A domestication gene links plant architecture and nitrogen metabolism to enhance yield in foxtail millet.

Foxtail millet (Setaria italica) has lagged in yield compared with major cereals, largely due to suboptimal architecture. Here, we report a telomere-to-telomere (T2T) genome assembly of the dwarf, multi-tillering cultivar Dungu and identify SiSD1 (Semi Dwarf 1) via map-based cloning. This gene encodes GA20ox2, the rice SD1 ortholog involved in gibberellin biosynthesis. The loss-of-function sisd1 reduces bioactive gibberellins and remodels nitrogen metabolism, conferring semi-dwarfism, increased tillering, and higher yield. Critically, the semi-dominantly heterozygous SiSD1/sisd1 in F1 hybrids

Light regulates hydrogen sulfide signalling during skoto- and photo-morphogenesis in foxtail millet.

Signal transduction mediated by photoreceptors regulates many physiological processes during plant growth and development including seed germination, flowering and photosynthesis, which are also regulated by hydrogen sulfide (H2S). However, studies of the connection between the vital environmental factors - light and the significant endogenous gasotransmitter - H2S, is lacking. Here, the seedlings of foxtail millet were used to reveal the mechanism of light regulation in H2S generation. Results showed that seedling hypocotyl elongation was promoted by H2S, but inhibited by HA under dark or whi

Involvement of mitogen-activated protein kinase signalling in pearl millet-downy mildew interaction.

Mitogen-activated protein kinase (MAPK) cascade-mediated signalling is essential in the establishment of resistance towards pathogens. The present study compared MAPK activities in a compatible and incompatible interaction between pearl millet [Pennisetum glaucum (L.) R. Br.] and downy mildew pathogen Sclerospora graminicola. Differential expression was observed with rapid and increased activation of MAPKs, PgMPK1 (48kDa) and PgMPK2 (44kDa), in the incompatible interaction; with a weak activity of only PgMPK1 in the compatible interaction. Immunoblot analysis showed PgMPK1 and PgMPK2 to be ort

Effects of dietary protein of proso millet on liver injury induced by D-galactosamine in rats.

In this paper, we examined the effects of dietary protein from proso millet on liver injury induced by D-galactosamine or carbon tetrachloride in rats using serum enzyme activities as indices. D-galactosamine-induced elevations of serum activities of aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase were significantly suppressed by feeding the diet containing 20% protein of proso millet for 14 days as compared with those of rats fed a 20% casein diet, but not in the case of carbon tetrachloride. The results showed that proso millet protein is effective at lower di

Genome-Wide Identification of LBD Genes in Foxtail Millet (Setaria italica) and Functional Characterization of SiLBD21.

Plant-specific lateral organ boundaries domain (LBD) proteins play important roles in plant growth and development. Foxtail millet (Setaria italica) is one new C4 model crop. However, the functions of foxtail millet LBD genes are unknown. In this study, a genome-wide identification of foxtail millet LBD genes and a systematical analysis were conducted. A total of 33 SiLBD genes were identified. They are unevenly distributed on nine chromosomes. Among these SiLBD genes, six segmental duplication pairs were detected. The thirty-three encoded SiLBD proteins could be classified into two classes an

Characterization of Antifungal Natural Products Isolated from Endophytic Fungi of Finger Millet (Eleusine coracana).

Finger millet is an ancient African-Indian crop that is resistant to many pathogens including the fungus, Fusarium graminearum. We previously reported the first isolation of putative fungal endophytes from finger millet and showed that the crude extracts of four strains had anti-Fusarium activity. However, active compounds were isolated from only one strain. The objectives of this study were to confirm the endophytic lifestyle of the three remaining anti-Fusarium isolates, to identify the major underlying antifungal compounds, and to initially characterize the mode(s) of action of each compoun

Evaluation of the effect of processing techniques on the nutrient and antinutrient contents of pearl millet (Pennisetum glaucum) seeds.

This work was undertaken to evaluate the effect of soaking, sprouting, cooking and fermentation or their combinations on the nutrient and antinutrient contents of pearl millet seeds. Standard assay procedures were adopted to determine both the nutrient and the antinutrient concentrations of the products. The synergistic effect of cooking and fermentation improved the nutrient quality. The antinutrients were reduced to safe levels to a greater extent than did any of the other processing techniques or their combinations employed.

Phenotyping for lodging tolerance in pearl millet: identification of optimal environment, plant stage, and potential traits.

Crop lodging poses a significant threat to global food security by reducing crop yields in terms of quantity and quality and causing difficulties in the harvesting process. Rainy-season crops like pearl millet encounter high-velocity winds and rain during different stages of crop growth, thus leading to drastic reductions in grain yields due to crop lodging. The aim of this study was (i) to identify the optimal environment for phenotyping, (ii) to identify trait linkages with lodging tolerance, and (iii) to identify the appropriate plant growth stages for lodging phenotyping. Hence, a four-yea

Millet cultivation mitigates soil erosion in sloping farmland by regulating near-soil surface traits.

INTRODUCTION: Crop growth induces seasonal variations in the near-soil surface traits, including vegetation features, root systems, and soil properties. However, the influencing mechanisms of variations in the near-soil surface traits on soil erosion remain unclear in sloping farmland. METHODS: Rainfall simulation experiments with slope gradients of 5.24%, 8.75%, 17.63%, and 26.79% were carried out on bare soil control (CK) and millet cultivation plots at different growth stages. Near-soil surface traits such as soil bulk density (BD), aggregate mean weight diameter (MWD), soil cohesion (Coh)
