Selective access to individual cells after assembling for advanced cell-cell interaction studies.

Understanding the mechanisms underlying cell-cell interactions necessitates a versatile method to assemble cells in a precisely-controlled, cell-specific environment, enabling analysis with high spatiotemporal resolution. Here, we introduce a microfluidic technique using stacked flows to create "virtual" channels under constant or pulsatile flow conditions, supporting various assembly geometries, e.g., doublets and triplets of cells with similar or different sizes, while maintaining independent access to each cell. We demonstrate the method using cell lines and primary human cells within the p

Versatile Compartmentalization of Hydrogel-Medium Interfaces in Microfluidic Channels for Reconstructing Complex Tissue Architectures.

Replicating in vivo tissues with complex, branched, and tortuous geometries remains a challenge in engineering physiologically relevant tissue models. Hydrogel compartmentalization in microfluidic chips can spatially organize cells and microenvironments, but micropillar-based confinement relies on discrete structures that generate segmented interfaces and can limit the design of continuous, nonlinear compartment boundaries. Here, we develop a weir-based microfluidic platform that forms continuous hydrogel-medium boundaries and enables stable hydrogel patterning across diverse, tissue-relevant

A Reusable Capillary Flow-Driven Microfluidic System for Abscisic Acid Detection Using a Competitive Immunoassay.

Point-of-care (PoC) devices offer a promising solution for fast, portable, and easy-to-use diagnostics. These characteristics are particularly relevant in agrifood fields like viticulture where the early detection of plant stresses is crucial to crop yield. Microfluidics, with its low reagent volume requirements, is well-suited for such applications. Self-driven microfluidic devices, which rely on capillary forces for fluid motion, offer an attractive alternative by eliminating the need for external pumps and complex fluid control systems. However, traditional microfluidic prototyping material

Novel 3D-Printed Microfluidic Magnetic Platform for Rapid DNA Isolation.

This study presents a novel miniaturized device as a 3D-printed microfluidic magnetic platform specifically designed to manipulate magnetic microparticles in a microfluidic chip for rapid deoxyribonucleic acid (DNA) isolation. The novel design enables the movement of the magnetic particles in the same or opposite directions with the flow or suspends them in continuous flow. A computational model was developed to assess the effectiveness of the magnetic manipulation of the particles. Superparamagnetic monodisperse silica particles synthesized in-house are utilized for the isolation of fish sper

A Review on Microfluidics: An Aid to Assisted Reproductive Technology.

Infertility is a state of the male or female reproductive system that is defined as the failure to achieve pregnancy even after 12 or more months of regular unprotected sexual intercourse. Assisted reproductive technology (ART) plays a crucial role in addressing infertility. Various ART are now available for infertile couples. Fertilization in vitro (IVF), intracytoplasmic sperm injection (ICSI) and intrauterine insemination (IUI) are the most common techniques in this regard. Various microfluidic technologies can incorporate various ART procedures such as embryo and gamete (sperm and oocyte)

Thinking big by thinking small: application of microfluidic technology to improve ART.

In Vitro Fertilization (IVF) laboratories often carry a penchant to resist change while in the pursuit of maintaining consistency in laboratory conditions. However, implementation of new technology is often critical to expand scientific discoveries and to improve upon prior successes to advance the field. Microfluidic platforms represent a technology that has the potential to revolutionize the fundamental processes of IVF. While the focus of microfluidic application in IVF has centered on embryo culture, the innovative platforms carry tremendous potential to improve other procedural steps and

Microfluidic culture platform for neuroscience research.

This protocol describes the fabrication and use of a microfluidic device to culture central nervous system (CNS) and peripheral nervous system neurons for neuroscience applications. This method uses replica-molded transparent polymer parts to create miniature multi-compartment cell culture platforms. The compartments are made of tiny channels with dimensions of tens to hundreds of micrometers that are large enough to culture a few thousand cells in well-controlled microenvironments. The compartments for axon and somata are separated by a physical partition that has a number of embedded microme

Label-Free Impedance-Activated Acoustic Sorting of Cells From Structural to Molecular Scale in Microfluidics.

Label-free, noninvasive, and high-throughput analysis and sorting of cells are essential capabilities in modern biomedicine, particularly for investigating cellular heterogeneity, functional phenotyping, and precision medicine. In this study, we present an integrated and broadly applicable microfluidic system capable of real-time electrical profiling and on-demand acoustic sorting across multiple physical and functional scales, from cell type, cellular biomechanics to molecularly regulated membrane-functional states. The system achieved up to 98% purity in size-based sorting, and accurately di

Self-grown mycelium in confined geometries as nanofluidic devices.

Precise control of ion and molecular transport at the nanoscale underpins next-generation nanofluidic technologies. However, current approaches such as top-down fabrication and bottom-up assembly remain constrained by cost, scalability, or limited programmability. Fungal mycelium-the largest natural ion transport network in soil-offers a living bio-derived route to nanofluidics. Here, we harness mycelium's self-growth and hyphal anastomosis to construct nanofluidic structures that autonomously conform to confined geometries. With interconnected fibrous networks, nanoscale porosity, and negativ

FloCyT: A Flow-Aware Centroid Tracker for Cell Analysis in High-Speed Capillary-Driven Microfluidic Flow.

Capillary-driven microfluidic chips have emerged as promising platforms for point-of-care diagnostics, offering portable, inexpensive, and pump-free operation. Accurate tracking of cell flow in these systems is vital for quantitative applications such as on-chip cytometry, cell counting, and biomechanical analysis. However, tracking in capillary-driven devices is challenging due to rapid cell displacements, flow instabilities, and visually similar cells. Under these conditions, conventional tracking algorithms such as TrackPy, TrackMate, SORT, and DeepSORT exhibit frequent identity switches an

Antibody-functionalized aluminum oxide-coated particles targeting neutrophil receptors in a multifrequency microfluidic impedance cytometer.

