Loss of CsCLV2 function causes dwarfism and determinates growth in cucumber.

Cucumber (Cucumis sativus L.) is a globally important vegetable crop. Ideal plant architecture optimizes spatial utilization, enhances economic coefficient, and facilitates mechanized cultivation. In this study, we identified a dwarf mutant, csdw3, exhibiting reduced plant height, shortened internodes, and fewer internodes. Genetic analysis showed that this dwarf phenotype is controlled by a single recessive gene. Fine-mapping localized the causal locus to an 80 kb region on chromosome 1, where we discovered a 102 bp deletion in CsCLV2, a gene encoding a leucine-rich repeat receptor-like prote

AtCAF-1 mutants show different DNA damage responses after ultraviolet-B than those activated by other genotoxic agents in leaves.

Chromatin assembly factor-1 (CAF-1) is a histone H3/H4 chaperone that participates in DNA and chromatin interaction processes. In this manuscript, we show that organs from CAF-1 deficient plants respond differently to ultraviolet-B (UV-B) radiation than to other genotoxic stresses. For example, CAF-1 deficient leaves tolerate better UV-B radiation, showing lower cyclobutane pyrimidine dimer (CPD) accumulation, lower inhibition of cell proliferation, increased cell wall thickness, UV-B absorbing compounds, and ploidy levels, whereas previous data from different groups have shown that CAF-1 muta

Organic acids counteract the determinate root development induced by phosphate deprivation in Arabidopsis thaliana.

Phosphate (Pi) deficiency impairs plant development and decreases productivity owing to its critical functions in terms of cell structure, metabolism, and photosynthesis. Here, we assessed the determinate growth program of Arabidopsis primary roots exposed to Pi scarcity to clarify the role of citrate and malate in the expression of the mitotic cyclin CycB1 and changes in the meristem, transition zone, and elongation zone. The transfer of seedlings from high-Pi medium to low-Pi medium, but not to high-Pi medium, inhibits root growth, cell division, and elongation, aspects that are restored upo

DipTRANS: an improved method for in planta transformation and genome engineering in Nicotiana benthamiana.

Plant transformation remains constrained by labor-intensive tissue culture. Our previous work showed that direct delivery of developmental regulators (DRs) can induce de novo meristems on plants, offering a promising transformation approach. In this resource article, we introduced DipTRANS (Direct in planta Transformation), an optimized, soil-based heritable transformation platform for Nicotiana benthamiana that bypasses sterile culture entirely. DipTRANS is built on DR-induced de novo meristem formation. After optimizing parameters, including regulator combinations, Agrobacterium strain, and

An FTIR study of the induction and release of kiwifruit buds from dormancy.

BACKGROUND: Many deciduous, perennial fruit crops require winter chilling for adequate budbreak and flowering. Recent research has shown that changes in sugar and amino acid profiles are associated with the release of buds from dormancy. This paper uses FTIR spectrometry to provide an alternative mechanism for tracking metabolic changes in the meristems of kiwifruit buds during winter dormancy. The results suggest that the application of multivariate analysis to FTIR spectra has the potential to be a reliable and fast method for detecting structural and compositional changes in fruit crops. LBD37, LBD38, and LBD39 are redundantly involved in regulating lateral root development and elongation in Arabidopsis thaliana.

The LATERAL ORGAN BOUNDARIES DOMAIN (LBD) genes encode a family of transcription factors harboring a conserved LOB domain that regulate diverse aspects of plant growth and development. Arabidopsis LBD proteins are grouped into two classes according to their domain structures. Class I proteins, which function as typical DNA-binding transcription factors, are relatively well characterized, whereas the biological functions of class II proteins remain largely unknown. Here, we show that LBD37, LBD38, and LBD39-three class II LBD genes-redundantly regulate lateral root (LR) formation and elongation

Ectopic expression of a tobacco homeobox gene, NTH15, dramatically alters leaf morphology and hormone levels in transgenic tobacco.

