A Microgel-Based Platform for Tunable Expansion and Function of γδ T-cells.

Current γδ T-cell expansion protocols often sacrifice functionality for yield and largely ignore the context of activation. Here we utilize a tunable alginate microgel system functionalized with anti-CD3 and co-stimulatory antibodies (αCD28 or αCD2) to investigate the impact of biochemical signaling and substrate mechanics on γδ T-cell activation. Microgel-mediated expansion was compared to conventional soluble antibodies and TransAct beads. The microgels enhanced γδ T-cell expansion compared to soluble antibodies, allowed for controlled tuning of differentiation state, and promoted higher NKG

Soft microgel networks stabilize and extend nozzle-free water jets.

The stability of high-speed liquid jets is crucial for applications ranging from precision printing to needle-free drug delivery, yet it is fundamentally limited by capillary-driven breakup. We find that tuning the nanoscale softness of Poly(N-Isopropylacrylamide) (PNIPAM) microgels provides a robust, biocompatible strategy to overcome this limitation. Soft, low-cross-linker-density microgels form elastic interfacial networks at the air-water interface that suppress surface tension recovery, delay Rayleigh-Plateau instabilities, and extend Surface Acoustic Wave (SAW)-driven jet lengths by up t

Effect of Internal Architecture on the Elasticity of Microgel Monolayers at the Air/Water Interface.

Soft microgels can deform and adsorb at liquid interfaces, forming monolayers with tunable compressibility and elasticity. Their deformabilityor softnessis governed by the internal architecture and inhomogeneity of the polymeric network. However, establishing a direct correlation between single-particle properties and their collective interfacial behaviorssuch as self-assembly and mechanical responseremains a fundamental challenge. In this work, we employ core-shell microgels with controllable internal architectures to investigate how single-particle softness influences the elasticity of m

A Protocol for Preparing Mucoadhesive Emulsion Microgels and Assessing Their Mucoadhesion Properties In Vitro.

Intravesical instillation is an efficient therapeutic technique based on targeted administration of a drug directly into the lesion for the treatment of bladder diseases. This is an alternative to traditional systemic administration of drugs. However, this technique requires repeated procedures, which can lead to even greater inflammation and infection of the urethra. To date, novel systems that allow prolonged drug retention in the bladder cavity are actively being developed. We recently reported a targeted drug delivery system based on the mucoadhesive emulsion microgels consisting of the na

Conductive Microgel Annealed Scaffolds Enhance Myogenic Potential of Myoblastic Cells.

Conductive biomaterials may capture native or exogenous bioelectric signaling, but incorporation of conductive moieties is limited by cytotoxicity, poor injectability, or insufficient stimulation. Microgel annealed scaffolds are promising as hydrogel-based materials due to their inherent void space that facilitates cell migration and proliferation better than nanoporous bulk hydrogels. Conductive microgels are generated from poly(ethylene) glycol (PEG and poly(3,4-ethylenedioxythiophene) polystyrene sulfonate (PEDOT: PSS) to explore the interplay of void volume and conductivity on myogenic dif

Metal-polyphenol microgels for oral delivery of puerarin to alleviate the onset of diabetes.

Puerarin (Pue) is a naturally bioactive compound with many potential functions in regulating blood glucose and lipid metabolism. However, the low bioavailability and rapid elimination in vivo limit the application of Pue in diabetic treatment. Here, we developed a metal-polyphenol-functionalized microgel to effectively deliver Pue in vivo and eventually alleviate the onset of diabetes. Pue was initially encapsulated in alginate beads through electrospray technology, and further immersed in Fe3+ and tannic acid solution from tannic acid (TA)-iron (Fe) coatings (TF). These constructed Pue@SA-TF

Microfluidic Synthesis of Microgel Building Blocks for Microporous Annealed Particle Scaffold.

The microporous annealed particle (MAP) scaffold platform is a subclass of granular hydrogels. It is composed of an injectable slurry of microgels that can form a structurally stable scaffold with cell-scale porosity in situ following a secondary light-based chemical crosslinking step (i.e., annealing). MAP scaffold has shown success in a variety of regenerative medicine applications, including dermal wound healing, vocal fold augmentation, and stem cell delivery. This paper describes the methods for synthesis and characterization of poly(ethylene glycol) (PEG) microgels as the building blocks

Microgel dynamics within the 3D porous structure of transparent PEG hydrogels.

We report an investigation on the effects of the confinement imposed by application-relevant poly(ethylene glycol) (PEG) hydrogel matrices with controlled porosity on the dynamics of soft microgels. Through a detailed characterization of the internal structure of the hydrogels at the nano and microscale, we were able to link the microgel dynamics, measured by particle tracking, to the 3D geometrical confinement imposed by the porous matrices. PEG hydrogels with a high degree of transparency and tunable pore sizes and volume fractions were obtained using freeze-thawing. We found that the porosi

The fluorescence scanning of microgels stained for glycoproteins with fluorescein isothiocyanate-labelled concanavalin A.

