Postnatal Gene Therapy Improves Spatial Learning Despite the Presence of Neuronal Ectopia in a Model of Neuronal Migration Disorder.

Patients with type II lissencephaly, a neuronal migration disorder with ectopic neurons, suffer from severe mental retardation, including learning deficits. There is no effective therapy to prevent or correct the formation of neuronal ectopia, which is presumed to cause cognitive deficits. We hypothesized that learning deficits were not solely caused by neuronal ectopia and that postnatal gene therapy could improve learning without correcting the neuronal ectopia formed during fetal development. To test this hypothesis, we evaluated spatial learning of cerebral cortex-specific protein O-mannos

Reversal learning impairments in the maternal immune activation rat model of schizophrenia.

One of the cognitive symptoms observed in schizophrenia is decreased flexibility in several tasks, including reversal learning. Reversal learning has previously been tested in rats following maternal immune activation (MIA), a risk factor for schizophrenia, with varying results. Whereas some previous studies have shown that MIA rats are slower to learn a reversal, others have reported more rapid learning compared with controls. Several of these latter studies have, however, used a T-maze task with aversive, negative reinforcement as a motivating factor. Because most human studies use positive

Impaired learning with enhanced hippocampal long-term potentiation in PTPdelta-deficient mice.

Protein tyrosine phosphatase delta (PTPdelta) is a receptor-type PTP expressed in the specialized regions of the brain including the hippocampal CA2 and CA3, B lymphocytes and thymic medulla. To elucidate the physiological roles of PTPdelta, PTPdelta-deficient mice were produced by gene targeting. It was found that PTPdelta-deficient mice were semi-lethal due to insufficient food intake. They also exhibited learning impairment in the Morris water maze, reinforced T-maze and radial arm maze tasks. Interestingly, although the histology of the hippocampus appeared normal, the magnitudes of long-t

Effects of hypothermia and hyperthermia on attentional and spatial learning deficits following neonatal hypoxia-ischemic insult in rats.

We previously reported that rats exposed to neonatal hypoxic-ischemic (HI) insult showed selective and long-lasting learning and memory impairments in the plus maze, 8-arm radial maze, choice reaction time (CRT) task, and water maze, and that they showed severe brain injury to areas such as parietal cortex, hippocampus, striatum and thalamus. In this study, we examined the effects of hypothermia and hyperthermia on learning and memory deficits following neonatal HI insult. Seven-day-old Wistar rats were subjected to left carotid artery ligation followed by 2 h of hypoxia (8% O2/92% N2) under t

Deficits across multiple cognitive domains in a subset of aged Fischer 344 rats.

Rodent models of cognitive aging routinely use spatial performance on the water maze to characterize medial temporal lobe integrity. Water maze performance is dependent upon this system and, as in the aged human population, individual differences in learning abilities are reliably observed among spatially characterized aged rats. However, unlike human aging in which cognitive deficits rarely occur in isolation, few non-spatial learning deficits have been identified in association with spatial impairment among aged rats. In this study, a subset of male aged Fischer 344 rats was impaired both in

A review of behavioral methods for the evaluation of cognitive performance in animal models: Current techniques and links to human cognition.

INTRODUCTION: Memory is defined as the ability to store, maintain and retrieve information. Learning is the acquisition of information that changes behavior and memory. Stress, dementia, head trauma, amnesia, Alzheimer's, Huntington, Parkinson's, Wernicke-Korsakoff syndrome (WKS) may be mentioned among the diseases in which memory and learning are affected. The task of understanding deficits in memory and learning in humans is daunting due to the complexity of neural and cognitive mechanisms in the nervous system. This job is made more difficult for clinicians and researchers by the fact that

Theta oscillations support active exploration in human spatial navigation.

Active navigation seems to yield better spatial knowledge than passive navigation, but it is unclear how active decision-making influences learning and memory. Here, we examined the contributions of theta oscillations to memory-related exploration while testing theories about how they contribute to active learning. Using electroencephalography (EEG), we tested individuals on a maze-learning task in which they made discrete decisions about where to explore at each choice point in the maze. Half the participants were free to make active decisions at each choice point, and the other half passivel

Spatial learning deficits in amyloid precursor protein 770 transgenic mice.

OBJECTIVE: To determine whether learning deficits could be seen in transgenic mice expressing human amyloid precursor protein 770 (APP770). METHODS: Female heterozygous transgenic and nontransgenic mice aged 3, 6 and 9 months at the start of testing were used, with eight mice in each age group. All mice were subjected to various behavioral tasks including the Y-maze task and the Morris water maze. After behavioral testing, the mice were sacrificed, and their brain tissues were used for measuring the choline acetyltransferase (ChAT) activity. RESULTS: Nine-month-old transgenic mice exhibited

[Reverse learning in WAG/Rij rats with depression-like behavior].