The shoot apical meristem functions to generate the lateral organs of a plant throughout the vegetative and reproductive phases. Homeobox genes play key roles in controlling such developmental programs, but their modes of action have not been well defined. Here we describe isolation and biological functions of a novel tobacco homeobox gene, designated NTH15 (Nicotiana tabacum homeobox 15), from a tobacco shoot apex cDNA library. NTH15 encodes a polypeptide of 342 amino acids, its homeodomain is very similar to the class 1 KNOTTED-type homeodomains. NTH15 mRNA is mainly localized in corpus cell

A Microscopy Toolkit for Root Studies in B. distachyon.

The study of plant organogenesis presents significant challenges due to the small number of cells involved in its initial stages. Observing the initial cell divisions becomes increasingly challenging when transitioning from Arabidopsis thaliana (Arabidopsis) to other species. Lateral root (LR) initiation is an essential process for enhancing a plant's ability to access water and nutrients. In most cases, LR formation starts in the pericycle of the parent root, giving rise to a new meristem. Understanding the mechanisms coordinating LR development is important for improving plant resilience to

The Striking Flower-in-Flower Phenotype of Arabidopsis thaliana Nossen (No-0) is Caused by a Novel LEAFY Allele.

The transition to reproduction is a crucial step in the life cycle of any organism. In Arabidopsis thaliana the establishment of reproductive growth can be divided into two phases: Firstly, cauline leaves with axillary meristems are formed and internode elongation begins. Secondly, lateral meristems develop into flowers with defined organs. Floral shoots are usually determinate and suppress the development of lateral shoots. Here, we describe a transposon insertion mutant in the Nossen accession with defects in floral development and growth. Most strikingly is the outgrowth of stems from the a

Auxin and cytokinin regulate growth dynamics underlying carpel initiation in Arabidopsis.

Plant organ initiation requires precise spatial and temporal coordination of cellular behaviors. In Arabidopsis, the gynoecium, the female reproductive structure formed by two fused carpels, is initiated after the termination of the floral meristem. Proper initiation of the carpels is crucial for ovule protection, successful fertilization, and the formation of diverse fruit structures. While the phytohormones auxin and cytokinin are known to regulate organogenesis, their interplay during the earliest stages of carpel development is not fully understood. In particular, how auxin-cytokinin cross

New Insights on the Role of ß-Cyanoalanine Synthase CAS-C1 in Root Hair Elongation through Single-Cell Proteomics.

(1) Background: Root hairs are specialized structures involved in water and plant nutrient uptake. They elongate from epidermal cells following a complex developmental program. ß-cyanoalanine synthase (CAS), which is mainly involved in hydrogen cyanide (HCN) detoxification in Arabidopsis thaliana, plays a role in root hair elongation, as evidenced by the fact that cas-c1 mutants show a severe defect in root hair shape. In addition to root hairs, CAS C1 is expressed in the quiescent center and meristem. (2) Methods: To identify its role in root hair formation, we conducted single-cell proteomic

A Constitutively Active Cytokinin Receptor Variant Increases Cambial Activity and Stem Growth in Poplar.

The cambial meristem is responsible for bark and wood formation in woody plants. The activity of the cambial meristem is controlled by various factors; one of them is the plant hormone cytokinin. Here, we have explored different approaches to genetically engineering cambial activity in poplar plants by the ectopic expression of a cytokinin biosynthesis gene with enhanced activity (named ROCK4) or of a gene encoding a constitutively active cytokinin receptor variant (ROCK3). Both genes are derived from Arabidopsis thaliana and were expressed in poplar trees under the control of their own promot

Winter dormancy in trees.

Plants growing in temperate and boreal regions of the world have to face strikingly different environmental conditions during summer and winter. Being sessile organisms, plants have had to develop various strategies to adapt to these changes in light, temperature, and water availability, thereby optimizing their 'economy of growth'. While annual plants can endure unfavorable winter conditions in the form of a seed, or under a protective cover of thick snow, perennial plants such as trees adapt by going into a stage of deep sleep called winter dormancy. To enter dormancy, vegetative growth is s

Effects of bud-flushing strategies on tree growth.

