Adaptation and growth kinetics study of an Indian isolate of virulent duck enteritis virus in Vero cells.

Duck virus enteritis, also known as duck plague, is a viral infection of ducks caused by duck enteritis virus (DEV). The control of the disease is mainly done by vaccination with chicken embryo adapted live virus that is known to be poorly immunogenic and elicits only partial protection. Further, the embryo propagated vaccine virus pose a threat of harboring other infectious agents. Seeing these limitations, the present study reports for the first time regarding propagation and adaptation of a virulent Indian isolate of duck enteritis virus in Vero cell line. In this study isolation of an outb

The comprehensive diagnosis and prevention of duck plague in northwest Shandong province of China.

Here, we report the first outbreak of duck plague (DP) confirmed in 4 tissue samples that were collected since August 2012 from the northwestern region of Shandong province, China. Among these, 3 were collected from commercial Jin-ding variety layer ducks and one from Cherry Valley meat-breeding ducks. The sick ducks (7 to 49 wk old) were characterized by typical DP symptoms and necroscopic features. The flocks experienced high morbidity and mortality rates, and decreased production performance, which led to tremendous economic losses. The diagnosis of DP infection was confirmed by comprehensi

First survey of the occurrence of duck enteritis virus (DEV) in free-ranging Polish water birds.

Duck plague (DP) caused by anatid herpesvirus 1, also called duck enteritis virus (DEV), presents one of the most important concerns in mass waterfowl production. Apart from geese and ducks, free-ranging water birds are the most notorious infection carriers. The epidemiology of DEV in Western Europe remains unknown. Therefore, it was reasonable to conduct a study on its occurrence using modern but simple real-time loop-mediated isothermal amplification (LAMP). Analysis of 132 field isolates showed the presence of DEV in 96 birds (72.7 %), and it was found predominantly in wild ducks (Anas plat

Downregulation of the host ribonucleotide reductase subunit M2 via the Wnt/β-catenin pathway is a novel mechanism for duck plague virus replication and a potential antiviral target.

Duck plague virus (DPV), a highly contagious alphaherpesvirus, poses a significant threat to the global waterfowl industry. A comprehensive understanding of virus-host interactions is critical for developing effective control strategies. While DPV replication is known to depend on host cellular machinery, the mechanisms by which it manipulates host metabolism to its advantage remain poorly defined. Using RNA-seq analysis of DPV-infected duck embryo fibroblasts (DEFs), we identified a significant downregulation of genes involved in nucleotide metabolism, particularly the ribonucleotide reductas

ERAD inhibitor Eeyarestatin I (EerI) suppresses duck plague virus by restoring STING-mediated antiviral immunity.

Duck plague (DP), caused by the duck plague virus (DPV), is a highly contagious disease characterized by high morbidity and mortality, causing significant economic losses and biosecurity risks to the duck farming industry. Since current vaccines do not provide complete protection, the development of antiviral drugs is urgently needed. In this study, we evaluated the anti-DPV activity of the ER-associated degradation (ERAD) pathway inhibitor Eeyarestatin I (EerI) in vitro. Our results demonstrated that EerI suppressed DPV proliferation in duck embryo fibroblast (DEF) cells in a dose-dependent m

The UL24 gene affects duck plague virus replication, and its deletion attenuates virulence, enabling use as a vaccine.

Duck plague virus (DPV) is the causative agent of duck plague (DP), which poses a significant threat to production safety in the duck industry. Little is known about the UL24 protein (pUL24) encoded by the DPV UL24 gene. In this study, pUL24, when transfected into duck embryo fibroblasts, localized to the nucleus, as demonstrated by immunofluorescence techniques. We discovered that UL24 contains two nuclear localization signals (NLS), with the first also serving as a nucleolar localization signal (NoLS). To further analyse the function of pUL24, two viral strains were constructed: a UL24 delet

Pioneering discovery: US3-phosphorylated sites on VP22 orchestrate duck plague virus release and pathogenicity.

Duck plague virus (DPV), a member of the Alphaherpesvirinae subfamily, causes severe infectious disease in waterfowl and Anseriformes birds. Although the VP22 protein (encoded by the UL49 gene) is known to play a pivotal role in secondary viral envelopment, its post-translational modifications and regulatory mechanisms during the viral life cycle remain poorly understood. In this study, we found that the conserved serine/threonine kinase US3 of DPV phosphorylates VP22. We identified eight serine residues (Ser37, Ser66, Ser68, Ser82, Ser114, Ser198, Ser226, and Ser227) as phosphorylation target

Construction and identification of a cDNA library for use in the yeast two-hybrid system from duck embryonic fibroblast cells post-infected with duck enteritis virus.

