Methods in molecular biology (Clifton, N.J.)2026eng
Elucidating the spatial and temporal regulation of gene expression during plant organogenesis is crucial for enabling precise crop improvement strategies that incorporate beneficial traits into crops while avoiding adverse effects. Root nodules, specialized organs formed in symbiosis with nitrogen-fixing bacteria, provide a valuable system to study cell-type-specific gene networks in a symbiosis-induced developmental context. However, capturing these dynamics at cellular resolution in intact plant tissues remains technically challenging. Spatial transcriptomics technologies developed for anima

Flooding induces hypoxia in plant tissues, impacting various physiological and biochemical processes. This study investigates the adaptive response of the roots and nitrogen-fixing nodules of Medicago truncatula in symbiosis with Sinorhizobium meliloti under short-term hypoxia caused by flooding. Four-week-old plants were subjected to flooding for 1-4 days. Physiological parameters as well as the expression of the senescence marker gene MtCP6 remained unchanged after 4 days of flooding, indicating no senescence onset. Hypoxia was evident from the first day, as indicated by the upregulation of

The ornithine-urea cycle (urea cycle) makes a significant contribution to the metabolic responses of lower photosynthetic eukaryotes to episodes of high nitrogen availability. In this study, we compared the role of the plant urea cycle and its relationships to polyamine metabolism in ammonium-fed and nitrate-fed Medicago truncatula plants. High ammonium resulted in the accumulation of ammonium and pathway intermediates, particularly glutamine, arginine, ornithine, and putrescine. Arginine decarboxylase activity was decreased in roots, suggesting that the ornithine decarboxylase-dependent produ

Reductions in sulfur dioxide emissions and the use of sulfur-free mineral fertilizers are decreasing soil sulfur levels and threaten the adequate fertilization of most crops. To provide knowledge regarding legume adaptation to sulfur restriction, we subjected Medicago truncatula, a model legume species, to sulfur deficiency at various developmental stages, and compared the yield, nutrient allocation and seed traits. This comparative analysis revealed that sulfur deficiency at the mid-vegetative stage decreased yield and altered the allocation of nitrogen and carbon to seeds, leading to reduced

The present study was conducted to determine whether lyophilized aqueous extract of alfalfa, or Medicago sativa L. could exert antioxidant activity against carbon tetrachloride-induced oxidative stress and liver injury in rats. The hepatoprotective activity of alfalfa extract was determined by assessing the levels of serum transaminases, ALP, bilirubin and lipid profile. Further, the effect of the test substance on malondialdehyde (MDA), an end product of lipid peroxidation; antioxidant liver enzyme non-protein sulfhydryl (NP-SH); and total protein (TP) were also studied. Serum transaminase, A

Terpenoids constitute one of the largest and most structurally diverse classes of plant specialized metabolites, with diversity generated by terpene synthases (TPSs) and downstream tailoring enzymes. In Medicago truncatula, the TPS-b subfamily comprises five putative synthases, two of which are embedded within a previously uncharacterized genomic locus containing a cytochrome P450 (CYP) and a BAHD-type acyltransferase. Here, we present a comprehensive biochemical analysis of the M. truncatula TPS-b subfamily and define a cineole-centered monoterpene biosynthetic module. Heterologous expression

Elevated CO(2) compromises the resistance of leguminous plants against chewing insects, but little is known about whether elevated CO(2) modifies the resistance against phloem-sucking insects or whether it has contrasting effects on the resistance of legumes that differ in biological nitrogen fixation. We tested the hypothesis that the physical and chemical resistance against aphids would be increased in Jemalong (a wild type of Medicago truncatula) but would be decreased in dnf1 (a mutant without biological nitrogen fixation) by elevated CO(2). The non-glandular and glandular trichome density

Integrated multi-omics analyses reveal that novel miR65 represses MsbZIP11 and negatively regulates saline-alkali tolerance in alfalfa by impairing photosynthetic maintenance and disrupting redox homeostasis. MicroRNA-mediated post-transcriptional regulation plays an important role in plant adaptation to saline-alkali stress; however, the underlying regulatory modules remain poorly understood in alfalfa (Medicago sativa L.). Here, integrated profiling of small RNAs, the transcriptome, and degradome identified 175 stress-responsive miRNAs and 418 degradome-supported miRNA-mRNA interactions. We

