Manganese Is Essential for Neuronal Health.

The understanding of manganese (Mn) biology, in particular its cellular regulation and role in neurological disease, is an area of expanding interest. Mn is an essential micronutrient that is required for the activity of a diverse set of enzymatic proteins (e.g., arginase and glutamine synthase). Although necessary for life, Mn is toxic in excess. Thus, maintaining appropriate levels of intracellular Mn is critical. Unlike other essential metals, cell-level homeostatic mechanisms of Mn have not been identified. In this review, we discuss common forms of Mn exposure, absorption, and transport v

Modulation of cholinergic systems by manganese.

Information on changes in the central nervous system (CNS) cholinergic systems following exposure to manganese are considerably less extensive than that associated with other neurotransmitter systems. However, experimental and clinical evidence support the notion that cholinergic activity plays a key role in the pathophysiology of manganese-induced neurotoxicity. Manganese acts as a chemical stressor in cholinergic neurons in a region-specific manner causing breakdown of the cellular homeostatic mechanisms. In fact, a number of cholinergic synaptic mechanisms are putative targets for manganese

[The role of manganese in the human organism].

Manganese is an essential element to be found in all tissues. It is needed for metabolism of amino acids, lipids, proteins and carbohydrates. Manganese takes part directly in the defense of red blood cells, in the metabolism of iron, in the synthesis of cholesterol of the membranes and oligosaccharides. Manganese plays significant role in the free radical defense system as MnSOD, which protects the endothelial and red blood cells and mitochondria from the damage caused by superoxide radicals. The metabolism of manganese (absorption, transport) is determined by the chemical form and quantity of

Peripheral biomarkers and exposure to manganese.

Biochemical mechanisms underlying manganese (Mn) toxicity include dopamine (DA) auto-oxidation and free radical generation with subsequent neuronal damage. A neuroendocrine approach based on the measurement of serum prolactin (PRL) has been proposed to assess the tonic inhibition of pituitary lactotrope cells by the tubero-infundibular DA system. Low level exposure to Mn oxides in industrial settings is associated with a shift in the distribution of serum PRL towards higher levels as compared to matched controls. The follow-up of a small cohort of workers from a ferro-manganese plant showed th

Manganese and Movement Disorders: A Review.

Scientific and technological advances achieved with industrial expansion have led to an ever-increasing demand for heavy metals. This demand has, in turn, led to increased contamination of soil, water and air with these metals. Chronic exposure to metals may be detrimental not only to occupational workers but also to the nonoccupational population exposed to these metals. Manganese (Mn), a commonly used heavy metal, is an essential cofactor for many enzymatic processes that drive biological functions. However, it is also a potential source of neurotoxicity, particularly in the field of movemen

Manganese-enhanced MRI of the myocardium.

Gadolinium-based contrast media are widely used in cardiovascular MRI to identify and to highlight the intravascular and extracellular space. After gadolinium, manganese has the second highest paramagnetic moment and was one of the first MRI contrast agents assessed in humans. Over the last 50 years, manganese-enhanced MRI (MEMRI) has emerged as a complementary approach enabling intracellular myocardial contrast imaging that can identify functional myocardium through its ability to act as a calcium analogue. Early progress was limited by its potential to cause myocardial depression. To overcom

Prions and manganese: A maddening beast.

The prion protein is well known because of its association with prion diseases. These diseases, which include variant CJD, are unusual because they are neurodegenerative diseases that can be transferred between individuals experimentally. The prion protein is also widely known as a copper binding protein. The binding of copper to the prion protein is possibly necessary for its normal cellular function. The prion protein has also been suggested to bind other metals, and among these, manganese. Despite over ten years of research on manganese and prion disease, this interaction has often been dis

Neuropsychiatric effects of manganese on mood.

Adverse mood effects of overexposure to Manganese (Mn) have been described in 15 studies which frequently report an association of Mn exposure with adverse effects in six dimensions of mood: 1) anxiety, nervousness, irritability; 2) psychotic experiences; 3) emotional disturbance; 4) fatigue, lack of vigor, sleep disturbance; 5) impulsive/compulsive behavior; 6) aggression hostility. Only 1:15 studies used a standardized psychological measure of mood, while the current study of environmental Mn exposure used two standardized mood scales in evaluating low levels of Mn exposure and mood sequelae

Simultaneous removal of nitrate, manganese, zinc, and bisphenol a by manganese redox cycling system: Performance and mechanism.

