Herpesvirus genome mapping: a rapid generic approach.

A protocol for mapping the genome of the alphaherpesvirus macropodid herpesvirus 1 is described. This protocol greatly simplifies a similar protocol that was used to map the genome of the poxvirus molluscum contagiosum virus. A single restriction digestion is carried out on the viral DNA, and the fragments cloned into a plasmid vector. The ends of each cloned fragment are sequenced, translated, and used to search peptide sequence databases. Putative genomic maps are constructed by assembling contiguous fragments identified by the sharing of common open reading frames and through the demonstrat

Diverse Tongue Base Retraction Strategies Drive Bolus Propulsion during Mammalian Swallowing.

During swallowing, a diverse range of mammals-from opossums to humans-propel food boluses out of the oropharynx via tongue base retraction (TBR). The widespread distribution of TBR behavior implies an ancient evolutionary origin, but the biomechanical mechanisms of TBR remain poorly understood. The evolution of TBR behavior is further complicated by the diversity of hyoid and tongue anatomy across mammals: to what extent does hyolingual morphology shape TBR mechanism? Using biplanar videoradiography and the XROMM workflow, we collected high-resolution 3D kinematic data in opossums (Marsupialia

Comparative patterns of postcranial ontogeny in therian mammals: an analysis of relative timing of ossification events.

Data on the relative sequence of ossification of postcranial elements for eight therian mammals (Myotis lucifugus, Homo sapiens, Rattus norvegicus, Mus musculus, Mesocricetus auratus, Cavia porcellus, Didelphis albiventris, and Sminthopsis macroura) and three outgroups (Chelydra serpentina, Alligator mississippiensis, and Lacerta vivipara) were taken from the literature. For each species, a matrix was constructed in which the relative timing of the onset of ossification in 24 elements was summarized. This resulted in 276 event pairs (characters) for each species. Thirty-three (33.3)% of the ch

Seeing through the eyes of the sabertooth Thylacosmilus atrox (Metatheria, Sparassodonta).

The evolution of mammalian vision is difficult to study because the actual receptor organs-the eyes-are not preserved in the fossil record. Orbital orientation and size are the traditional proxies for inferring aspects of ocular function, such as stereoscopy. Adaptations for good stereopsis have evolved in living predaceous mammals, and it is reasonable to infer that fossil representatives would follow the same pattern. This applies to the sparassodonts, an extinct group of South American hypercarnivores related to marsupials, with one exception. In the sabertooth Thylacosmilus atrox, the bony

A review of the function and evolution of the cecal appendix.

Since its initial discovery in the 1500s, the cecal appendix has been an anatomical structure of great intrigue, notorious for its inconvenient tendency to become inflamed and often require surgical intervention. Appendicitis is one of the most common indications for emergency abdominal surgery, costing healthcare systems billions of dollars globally and causing tens of thousands of deaths annually. Yet, recent studies have indicated that the appendix may serve important protective functions in fortifying the body's immune response against invading pathogens and re-inoculating the gut with com

Comparative anatomy of neonates of the three major mammalian groups (monotremes, marsupials, placentals) and implications for the ancestral mammalian neonate morphotype.

The existing different modes of reproduction in monotremes, marsupials and placentals are the main source for our current understanding of the origin and evolution of the mammalian reproduction. The reproductive strategies and, in particular, the maturity states of the neonates differ remarkably between the three groups. Monotremes, for example, are the only extant mammals that lay eggs and incubate them for the last third of their embryonic development. In contrast, marsupials and placentals are viviparous and rely on intra-uterine development of the neonates via choriovitelline (mainly marsu

Convergence vs. Specialization in the ear region of moles (Mammalia).

We investigated if and how the inner ear region undergoes similar adaptations in small, fossorial, insectivoran-grade mammals, and found a variety of inner ear phenotypes. In our sample, afrotherian moles (Chrysochloridae) and the marsupial Notoryctes differ from most other burrowing mammals in their relatively short radii of semicircular canal curvature; chrysochlorids and fossorial talpids share a relatively long interampullar width. Chrysochlorids are unique in showing a highly coiled cochlea with nearly four turns. Extensive cochlear coiling may reflect their greater ecological dependence

The arrangement of gut-associated lymphoid tissues and lymph pathways in the koala (Phascolarctos cinereus).

