To determine how alterations of megakaryocyte proliferation will affect platelet production, we measured mean platelet volume (MPV), platelet volume heterogeneity, platelet count, and mean megakaryocyte ploidy in 42 patients. In normal subjects, mean platelet volume and megakaryocyte ploidy were related inversely but nonlinearly to platelet count, whereas mean platelet volume and platelet volume heterogeneity were related directly. In patients with immune thrombocytopenic purpura (low platelet count, MPV above normal, and increased megakaryocyte ploidy), and in those with reactive thrombocytos

A high incidence of acute megakaryoblastic leukemia (AMKL) in Down syndrome patients implies that chromosome 21 genes have a pivotal role in AMKL development, but the functional contribution of individual genes remains elusive. Here, we report that SON, a chromosome 21-encoded DNA- and RNA-binding protein, inhibits megakaryocytic differentiation by suppressing RUNX1 and the megakaryocytic gene expression program. As megakaryocytic progenitors differentiate, SON expression is drastically reduced, with mature megakaryocytes having the lowest levels. In contrast, AMKL cells express an aberrantly

OBJECTIVE: Umbilical cord blood (CB) provides an alternative source of hematopoietic progenitor cells for transplantation; however, prolonged thrombocytopenia remains a major obstacle due to the low numbers of megakaryocyte progenitor (Mk-prog) cells and their subsequent delayed engraftment. In this study, we improved techniques for enrichment, cryopreservation, and ex vivo expansion of Mk-prog cells from CB.
MATERIALS AND METHODS: CB mononuclear cells (MNC) were isolated and Mk-prog enriched by sedimentation on gelatin followed by centrifugation with Ficoll-Hypaque and cryopreserved. The cap

Megakaryocyte polyploidization is a specialized process essential for platelet production, orchestrated by tightly regulated transcriptional programs, cell-cycle control, and signaling pathways. Disruption of this process - characterized by incomplete endomitosis, low ploidy, and maturation arrest - contributes to leukemic transformation, particularly in acute megakaryoblastic leukemia and related myeloid malignancies. Aberrant polyploidization arises from transcription factor dysregulation, cell-cycle abnormalities, hyperactive proliferative signaling, microenvironmental alterations, and geno

BACKGROUND: The mechanisms that regulate platelet biogenesis remain unclear; factors that trigger megakaryocytes (MKs) to initiate platelet production are poorly understood. Platelet formation begins with proplatelets, which are cellular extensions originating from the MK cell body.
OBJECTIVES: Proplatelet formation is an asynchronous and dynamic process that poses unique challenges for researchers to accurately capture and analyze. We have designed an open-source, high-content, high-throughput, label-free analysis platform.
METHODS: Phase-contrast images of live, primary MKs are captured ov

Megakayocytes (Megakaryocytes) are among the rarest type of haematopoietic cells and highly specialized precursors for platelets. Normal mature megakaryocytes are, perhaps, the largest cells in the marrow. In contrast, their annucleated platelets progeny are the smallest subcellular fragments in the circulation, which in spite of their size, play crucial roles in thrombostasis and haemostasis. Megakaryopoiesis involves complicated and multi-step biological processes. Research over the last decade has resulted in certain important new discoveries, such as the specific megakaryocyte-forming haem

We developed a new microfluorometric method to measure the DNA and RNA contents of individual megakaryocytes using acridine orange (AO) staining in human bone marrow smears. Some bone marrow smears were fixed with ethanol and treated with pretreatment solution and then stained with an AO staining solution. The DNA content was assayed by measuring the green fluorescence and the RNA content was assayed by measuring the red fluorescence under B excitation with microfluorometer using peripheral lymphocytes as the control. The ploidy peak was shown to be 16N in all of 5 normal controls, but it was

Dysmegakaryocytopoiesis is an integral component of myelodysplastic syndrome (MDS), and has been shown in some studies to be an independent prognostic factor. Megakaryocytic hypogranulation, a feature we have noticed for some time to be fairly common in MDS and acute myeloid leukaemia (AML), has received little attention in the literature as a dysplastic feature of megakaryocytes. This study was performed to determine how frequently this feature was observed in MDS and the specificity of its occurrence. On review of archival materials, hypogranular megakaryocytes were observed in 80.3% of MDS,