The manganese(Mn) redox cycling system in this work was created by combining Mn(IV)-reducing bacteria MFG10 with Mn(II)-oxidizing bacteria HY129. The biomanganese oxides (BMO) generated by strain HY129 were transformed by strain MFG10 to Mn(II), finishing the Mn redox cycling, in which nitrate (NO3--N) was converted to nitrite, which was further reduced to nitrogen gas. The system could achieve 85.7 % and 98.8 % elimination efficiencies of Mn(ⅠⅠ) and NO3--N, respectively, at Mn(ⅠⅠ) = 20.0 mg/L, C/N = 2.0, pH = 6.5, and NO3--N = 16.0 mg/L. The removal of bisphenol A (BPA) and zinc (Zn(II)) at 3

Blood manganese and non-alcoholic fatty liver disease in a high manganese exposure area in China.

BACKGROUND AND AIMS: Manganese (Mn) deficiency and intoxication may affect nonalcoholic fatty liver disease (NAFLD) risk differently. We aimed to explore the association between blood Mn and NAFLD in an area with high Mn exposure in drinking water. METHODS: We conducted a case-control study among 1407 patients with NAFLD and 1774 sex- and age-matched healthy controls in a physical examination population in Zhoushan hospital, Zhejiang province in China. We used the restricted cubic splines method to investigate the dose-response relationship. Logistic regression models were applied to determin

Tuning the redox properties of manganese(II) and its implications to the electrochemistry of manganese and iron superoxide dismutases.

Superoxide dismutases (SODs) catalyze the disproportionation of superoxide to dioxygen and hydrogen peroxide. The active metal sites of iron and manganese superoxide dismutases are structurally indistinguishable from each other. Despite the structural homology, these enzymes exhibit a high degree of metal selective activity suggesting subtle redox tuning of the active site. The redox tuning model, however, up to now has been challenged by the existence of so-called cambialistic SODs that function with either metal ion. We have prepared and investigated two sets of manganese complexes in which

Activity-induced manganese-dependent functional MRI of the rat visual cortex following intranasal manganese chloride administration.

Manganese-enhanced MRI (MEMRI) of the brain requires delivery of manganese into the target brain regions. It was previously shown that, following intranasal application, ongoing olfactory stimulation facilitates manganese transport along the olfactory nerve into the olfactory bulb, so bypassing the blood-brain barrier (BBB). We report on experiments to evaluate whether visual stimulation can permit manganese transport onwards from the olfactory bulb to the visual cortex. Rats in intact olfactory bulb group were reserved intact olfactory bulb, while those in olfactory bulbectomy group received

Manganese inhibits poly(ADP-ribosyl)ation in human cells: a possible mechanism behind manganese-induced toxicity?

For humans manganese is both an essential trace element and, at higher doses, a toxic metal. Due to the ubiquitous occurrence of manganese in foodstuff, in industrial countries daily dietary uptake is higher as compared to the estimated daily requirement. Therefore manganese deficiency is extremely rare. In contrast chronic manganese toxicity, affecting primarily the central nervous system, is more prevalent. Thus manganese occupational and dietary overexposure has been shown to cause progressive, permanent, neurodegenerative damage, resulting in syndromes similar to idiopathic Parkinson's dis

Changes in brain regional manganese and magnesium levels during postnatal development: modulations by chronic manganese administration.

Manganese (Mn) and magnesium (Mg) levels in hypothalamus, cerebellum, pons and medulla, striatum, midbrain, and cerebral cortex of control and Mn-treated (10 mg MnCl2.4H2O per ml of drinking water) rats during postnatal development were studied using instrumental neutron activation analysis. The age-dependent Mn accumulation showed regional variation: at day 5, this accumulation was most marked in striatum (12.05 micrograms/g wet weight) but least marked in cerebral cortex (0.85 micrograms/g wet weight). By day 10, pons and medulla, and hypothalamus were regions with, respectively, the highest
