Distinct patterns of control and expression amongst members of the PEP carboxylase kinase gene family in C4 plants.

We have examined the complexity of the phosphoenolpyruvate carboxylase kinase (PPCK) gene family in the C(4) monocots maize and sorghum. Maize contains at least four PPCK genes. The encoded proteins are similar to other phosphoenolpyruvate carboxylase (PEPC) kinases, in that they comprise a protein kinase domain with minimal extensions, except that two of the proteins contain unusual acidic insertions. The spatial and temporal expression patterns of the genes provide information about their presumed functions. Expression of ZmPPCK1 in leaves is mesophyll cell-specific and light-induced, indica

Abscisic acid-triggered guard cell l-cysteine desulfhydrase function and in situ hydrogen sulfide production contributes to heme oxygenase-modulated stomatal closure.

Recent studies have demonstrated that hydrogen sulfide (H2 S) produced through the activity of l-cysteine desulfhydrase (DES1) is an important gaseous signaling molecule in plants that could participate in abscisic acid (ABA)-induced stomatal closure. However, the coupling of the DES1/H2 S signaling pathways to guard cell movement has not been thoroughly elucidated. The results presented here provide genetic evidence for a physiologically relevant signaling pathway that governs guard cell in situ DES1/H2 S function in stomatal closure. We discovered that ABA-activated DES1 produces H2 S in gua

Salinity Effects on Photosynthesis and Growth in Alternanthera philoxeroides (Mart.) Griseb.

Alternanthera philoxeroides, alligator weed, was grown at five different NaCl concentrations to determine the effect of salinity on factors related to the net rate of CO(2) uptake (P(n)). Over the range of 0 to 400 millimolar NaCl, P(n) declined 51%. Stomatal conductance declined in parallel with P(n) and as a result there was no reduction in intercellular CO(2) concentration and therefore no reduction in the amount of CO(2) available for photosynthesis. The CO(2) compensation point did not change with salt stress. Increases in leaf thickness tended to compensate slightly for the negative effe

Anatomical injury induced by the eriophyid mite Aceria anthocoptes on the leaves of Cirsium arvense.

Anatomical injury of the leaves of the invasive species, Cirsium arvense (L.) Scop., caused by the eriophyid mite Aceria anthocoptes (Nal.), which is the only eriophyid mite that has been recorded on C. arvense worldwide, is described. The injury induced by the mite feeding on the leaves of C. arvense results in visible russeting and bronzing of the leaves. Other conspicuous deformations are folding and distortion of the leaf blade and curling of leaf edge, as well as gradual drying of leaves. The anatomical injury of the mature leaves of field-collected plants was limited to the epidermis of

Toxicity of cadmium to wheat seedling roots in the presence of graphene oxide.

The effects of cadmium (Cd) on wheat seedlings in the presence of graphene oxide (GO) were investigated. Parameters evaluated include root morphology, microtubule protein content, cytochrome P450 activity, and the microcellular structure of wheat seedlings. Compared with treatments with Cd or GO in isolation, treatments combining GO and Cd inhibited the total root length, total root surface area, average root diameter, and number of root hairs. GO combined with Cd also increased cytochrome P450 activity and reduced tubulin content. Cotransport of GO-loading Cd entered root tissues and was then

Altered cell wall properties in domesticated cotton enhance CO2 diffusion conductance.

The cell wall plays a key role in determining mesophyll conductance (gm) and photosynthetic capacity. While the impact of wall thickness (Tcw) on gm is well understood, the influence of wall composition and structural interactions on Tcw and gm remains unclear, and it is unknown whether these factors have been affected during crop domestication. In this study, we examined 25 wild and 13 domesticated Gossypium genotypes to investigate whether variations in Tcw, composition, and structure affected gm and photosynthesis. X-ray diffraction was used to analyze internal cell wall structure. Cotton d

The RNA-dependent RNA polymerase of cowpea is not involved in cowpea mosaic virus RNA replication: immunological evidence.

The RNA-dependent RNA polymerase (RdRp) of plants has been proposed to act as core RNA polymerase in plant viral RNA replication. An antiserum directed against the host-encoded RdRp from cowpea leaves was applied to examine a possible role of this enzyme in cowpea mosaic virus (CPMV) RNA replication. The amount of host-encoded RdRp increased more than 20-fold in cowpea leaf cells infected with CPMV. In striking contrast, no increase of RdRp was found to accompany CPMV RNA replication in cowpea mesophyll protoplasts after infection with CPMV, indicating that an increased amount of RdRp is not a

Intercellular Concentration Gradients of 3-Phosphoglycerate and Triose-Phosphate Demonstrate Operation of an Energy Shuttle in NAD-Malic Enzyme and Phosphoenolypyruvate Carboxykinase C4 Subtypes.