A technique is presented which allows one to label and quantitate glycoproteins. Small amounts of protein from biological samples (0.5-2.5 microgram for mixtures and less for individual proteins) are separated by sodium dodecyl sulfate gel electrophoresis on 1-30% polyacrylamide gradient microgels. The gels are stained with Co-omassie Brillant Blue R250 to evaluate relative migration or fixed in 2-propanol/acetic acid and stained with fluorescein isothiocyanate-labelled concanavalin A. The microgels are then scanned using a fluorescence microscope controlled by a computer, although simpler con

Nanofibrous Interface-Engineered Microgel Array Enabling Streamlined Digital PCR.

Digital polymerase chain reaction (dPCR) is increasingly used due to its key advantageshigh sensitivity, absolute quantification, and tolerance to contaminantsthat contribute to its overall robustness. However, it typically relies on complex droplet or microfluidic systems requiring pumps or vacuum systems for partitioning, limiting its accessibility and portability. Here, we present a microgel array chip comprising 2003 porous microgels (1 nL) in a 1 cm2 area, offering simple and rapid partitioning. Controlling internal porosity and surface structure of microgels is crucial to achieving rel

A Free Energy Model for the Plateau Shear Modulus in Thermosensitive Microgel Suspensions.

Polymer microgels exhibit intriguing macroscopic flow properties arising from their unique microscopic structure. Microgel colloids usually comprise a cross-linked polymer network with a radially decaying density profile, resulting in a dense core surrounded by a fuzzy corona. Notably, microgels synthesized from poly(N-isopropylacrylamide) (PNIPAM) are thermoresponsive and capable of adjusting their size and density profile based on temperature. Above the lower critical solution temperature (TLCST∼33 °C), the microgel's polymer network collapses, expulsing water through a reversible process. C

Microgel reinforced zwitterionic hydrogel coating for blood-contacting biomedical devices.

Zwitterionic hydrogels exhibit eminent nonfouling and hemocompatibility. Several key challenges hinder their application as coating materials for blood-contacting biomedical devices, including weak mechanical strength and low adhesion to the substrate. Here, we report a poly(carboxybetaine) microgel reinforced poly(sulfobetaine) (pCBM/pSB) pure zwitterionic hydrogel with excellent mechanical robustness and anti-swelling properties. The pCBM/pSB hydrogel coating was bonded to the PVC substrate via the entanglement network between the pSB and PVC chain. Moreover, the pCBM/pSB hydrogel coating ca

Aqueous Lubrication, Structure and Rheological Properties of Whey Protein Microgel Particles.

Aqueous lubrication has emerged as an active research area in recent years due to its prevalence in nature in biotribological contacts and its enormous technological soft-matter applications. In this study, we designed aqueous dispersions of biocompatible whey-protein microgel particles (WPM) (10-80 vol %) cross-linked via disulfide bonding and focused on understanding their rheological, structural and biotribological properties (smooth polydimethylsiloxane (PDMS) contacts, Ra < 50 nm, ball-on-disk set up). The WPM particles (Dh = 380 nm) displayed shear-thinning behavior and good lubricating

Fabrication of gelatin methacrylate/nanohydroxyapatite microgel arrays for periodontal tissue regeneration.

INTRODUCTION: Periodontitis is a chronic infectious disease and is the major cause of tooth loss and other oral health issues around the world. Periodontal tissue regeneration has therefore always been the ultimate goal of dentists and researchers. Existing fabrication methods mainly focused on a top-down tissue engineering strategy in which several drawbacks remain, including low throughput and limited diffusion properties resulting from a large sample size. Gelatin methacrylate (GelMA) is a kind of photocrosslinkable and biocompatible hydrogel, with the capacities of enabling cell encapsulat

Engineering neuronal networks in granular microgels to innervate bioprinted cancer organoids on-a-chip.

Organoid models are invaluable for studying organ processes in vitro, offering an unprecedented ability to replicate organ function. Despite recent advancements that have increased their cellular complexity, organoids generally lack key specialized cell types, such as neurons, limiting their ability to fully model organ function and dysfunction. Innervating organoids remains a significant challenge due to the asynchronous biological cues governing neural and organ development. Here, we present a versatile organ-on-a-chip platform designed to innervate organoids across diverse tissue types. Our

Capturing Structure and Morphology in Responsive Microgels: From Intrinsic Free Energy to Collective Behavior.

We develop a coarse-grained theoretical and computational framework based on responsive effective pair potentials to describe the compression behavior of core-shell microgels in a good solvent. Our approach accounts for the intrinsic morphological heterogeneity of the particles by decomposing the total free energy into core and shell contributions, each governed by a Flory-Rehner-type model with distinct mechanical properties. Mechanical equilibrium between both regions is imposed to capture the swelling behavior self-consistently. Interparticle interactions are modeled using a four-component,

Dual role of injectable curcumin-loaded microgels for efficient repair of osteoarthritic cartilage injury.

Curcumin has been widely used for the treatment of age-associated diseases, and showed chondroprotective potential for post-traumatic osteoarthritis (OA). However, due to the irregular-shaped and large-sized defects on joint cartilage in degenerated OA, the in vivo delivery and therapeutic effect of curcumin for effective repair remain challenging. In this study, we first present a PEG-GelMA [Poly(Ethylene Glycol) Dimethacrylate-Gelatin Methacrylate, PGMs] hydrogel microgel-based curcumin delivery system for both improved anti-inflammatory and pro-regenerative effects in treatment for cartilag