Medicago sativa Linnaeus, 1753 (Leguminosae) is a globally important autotetraploid forage crop with a complex genome. Local populations adapted to specific ecological niches, such as those in Türkiye's Lake Region, represent valuable genetic resources for breeding. However, their cytogenetic diversity remains poorly characterized. This study aimed to perform detailed karyotype analyses of eight alfalfa populations from this region to assess chromosome number, morphology, asymmetry, and structural variants. Root tips from eight populations collected from Afyonkarahisar, Burdur, Isparta, and Ko

The recent discovery of the lanthanide (Ln)-dependent methanol dehydrogenase (Ln-MDH) XoxF has expanded the spectrum of bacteria recognized for methylotrophic metabolism. Many bacteria, including rhizobia, have historically escaped being categorized as methylotrophs because they exclusively produce XoxF-type Ln-MDHs and entirely lack the long-studied calcium-dependent MDH MxaFI. We report that the XoxF-type Ln-MDH encoded by the smb20173 gene is the sole MDH that supports methylotrophic growth of Sinorhizobium meliloti. The lanthanides that consistently supported growth of S. meliloti in minim

Sesquiterpenoid chemical diversity is frequently generated through cytochrome P450 (CYP)-mediated oxidation of terpene scaffolds. In Medicago truncatula, the sesquiterpene alcohol himachalol, produced by the terpene synthase MtTPS10, has been implicated in pathogen defense, yet the enzymes responsible for its downstream modification remain undefined. Here, we characterize a compact locus containing two sesquiterpene synthases and three CYP71D genes. Using heterologous expression, we show that two duplicated CYP71D enzymes catalyze regiospecific oxidations of himachalol. CYP71D81 hydroxylates t

UNLABELLED: Recently, molecular hydrogen (H2) has emerged as a bio-regulator both in animals and plants. Normally, functions of endogenous generated H2 could be mimicked by exogenously applied hydrogen-rich water (HRW) or hydrogen-rich saline (particularly in animals). Although alfalfa seedlings showed more cadmium (Cd) resistance after the administration with HRW, corresponding molecular mechanism is still elusive. To address this gap, iTRAQ-based quantitative proteomics was used. The results showed that a total of 2377 proteins were identified with <1% FDR, and 1254 protein abundance perturb

Nitric oxide (NO) is a bioactive, diffusible molecule involved in a multitude of physiological and developmental processes in plants, which has been reported to display both antioxidant and pro-oxidant properties in plants. Several reports exist highlighting the protective action of sodium nitroprusside (SNP), an NO donor, which demonstrate its important role as a signal molecule in plants responsible for the expression regulation of antioxidant and other defense enzymes. However, the mode of application of this compound varies greatly between studies. The present study provides a comprehensiv

To reduce S emissions, petroleum coke with a high concentration of S was combusted with limestone in a circulating fluidized bed (CFB) boiler. The combustion process creates a bed product that has potential for agricultural uses. This CFB product is often alkaline and enriched in S and other essential plant nutrients, but also contains high concentrations of Ni and V. Agricultural land application of CFB product is encouraged, but little information is available related to plant responses and environmental impacts. CFB product and agricultural lime (ag-lime) were applied at rates of 0, 0.5, 1.

Plant A/T-rich sequence and zinc-binding protein (PLATZ) transcription factors are pivotal regulators of plant growth, development, and abiotic stress responses. Although PLATZ genes have been extensively studied and functionally characterized in various plant species, little information is available regarding these genes in the legume model plant Medicago truncatula. In this study, a total of 16 PLATZ genes (designated MtPLATZ1 to MtPLATZ16) were identified from the M. truncatula genome and distributed across six of the eight chromosomes. Subcellular localization prediction analysis indicated

BACKGROUND: Structural variants (SVs) constitute a large proportion of the genomic variation that results in phenotypic variation in plants. However, they are still a largely unexplored feature in most plant genomes. Here, we present the whole-genome landscape of SVs between two model legume Medicago truncatula ecotypes-Jemalong A17 and R108- that have been extensively used in various legume biology studies.
RESULTS: To catalogue SVs, we first resolved the previously published R108 genome assembly (R108 v1.0) to chromosome-scale using 124 × Hi-C data, resulting in a high-quality genome assemb