In C4 photosynthesis, incoming CO2 is incorporated in mesophyll cells (MC) into 4-carbon acids that diffuse to bundle sheath cells (BSC) and are decarboxylated to generate a high CO2 concentration that suppresses the oxygenation reaction of Rubisco. Decarboxylation can occur by NADP-malic enzyme, (NADP-ME), NAD-malic enzyme (NAD-ME) or phosphoenolpyruvate carboxykinase (PEPCK). NADP-ME generates NADPH in the BSC chloroplast and species that use it as the major route for decarboxylation typically have dimorphic BSC chloroplasts with little or no photosystem II. They operate an energy shuttle: m

Uptake, distribution, and speciation of chromium in Brassica juncea.

Brassica juncea (Indian mustard) has been widely used in phytoremediation because of its capacity to accumulate high levels of chromium (Cr) and other metals. The present study was conducted to investigate mechanism(s) involved in Cr binding and sequestration by B. juncea. The plants were grown under greenhouse conditions in field-moist or air-dried soils, amended with 100 mg kg(-1) of Cr (III) or VI). The plant concentrated Cr mainly in the roots. B. juncea removed an average of 48 and 58 microg Cr per plant from Cr (III) and Cr (VI)-treated soils, respectively. The uptake of Cr was not affec

Cell-type-specific gating of gene regulatory modules as a hallmark of early immune responses in Arabidopsis leaves.

In plants, multiple cell types contribute to immunity, but what division of labor exists among cell types when immunity is activated? We compared, at single-cell resolution, the response of Arabidopsis thaliana leaf cells during pattern-triggered and effector-triggered immunity (PTI/ETI), sampled at 3 and 5 h after infection with Pseudomonas syringae DC3000. Core defense modules were broadly shared across cell clusters, but their activation varied in timing and intensity, with key immune receptors also showing cell type-specific expression dynamics. Mesophyll cell populations could be distingu

Systematic Evaluation of Mesophyll Protoplast Isolation and PEG-Mediated Transient Expression in Rosa chinensis 'Old Blush'.

Stable transformation in rose is slow and genotype-dependent, motivating rapid cellular assays for candidate-gene testing. We systematically evaluated mesophyll protoplast isolation and PEG-mediated transient expression in Rosa chinensis 'Old Blush'. Sequential experiments compared enzyme preparations, digestion time, D-mannitol concentration, cultivar, and donor cultivation regime. Under the tested conditions, Yakult preparations containing 3.0% Cellulase R-10 and 2.0% Macerozyme R-10, 16 h digestion, and 0.4 mol/L D-mannitol yielded 4.641 ± 0.102 × 106 protoplasts/g fresh weight with 94.26 ±

Negative regulation of defense responses in Arabidopsis by two NPR1 paralogs.

NPR1 is required for systemic acquired resistance, and there are five NPR1 paralogs in Arabidopsis. Here we report knockout analysis of two of these, NPR3 and NPR4. npr3 single mutants have elevated basal PR-1 expression and the npr3 npr4 double mutant shows even higher expression. The double mutant plants also display enhanced resistance against virulent bacterial and oomycete pathogens. This enhanced disease resistance is partially dependent on NPR1, can be in part complemented by either wild-type NPR3 or NPR4, and is not associated with an elevated level of salicylic acid. NPR3 and NPR4 int

Guard cell-specific glycine decarboxylase manipulation affects Arabidopsis photosynthesis, growth and stomatal behavior.

Photorespiration is a mandatory metabolic repair shunt of carbon fixation by the Calvin-Benson cycle in oxygenic phototrophs. Its extent depends mainly on the CO2 : O2 ratio in chloroplasts, which is regulated via stomatal movements. Despite a comprehensive understanding of the role of photorespiration in mesophyll cells, its role in guard cells (GC) is unknown. Therefore, a key enzyme of photorespiration, glycine decarboxylase (GDC), was specifically manipulated by varying glycine decarboxylase H-protein (GDC-H) expression in Arabidopsis GC. Multiple approaches were used to analyze the transg

Physiological Responses and Tolerance Mechanisms to Cadmium in Conyza canadensis.

