Canine peritoneal and pleural larval mesocestoidosis with sarcoma development.

A 5-year-old, spayed female Anatolian Shepherd dog adopted from Turkey presented for an ultrasound. A splenic mass, abdominal cysts, and peritoneal effusion were incidentally found. A presumptive diagnosis of peritoneal cestodosis was made. Despite antiparasitic treatment, the patient developed lethargy, hyporexia, tachypnea, dyspnea, and progressive peritoneal effusion. Due to the declining condition and poor prognosis, humane euthanasia was elected, and a necropsy was performed. The main gross findings were severe peritoneal and pleural effusion with a large amount of tan, turbid fluid, fibr

New evidence from the northern Apennines, Italy, suggests a southward expansion of Echinococcus multilocularis range in Europe.

Echinococcus multilocularis (Em) is a neglected zoonotic cestode circulating among wild canids and voles across the northern hemisphere, and is the aetiological agent of alveolar echinococcosis in humans. The expansion of the European distribution of this parasite has been raising public health concerns in recent decades. We aimed to investigate the occurrence of Em and other taeniids in wild carnivore faeces in the Apuan Alps Regional Park and the Monte Pisano mountain chain (northern Tuscany), a few hundred kilometres from the nearest report of Em in Italy. Using standard flotation-sieving a

New records of helminths of Sceloporus pyrocephalus Cope (Squamata, Phrynosomatidae) from Guerrero and Michoacán, Mexico, with the description of a new species of Thubunaea Seurat, 1914 (Nematoda, Physalopteridae).

A total of 61 specimens of the Red-headed Spiny Lizard Sceloporus pyrocephalus Cope (Phrynosomatidae) collected during the breeding season (June/July 2003, 2004 and 2005) from Western Mexico were examined for helminths. The morphological characterization of the helminths found was made through light microscopy and scanning electron microscopy. Nine taxa of helminths were identified, two cestodes: Mesocestoides sp. and Oochoristica sp., and seven nematodes: Parapharyngodon ayotzinapaensis Garduño-Montes de Oca, Mata-López & León-Règagnon, 2016, Parapharyngodon tikuinii Garduño-Montes de Oca, Ma

Levels of infection of intestinal helminth species in the golden jackal Canis aureus from Serbia.

During the past decade, golden jackal populations have substantially increased, yet little is known of their potential for transmitting parasites within animal and human hosts. In the present study, between 2005 and 2010, 447 jackals from six localities in Serbia were examined for intestinal parasites. Two species of trematodes (Alaria alata, Pseudamphistomum truncatum), three nematodes (Toxocara canis, Ancylostoma caninum, Gongylonema sp.), and seven cestodes (Taenia pisiformis, Taenia hydatigena, Multiceps multiceps, Multiceps serialis, Mesocestoides lineatus, Mesocestoides litteratus, Dipyl

[The effect of anthelmintics on the serotonin content of cestodes].

The effects of some anthelminthic agents, such as praziquantel, fenasal, albendazole, on the levels of the neurotransmitter serotonin in the tissues of Hymenolepis diminuta cestodes and Mesocestoides corti tetrathyridia were examined in vivo and in vitro. The finding showed lower serotonin levels in H.diminuta after praziquantel and albendazole and in M. corti after fenasal. It is suggested that the serotonin neurotransmitter system of helminths is involved in the mechanism of action of these agents.

microRNA silencing in a whole worm cestode model provides insight into miR-71 function.

Parasites belonging to the class Cestoda include zoonotic species such as Echinococcus spp. and Taenia spp. that cause morbidity and mortality in endemic areas, mainly affecting pastoral and rural communities in low income countries but also upper middle income countries. Cestodes show remarkable developmental plasticity, implying tight regulation of gene expression throughout their complex life cycles. Despite the recent availability of genomic data for cestodes, little progress was made on postgenomic functional studies. MicroRNAs (miRNAs) are key components of gene regulatory systems that g

Distribution of Parasitic Helminths in the Small Intestine of the Red Fox (Vulpes vulpes).

The aim of the study was to analyze the distribution of the main groups of parasitic helminths within the small intestine of the red fox on the example of animals coming from eastern Poland. Two hundred and sixteen red foxes shot in eastern Poland were used in the investigation. Before examination, each small intestine was divided into three equal parts: anterior (A), middle (M), and posterior (P). Each part was examined separately with the sedimentation and counting technique. Six different types of intestinal parasites were detected: Alaria alata (78.7%), Mesocestoides spp. (78.2%), hookworm

STAT6⁻/⁻ mice exhibit decreased cells with alternatively activated macrophage phenotypes and enhanced disease severity in murine neurocysticercosis.