Gut-associated lymphoid tissues are poorly developed in koalas. They comprise paired caecocolic lymphoid patches, and a few small mesenteric lymph nodes. The patches lie opposite one another in the lateral gut wall at the junction of the ileum, caecum and proximal colon. The lymphoid parenchyma of the patches consists of a layer of nodules and internodular parenchyma in the submucosa. Apoptosis is common in the nodules. The mucosa and lymphoid tissue of each patch is continuous over a caecocolic recess, formed by the coalescence of laminae which extend along the large intestine. Lymph sinuses

New Cretaceous marsupial from Mongolia and the early radiation of Metatheria.

A marsupial mammal from Mongolia representing a previously unreported group, Asiadelphia, offers unequivocal evidence that metatherians were represented on the continent of Asia during the Late Cretaceous (Campanian), even in the northern arid interior. Asiatherium is known by skull, left and right mandibles, and most of its postcranial skeleton. Comparisons of the dentition, ear region, and aspects of the postcranium not only allow its diagnosis as an Asian group of metatherians but also permit a diagnosis of the Metatheria based on the unequivocally apomorphous condition of the dental formul

Returning an extirpated native carnivore to its former range: a habitat suitability analysis of Kangaroo Island for spotted-tailed quolls (Dasyurus maculatus).

Native predators have declined significantly in range and abundance globally. In Australia, the spotted-tailed quoll's (Dasyurus maculatus) range has contracted by up to 30% since Europeans arrived in the late 1700s, with its extirpation from Kangaroo Island sometime after 1837. A feral cat eradication program is currently underway on Kangaroo Island which, if successful, will leave a vacant predator niche. The spotted-tailed quoll has been proposed as the most suitable species to reoccupy this niche, however following its local extirpation, over 50% of the island's vegetation was cleared, mos

Suspected iron deficiency anaemia in two hand-reared, suckling bare-nosed wombats (Vombatus ursinus): a case report.

This case report describes clinical and haematological findings suggestive of iron deficiency anaemia in two approximately 190-200-day-old, hand-reared, suckling bare-nosed wombats (Vombatus ursinus) at a single wildlife rehabilitation facility. Both exhibited microcytic, hypochromic anaemia, with serum iron concentrations measured later in the clinical course that were suggestive of iron deficiency. Although the aetiology remains unclear, factors such as low iron bioavailability in milk formula and lack of environmental iron are discussed. Due to the lack of validated reference intervals for

Adaptive interventions for advancing in situ wildlife disease management.

There is a critical need for advancements in disease management strategies for wildlife, but free-living animals pose numerous challenges that can hinder progress. Most disease management attempts involve fixed interventions accompanied by post hoc outcome assessments focused on success or failure. Though these approaches have led to valuable management advances, there are limitations to both the rate of advancement and amount of information that can be gained. As such, strategies that support more rapid progress are required. Sarcoptic mange, caused by epidermal infection with Sarcoptes scabi

Resolving when (and where) the Thylacine went extinct.

Like the Dodo and Passenger Pigeon before it, the predatory marsupial Thylacine (Thylacinus cynocephalus), or 'Tasmanian tiger', has become an iconic symbol of anthropogenic extinction. The last captive animal died in 1936, but even today reports of the Thylacine's possible ongoing survival in remote regions of Tasmania are newsworthy and capture the public's imagination. Extirpated from mainland Australia in the mid-Holocene, the island of Tasmania became the species' final stronghold. Following European settlement in the 1800s, the Thylacine was relentlessly persecuted and pushed to the marg

Verification of the Spotted-Tail Quoll, Dasyurus maculatus, as a Definitive Host for the Pentastomid Linguatula sp. in Australia.

PURPOSE: Specimens of a pentastomid, identified as a nymphal Linguatula serrata, have previously been reported from the nasal cavity of the endangered Australian marsupial, the spotted-tail quoll, Dasyurus maculatus. These specimens were made available for morphological and molecular characterisation to confirm their species identity. METHODS: Specimens were examined by light microscopy. Molecular sequencing attempts were unsuccessful. RESULTS: Examination showed that the specimens were adult pentastomes of the genus Linguatula. Morphological differences are reported between these specimens

Leptospirosis in Tasmanian Devils ( Sarcophilus harrisii ) in Tasmania, 2008-12.

In 2014, we performed a diagnostic study of leptospirosis in Tasmanian devil ( Sarcophilus harrisii ) samples collected between 2008 and 2012 from wild and captive animals. Tasmanian devil populations have been declining because of a facial tumor disease since the 1990s, with ongoing investigations examining potential causative agents. Identifying other causative pathogens that may contribute additively to their decline is important to preserve current and future populations. We tested 81 Tasmanian devil serum samples and two tissue samples using PCR, microscopic agglutination test (MAT), and