Allocation of carbohydrates between competing organs is fundamental to plant development, growth and productivity. Carbohydrates are synthesized in mature leaves and distributed via the phloem vasculature to developing buds where they are consumed to produce new biomass. The distribution and mass-allocation processes within the plant remain poorly understood and may involve complex feedbacks between different plant functions, with implications for the emergent structure of the plant. Here, we investigate how the order in which dormant buds are flushed affects the development of tree size and r

The Arabidopsis ARF3-AIP1/2-SAP18 module specifies the root stem cell niche in response to auxin.

In the root apical meristem, the stem cell niche (SCN) comprises a mitotically inactive quiescent center (QC) and adjacent, mitotically active stem cells that divide to form root tissues. Auxin dynamics are essential for specification and maintenance of the root SCN; however, the underlying mechanisms remain to be explored. Here, we report that Arabidopsis (Arabidopsis thaliana) AUXIN RESPONSE FACTOR3 (ARF3), ARF3-INTERACTING PROTEIN1/2 (AIP1/2), and SIN3-ASSOCIATED POLYPEPTIDE OF 18 KDA (SAP18) form a protein complex that specifies root SCN cell fate in response to auxin level. Single-nucleus transcriptomics reveals cell fate transitions and transcriptional programs driving bulblet formation in Lilium.

Bulblet formation is a critical type of direct vegetative regeneration in plants, where modified leaf bases undergo a developmental transition to generate new bulb structures. However, the cellular trajectories driving this scale-to-bulblet transition remain poorly understood. Here, we applied single-nucleus sequencing to construct a high-resolution cellular atlas of lily bulblet formation. We identified a population of meristem-like cells located adjacent to vascular bundle sheath cells that exhibits progenitor-like features and transcriptional associations with vascular and parenchyma cell s

Water hyacinth (Eichhornia crassipes) to biomonitor genotoxicity of low levels of mercury in aquatic environment.

The mitotic cell-cycle duration of root meristematic cells of Eichhornia crassipes as determined by the colchicine labelling method was approximately 24 h at 30 +/- 1 degrees C. In one experiment the intact root meristems of E. crassipes were subjected to 1 h acute exposure to water contaminated with maleic hydrazide (MH), 56 ppm, or methyl mercuric chloride (MMCl), 0.1-0.5 ppm, followed by recovery in tap water for 4-48 h. In a second experiment the roots were subjected to 96 h exposure to water contaminated with MH, 56 ppm, or MMCl, 0.0001-0.1 ppm. In both experiments the cytological end-poi

Structure and regulation of the Asr gene family in banana.

Abscisic acid, stress, ripening proteins (ASR) are a family of plant-specific small hydrophilic proteins. Studies in various plant species have highlighted their role in increased resistance to abiotic stress, including drought, but their specific function remains unknown. As a first step toward their potential use in crop improvement, we investigated the structure and regulation of the Asr gene family in Musa species (bananas and plantains). We determined that the Musa Asr gene family contained at least four members, all of which exhibited the typical two exons, one intron structure of Asr ge

Mutations in a plastid-localized elongation factor G alter early stages of plastid development in Arabidopsis thaliana.

BACKGROUND: Proper development of plastids in embryo and seedling tissues is critical for plant development. During germination, plastids develop to perform many critical functions that are necessary to establish the seedling for further growth. A growing body of work has demonstrated that components of the plastid transcription and translation machinery must be present and functional to establish the organelle upon germination. RESULTS: We have identified Arabidopsis thaliana mutants in a gene that encodes a plastid-targeted elongation factor G (SCO1) that is essential for plastid developmen

A Leaf-Expressed TERMINAL FLOWER1 Homolog from Coffee with Alternative Splice Forms Alters Flowering and Branching in Arabidopsis.

Coffee is a perennial plant that exhibits asynchronous flowering while maintaining concomitant vegetative growth. This growth dichotomy affects fruit development and maturation time. To better understand flowering in coffee, we characterized a phosphatidylethanolamine binding protein (PEBP) homolog with high similarity to Arabidopsis thaliana TERMINAL FLOWER1 (TFL1). The interaction of TFL1 with floral regulator bZIP transcription factor, FD, forms a floral repressor complex that maintains inflorescence meristems in an indeterminate state. Arabidopsis TFL1 is expressed only in shoot apical mer
