The Spatial Biology of HIV Transmission and Infection: Imaging the Female and Male Reproductive Tracts.

HIV transmission through the female and male reproductive tracts is governed by spatially organized interactions between virions, epithelial barriers, and tissue-resident immune cells. Imaging approaches have been central to defining these earliest events, progressing from classical histology and immunohistochemistry (IHC) to modern fluorescence, multiphoton microscopy, tissue clearing, and spatially resolved molecular platforms. Early FFPE-based IHC and ISH established foundational maps of infected cells within intact mucosa, revealing regional immune heterogeneity and linking susceptibility

Evolution of neuronal and astroglial disruption in the peri-contusional cortex of mice revealed by in vivo two-photon imaging.

In traumatic brain injury mechanical forces applied to the cranium and brain cause irreversible primary neuronal and astroglial damage associated with terminal dendritic beading and spine loss representing acute damage to synaptic circuitry. Oedema develops quickly after trauma, raising intracranial pressure that results in a decrease of blood flow and consequently in cerebral ischaemia, which can cause secondary injury in the peri-contusional cortex. Spreading depolarizations have also been shown to occur after traumatic brain injury in humans and in animal models and are thought to accelerat

Seeing is believing: unraveling kidney function and remodeling in real time with in vivo imaging.

PURPOSE OF REVIEW: Live imaging has become a transformative tool in kidney research, enabling real-time visualization of renal structure and function in vivo. With rapid technological progress, particularly in multiphoton microscopy, this review is reassessing how intravital imaging advances our understanding of kidney physiology and disease mechanisms. RECENT FINDINGS: Recent studies have expanded intravital imaging beyond static morphology to include dynamic functional biomarkers. These approaches now allow simultaneous assessment of cellular behavior, microvascular function, and tissue rem

Adapting a Two-Photon Scanning Microscope for Simultaneous Single-Photon Imaging of an Infrared Dopamine Sensor.

We describe a novel method for adapting a two-photon scanning microscope to enable simultaneous detection of two-photon-generated visible fluorescence and single-photon-generated near-infrared (nIR) fluorescence. In this configuration, nIR fluorescence is routed through a single-mode optical fiber before detection by a photomultiplier tube. This fiber coupling offers two advantages: first, the optical fiber functions as a pinhole aperture, allowing for improved optical sectioning of the nIR signal; second, it minimizes nIR background fluorescence. To validate the effectiveness of this design,

Hundred-Nanosecond Equivalent Pixel Dwell Time for Deep-Tissue 3D Three-Photon Fluorescence Microscopy via Sparse X-Y-Z Reconstruction.

Three-photon fluorescence microscopy (3PFM) enables high-resolution volumetric imaging in deep tissues but is fundamentally constrained by a trade-off among imaging speed and spatial resolution, due to low photon flux and prolonged laser exposure. We present DeepR-SXYZ, a deep learning framework that achieves pixel dwell time equivalent to hundreds of nanoseconds (0.168-0.38 µs) through sparse X-Y-Z reconstruction for 3PFM. Trained on paired datasets consisting of sparsely acquired low-resolution volumetric scans (e.g., 256 × 256 pixels, 1.2 µs per pixel, Z step: 16 µm) and their corresponding

Quantitative second harmonic generation microscopy for characterizing collagen remodeling in papillary thyroid carcinoma.

SIGNIFICANCE: Thyroid cancer is the most common endocrine malignancy. Although fine needle aspiration (FNA) is the gold standard for diagnosis, 10% to 20% of FNAs yield indeterminate cytology, of which 20% to 30% prove malignant. Such diagnostic uncertainty drives repeat FNAs, costly molecular tests, and unnecessary surgeries, leading to increased healthcare costs, reduced quality of life, and emotional distress to patients. Current methods provide limited structural information about the extra-cellular matrix, despite collagen remodeling being a hallmark of malignancy. A rapid, label-free, an

The Use of Intravital Two-Photon and Thick Section Confocal Imaging to Analyze B Lymphocyte Trafficking in Lymph Nodes and Spleen.