Learning and reverse learning in a complex maze, behavior in the open field test, novelty-suppressed feeding test, and forced swimming test were studies in WAG/Rij and Wistar rats. As compared with Wistar rats, WAG/Rij rats more slowly learned the spatial task, more slowly performed in the learning and reverse learning tasks, and made more errors in the complex maze (18% of WAG/Rij rats didn't reach learning criterion). Moreover, WAG/Rij rats exhibited reduced grooming reactions in the open field test, longer latency of approaching to food in the novel open field, reduced amount of food consum

Neural network classification of Barnes maze search strategy utilization.

The Barnes maze is a commonly used test of allocentric spatial reference memory, consisting of an elevated circular table with holes around the perimeter. Spatial cues surrounding the maze are intended to allow the animal on the maze to locate a target hole from which they can escape and return to their home cage during a period of acquisition trials. Following the acquisition period, the target box under the target hole is removed and a probe trial is performed to test spatial memory. One of the limitations of Barnes maze testing is that non-spatial strategies can be employed to locate the ta

Effects of BN-50730 (PAF receptor antagonist) and physostigmine (AChE inhibitor) on learning and memory in mice.

The present study was designed to investigate the effect of BN-50730, a PAF receptor antagonist, on learning and memory in mice using elevated plus-maze and to delineate the role of acetylcholine in modulating the effect of PAF receptor antagonist on learning and memory. BN-50730 administered immediately after plus-maze training on day 1 induced retrograde amnesia as indicated by a dose-dependent increase in transfer latency (TL) measured on day 2 whereas no such increase in TL was noted when BN-50730 (2.5 mg/kg, i.p.) was administered prior to plus-maze training. Physostigmine (0.5 mg/kg; 1.0

Learning and laterality in wild-caught Namaqua rock mice (Micaelamys namaquensis).

Individuals often exhibit differences in cognition and/or preferences for one side over the other. Such differences have been attributed to the mating system and lateralization of the brain hemispheres that can differ between the sexes, respectively. Despite the hypothesized significant effects on fitness, only a limited number of rodent studies addresses sex differences in laterality and most focus on laboratory rodents. Here we examined whether wild-caught Namaqua rock mice (Micaelamys namaquensis), a rodent widespread in sub-Saharan Africa, exhibit sex differences in learning and laterality

Streptozotocin-induced diabetes during gestation and post-partum: effects on behavior and hippocampal plasticity in female Wistar rats.

Gestational Diabetes (GD) is associated with cognitive dysfunction, anxiety, and post-partum depression. The mechanisms underlying such changes are poorly understood. It is not known whether animal models of GD can replicate the dysfunction observed in humans. In this pilot study, we evaluated whether a streptozotocin (STZ)-induced model of gestational diabetes reproduces postpartum anxiety-like behavior and learning and memory impairments. Female Wistar rats were assigned to the control (n = 14) and STZ (n = 10) groups. After weaning, maternal anxiety behavior was evaluated using the Elevated

Loganin enhances long-term potentiation and recovers scopolamine-induced learning and memory impairments.

Although the incidence rate of dementia is rapidly growing in the aged population, therapeutic and preventive reagents are still suboptimal. Various model systems are used for the development of such reagents in which scopolamine is one of the favorable pharmacological tools widely applied. Loganin is a major iridoid glycoside obtained from Corni fructus (Cornusofficinalis et Zucc) and demonstrated to have anti-inflammatory, anti-tumor and osteoporosis prevention effects. It has also been found to attenuate Aβ-induced inflammatory reactions and ameliorate memory deficits induced by scopolamine

Maze design: size and number of choices impact fish performance in cognitive assays.

Although studies on fish cognition are increasing, consideration of how methodological details influence the ability to detect and measure performance is lagging. Here, in two separate experiments the authors compared latency to leave the start position, latency to make a decision, levels of participation and success rates (whether fish entered the rewarded chamber as first choice) across different physical designs. Experiments compared fish performance across (a) two sizes of T-mazes, large and standard, and a plus-maze, and (b) open choice arenas with either two or four doors. Fish in T-maze

ECT-induced anterograde amnesia: can the deficits be minimized?

To date, no pharmacological agent has been confirmed to lessen electroconvulsive therapy (ECT)-induced memory deficits. BR-16A is an herbal preparation, containing various organic extracts, used in India for the enhancement of cognition (among other applications). In the present study, adult male Sprague-Dawley rats received six once-daily electroconvulsive shocks (ECSs). Half the animals were treated with BR-16A (200 mg/kg/day) for 1 week before ECS, during the ECS course, and during the post-ECS learning assessment phase; the remaining animals received vehicle alone. In experiment 1, rats (n