Personalized diagnostics of infectious diseases require monitoring disease progression due to their ever-changing physiological conditions and the multi-faceted organ system mechanisms involved in disease pathogenesis. In such instances, the recommended clinical strategies involve multiplexing data collection from critical biomarkers related to a patient's conditions along with longitudinal frequent patient monitoring. Numerous detection technologies exist both in research and commercial settings to monitor these conditions, however, they fail to provide biomarker multiplexing ability with des

An automated centrifugal microfluidic assay for whole blood fractionation and isolation of multiple cell populations using an aqueous two-phase system.

Fractionating whole blood and separating its constituent components one from another is an essential step in many clinical applications. Currently blood sample handling and fractionation processes remain a predominantly manual task that require well-trained operators to produce reliable and reproducible results. Herein, we demonstrate an advanced on-chip whole human blood fractionation and cell isolation process combining (i) an aqueous two-phase system (ATPS) to create complex separation layers with (ii) a centrifugal microfluidic platform (PowerBlade) with active pneumatic pumping to control

Generation of spatially patterned human neural tube-like structures using microfluidic gradient devices.

The functional complexity and anatomical organization of the nervous system are established during regional patterning of its embryonic precursor-the neural tube. Human pluripotent stem (hPS) cell-based models have emerged as valuable complements to animal models for studying neural development. Here we present the design and implementation of a microfluidic gradient device for modeling human neural tube formation and regional patterning with hPS cells. The microfluidic device enables the formation of tubular or spherical colonies of hPS cells at prescribed locations within microfluidic channe

Electrochemical Biosensor on a Microfluidic Coculture Chip for Ovalbumin Detection.

In this study, a microfluidic coculture electrochemical sensing platform was developed for the sensitive detection of ovalbumin (OVA). The chip integrated dual-layer serpentine microchannels separated by a porous PET membrane, enabling coculture and communication between Caco-2 and RBL-2H3 cells. OVA was quantified indirectly by monitoring interleukin-6 (IL-6) released during the cellular allergic response. The screen-printed electrode was modified by diazonium grafting, followed by AuNP electrodeposition for antibody immobilization and signal amplification. The integrated platform exhibited a

A Data-Driven Multifunctional Microfluidic Platform for in Vitro Safety Assessment of 5-Hydroxymethylfurfural.

5-Hydroxymethylfurfural (5-HMF), an important Maillard reaction product formed during food processing, has received increasing attention. However, its safety and efficacy are still debated, likely due to inconsistent findings from existing evaluation models. To address these controversies, we established a multifunctional microfluidic platform integrated with a data-driven weighting strategy. This platform consisted of a concentration-gradient generator, a gut-on-a-chip module, and aptamer-based online monitoring sensors. It enabled high-throughput, sensitive analysis under physiologically rel

Microfluidic flow-cell with passive flow control for microscopy applications.

We present a fast, inexpensive and robust technique for constructing thin, optically transparent flow-cells with pump-free flow control. Using layers of glass, patterned adhesive tape and polydimethylsiloxane (PDMS) connections, we demonstrate the fabrication of planar devices with chamber height as low as 25 μm and with millimetre-scale (x,y) dimensions for wide-field microscope observation. The method relies on simple benchtop equipment and does not require microfabrication facilities, glass drilling or other workshop infrastructure. We also describe a gravity perfusion system that exploits

Injection system for fast capillary electrophoresis based on pressure regulation with flow restrictors.

A fast automated system for rapid electrophoretic separations in short conventional capillaries employing contactless conductivity detection is presented. The instrument is based on pneumatic pressurization and does not require a conventional pump. The required pressures and flow rates for the different steps of the injection and flushing processes are produced with the help of two flow restrictors. The device is implemented on a microfluidic breadboard with dimensions of ca. 13 × 20 cm and employs miniature valves. Nine inorganic cations, namely NH4+, K+ , Na+ , Ca2+ , Mg2+ , Mn2+ , Sr2+ , Li

A switchable digital microfluidic droplet dye-laser.

Digital microfluidic devices allow the manipulation of droplets between two parallel electrodes. These electrodes can act as mirrors generating a micro-cavity, which can be exploited for a droplet dye-laser. Three representative laser-dyes with emission wavelengths spanning the whole visible spectrum are chosen to show the applicability of this concept. Sub-microlitre droplets of laser-dye solution are moved in and out of a lasing site on-chip to down-convert the UV-excitation light into blue, green and red laser-pulses.

A simple, valveless microfluidic sample preparation device for extraction and amplification of DNA from nanoliter-volume samples.

A glass microdevice has been constructed for the on-line integration of solid-phase extraction (SPE) of DNA and polymerase chain reaction (PCR) on a single chip. The chromatography required for SPE in the microfluidic sample preparation device (muSPD) was carried out in a silica bead/sol-gel SPE bed, where the purified DNA was eluted directly into a downstream chamber where conventional thermocycling allowed for PCR amplification of specific DNA target sequences. Through rapid, simple passivation of the PCR chamber with a silanizing reagent, reproducible DNA extraction and amplification was de

Rotating sample system: an equivalent of a rotating electrode for microliter samples.

Voltammetric experiments in sample droplets of 20 microL volume were carried out with gold ring electrodes microfabricated on Pyrex substrates. The droplet studied was centered and kept in place by a hydrophobic ring deposited on the substrate around the ring electrode, which also ensured that each sample assume a semispherical shape. A small hole in the center of the substrate filled with an agar gel membrane served as a junction. A mild jet of humidified nitrogen gas directed tangentially at the droplet caused it to rotate at a high rate. Cyclic voltammetry and constant-potential electrolysi