Intravital two-photon laser scanning microscopy (TP-LSM) has allowed the direct observation of immune cells in intact organs of living animals. In the B cell biology field TP-LSM has detailed the movement of B cells in high endothelial venules and during their transmigration into lymph organs; described the movement and positioning of B cells within lymphoid organs; outlined the mechanisms by which antigen is delivered to B cells; observed B cell interacting with T cells, other cell types, and even with pathogens; and delineated the egress of B cells from the lymph node (LN) parenchyma into th

High bacterial colonization and lipase activity in microcomedones.

BACKGROUND: Although acne vulgaris has a multifactorial aetiology, comedogenesis and bacteria colonization of the pilosebaceous unit are known to play a major role in the onset of inflammatory acne lesions. However, many aspects remain poorly understood such as where and when is the early stage of the Propionibacterium acnes colonization in follicular unit? Our research aimed at providing a precise analysis of microcomedone's structure to better understand the interplay between Propionibacterium acnes and follicular units, and therefore, the role of its interplay in the formation of acne lesio

Identifying Two Common Types of Breast Benign Diseases Based on Multiphoton Microscopy.

Multiphoton microscopy has attracted increasing attention and investigations in the field of breast cancer, based on two-photon excited fluorescence (TPEF) and second-harmonic generation (SHG). However, the incidence of breast benign diseases is about 5 to 10 times higher than breast cancer; up to 30% of women suffer from breast benign diseases and require treatment at some time in their lives. Thus, in this study, MPM was applied to image fibroadenoma and fibrocystic lesion, which are two of the most common breast benign diseases. The results show that MPM has the capability to identify the m

Visualization of lymphatic vascular invasion in breast cancer by multiphoton microscopy.

Lymphatic vascular invasion (LVI) is regarded as one of the independent factors which affect the prognosis of breast cancer. Once LVI is formed, it indicates the tumor has metastasized or has the possibility of metastasis. In this work, multiphoton microscopy (MPM), which relies on the two-photon excited fluorescence (TPEF) and second harmonic generation (SHG), was applied to identify the typical morphology of LVI and also visualize the histological features of LVI. Furthermore, the pixel density of collagen fibers was extracted as a quantitative parameter to differentiate LVI from the ductal

Intravital multiphoton imaging of the selective uptake of water-dispersible quantum dots into sinusoidal liver cells.

Although many studies reporting the organ-level biodistribution of nanoparticles (NPs) in animals, very few have addressed the fate of NPs in organs at the cellular level. The liver appears to be the main organ for accumulation of NPs after intravenous injection. In this study, for the first time, the in vivo spatiotemporal disposition of recently developed mercaptosuccinic acid (MSA)-capped cadmium telluride/cadmium sulfide (CdTe/CdS) quantum dots (QDs) is explored in rat liver using multiphoton microscopy (MPM) coupled with fluorescence lifetime imaging (FLIM), with subcellular resolution (∼

Extended field-of-view two-photon imaging in the deep brain of freely behaving mice using lightweight microfabricated corrective optics.

Investigating neural circuits underlying complex behavior requires high-resolution imaging of large neuronal populations during naturalistic conditions. Miniaturized two-photon microscopes (MINI2Ps) enable deep brain imaging in freely moving animals when coupled with gradient index (GRIN)-based microendoscopes. However, intrinsic optical aberrations in GRIN-based microendoscopes degrade spatial resolution and limit the effective field-of-view (FOV). Here, we address these limitations using lightweight microfabricated optics to correct GRIN aberrations during deep brain MINI2P imaging. The aber

Liver fibrosis analysis using digital pathology.