In this study, using a murine model for neurocysticercosis, macrophage phenotypes and their functions were examined. Mesocestoides corti infection in the central nervous system (CNS) induced expression of markers associated with alternatively activated macrophages (AAMs) and a scarcity of iNOS, a classically activated macrophage marker. The infection in STAT6(-/-) mice resulted in significantly reduced accumulation of AAMs as well as enhanced susceptibility to infection coinciding with increased parasite burden and greater neuropathology. These results demonstrate that macrophages in the helmi

The efficacy of praziquantel against cestodes in cats, dogs and sheep.

Praziquantel is a new type of acylated isoquinoline-pyrazine. A single, low oral or subcutaneous dose of the compound is reliably effective against all tested juvenile and adult cestodes in cats, dogs and sheep. Praziquantel is the first cestodicide which is also effective on bile duct cestodes. In cats and dogs, 5 mg praziquantel per kg is completely effective on all stages of Taenia hydatigena, T pisiformis, T ovis, T taeniaeformis, Dipylidium caninum, Mesocestoides corti, Echinococcus multilocularis and E granulosus. Because of its very wide therapeutic index praziquantel is thus particular

Squamata reptiles as a potential source of helminth infections when preyed on by companion animals.

BACKGROUND: Squamate reptiles cohabiting with companion animals may represent a source of helminth infections, especially through predation by dogs and cats with an outdoor lifestyle. METHODS: In order to assess the role of reptiles as intermediate/paratenic hosts of trophically transmitted helminths, synanthropic reptiles (n = 245) captured from different ecological settings (i.e., households, dog shelters, urban, peri-urban and rural areas or natural parks) of southern Italy were examined for endoparasites. Parasitic cysts (i.e., larval forms of acanthocephalans, cestodes and nematodes) and

Successful treatment of pleural and peritoneal larval mesocestoidosis with fenbendazole in two dogs.

Mesocestoides spp. are common intestinal tapeworms of dogs and cats. The presence of numerous larval stages in the pleural or peritoneal cavities causes larval mesocestoidosis, that is a potentially life-threatening disease in massive infection. In vivo diagnosis of larval mesocestoidosis is challenging and confirmed diagnosis is often made post-mortem. For this reason, therapeutic recommendations are empirical and there is currently no data on the best treatment options in dogs and cats. Both fenbendazole and praziquantel have been used to treat canine larval mesocestoidosis but failure of co

[The efficacy of praziquantel against experimental cysticercosis and hydatidosis (author's transl)].

Praziquantel is active against cysticerci in experimentally infected mice, rabbits, sheep and cattle. After single or repeated application the infectious stages of Hymenolepis nana, Cysticercus fasciolaris, C. pisiformis, C. tennuicollis and C. bovis were killed. The compound is more active against older than against younger stages as demonstrated with H. nana-cystecercoids and C. fasciolaris in mice. Against the tetrahyridia of Mesocestoides corti in mice praziquantel is only effective after an intraperitoneal application of an oily suspension. Praziquantel shows also activity in rodents agai

The complex role of pro- and anti-inflammatory cytokines in cysticercosis: immunological lessons from experimental and natural hosts.

Parasitic helminthes have developed complex mechanisms to evade or modulate hosts responses. Studies on cysticercosis are solid, but scarce. The most studied immunological models of cysticercosis are Taenia crassiceps infecting mice and T. solium infecting pigs. These parasites, despite being widely exposed to the host, are able to modulate the host immune system. Taenia metacestodes, much like other helminthes parasites, have developed complicated strategies in order to infect and successfully colonize their hosts. We focus here on the accumulated evidence from experimental models that have b

Histone deacetylase enzymes as potential drug targets of Neglected Tropical Diseases caused by cestodes.

Cestode parasites cause neglected diseases, such as echinococcosis and cysticercosis, which represent a significant problem in human and animal health. Benzimidazoles and praziquantel are the only available drugs for chemotherapy and it is therefore important to identify new alternative drugs against cestode parasites. Histone deacetylases (HDACs) are validated drug targets for the treatment of cancer and other diseases, including neglected diseases. However, knowledge of HDACs in cestodes is very scarce. In this work, we investigated cestode HDACs as potential drug targets to develop new ther

Differential release and phagocytosis of tegument glycoconjugates in neurocysticercosis: implications for immune evasion strategies.

Neurocysticercosis (NCC) is an infection of the central nervous system (CNS) by the metacestode of the helminth Taenia solium. The severity of the symptoms is associated with the intensity of the immune response. First, there is a long asymptomatic period where host immunity seems incapable of resolving the infection, followed by a chronic hypersensitivity reaction. Since little is known about the initial response to this infection, a murine model using the cestode Mesocestoides corti (syn. Mesocestoides vogae) was employed to analyze morphological changes in the parasite early in the infectio