Normal human platelets and megakaryocytes were examined for peroxidase activity by the diaminobenzidine (DAB) cytochemical technic. When the fixation and the incubation were adequate, a strong reaction was present in the dense tubular system of platelets suspended in plasma or spread on carbon. The black reaction product was ascribed to enzyme activity, since the reaction was completely eliminated when H(2)O(2) or DAB were omitted, or when H(2)O(2) was in excess. In addition, the reaction was inhibited by aminotriazole, cyanide and azide. In the human megakaryocytes, the reaction was localized

By endothelial markers such as FVIII, UEA-I, CD31 and QBEND/10, the Authors have investigated the immunohistochemical pattern of bone marrow megakaryocytes in myelodysplastic (8 cases) and myeloproliferative (35 cases) disorders as well as in control biopsies (8 cases). An evident cytoplasmic positivity with FVIII and UEA-I was encountered in all normal and pathological specimens, whereas the immunolocalization of CD31 was limited to megakaryocytes present in normal bone marrow biopsies or in cases of myelofibrosis. No immunostaining with QBEND/10 was observed in any case, although the most se

The relation between polyploidy and morphological classification of human megakaryocytes was studied in bone marrow aspirates from five normal individuals. On a Wright-Giemsa stained smear, megakaryocytes were morphologically classified into four groups according to a modification of Feinendegen's classification which is considered to reflect megakaryocyte maturation. The DNA of the morphologically classified cell is measured by microcytofluorometry using DAPI (4',6-diamidino-2-phenylindole) staining after removing the Wright-Giemsa stain. Most of the normal megakaryocytes were classified into

Antibodies against megakaryocytes into the bone-marrow were investigated with the aid of direct immunofluorescence in 56 children with chronic-idiopathic thrombocytopenic purpura (ITP) and in 10 children of a control group. The megakaryocyte concentrate of the bone-marrow was produced by using a density grade separation with percoll. In children of the control group, small amounts of fixed immunoglobulins of all three main classes (G, A and M) could only be found on the membrane of megakaryocytes. It was only in some cases that illuminating granules could be identified on the membrane. In pati

Platelets and megakaryocytes from 11 patients with the carcinoid syndrome have been studied by transmission electron microscopy. Cells fixed in phosphate-buffered glutaraldehyde are oval to discoid, with pseudopods, a dilated open-channel system, and a prominent dense tubular system as defined by peroxidase activity and alkaline bismuth stain. Atypical with hexagonal lattices and treaded substructures and large (diameter greater than 0.5 mum), phosphatase-positive, debris-containing vacuoles are four times more numerous than in normal platelets. Incubation of platelets in a 0.05% suspension of

Pathological accumulation of reactive oxygen species (ROS) is implicated in several diseases, including cancer, cardiovascular diseases, and aging. However, ROS play essential roles in cellular functions, including proliferation, differentiation, and immune responses, at physiological levels. In megakaryocytes, the cells responsible for producing platelets, ROS exert context-dependent effects, either promoting or impairing maturation depending on developmental stage and subcellular localization. In this review, we summarize current evidence demonstrating that balanced ROS signaling is required

Sunitinib is associated with a higher incidence of thrombocytopenia than other molecularly targeted drugs. However, the underlying mechanism remains unclear. Because platelets are derived from megakaryocytes originating from hematopoietic progenitor cells in the bone marrow, sunitinib is presumed to interfere with megakaryocyte proliferation, maturation, or platelet production. Therefore, megakaryocytes play a crucial role in elucidating the mechanism of sunitinib-induced thrombocytopenia. However, in vivo sampling of megakaryocytes is challenging due to the invasiveness of bone marrow collect

Runt-related transcription factor 1/acute myeloid leukemia 1 (RUNX1/AML1) is essential for the generation of myeloid progenitor cells and differentiation of megakaryocytes (MKs). During megakaryocyte differentiation and maturation, RUNX1 forms transcriptional complexes with transcription factors such as GATA-1, Ets-1, Fli-1, and NF-E2 to promote platelet production. Disruption of RUNX1 is implicated in hematopoietic neoplasms like familial platelet disorder (FPD/AML), myelodysplastic syndrome (MDS) and acute myeloid leukemia. Here, we review the transcriptional regulation of RUNX1 in megakaryo