Digital pathology has enabled the noninvasive quantification of pathological parameters. In addition, the combination of digital pathology and artificial intelligence has enabled the analysis of a vast amount of information, leading to the sharing of much information and the elimination of knowledge gaps. Fibrosis, which reflects chronic inflammation, is the most important pathological parameter in chronic liver diseases, such as viral hepatitis and metabolic dysfunction-associated steatotic liver disease. It has been reported that the quantitative evaluation of various fibrotic parameters by

Two-Photon Excited Polarization-Dependent Autofluorescence of Amyloids as a Label-Free Method of Fibril Organization Imaging.

Amyloids are broadly investigated protein misfolding products with characteristic β-sheet assemblies that have an important role in neurodegenerative diseases (e.g., Alzheimer's disease). While they are usually visualized by staining with Thioflavin-T, Congo Red, or other fluorescent markers, it still arouses a controversy over possible staining molecule influence on the amyloid structure or aggregation process. In this work we present, for the first time, the polarization analysis of two-photon excited autofluorescence of amyloids and confirm that polarization dependence of the observed emiss

In vivo multimodal optical imaging of dermoscopic equivocal melanocytic skin lesions.

There is a wide range of equivocal melanocytic lesions that can be clinically and dermoscopically indistinguishable from early melanoma. In the present work, we assessed the possibilities of combined using of multiphoton microscopy (MPM) and optical coherence angiography (OCA) for differential diagnosis of the equivocal melanocytic lesions. Clinical and dermoscopic examinations of 60 melanocytic lesions revealed 10 benign lesions and 32 melanomas, while 18 lesions remained difficult to diagnose. Histopathological analysis of these lesions revealed 4 intradermal, 3 compound and 3 junctional nev

In vivo two-photon imaging of neuronal and brain vascular responses in mice chronically exposed to ethanol.

The effects of ethanol on brain function have been extensively studied using a variety of in vitro and in vivo techniques. For example, electrophysiological studies using brain slices from rodents and non-human primates have demonstrated that acute and chronic exposure to ethanol alters the intrinsic excitability and synaptic signaling of neurons within cortical and sub-cortical areas of the brain. In humans, neuroimaging studies reveal alterations in measures of brain activation and connectivity in subjects with alcohol use disorder. While complementary, these methods are inherently limited d

Fast Fourier transform-based analysis of second-harmonic generation image in keratoconic cornea.

PURPOSE: To qualify and quantify structural alterations in keratoconic corneas ex vivo by the use of second-harmonic generation (SHG) microscopy and two-dimensional fast Fourier transform (2D-FFT) analysis. METHODS: Two keratoconic and three normal human corneal specimens were imaged with a multiphoton microscope. Forward and backward second-harmonic generation images (FSHG and BSHG) of corneal stroma were obtained at high resolution and at different depths. The SHG images were further analyzed with a 2D-FFT algorithm to quantify the texture and orientation changes of collagen fibers. RESULT

Deep-tissue two-photon brain imaging enabled by a tunable fiber-optic dispersive wave generator.

Here, we present a fiber-optic dispersive wave generator for highly-efficient, wavelength-tunable ultrashort pulse generation, enabling multicolor deep-tissue two-photon imaging of neuronal and vascular structures in fixed, labeled mouse brain. Guided by comprehensive numerical simulations, a compact Yb: fiber laser-driven system is constructed that utilizes precisely parameter- and phase-matching-controlled dispersive wave generation in a photonic crystal fiber. The system delivers sub-100 fs pulses with over ~ 6.7 nJ of energy across a continuously tunable spectral range of 880-950 nm, achie

A pipeline for mouse brain registration to an atlas after in vivo neurophysiological recordings.

Neurophysiological recordings and histological examinations, along with behavioral observations, are interconnected methodological dimensions of systems neuroscience. Current progresses in the neurophysiological data acquisition and machine learning-based data-driven behavioral analysis emphasize the need for precise anatomical localization of recorded neurons. Here, we describe an integrated pipeline for mapping mouse brain regions expressing genetically encoded calcium indicator imaged with two-photon microscopy, and high-density multichannel electrode positions marked with lipophilic dye, t