Experiments were conducted to examine the effects of different concentrations of Cd on the performance of the Cd accumulator Conyza canadensis. Cd accumulation in roots and leaves (roots>leaves) increased with increasing Cd concentration in soil. High Cd concentration inhibited plant growth, increased the membrane permeability of leaves, and caused a significant decline in plant height and chlorophyll [chlorophyll (Chl) a, Chl b, and total Chl] content. Leaf ultrastructural analysis of spongy mesophyllic cells revealed that excessive Cd concentrations cause adverse effects on the chloroplast a

Morphometric analysis of young petiole galls on the narrow-leaf cottonwood, Populus angustifolia, by the sugarbeet root aphid, Pemphigus betae.

An insect-induced gall is a highly specialized structure resulting from atypical development of plant tissue induced by a reaction to the presence and activity of an insect. The insect induces a differentiation of tissues with features and functions of an ectopic organ, providing nutrition and protection to the galling insect from natural enemies and environmental stresses. In this anatomical and cytological study, we characterized how the gall-inducing aphid Pemphigus betae reshapes the leaf morphology of the narrow-leaf cottonwood Populus angustifolia to form a leaf fold gall. Young galls di

Subcellular Localization of Iron in Rhizophora mangle Leaves Revealed by Integrated Perls Reaction, TEM, STEM-HAADF, and EDS Analyses.

Mangrove ecosystems are frequently exposed to high concentrations of iron (Fe) in sediments, resulting in Fe accumulation in plant tissues. Although Fe is an essential micronutrient involved in several metabolic processes, its excess requires efficient mechanisms of compartmentalization and storage to maintain cellular homeostasis. Histochemical detection using the Perls reaction has usually been applied to identify ferric iron (Fe3+) in biological tissues; however, the combination of this technique with ultrastructural and elemental analyses remains relatively unexplored in plant cells. Single-Cell Insights into Medicinal Plant Development and Metabolism.

Medicinal plants are major sources of therapeutic natural products, yet the cell-type-specific organization that governs metabolite biosynthesis, transport, and storage remains imperfectly resolved by organ-level omics. This review synthesizes studies published up to June 2026 that used single-cell, single-nucleus, spatial, metabolomic, and epigenomic approaches to medicinal plant systems, following a PRISMA-guided literature search across PubMed, Web of Science, Scopus, and CNKI. Emerging evidence shows that specialized metabolism is organized through discrete and often rare cell populations,

Rapid dehydration drives a nondiffusional drop in C3 photosynthesis that aligns with phosphate limitation.

Drought is an abnormally prolonged water deficit posing major challenges to plants. Stomatal closure has long been considered the primary factor limiting photosynthesis during the early stages of drought. However, emerging evidence suggests that nonstomatal limitation may also arise, particularly under rapid dehydration. We hypothesised that under rapid dehydration, photosynthesis is constrained by an impeded supply of CO2 to the chloroplasts. To test this, we conducted an innovative experiment inducing rapid but controlled dehydration in hydroponically grown wheat and sunflower. We measured w

Starch Degradation and Soluble Sugar Accumulation Are Associated with Cold Tolerance in Winter Rapeseed (Brassica napus L.).

Winter rapeseed suffers severe damage under cold stress, which seriously restricts its growth and yield. However, differences among cultivars in leaf starch and soluble sugar, as well as their underlying molecular regulatory mechanisms under low-temperature stress, remain poorly understood. We hypothesized that winter rapeseed cultivars with differences in cold tolerance would vary in starch accumulation, soluble sugar contents, and expression of the starch and sugar metabolism-related genes under cold stress. This study aimed to investigate differences in starch accumulation, soluble sugar co

Cytological basis and key metabolite profiling of fresh tobacco leaves with different oiliness indices in Yunyan 87.

INTRODUCTION: Tobacco oiliness index is a key chemical trait that significantly influences tobacco quality. METHODS: To elucidate the cytological features and key metabolites underlying varying relative oiliness indices, fresh Yunyan 87 tobacco leaves were analyzed by microscopic and metabolomic profiling. RESULTS: The results indicated that leaves with high oiliness exhibited a tighter tissue architecture, characterized by closely packed mesophyll cells and reduced intercellular spaces; conversely, leaves with low oiliness showed a looser arrangement. The levels of cembratriene-diols, methy