To explore and isolate genes related to duck embryonic fibroblast cells (DEFs) post-infected with duck enteritis virus (DEV), a cDNA library was established using SMART (Switching Mechanism At 5' end of the RNA Transcript) technique coupling with a homologous recombination method. The cells were harvested and total RNA was extracted at 48 h post infection. Then the mRNAs were purified and reverse transcribed to first-strand cDNAs using oligo (dT) primers (CDS III). Subsequently, long distance-PCR was performed, the double-stranded cDNAs were purified, and a transformation assay was carried out

Late Marek's disease in adult chickens inoculated with virulent Marek's disease virus.

Recently, excessive losses from Marek's disease (MD) have been noted in adult laying flocks over the age of 40 weeks. We defined these late outbreaks in adult chickens as "late MD", and experimentally reproduced the disease in adult SPF P2 line (50-week-old) or commercial line (74-week-old) chickens inoculated with a virulent strain of Marek's disease virus (MDV). Commercial line chickens were given MDV vaccines (HVT and CVI 988) at hatch. The occurrence of MD was evaluated periodically by the evidence of neurologic signs such as paralysis, torticollis, ataxia, and/or nervous tics, as well as

Cloning, expression and characterization of gE protein of duck plague virus.

BACKGROUND: The gE protein of duck plague virus is the important membrane glycoprotein, its protein characterization has not been reported. In this study, we expressed and presented the characterization of the DPV gE product. RESULTS: According to the sequence of the gE gene, a pair of primers were designed, and the DNA product with 1490bp in size was amplified by using the polymerase chain reaction (PCR). The PCR product was cloned into pMD18-T vector, and subcloned into pET32a(+), generating the recombinant plasmid pET32a/DPV-gE. SDS-PAGE analysis showed that the fusion pET32a/DPV-gE protei

Multidecadal High Mortality Disease Events in Australian Domestic Geese Associated with a Novel Alphaherpesvirus, Designated Anatid Alphaherpevirus 2.

Herpesviruses are ubiquitous viruses which infect a wide range of hosts. Novel herpesviruses are being increasingly detected in free-ranging bird populations and there are growing concerns for cross-species infection and spillover events. Herein, multiple sporadic outbreaks of mortality caused by a herpesvirus are described in domestic geese in Queensland, Australia. Goose herpesvirus was initially detected in 1989 in south-east Queensland, and this article details four further recent outbreaks and reports novel genome sequencing and phylogeny of the preliminarily designated anatid alphaherpes

Pathogenic mechanisms and molecular features of a novel UL2 gene-deficient duck enteritis virus endemic to China.

Duck enteritis virus (DEV) was identified as the etiological agent responsible for an outbreak of morbidity and mortality in adult ducks on a farm in Jiangsu, China. Diagnostic approaches confirmed that the outbreak was caused by the highly pathogenic DEV-JS2024 isolate. The clinical progression of the disease, characterized by lethargy, anorexia, ocular discharge, and high mortality, was accompanied by extensive hemorrhagic lesions in critical organs such as the liver, spleen, lungs, and bursa of Fabricius, consistent with known signs of DEV infection. Genomic analysis of DEV-JS2024 revealed

Duck plague virus-encoded microRNA dev-miR-D28-3p inhibits viral replication via targeting UL27.

Herpesviruses-encoded microRNAs (miRNAs) have been discovered to be essential regulators in viral life cycle, participating in viral replication, latent or lytic infection, and immunological escape. However, the roles of miRNAs encoded by duck plague virus (DPV) are still unknown. Dev-miR-D28-3p is a miRNA uniquely encoded by DPV CHv strain. The aim of this study was to explore the effect of dev-miR-D28-3p on DPV replication and explore the potential mechanisms involved. Our findings demonstrated that transfection of dev-miR-D28-3p mimic into duck embryo fibroblasts (DEFs) effectively suppress

The intracellular domain of duck plague virus glycoprotein E affects UL11 protein incorporation into viral particles.

