle types are defined by the relationshi

Silverleaf Nightshade (Solanum elaeagnifolium) Undergoes Guild-Specific Transcriptomic Reprogramming Against Chewing Versus Piercing Sucking Herbivory.

Solanum elaeagnifolium (silverleaf nightshade, SLN) is a globally invasive perennial weed with reproductive plasticity and complex defence systems that significantly impact crop yields by invading fields of staple, commercial and ornamental crops. Despite being a host to multiple insect herbivores, the molecular programming enabling its rapid adaptation to defend against diverse feeding guilds remains poorly resolved. Here, we report a comprehensive de novo transcriptomic assembly and comparative analysis of S. elaeagnifolium challenged by two distinct herbivores: the piercing-sucking generali

An enteric ultrastructural surface atlas of the model insect Manducasexta.

The tobacco hornworm is a laboratory model that is particularly suitable for analyzing gut inflammation, but a physiological reference standard is currently unavailable. Here, we present a surface atlas of the healthy hornworm gut generated by scanning electron microscopy and nano-computed tomography. This comprehensive overview of the gut surface reveals morphological differences between the anterior, middle, and posterior midgut, allowing the screening of aberrant gut phenotypes while accommodating normal physiological variations. We estimated a total resorptive midgut surface of 0.42 m2 for

Vacuolar-type proton pumps in insect epithelia.

Active transepithelial cation transport in insects was initially discovered in Malpighian tubules, and was subsequently also found in other epithelia such as salivary glands, labial glands, midgut and sensory sensilla. Today it appears to be established that the cation pump is a two-component system of a H(+)-transporting V-ATPase and a cation/nH(+) antiporter. After tracing the discovery of the V-ATPase as the energizer of K(+)/nH(+) antiport in the larval midgut of the tobacco hornworm Manduca sexta we show that research on the tobacco hornworm V-ATPase delivered important findings that emer

Flexible strategies for flight control: an active role for the abdomen.

Moving animals orchestrate myriad motor systems in response to multimodal sensory inputs. Coordinating movement is particularly challenging in flight control, where animals deal with potential instability and multiple degrees of freedom of movement. Prior studies have focused on wings as the primary flight control structures, for which changes in angle of attack or shape are used to modulate lift and drag forces. However, other actuators that may impact flight performance are reflexively activated during flight. We investigated the visual-abdominal reflex displayed by the hawkmoth Manduca sext