Studies have shown that proteins in the tegument (located between the viral capsid and envelope layer) play critical roles in the assembly and budding of herpesviruses. The UL11 protein of herpesviruses is important in the process of virus particle cell entry, release, assembly and secondary envelopment. Herpesvirus glycoprotein E (gE) is involved in syncytia formation, transmission between cells and nerve invasion. In herpes simplex virus, UL11 has been shown to interact with gE. However, little is known about the relationship of duck plague virus (DPV) pUL11 and gE. In this study, we constru

Isolation and Selection of Duck Primary Cells as Pathogenic and Innate Immunologic Cell Models for Duck Plague Virus.

Duck plague virus (DPV) is a representative pathogen transmitted among aquatic animals that causes gross lesions and immune inhibition in geese and ducks. The mechanism of organ tropism and innate immune evasion of DPV has not been completely deciphered due to a lack of cell models to study the innate immune manipulation and pathogenicity of aquatic viruses. In the present study, we isolated five types of duck primary cells [duck embryo fibroblasts (DEFs), neurons, astrocytes, peripheral blood mononuclear cells (PBMCs), and monocytes/macrophages] to identify appropriate cell models for DPV, us

Evaluation of factors influencing the development of late Marek's disease virus-induced immunosuppression: virus pathotype and host sex.

Marek's disease virus (MDV) is a herpesvirus that induces lymphoma and immunosuppression in chickens. MDV-induced immunosuppression (MDV-IS) is complex and can be divided into two phases: early-MDV-IS associated with cytolytic infection in the lymphoid organs in chickens lacking maternal antibodies against MDV (MAbs) and late-MDV-IS that appears later in the pathogenesis and occurs even in chickens bearing MAbs. We have recently developed a model to reproduce late-MDV-IS under laboratory conditions. This model evaluates late-MDV-IS indirectly by assessing the effect of MDV infection on the eff

Evaluation of the Protection Efficacy of a Serotype 1 Marek's Disease Virus-Vectored Bivalent Vaccine Against Infectious Laryngotracheitis and Marek's Disease.

Laryngotracheitis (LT) is a highly contagious respiratory disease of chickens that produces significant economic losses to the poultry industry. Traditionally, LT has been controlled by administration of modified live vaccines. In recent years, the use of recombinant DNA-derived vaccines using turkey herpesvirus (HVT) and fowlpox virus has expanded, as they protect not only against the vector used but also against LT. However, HVT-based vaccines confer limited protection against challenge, with emergent very virulent plus Marek's disease virus (vv+MDV). Serotype 1 vaccines have been proven to

Differential expression profile and in-silico functional analysis of long noncoding RNA and mRNA in duck embryo fibroblasts infected with duck plague virus.

BACKGROUND: Duck plague virus (DPV), belonging to herpesviruses, is a linear double-stranded DNA virus. There are many reports about the outbreak of the duck plague in a variety of countries, which caused huge economic losses. Recently, increasing reports revealed that multiple long non-coding RNAs (lncRNAs) can possess great potential in the regulation of host antiviral immune response. Furthermore, it remains to be determined which specific molecular mechanisms are responsible for the DPV-host interaction in host immunity. Here, lncRNAs and mRNAs in DPV infected duck embryonic fibroblast (DE

Induction of immune responses in ducks with a DNA vaccine encoding duck plague virus glycoprotein C.

BACKGROUND: A DNA vaccine expressing glycoprotein C (gC) of duck plague virus (DPV) was evaluated for inducing immunity in ducks. The plasmid encoding gC of DPV was administered via intramuscular (IM) injection and gene gun bombardment. RESULTS: After immunization by both routes virus-specific serum antibody and T-cell responses developed. Vaccination of ducks by IM injection induced a stronger humoral, but weaker cell-mediated immune response. In contrast, a better cell-mediated immune response was achieved by using a gene gun to deliver DNA-coated gold beads to the epidermis with as little

Marek's disease virus infection in the eye: chronological study of the lesions, virus replication, and vaccine-induced protection.

Marek's disease virus (MDV) infection in the eye was studied chronologically after inoculating 1-day-old chickens with a very virulent MDV strain, Md5. The ocular lesions could be classified as early lesions (6-11 days postinoculation [dpi]) and late lesions (26 and 56 dpi), based upon the location and severity of the lesions. The early lesions involved iris, ciliary body, and choroid layer, and were characterized by endothelial cell hypertrophy, vasculitis, and infiltration of lymphocytes (mainly CD8+), plasma cells, macrophages, and heterophils.
